Hydrogen cyanamide breaks grapevine bud dormancy in the summer through transient activation of gene expression and accumulation of reactive oxygen and nitrogen species.

BACKGROUND: Hydrogen cyanamide (HC) and pruning (P) have frequently been used to break dormancy in grapevine floral buds. However, the exact underlying mechanism remains elusive. This study aimed to address the early mode of action of these treatments on accumulation of reactive oxygen species (ROS) and reactive nitrogen species (RNS) and expression of related genes in the dormancy breaking buds of grapevine in the summer. RESULTS: The budbreak rates induced by pruning (P), hydrogen cyanamide (HC), pruning plus hydrogen cyanamide (PHC) and water (control) after 8 days were 33, 53, 95, and 0 %

Long-Chain Hydrocarbons (C21, C24, and C31) Released by Bacillus sp. MH778713 Break Dormancy of Mesquite Seeds Subjected to Chromium Stress.

Volatile organic compounds (VOCs) produced by rhizobacteria have been proven to stimulate plant growth during germination and seedling stages. However, the modulating effect of bacterial volatiles on the germination of seeds subjected to heavy metal stress is scarcely studied. In this work, the ability of volatiles released by Bacillus sp. MH778713 to induce seed dormancy breakage in Prosopis laevigata and Arabidopsis thaliana seeds were examined. The minimal inhibitory concentration of chromium (Cr) VI that prevents seed germination of P. laevigata and A. thaliana on water-Cr-agar plates was

Convergent innovation of (crypto)vivipary via asymmetric dismantling of seed desiccation and dormancy machinery.

Cryptovivipary and vivipary, referred to as (crypto)vivipary, occur across multiple angiosperm lineages and enable embryos to continue developing before dispersal. Yet, this trait raises a fundamental question: how are seed-specific abscisic acid (ABA) responses modulated to support continuous embryonic growth without compromising ABA-dependent vegetative stress tolerance? Here, integrating phylogenomic analyses of independently evolved (crypto)viviparous lineages with spatiotemporal transcriptomics and functional validation, we show that (crypto)vivipary evolved via a convergent "less-is-more

Seed dormancy and germination of three Corydalis species, disjunct relict species in East Asia.

Disjunct congeneric species are common in the temperate flora of Europe, East Asia and eastern North America. However, it is unclear whether the kind of seed dormancy is maintained after divergence. This study aimed to determine the type of seed dormancy of three Corydalis species native to Korea and examine whether the dormancy trait has been conserved after long-term separation. We examined embryo growth and germination characteristics of C. remota, C. incisa and C. speciosa through phenology experiments, controlled temperature experiments, stratification treatments and GA3 application. Redox metabolism and cell wall modifications as global and local targets respectively, of cyanide induced dormancy release of walnut kernels.

The HCN-induced seed dormancy release necessitates alterations in reactive oxygen species (ROS) metabolism and radicle cell wall loosening. Little is known about the interaction of ROS metabolism with cell wall hydrolytic enzymes during HCN-induced seed dormancy release. Thus dormant walnut (Juglans regia L.) kernels were exposed to HCN (4 h) and studied for redox metabolism and cell wall-modifying enzymes during 10 days of incubation (DI) i.e. before radicle emergence. HCN increased ROS especially in the embryonic axes (EAs) but decreased ROS-generating NADPH oxidase and ROS scavenging supero

Genomic and evolutionary analysis reveals dynamic variations of MKK3 gene, a key regulator for seed dormancy in barley.

Barley (Hordeum vulgare L.) is an important crop in the world, and its seed dormancy is primarily controlled by a mitogen-activated protein kinase kinase 3 (MKK3) gene. Although kinase activity of MKK3 and its roles in barley post-domestication have been widely studied, the pre-domestication evolution of MKK3 and the spread of nondormant alleles among global barley varieties remain largely unexplored. In this study, we analyzed MKK3 sequences in barley and its wild progenitor (Hordeum spontaneum K. Koch) and identified two polymorphic miniature inverted-repeat transposable elements (MITEs). Co

Butenolides from plant-derived smoke: natural plant-growth regulators with antagonistic actions on seed germination.

Smoke plays an intriguing role in promoting the germination of seeds of many species following a fire. Recently, a bicyclic compound containing a condensed butenolide moiety, 3-methyl-2H-furo[2,3-c]pyran-2-one (1), was reported as a potent germination promoter from plant-derived smoke. In this study, a related butenolide, 3,4,5-trimethylfuran-2(5H)-one (2), which inhibits germination and significantly reduces the effect of 1 when applied simultaneously, was also isolated from plant-derived smoke. The interaction of these compounds with opposing actions on seed germination may have important ec

Nitric Oxide Pre-Treatment Advances Bulblet Dormancy Release by Mediating Metabolic Changes in Lilium.

The lily is a globally popular cut flower, and managing dormancy in lily bulblets is essential for continuous, year-round production. While nitric oxide (NO) has been shown to influence seed dormancy and germination, its role in dormancy release in lilies was previously unconfirmed. In this study, we investigated the effects of NO on dormancy release in lily bulblets using SNP and c-PTIO. Results showed that SNP treatment promoted dormancy release, while c-PTIO inhibited it. Measurement of endogenous NO levels in the bulbs, along with enzyme activities of NOS-like and NR and gene expression le

Roles of auxin in the inhibition of shoot branching in 'Dugan' fir.

Shoot branching substantially impacts vegetative and reproductive growth as well as wood characteristics in perennial woody species by shaping the shoot system architecture. Although plant hormones have been shown to play a fundamental role in shoot branching in annual species, their corresponding actions in perennial woody plants are largely unknown, in part due to the lack of branching mutants. Here, we demonstrated the role of plant hormones in bud dormancy transition toward activation and outgrowth in woody plants by comparing the physiological and molecular changes in the apical shoot ste

N-terminal truncated RHT-1 proteins generated by translational reinitiation cause semi-dwarfing of wheat Green Revolution alleles.

The unprecedented wheat yield increases during the Green Revolution were achieved through the introduction of the Reduced height (Rht)-B1b and Rht-D1b semi-dwarfing alleles. These Rht-1 alleles encode growth-repressing DELLA genes containing a stop codon within their open reading frame that confers gibberellin (GA)-insensitive semi-dwarfism. In this study, we successfully took the hurdle of detecting wild-type RHT-1 proteins in different wheat organs and confirmed their degradation in response to GAs. We further demonstrated that Rht-B1b and Rht-D1b produce N-terminal truncated proteins throug

Low Temperature-ROS-Hormones Co-Regulation of Seed Dormancy Release and Germination in Xanthoceras sorbifolium.

Seed dormancy is an important adaptive mechanism in plants. However, some seeds, such as those of Xanthoceras sorbifolium (a valuable economic and medicinal species), exhibit strong dormancy, resulting in a very low natural germination rate. This work investigated the mechanism by which low temperatures (LT: -20 °C storage for 60 days) release seed dormancy and promote germination. This was achieved by examining seed germination conditions, applying scanning electron microscopy (SEM), and measuring physiological indicators of seeds and the hormone levels. Targeted metabolomic analysis of sugar

Towards an integrated molecular understanding of plant hormones.

From seed dormancy to pathogen attacks and senescence, many events in a plant's life are critically controlled by low-molecular-weight signaling molecules, called plant hormones. These master regulators include abscisic acid, auxins, brassinosteroids, cytokinins, ethylene, gibberellins, jasmonates, salicylates, and strigolactones. Years of research have unveiled multiple aspects of their biosynthesis, transport, signaling, functions, and interactions. With this knowledge, complex molecular networks have been established and refined over time in an attempt to explain how plant hormones collecti

Maternal and Cold-Stratification Warming Outweigh Germination-phase Warming in Regulating Seed Germination of Alpine Plants.

BACKGROUND AND AIMS: Global warming impacts on key plant life-history stages represent a central focus in ecology. Seed germination, the critical starting point in the plant life cycle, lacks a systematic understanding of its response mechanisms to climate warming. METHODS: Here, we examined how warming at the maternal, cold stratification, and germination stages affects seed germination in ten alpine plant species. KEY RESULTS: Cold stratification (absence vs. presence) is decisive in regulating seed dormancy, but its effect is modulated by temperatures experienced during both the maternal

A Protocol for Accelerating Homozygous Line Screening in Arabidopsis thaliana.

Obtaining homozygous mutant and transgenic lines is a critical yet time-consuming step in Arabidopsis thaliana research. Conventional breeding procedures require seeds to undergo complete maturation and natural desiccation before harvest, followed by cold stratification to overcome seed dormancy. This process substantially prolongs generation turnover and delays genetic screening. Here, we describe a rapid germination strategy based on the use of partially dehydrated seeds collected approximately 15 days after pollination (DAP). At this developmental stage, embryos have reached physiological m

Genome-wide analysis of the HDAC gene family in Pyrus betulifolia and functional validation of PcHDAC5 and PcHDT1.2 in Pyrus calleryana seed dormancy release.

Histone deacetylation, catalyzed by histone deacetylases (HDACs), is a core epigenetic regulatory mechanism in plants. However, no systematic characterization of the HDAC gene family has been reported in pear (Pyrus spp.), a globally important temperate fruit crop. In this study, we performed genome-wide identification and functional characterization of HDAC genes in pear, with a specific focus on their regulatory roles in seed dormancy release. We identified 22 PbHDAC genes from the genome of Pyrus betulifolia Bunge, which were classified into three conserved subfamilies. Evolutionary analysi

Cadmium stress suppresses the tillering of perennial ryegrass and is associated with the transcriptional regulation of genes controlling axillary bud outgrowth.

Perennial ryegrass (Lolium perenne L.), a grass species with superior tillering capacity, plays a potential role in the phytoremediation of cadmium (Cd)-contaminated soils. Tiller production is inhibited in response to serious Cd stress. However, the regulatory mechanism of Cd stress-induced inhibition of tiller development is not well documented. To address this issue, we investigated the phenotype, the expression levels of genes involved in axillary bud initiation and bud outgrowth, and endogenous hormone biosynthesis and signaling pathways in seedlings of perennial ryegrass under Cd stress.

Metalliferous habitats and seed microbes affect the seed morphology and reproductive strategy of Arabidopsis halleri.

PURPOSE: Plant reproduction in metalliferous habitats is challenged by elevated concentrations of metal trace elements in soil. As part of their survival strategy, metal-tolerant plants have adjusted reproductive traits, including seed morphology, dormancy, and germination rate. These traits are particularly relevant, yet poorly understood, in metal hyperaccumulators that are promising candidates for phytoremediation. METHODS: We assessed seed shape characteristics, dormancy, and germination rate in the hyperaccumulating model species Arabidopsis halleri. Seed morphological parameters were ev

Dormancy alleviation by NO or HCN leading to decline of protein carbonylation levels in apple (Malus domestica Borkh.) embryos.

Deep dormancy of apple (Malus domestica Borkh.) embryos can be overcome by short-term pre-treatment with nitric oxide (NO) or hydrogen cyanide (HCN). Dormancy alleviation of embryos modulated by NO or HCN and the first step of germination depend on temporary increased production of reactive oxygen species (ROS). Direct oxidative attack on some amino acid residues or secondary reactions via reactive carbohydrates and lipids can lead to the formation of protein carbonyl derivatives. Protein carbonylation is a widely accepted covalent and irreversible modification resulting in inhibition or alter

Regulation of seed germination and seedling growth by chemical signals from burning vegetation.

It is well known that burning of vegetation stimulates new plant growth and landscape regeneration. The discovery that char and smoke from such fires promote seed germination in many species indicates the presence of chemical stimulants. Nitrogen oxides stimulate seed germination, but their importance in post-fire germination has been questioned. Cyanohydrins have been recently identified in aqueous smoke solutions and shown to stimulate germination of some species through the slow release of cyanide. However, the most information is available for karrikins, a family of butenolides related to

DOF Transcription Factors: Regulatory Hubs Coordinating Plant Growth, Development and Stress Tolerance.

Under global climate warming, plants evolve unique growth and developmental strategies to cope with frequent abiotic stresses. The plant-specific DNA-BINDING WITH ONE FINGER (DOF) transcription factors (TFs) originated in green algae and are phylogenetically divided into four subfamilies (A, B, C, D), governing plant growth development and stress adaptation. Subfamily B mainly regulates vegetative growth, subfamily C is predominantly involved in seed development and vegetative growth, and subfamily D1 (CYCLING DOF FACTOR, CDF) participates extensively in vegetative growth, flowering, and stres
