Enhancing Rubisco gene expression and metabolites accumulation for better plant growth in Ficus deltoidea under drought stress using hydrogen peroxide.

Growth improvement of the medicinal plant, Ficus deltoidea (Mas Cotek) under drought conditions is a vital issue in Malaysia since it is a slow-growing plant and disposed to leaf damage under the stresses of drought. Therefore, investigation was done to examine the outcomes of hydrogen peroxide (H2O2) application on Rubisco gene expression and metabolites accumulation of stressed F. deltoidea plants, and thereby to record the changes in leaf histology, photosynthesis, biochemical properties, and the growth of the plant. H2O2 at the rates of 0, 5, 10, 15, and 20 mM were foliar sprayed biweekly

Opportunities for improving intrinsic water use efficiency in C4 plants under climate change.

C4 photosynthesis is inherently efficient, saturating at low intercellular CO2 (Ci) and operating under low stomatal conductance (gs), resulting in high intrinsic water use efficiency (iWUE = assimilation rates, (A)/ stomatal conductance (gs)). While iWUE is generally higher in C4 than in C3 plants, future climate scenarios, marked by increasing atmospheric CO2, temperatures, and aridity, present ongoing pressures on crop productivity in C4-dominated environments. This review explores the physiological, anatomical, and environmental factors controlling iWUE in C4 plants, with a focus on balanc

Uncovering the core genetic programs governing plant guard cell biology.

Guard cells (GCs) regulate gas exchange and water loss in plants and have been extensively studied in Arabidopsis thaliana. However, cross-species comparisons at single-cell resolution remain limited. To address this, we aimed to define conserved and divergent transcriptomic signatures of GCs by generating a comparative single-cell atlas encompassing five species: A. thaliana, soybean (Glycine max), tomato (Solanum lycopersicum), rice (Oryza sativa), and sorghum (Sorghum bicolor). We performed single-nucleus RNA sequencing (sNucRNA-seq) on leaf tissues from each species, followed by integrativ

The combined effect of decreased stomatal density and aperture increases water use efficiency in maize.

Stomata play a crucial role in balancing carbon dioxide uptake and water vapor loss, thereby regulating plant water use efficiency (WUE). Enhancing WUE is important for sustainable agriculture and food security, particularly for crops such as maize (Zea mays L.), as climate change and growing global food demand exacerbate limitations on water availability. Genetic factors controlling stomatal density and levels of the plant hormone abscisic acid (ABA) in leaves, which affect stomatal aperture, are key determinants of stomatal conductance (gs) and intrinsic WUE (iWUE). In this study, we demonst

Dovetailing the Genetic, Developmental and Functional Bases of the Adaptation of Stomatal Traits to Climate.

Stomata, the micro-valves on leaves, regulate CO2 uptake and water loss. Thus, the anatomical maximum stomatal conductance, gmax, is a key influence on gas exchange, growth and productivity. For 145 geographically diverse ecotypes of Arabidopsis thaliana grown in a common garden we tested hypotheses for the drivers of variation in gmax and a stomatal size-density trade-off, considering epidermal optimization, epidermal development and climate adaptation. Ecotypes native to colder and drier macroclimates had higher gmax, consistent with leaf-scale stress-avoidance, despite having longer times t

Growth and nutritive value of cassava (Manihot esculenta Cranz.) are reduced when grown in elevated CO.

Global food security in a changing climate depends on both the nutritive value of staple crops as well as their yields. Here, we examined the direct effect of atmospheric CO(2) on cassava (Manihot esculenta Cranz., manioc), a staple for 750 million people worldwide. Cassava is poor in nutrients and contains high levels of cyanogenic glycosides that break down to release toxic hydrogen cyanide when damaged. We grew cassava at three concentrations of CO(2) (C(a): 360, 550 and 710 ppm) supplied together with nutrient solution containing either 1 mM or 12 mM nitrogen. We found that total plant bio

Stomatal biology meets multimodal foundation model-driven AI.

Stomata regulate plant growth and environmental responses through their control of gas exchange and transpiration. However, high-throughput stomatal phenotyping and the integration of phenotypic, genetic, and multiomics data remain major challenges for biological discovery and for understanding stomatal function in plants. Here, we review emerging imaging technologies and foundation models for stomatal biology and discuss how advances in AI could transform plant stomatal research. The rapid accumulation of large-scale phenotypic, genetic, and omics datasets requires systematic integration of h

Cerato-platanin induces resistance in Arabidopsis leaves through stomatal perception, overexpression of salicylic acid- and ethylene-signalling genes and camalexin biosynthesis.

Microbe-associated molecular patterns (MAMPs) lead to the activation of the first line of plant defence. Few fungal molecules are universally qualified as MAMPs, and proteins belonging to the cerato-platanin protein (CPP) family seem to possess these features. Cerato-platanin (CP) is the name-giving protein of the CPP family and is produced by Ceratocystis platani, the causal agent of the canker stain disease of plane trees (Platanus spp.). On plane tree leaves, the biological activity of CP has been widely studied. Once applied on the leaf surface, CP acts as an elicitor of defence responses.

Prevention of Stomatal Entry as a Strategy for Plant Disease Control against Foliar Pathogenic Pseudomonas Species.

The genus Pseudomonas includes some of the most problematic and studied foliar bacterial pathogens. Generally, in a successful disease cycle there is an initial epiphytic lifestyle on the leaf surface and a subsequent aggressive endophytic stage inside the leaf apoplast. Leaf-associated bacterial pathogens enter intercellular spaces and internal leaf tissues by natural surface opening sites, such as stomata. The stomatal crossing is complex and dynamic, and functional genomic studies have revealed several virulence factors required for plant entry. Currently, treatments with copper-containing

Presence of triploids among oak species.

BACKGROUND AND AIMS: Polyploids, organisms that have multiple sets of chromosomes, are common in certain plant and animal taxa. However, there are only a few reports of intraspecific ploidy variation within the genus Quercus. The aim of the study was to investigate the suspected ploidy level of two oaks that have unusual microsatellite banding patterns. METHODS: Polyploidy was investigated by using microsatellite analysis, stomata length measurements and nuclear DNA content estimation by flow cytometry. KEY RESULTS: Each putative triploid tree has patterns of microsatellite variation unexpec

Carbon dioxide or photosynthetically active radiation? Evaluation of the significance of individual environmental factors that control leaf stomatal development.

BACKGROUND AND AIMS: Stomata are pores in the plant epidermis that regulate gas exchange by controlling CO2 uptake and water loss through transpiration. Stomatal conductance, which determines the rate of this gas exchange, is influenced by both the dynamic aperture of individual stomata and stomatal density (StD). It is a matter of discussion whether a single dominant factor that controls StD exists or a combination of several independent factors results in the final StD. METHODS: We evaluated the role of photosynthetic photon flux density (PPFD) and internal CO2 concentration (Ci). Plants of

Stomatal density and aperture dynamics regulate drought response and yield in tomato.

In crops, key physiological traits such as transpiration, stomatal conductance, and photosynthesis are closely linked to productivity. However, these dynamic traits are often studied under steady-state conditions or modeled with limited data, failing to capture the plant's versatile responses to dynamic field conditions. We hypothesized that natural variation in stomatal morphological traits, specifically, abaxial-to-adaxial distribution and density regulate the temporal physiological response patterns in tomato introgression lines (ILs; crosses between WT, S. Pennellii, and M82 lines). We fur

Mapping CO2 fixation to two effective parameters: A framework toward data-informed species and model comparison.

To improve crop yield and resilience, it is essential to identify the steps limiting [Formula: see text] assimilation rate in plant leaves. The combined effect of multiple traits can be resolved by mechanistic models of the underlying diffusion, biochemistry, and geometry. Yet the widely used simple serial resistance models overlook tissue geometry, and detailed anatomical models are computationally heavy and rely on parameters that are difficult to measure. Here, we develop a framework for systematic species and model comparison, and find that the necessary level of model resolution is specie

Role of stomatal and leaf anatomical features in defining plant performance under elevated carbon dioxide and ozone, in the changing climate scenario.

This research investigates the interactive effects of elevated ozone (eO3) and carbon dioxide (eCO2) on stomatal morphology and leaf anatomical characteristics in two wheat cultivars with varying O3 sensitivities. Elevated O3 increased stomatal density and conductance, causing oxidative stress and cellular damage, particularly in the O3-sensitive cultivar PBW-550 (PW), compared to HUW-55 (HW). Conversely, eCO2 reduced stomatal density and pore size, mitigating O3-induced damage by limiting O3 influx. Ultrastructural analysis showed that eO3 increased plastoglobule density and damaged chloropla

Concurrent Temperature and Light Intensity Fluctuations Promote Stomatal Opening and Non-Steady State Photosynthesis.

In nature, fluctuations in light intensity (FL) are tightly coupled to rapid changes in leaf temperature (Tleaf), yet the short-term physiological effects of these concurrent drivers remain largely unresolved. Here, we combined rapid infrared induced Tleaf fluctuations with controlled step changes in light intensity for leaf gas exchange measurements to disentangle how each factor, and their interaction, affects stomatal conductance (gs) and net CO2 assimilation (A) in cucumber plants grown under constant light or FL. Modest but rapid increases in Tleaf (~3°C) alone triggered a pronounced wron

Coronatine promotes Pseudomonas syringae virulence in plants by activating a signaling cascade that inhibits salicylic acid accumulation.

Phytopathogens can manipulate plant hormone signaling to access nutrients and counteract defense responses. Pseudomonas syringae produces coronatine, a toxin that mimics the plant hormone jasmonic acid isoleucine and promotes opening of stomata for bacterial entry, bacterial growth in the apoplast, systemic susceptibility, and disease symptoms. We examined the mechanisms underlying coronatine-mediated virulence and show that coronatine activates three homologous NAC transcription factor (TF) genes, ANAC019, ANAC055, and ANAC072, through direct activity of the TF, MYC2. Genetic characterization

Genetic elucidation of hydrogen signaling in plant osmotic tolerance and stomatal closure via hydrogen sulfide.

Although ample evidence showed that exogenous hydrogen gas (H2) controls a diverse range of physiological functions in both animals and plants, the selective antioxidant mechanism, in some cases, is questioned. Importantly, most of the experiments on the function of H2 in plants were based on pharmacological approaches due to the synthesis pathway(s) in plants are still unclear. Here, we observed that the seedling growth inhibition of Arabidopsis caused by low doses of mannitol could progressively recover by recuperation, accompanied with the increased hydrogenase activity and H2 synthesis. Plant phenotypic adjustments in response to changes in atmospheric pCO2: insights from δ13C values and stomatal index in C3 plant leaves.

The commencement of the Industrial Revolution has resulted in an unprecedented increase in the concentration of atmospheric CO2 (pCO2). It is, therefore, important to understand how plant communities respond to increased levels of CO2 levels in the environment. To this end, we examined the effects of spatial variation in pCO2 on plant physiology using carbon isotope ratios (δ13C values) and stomatal index (SI) in C3 plant leaves along a transect from the central Ganga Plain to the foothills of the Himalayas with industrial and non-industrial zones. Our study shows that the plants adjacent to t
