Phytoremediation of trichloroethylene with hybrid poplars.

Axenic tumor cultures of poplar cells, clone H11-11, were grown in the presence of [14C]-trichloroethylene (TCE) (uniformly labeled). The cells were capable of metabolizing TCE to produce trichloroethanol, di- and trichloroacetic acid. Some of the carbon from TCE was found in insoluble, nonextractable cell residue, and small amounts were mineralized to [14C]CO2. Poplar cuttings grown in soil and exposed to TCE produced the same metabolites. In field trials, trees were planted in soil in test cells and exposed to TCE via underground water injection during the growing season. During the growing

Arbuscular mycorrhizal colonization does not improve root hydraulic supply in tomato and pea.

Arbuscular mycorrhizal (AM) fungi are known to enhance plant drought tolerance, but the physiological mechanism behind this benefit remains unclear. One explanation is that AM colonization improves root hydraulic conductance (Kr), thereby facilitating more efficient water uptake under soil drying, though this mechanism remains highly debated. Here, we measured Kr in tomato (Solanum lycopersicum L.) and pea (Pisum sativum L.) with and without AM using a noninvasive rehydration technique under soil drying, and this was complemented with the evaporative flux method under hydrated conditions. Anatomical Features of Foliar Vasculature as Flux-Capacity Proxies.

We present a procedure for preparing leaf tissue for minor-vein analysis using light and transmission electron microscopy that facilitates quantification of phloem and xylem features that correlate with photosynthetic capacity or transpiration rates, respectively, and thus provide proxies for foliar sugar- and/or water-transport capacity. We suggest here that the numbers and individual sizes of phloem cells of foliar minor veins (sometimes normalized by vein density) and transfer-cell wall ingrowths provide quantitative proxies for the capacity of the leaf to load and export photosynthates. Radial branch hydraulics and cellular injury thresholds in poplar: insights from controlled dehydration and high-resolution dendrometry.

Branch water release and radial exchange influence drought responses. Although mechanistic models describe radial hydraulic and storage dynamics, experimental quantification of hydraulic capacitance (C) and radial resistance (Rr) across broad ranges of xylem water-potential (Ψx), particularly under severe dehydration, remains limited. We quantified how C and Rr vary with Ψx in Populus albaL., assessed the value of coupling controlled osmotic dehydration with high-resolution dendrometry, and evaluated cellular damage and bud viability across increasing dehydration levels. Branch segments were s

Interactions between beech and oak seedlings can modify the effects of hotter droughts and the onset of hydraulic failure.

Mixing species with contrasting resource use strategies could reduce forest vulnerability to extreme events. Yet, how species diversity affects seedling hydraulic responses to heat and drought, including mortality risk, is largely unknown. Using open-top chambers, we assessed how, over several years, species interactions (monocultures vs mixtures) modulate heat and drought impacts on the hydraulic traits of juvenile European beech and pubescent oak. Using modeling, we estimated species interaction effects on timing to drought-induced mortality and the underlying mechanisms driving these impact

Responses of herbaceous plants to urban air pollution: effects on growth, phenology and leaf surface characteristics.

Vehicle exhaust emissions are a dominant feature of urban environments and are widely believed to have detrimental effects on plants. The effects of diesel exhaust emissions on 12 herbaceous species were studied with respect to growth, flower development, leaf senescence and leaf surface wax characteristics. A diesel generator was used to produce concentrations of nitrogen oxides (NO(x)) representative of urban conditions, in solardome chambers. Annual mean NO(x) concentrations ranged from 77 nl l(-l) to 98 nl l(-1), with NO:NO(2) ratios of 1.4-2.2, providing a good experimental simulation of

Plant water transport and aquaporins in oxygen-deprived environments.

Oxygen deprivation commonly affects plants exposed to flooding and soil compaction. The resulting root hypoxia has an immediate effect on plant water relations and upsets water balance. Hypoxia inhibits root water transport and triggers stomatal closure. The processes contributing to the inhibition of root hydraulic conductivity and conductance (hydraulic conductivity of the whole root system) are complex and involve changes in root morphology and the functions of aquaporins. Aquaporins (AQPs) comprise a group of membrane intrinsic proteins that are responsible for the transport of water, as w

A potential overestimation of CO2 physiological effects on evapotranspiration.

It is generally believed that CO2 physiological forcing can partially mitigate land surface drying under global warming by reducing stomatal conductance and evapotranspiration. Most of this type of study focuses on the direct regulation by vegetation physiology, overlooking interactive feedback from the atmosphere. Using fully coupled earth system model simulations, we find that the physiological benefit may have been optimistically overestimated. Vegetation-induced energy change may in turn further affect atmospheric vapor pressure deficit (VPD), exerting extra evapotranspiration demand indir

Loss of conductance between mesophyll symplasm and intercellular air spaces explains nonstomatal control of transpiration.

The conventional assumption is that stomatal conductance ([Formula: see text]) dominates the regulation of water and carbon dioxide fluxes between leaves and the atmosphere. Here, a nanoreporter of water status at the mesophyll cell surface and local xylem within intact maize leaves documents significant undersaturation of water vapor in the outside-xylem zone (OXZ) and a large loss of conductance of this zone ([Formula: see text]) at moderate xylem water stress, without stomatal closure or turgor loss. The ratio of the resistances [Formula: see text] serves as a predictive phenotype of unders

Interactions of mixed organic contaminants in uptake by rice seedlings.

Uptakes of o-chlorophenol (CP), 2,4-dichlorophenol (DCP), trichloroethylene (TCE), and atrazine (ATR), as single and mixed contaminants, by roots and shoots of rice seedlings (Oryza sativa L.) from hydroponic solutions were measured following a 48-h exposure of plant roots. As single contaminants, the concentrations of CP, DCP, and ATR in rice roots and shoots increased significantly with increasing concentrations in external solutions; however, TCE concentrations in rice roots and shoots decreased with increasing external TCE concentration or the exposure time. The observed bioconcentration f

Emergent feedforward and isohydric responses to soil and atmospheric aridity: insights from a time-dependent hydraulic model.

Drought and heat waves have synergistic effects on mortality as plants experience both supply and demand-side water stress. Which of these stresses ultimately controls stomatal conductance (gs) and whether patterns of regulation represent biological strategies or are imposed on the plant are addressed by way of a mechanistic hydraulic 'null' model. Dynamic losses of soil hydraulic conductivity are modeled in a soil domain fed by a maximum water flux from a deep-water source. A root-uptake plane extracts water from the soil in a two-node (root and leaf) plant model, with gs a function of leaf w

Excessive leaf oil modulates the plant abiotic stress response via reduced stomatal aperture in tobacco (Nicotiana tabacum).

High lipid producing (HLP) tobacco (Nicotiana tabacum) is a potential biofuel crop that produces an excess of 30% dry weight as lipid bodies in the form of triacylglycerol. While using HLP tobacco as a sustainable fuel source is promising, it has not yet been tested for its tolerance to warmer environments that are expected in the near future as a result of climate change. We found that HLP tobacco had reduced stomatal conductance, which results in increased leaf temperatures up to 1.5°C higher under control and high temperature (38°C day/28°C night) conditions, reduced transpiration, and redu

The role of evapotranspiration in the groundwater hydrochemistry of an arid coastal wetland (Península Valdés, Argentina).

Coastal wetlands are complex hydrogeological systems, in which saline groundwater usually occurs. Salinity can be attributed to many origins, such as dissolution of minerals in the sediments, marine contribution and evapotranspiration, among others. The aim of this paper is to evaluate the processes that condition the hydrochemistry of an arid marsh, Playa Fracasso, located in Patagonia, Argentina. A study of the dynamics and geochemistry of the groundwater was carried out in each hydrogeomorphological unit, using major ion and isotope ((18)O and (2)H) data, soil profiles descriptions and meas

Physiological responses of Vicia faba plants to sulfur dioxide.

Exposure of broad bean (Victa faba L.) plants to 270 +/- 32 and 670 +/- 45 micrograms m 3SO2 for 1.5 hr daily between 40 and 85 days of their ages resulted in an increase in their transpiration rate, water saturation deficit, phenol content, and peroxidase activity and a decrease in protein content. With the increase in number of exposures of plants to SO2, chlorotic and brown, necrotic visible injury signs were also developed in leaves. It was further noted that the magnitude of undesirable biochemical changes, which possibly helped in the formation of new pigment characteristic of necrotic t

Foliar abscisic acid content underlies genotypic variation in stomatal responsiveness after growth at high relative air humidity.

BACKGROUND AND AIMS: Stomata formed at high relative air humidity (RH) respond less to abscisic acid (ABA), an effect that varies widely between cultivars. This study tested the hypotheses that this genotypic variation in stomatal responsiveness originates from differential impairment in intermediates of the ABA signalling pathway during closure and differences in leaf ABA concentration during growth. METHODS: Stomatal anatomical features and stomatal responsiveness to desiccation, feeding with ABA, three transduction elements of its signalling pathway (H2O2, NO, Ca(2+)) and elicitors of thes

Photosynthetic responses of Monarch birch seedlings to differing timings of free air ozone fumigation.

To study the effects of different periods of ozone (O3) fumigation on photosynthesis in leaves of the Monarch birch (Betula maximowicziana), we undertook free air O3 fumigation to Monarch birch seedlings at a concentration of 60 nmol mol(-1) during daytime. Plants were exposed to O3 at early, late or both periods in the growing season. The light-saturated net photosynthetic rate (A(sat)) in July and August was reduced by O3 exposure through a reduction in the maximum rate of carboxylation (V(c,max)). In early September, on the other hand, despite a reduction in V(c,max), A(sat) was not reduced

Calcium localization and tipburn development in lettuce leaves during early enlargement.

Tissue concentrations of Ca, Mg, and K were determined across immature leaves of lettuce (Lactuca sativa L. 'Buttercrunch') at different stages of enlargement using electron microprobe x-ray analysis. The analysis was with a wavelength dispersive spectrometer to permit detection of low concentrations of Ca. Patterns of mineral accumulation in immature leaves that were exposed were compared to patterns of accumulation in leaves that were enclosed within a developing head. The leaves developing without enclosure were free to transpire and developed normally whereas leaves developing with enclosu

Seasonal dynamics and sun/shade heterogeneity of leaf gas exchange and VOC emissions inside a tall temperate forest canopy.

Leaf gas exchange is the key driver of forest carbon uptake and directly determines forest carbon sink activity. Additionally, plants release a variety of biogenic volatile organic compounds (VOCs) acting as stress signals of trees. However, continuous hourly resolved measurements of leaf gas exchange and VOC emissions in tall tree canopies are challenging and remain scarce. To this end, we developed a complex in-situ leaf gas exchange measurement system with 24 cuvettes deployed on mature Fagus sylvatica L. (n = 3) and Pseudotsuga menziesii (MIRBEL) FRANCO (n = 3) individuals in a mixed tempe

Canopy Carbon- and Water-Use Efficiencies in Response to Temperature and Water Deficit for Wheat.

The frequency and intensity of extreme climatic events increase the complexity in assessing climate change impacts on (agro)ecosystem functions and crop production. A better understanding of carbon and water fluxes for crop plants under climate change requires research based on direct canopy-scale measurements. By analysing a canopy gas exchange data set synthesised from 8 years' experimentation under semi-field conditions for the post-anthesis period of five wheat genotypes, we examined canopy carbon and water fluxes as well as carbon use efficiency (CUE) and water use efficiency (WUE) under

Exogenous Application of Cellulose-Based Materials for Improved Plant Fitness.

Exogenous application of bio-based nanomaterials provides a targeted strategy to modulate plant physiological and biochemical responses. This review synthesizes recent advancements in the foliar application of nanocellulose (NC), in particular, cellulose nanocrystals (CNC) and cellulose nanofibers (CNF), to enhance plant fitness. CNC-formed films provide physical and biochemical barriers that increase plant drought and cold stress tolerance. Topically applied CNC reduce non-stomatal transpiration and serve as insulators, allowing the flowering buds to successfully survive chilling, avoid freez
