The electrochemical behaviour of biomimetic monolayers of monogalactosyldiacylglycerol (MGDG) incorporating ubiquinone-10 (UQ) has been investigated. MGDG is the principal component in the thylakoid membrane and UQ seems a good substitute for plastoquinone-9, involved in photosynthesis chain. The monolayers have been performed using the Langmuir and Langmuir-Blodgett (LB) techniques and the redox behaviour of the LB films, transferred at several surface pressures on a glass covered with indium-tin oxide (ITO), has been characterized by cyclic voltammetry. The cyclic voltammograms show that UQ

In Arabidopsis thaliana, the main route of cyclic electron transport around PSI is sensitive to antimycin A, but the site of inhibition has not been clarified. We discovered that ferredoxin-dependent plastoquinone reduction in ruptured chloroplasts was less sensitive to antimycin A in Arabidopsis that overaccumulated PGR5 (PROTON GRADIENT REGULATION 5) originating from Pinus taeda (PtPGR5) than that in the wild type. Consistent with this in vitro observation, infiltration of antimycin A reduced PSII yields and the non-photochemical quenching (NPQ) of Chl fluorescence in wild-type leaves but no

Cyanobacteria achieve highly efficient photosynthesis using a CO2-concentrating mechanism relying on specialized Type I (NDH-1) complexes. Among these, NDH-13 and NDH-14 catalyze redox-coupled hydration of CO2 to bicarbonate, supporting carbon fixation in carboxysomes. The mechanism of coupling electron transfer to CO2-hydration by these variant NDH-1 complexes remains unknown. We engineered a Synechococcus PCC7942 strain that expresses exclusively the high flux/low affinity NDH-14 complex, enabling the observation of the coupling of CO2 hydration to cyclic electron flow in isolation from the

One of the major mechanisms of heavy metal toxicity is the induction of oxidative stress. Redox-active heavy metals, like chromium, can induce it directly, whereas redox-inactive metals, like cadmium, play an indirect role in the generation of reactive oxygen species (ROS). Living organisms defend themselves against oxidative stress taking advantage of low-molecular-weight antioxidants and ROS-detoxifying enzymes. Tocopherols and plastoquinol are important plastid prenyllipid antioxidants, playing a role during acclimation of Chlamydomonas reinhardtii to heavy metal-induced stress. However, pa

The increasing prevalence of herbicide-resistant weed biotypes and concerns regarding pesticide toxicity call for the discovery of new, potent, and environmentally benign pesticides. This process requires the identification of new target enzymes, including solanesyl diphosphate synthase (SPS). SPS catalyzes the production of solanesyl diphosphate, a precursor to plastoquinone and ubiquinone, which are essential for plant growth. Targeting the inhibition of SPS has enabled the identification of chemical compounds with agrochemical potential. Owing to its critical role in plant growth and its po

BACKGROUND: The need for new, efficacious, and safe crop protection chemistries to control evolving pest populations is ever-present to combat resistance derived from current crop protection offerings. Rimisoxafen is a novel pyrimidinyloxy benzene with herbicidal activity. Determination of mode of action (MoA) is key to de-risking newly introduced crop protection chemistries with regards to toxicology and the potential for field resistance.
RESULTS: Weeds treated with rimisoxafen display a distinct bleaching phenotype, leading to investigation of multiple herbicidal targets surrounding carote

Exposure of control (non-hardened) Arabidopsis leaves to high light stress at 5 °C resulted in a decrease of both photosystem II (PSII) (45 %) and Photosystem I (PSI) (35 %) photochemical efficiencies compared to non-treated plants. In contrast, cold-acclimated (CA) leaves exhibited only 35 and 22 % decrease of PSII and PSI photochemistry, respectively, under the same conditions. This was accompanied by an accelerated rate of P700(+) re-reduction, indicating an up-regulation of PSI-dependent cyclic electron transport (CET). Interestingly, the expression of the NDH-H gene and the relative abund

4-Hydroxyphenylpyruvate dioxygenase (HPPD) is an Fe(II)-dependent, non-heme oxygenase that catalyzes the conversion of 4-hydroxyphenylpyruvate to homogentisate. This reaction involves decarboxylation, substituent migration and aromatic oxygenation in a single catalytic cycle. HPPD is a member of the alpha-keto acid dependent oxygenases that typically require an alpha-keto acid (almost exclusively alpha-ketoglutarate) and molecular oxygen to either oxygenate or oxidize a third molecule. As an exception in this class of enzymes HPPD has only two substrates, does not use alpha-ketoglutarate, and

BACKGROUND: Cornea protects the eye against natural and anthropogenic ultraviolet (UV) damage and mechanical injury. Corneal incisions produced by UV lasers in ophthalmic surgeries are often complicated by oxidative stress and inflammation, which delay wound healing and result in vision deterioration. This study trialed a novel approach to prevention and treatment of iatrogenic corneal injuries using SkQ1, a mitochondria-targeted antioxidant approved for therapy of polyethiological dry eye disease.
METHODS: Rabbit models of UV-induced and mechanical corneal damage were employed. The animals w

According to one hypothesis, Parkinson's disease pathogenesis is largely caused by dopamine catabolism that is catalyzed on mitochondrial membranes by monoamine oxidase. Reactive oxygen species are formed as a byproduct of these reactions, which can lead to mitochondrial damage followed by cell degeneration and death. In this study, we investigated the effects of administration of the mitochondrial antioxidant SkQ1 on biochemical, immunohistochemical, and behavioral parameters in a Parkinson-like condition caused by protoxin MPTP injections in C57BL/6 mice. SkQ1 administration increased dopami

This study demonstrates the effect of high temperature stress on the heterogeneous behavior of PSII in Wheat (Triticum aestivum) leaves. Photosystem II in green plant chloroplasts displays heterogeneity both in the composition of its light harvesting antenna i.e. on the basis of antenna size (α, β and γ centers) and in the ability to reduce the plastoquinone pool i.e. the reducing side of the reaction centers (Q(B)-reducing centers and Q(B)-non-reducing centers). Detached wheat leaves were subjected to high temperature stress of 35°C, 40°C and 45°C. The chlorophyll a (Chl a) fluorescence trans

The semiquinone radical Q(A)- has been studied by electron spin echo envelope modulation (ESEEM) spectroscopy in Photosystem II membranes treated with CN- at various pH values. Two protein 14N nuclei (N(I) and N(II)) were found to be magnetically coupled with the Q(A)- spin. N(I) is assigned to an amide nitrogen from the protein backbone while N(II) is assigned to the amino nitrogen, N(epsilon), of an imidazole. Above pH 8.5 only the N(I) coupling is present while both N(I) and N(II) couplings are present at lower pH values. These results are interpreted in terms of a model based on the struct

Chloroplasts are equipped with a nuclear-encoded antioxidant defence system the components of which are usually expressed at high transcript and activity levels. To significantly challenge the chloroplast antioxidant system, Arabidopsis thaliana plants, acclimated to extremely low light slightly above the light compensation point or to normal growth chamber light, were moved to high light corresponding to a 100- and 10-fold light jump, for 6 h and 24 h in order to observe the responses of the water-water cycle at the transcript, protein, enzyme activity, and metabolite levels. The plants coped

Green microalgae, such as Chlamydomonas reinhardtii, show great potential for producing green hydrogen using only water and sunlight, with no carbon emissions. However, sustainable hydrogen production requires addressing the hydrogenase sensitivity to oxygen and enhancing electron allocation to this enzyme. Previous methods for hydrogen photoproduction rely on a brief nitrogen flushing followed by a dark incubation phase to establish anoxia prior to exposure to high light. In this study, we present a straightforward protocol involving a mixotrophic growth phase followed by a semi-autotrophic h

Responses of electron transport to three levels of irradiation (20, 200, and 1200 μmol photons m-2 s-1 PAR; exposures called LL, ML and HL, respectively) were investigated in eelgrass (Zostera marina L.) utilizing the chlorophyll a fluorescence technique. Exposure to ML and HL reduced the maximum quantum yield of photosystem II (PSII) (Fv/Fm) and the maximum slope decrease of MR/MRO (VPSI), indicating the occurrence of photoinhibition of both PSII and photosystem I (PSI). A comparatively slow recovery rate of Fv/Fm due to longer half-life recovery time of PSII and 40% lower descending amplitud

Solanesyl diphosphate synthases (SPSs), are key enzymes in the isoprenoid metabolic pathway, catalyze the biosynthesis of solanesyl diphosphate (SPP). In plants, the SPS family includes geranyl diphosphate synthase (GPS), farnesyl diphosphate synthase (FPS), geranylgeranyl diphosphate synthase (GGPPS), and SPS, catalyzing the stepwise condensation to form SPP. SPP is involved in the biosynthesis of solanesol and plastoquinone (PQ), both of which play important roles in plant growth, development, and the regulation of photosynthesis. Here, to elucidate the functions of the SPS subfamily in wood

The main reason for the maize genotype "DKC7221" to be heat tolerant is to have higher photosynthetic activity under heat stress conditions. The genotype "P3167" is sensitive to high temperature because of the heat-induced inhibition in photosynthetic electron transport reactions. In the present study, the effect of heat stress (45 ºC for 20 min) on some physiological changes was investigated through a chlorophyll afluorescence technique, and some endogenous resistance mechanisms (activities of some antioxidant enzymes, free proline, and reduced ascorbate contents) in two maize cultivars (Zea

Citrus sinensis seedlings were irrigated with nutrient solution at a concentration of 0 (Mg-deficiency) or 2 (Mg-sufficiency) mM Mg (NO3)2 for 16 weeks. Mg-deficiency-induced interveinal chlorosis, vein enlargement and corkiness, and alterations of gas exchange, pigments, chlorophyll a fluorescence (OJIP) transients and related parameters were observed in middle and lower leaves, especially in the latter, but not in upper leaves. Mg-deficiency might impair the whole photosynthetic electron transport, including structural damage to thylakoids, ungrouping of photosystem II (PSII), inactivation o

Lipid peroxidation plays an important role in various pathologies and aging, at least partially mediated by ferroptosis. The role of mitochondrial lipid peroxidation during ferroptosis remains poorly understood. We show that supplementation of exogenous iron in the form of ferric ammonium citrate at submillimolar doses induces production of reactive oxygen species (ROS) and lipid peroxidation in mitochondria that precede ferroptosis in H9c2 cardiomyocytes. The mitochondria-targeted antioxidant SkQ1 and the redox mediator methylene blue, which inhibits the production of ROS in complex I of the
