INTRODUCTION: In obese humans, Coleus forskohlii root extract (CF) protects against weight gain owing to the presence of forskolin, an adenylate cyclase (AC) activator. As AC increases intracellular cyclic adenosine monophosphate (cAMP) levels in osteoblasts that has an osteogenic effect, we thus tested the skeletal effects of a standardized CF (CFE) in rats.
METHODS: Concentrations of forskolin and isoforskolin were measured in CFE by HPLC. CFE and forskolin (the most abundant compound present in CFE) were studied for their osteogenic efficacy in vitro by alkaline phosphatase (ALP), cAMP and

Accessing plant resources to extract compounds of interest can sometimes be challenging. To facilitate access and limit the environmental impact, innovative cultivation strategies can be developed. Forskolin is a molecule of high interest, mainly found in the roots of Coleus forskohlii. The aim of this study was to develop aeroponic cultivation methods to provide a local source of Coleus forskohlii and to study the impact of abiotic stress on forskolin and bioactive metabolite production. Three cultivation itineraries (LED lighting, biostimulant, and hydric stress) along with a control itinera

The number of cases of failure in the treatment of infections associated with resistant bacteria is on the rise, due to the decreasing efficacy of current antibiotics. Notably, 7α-Acetoxy-6β-hydroxyroyleanone (AHR), a diterpene isolated from different Plectranthus species, showed antibacterial activity, namely against Methicillin-resistant Staphylococcus aureus (MRSA) strains. The high antibacterial activity and low cytotoxicity render this natural compound an interesting alternative against resistant bacteria. The aim of this study is to understand the mechanism of action of AHR on MRSA, usin

Plants are promising bioreactor for the sustainable and scalable production of high-value natural bioactive compounds, because they can synthesise phytochemicals from CO2, light, water and minerals through their innate photosynthetic carbon assimilation machinery. However, metabolic engineering in multicellular plants via stable transgene is challenged by the low accumulation of heterologous compounds. Forskolin is a labdane-type diterpenoid that accumulates in the roots of medicinal plant Coleus forskohlii. It activates the cyclic adenosine 3',5'-monophosphate (cAMP) signalling pathway and is

Coleus forskohlii supplements (FKSs) are widely consumed for their obesity-preventing effects, mainly related to forskolin, its main bioactive compound. However, these supplements are susceptible to adulteration and various types of frauds have been described. Therefore, the development of rapid and sensitive methodologies is of great interest in order to determine the quality of these products. In this work, a new method by Flow Injection Analysis-Mass Spectrometry (FIA-MS) has been developed and validated for the detection of potential frauds in FKSs. A Box-Behnken experimental design was us

Belgian endive-derived biostimulant (BEE) has been previously shown to enhance root and shoot growth of Arabidopsis thaliana and Plectranthus esculentus in in vitro culturing conditions. In this study, we evaluated the effect of BEE on A. thaliana subjected to abiotic stresses and assessed the translatability of its bioactivity to lettuce (Lactuca sativa) and sweet pepper (Capsicum annuum) grown in substrate and soil. A first set of experiments tested the impact of BEE on protection during and on recovery after osmotic or salt (NaCl) stress. BEE treatment had little to no restorative effect wh

INTRODUCTION: Glioblastoma (GB) is the most aggressive primary glioma, with a median survival of 15-18 months. Current treatments are often ineffective, largely due to tumor heterogeneity and recurrence. Advances in understanding GB's molecular landscape and microenvironment have highlighted new therapeutic strategies to fight this life-threatening tumor. Given the pivotal role of natural compounds in drug discovery, those with anti-inflammatory and cytotoxic/cytostatic properties are emerging as promising candidates for GB therapy.
METHODS: This study investigates the antitumor and immunomod

Fairness of skin, UV sensitivity and skin cancer risk all correlate with the physiologic function of the melanocortin 1 receptor, a Gs-coupled signaling protein found on the surface of melanocytes. Mc1r stimulates adenylyl cyclase and cAMP production which, in turn, up-regulates melanocytic production of melanin in the skin. In order to study the mechanisms by which Mc1r signaling protects the skin against UV injury, this study relies on a mouse model with "humanized skin" based on epidermal expression of stem cell factor (Scf). K14-Scf transgenic mice retain melanocytes in the epidermis and t
