The indices of blast transformation of lymphocytes to polyclonal mitogens of phytohemagglutinin (PHA) and Pokeweed mitogen (PWM) and to tick-borne encephalitis virus antigen (TBE) were studied in 80 individuals in the course of vaccination against tick-borne encephalitis. In the time course of vaccination reactions of blast transformation of lymphocytes was found to decrease to PHA and to increase to TBE and PWM antigens; spontaneous blast transformation also increased. We have compared the lymphocyte in vitro proliferative response induced by muscle extract, phytohaemagglutinin, and pokeweed mitogen in peripheral blood lymphocytes of patients with myasthenia gravis, polymyositis, dermatomyositis, and normal subjects. Twenty patients with sickle cell disease (14 black females and 6 black males, mean age 31.5 +/- 9.3) were studied by quantitating peripheral T and B lymphocyte percentages and measuring lymphocyte blast transformation (LBT) in response to phytohemagglutinin-P, concanavalin-A and pokeweed mitogen. Mitogen- and antigen-induced lymphocyte transformation was determined in peripheral blood lymphocytes from tobacco smokers and nonsmokers. Each group was evaluated by race, sex, and age subgroups. Male smokers as a group had depressed responses to phytohemagglutinin (PHA), concanavalin A, and pokeweed mitogen. The effect was most pronounced in all subjects on PHA stimulation. There was no apparent relationship to age or race. Antigenic stimulation of lymphocytes with mumps skin test antigen was not affected in any of the groups studied.

We report six Chinese boys with hypogammaglobulinaemia. All but one had very low or undetectable circulating B-lymphocytes, two had reversed CD4/CD8 ratios (in one of whom this latter became normal), one had reduced lymphocyte proliferative responses to concanavalin A and pokeweed mitogen and three had deficient responses to OKT3. Immunoglobulin production and its regulation have been investigated in 36 untreated patients with primary biliary cirrhosis and 14 healthy controls. Using a haemolytic plaque assay, the number of cells spontaneously producing IgG or IgM was normal, while in contrast, proliferation of both IgG and IgM producing cells in the presence of pokeweed mitogen was impaired (P less than 0.01). Six young adult women with recurrent rubella arthritis after HPV-77 DE/5 rubella immunization were studied.
