New insights into the regulatory effects of galloylated catechin on male reproduction in Camellia oleifera.

Flavonoids, particularly galloylated catechins, are known to regulate pollen tube growth, but their mode of action in woody plants remains poorly characterized. Here, we investigated the effects of epigallocatechin gallate (EGCG)-a major galloylated catechin-on Camellia oleifera pollen tubes using combined cytological and transcriptomic approaches. In vitro treatment with 100 μg mL-1 EGCG significantly reduced pollen germination and tube elongation. Confocal and fluorescence microscopy revealed that EGCG rapidly disrupted the apical Ca2+ gradient within 10 min, accompanied by disorganized acti

Identification and characterization of stretch-activated ion channels in pollen protoplasts.

Pollen tube growth requires a Ca2+ gradient, with elevated levels of cytosolic Ca2+ at the growing tip. This gradient's magnitude oscillates with growth oscillation but is always maintained. Ca2+ influx into the growing tip is necessary, and its magnitude also oscillates with growth. It has been widely assumed that stretch-activated Ca2+ channels underlie this influx, but such channels have never been reported in either pollen grains or pollen tubes. We have identified and characterized stretch-activated Ca2+ channels from Lilium longiflorum pollen grain and tube tip protoplasts. The channels

Chromosome doubling is associated with enhanced self-compatibility and transcriptional remodeling of pollen-pistil interactions in Lycium chinense.

Cytology and 72-sample pistil transcriptomes link chromosome doubling to self-compatibility in autotetraploid Lycium chinense through altered pollen-pistil responses and candidate S-RNase trafficking and detoxification pathways. Polyploidy can reshape plant reproductive traits and self-compatibility systems, yet the mechanisms underlying ploidy-associated transitions to self-compatibility remain poorly understood. In this study, diploid and autotetraploid Lycium chinense were compared using floral morphology, stigma receptivity, pollen viability, and pollen tube cytology, controlled pollinatio

Development of accessible and scalable maize pollen storage technology.

The inherent short lifespan of Zea mays (maize, corn) pollen hinders crop improvement and challenges the hybrid seed production required to produce food, fuel, and feed. Decades of scientific effort on maize pollen storage technology have been unable to deliver a widely accessible protocol that works for liters of pollen at a hybrid seed production scale. Here we show how suppressing the pollen cellular respiration rate through refrigeration and optimizing gas exchange within the storage environment are the critical combination of factors for maintaining pollen viability in storage. The common

A receptor heteromer mediates the male perception of female attractants in plants.

Sexual reproduction requires recognition between the male and female gametes. In flowering plants, the immobile sperms are delivered to the ovule-enclosed female gametophyte by guided pollen tube growth. Although the female gametophyte-secreted peptides have been identified to be the chemotactic attractant to the pollen tube, the male receptor(s) is still unknown. Here we identify a cell-surface receptor heteromer, MDIS1-MIK, on the pollen tube that perceives female attractant LURE1 in Arabidopsis thaliana. MDIS1, MIK1 and MIK2 are plasma-membrane-localized receptor-like kinases with extracell

Investigating morphological and biological reproductive traits in self-fertile and -infertile macadamia cultivars.

Self-fertility is a commercially valuable trait in crop species, enabling fruit set without reliance on pollinators or external pollinisers. In macadamia, most cultivars are self-infertile, though some can produce nuts with self-pollen. The mechanisms underlying this variation remain unclear. This study investigates herkogamy, dichogamy and in vivo pollen tube growth to investigate self-fertility. The traits were measured on cultivars from self-fertile and self-infertile groups. Herkogamy was assessed by pistil length (PL), stamen length (SmL), and stigma-anther distance (SAD). Significant int

Morphological characteristics of pollen from triploid watermelon and its fate on stigmas in a hybrid crop production system.

Hybrid crop production is more reliant on pollinators compared to open-pollinated crops because they require cross-pollination between a male-fertile and a male-sterile line. Little is known about how stigma receipt of pollen from male-sterile genotypes affects reproduction in hybrids. Non-viable and non-compatible pollen cannot fertilise plant ovules, but may still interfere with pollination success. Here we used seedless watermelon (Citrullus lanatus (Thunb.) Matsum. & Nakai) as a model hybrid plant, to evaluate the morphology, physiology, and movement of pollen from inter-planted genotypes

Winning in style: Longer styles receive more pollen, but style length does not affect pollen attrition in wild Clarkia populations.

PREMISE OF THE STUDY: One proposed function of long styles is to intensify selection among male gametophytes relative to short styles. If so, given sufficient competition, longer styles will have higher rates of pollen tube attrition (failure to reach the style base) within the style than shorter ones. Alternatively, style length may influence pollen receipt, which itself may affect attrition rates. METHODS: We tested these predictions by collecting senescing styles from wild populations of two insect-pollinated Clarkia species. We examined the number of pollen grains adhering to the stigma,

Central cell-produced salvagers promote fertilization recovery and interspecific hybridization.

Angiosperms face frequent fertilization failure in natural environments. To address this challenge, angiosperms have evolved fertilization recovery mechanisms to ensure successful reproduction. The central cell secretes pollen tube attractants to mediate fertilization recovery, but the molecular mechanisms remain poorly defined. In this study, we identified two central cell-produced peptides SALVAGER3 (SAL3) and SAL4 that attract pollen tubes when synergid cell-controlled attraction is compromised. Loss of SALs reduces the ovular capacity for fertilization recovery. These central cell-derived

Arabidopsis leucine-rich repeat malectin receptor-like kinases regulate pollen-stigma interactions.

Flowering plants contain tightly controlled pollen-pistil interactions required for promoting intraspecific fertilization and preventing interspecific hybridizations. In Arabidopsis (Arabidopsis thaliana), several receptor kinases (RKs) are known to regulate the later stages of intraspecific pollen tube growth and ovular reception in the pistil, but less is known about RK regulation of the earlier stages. The Arabidopsis RECEPTOR-LIKE KINASE IN FLOWERS1 (RKF1)/RKF1-LIKE (RKFL) 1-3 cluster of 4 leucine-rich repeat malectin (LRR-MAL) RKs was previously found to function in the stigma to promote

Cellular localization of ROS and NO in olive reproductive tissues during flower development.

BACKGROUND: Recent studies have shown that reactive oxygen species (ROS) and nitric oxide (NO) are involved in the signalling processes taking place during the interactions pollen-pistil in several plants. The olive tree (Olea europaea L.) is an important crop in Mediterranean countries. It is a dicotyledonous species, with a certain level of self-incompatibility, fertilisation preferentially allogamous, and with an incompatibility system of the gametophytic type not well determined yet. The purpose of the present study was to determine whether relevant ROS and NO are present in the stigmatic

Arabinogalactan Proteins in Pollen-Pistil Interactions: Molecular Regulators of Reproductive Barriers and Compatibility.

Arabinogalactan proteins (AGPs) are a family of highly glycosylated proteins that play an important role in the reproductive process of flowering plants. This review summarizes current knowledge of arabinogalactan proteins at various stages of male and female gametophyte development and in the regulation of their interactions-from pollen grain contact with the stigma, through pollen tube (PT) germination and its guided growth toward the embryo sac, to sperm cell delivery to the egg cell. Particular attention is given to the involvement of AGPs in pollen-pistil recognition, pollen tube growth a

From recognition to response: integrated signaling pathways determining pollen acceptance and rejection in Brassicaceae.

Generation of competent offspring is vital for the prosperity of flowering plants. The pistil not only functions as a conduit for pollen tubes to grow to the ovary but also provides a selective venue for facilitating the growth of compatible pollen tubes and discouraging invaders and incompatible pollen. This review integrates recent advances in pollen-pistil interactions on dry stigmas of the Brassicaceae in the domains of self-incompatibility (SI) and cross-compatibility. We first outline the initial recognition mechanisms that distinguish self from nonself pollen and then highlight how key

Assessment of Pollen Viability and Germination Through Histochemical, In Vivo and In Vitro Methods.

Pollen viability is a key parameter for understanding the functioning of plant reproductive systems and for supporting strategies aimed at genetic resource conservation and plant breeding. Accurate assessments of this attribute are essential for identifying genotypes with a higher reproductive rate, monitoring prezygotic barriers, testing compatibility between individuals, and planning controlled crosses, especially in cases of floral asynchrony or spatial isolation. Accordingly, this chapter compiles the main methods used for this purpose, providing detailed and up-to-date protocols. It cover

'Jinju' Pomelo pollen enhances fruit set and development in Citrus maxima 'Tomentosa': physiological and proteomic associations.

BACKGROUND: Citrus maxima (Burm.) Merr. 'Tomentosa' ('Luchuan Juhong') is a valuable medicinal and edible germplasm, yet its commercial cultivation is severely constrained by strong self-incompatibility, which results in poor fruit set and excessive physiological fruit drop. While artificial cross-pollination can alleviate this limitation, the relative efficacy of different pollen donors and the underlying molecular mechanisms remain largely unknown. RESULTS: We evaluated four pomelo cultivars as pollen donors and identified 'Jinju' Pomelo as the most effective. Following 'Jinju' pollination,

Electrostatic treatment enhances pollen viability and improves fruit quality in 'Xuxiang' kiwifruit.

Electrostatic treatment holds promise for enhancing pollen and seed viability, as well as improving crop quality. However, there are limited reports regarding the effects of electrostatically treated pollen on pollination and fruit quality in plants. In this study, an electrostatic pollination platform was established, treated pollen was used to pollinate the kiwifruit cultivar (Actinidia deliciosa cv. 'Xuxiang'), and in vitro germination experiments were conducted. In field experiment, style samples were collected every 0.5 hours over a continuous period of 20 hours, and pollen tube behavior

Air quality biomonitoring through pollen viability of Fabaceae.

In this study, pollen viability and germination of three plant species, Cercis siliquastrum L., Robinia pseudoacacia L., and Spartium junceum L., belonging to the Fabaceae family, was evaluated in sites with different intensity of road traffic, constantly monitored with continuous analysers for air pollutants (carbon monoxide (CO), sulphur dioxide (SO(2)), and nitrogen dioxide (NO(2))) by the Municipality of Catania. Two sites, in which road traffic was absent, were selected, too. The percentages of viable pollen by 2,3,5-trypheniltetrazolium chloride (TTC) test ranged from 59.0 to 90.2 % in C

To test if allergic rhinitis is associated with abnormal eustachian tube function (ET), juvenile rhesus monkeys were passively sensitized with serum samples from patients allergic to timothy grass or ragweed pollen. Tympanostomy tubes were inserted for direct measurement of ET function by inflation-deflation and forced-response testing before and after provocative intranasal challenge. Intranasal challenge with pollen (antigen) resulted in rhinorrhea and nasal obstruction. Slight increases in passive ET function values and significantly impaired active ET function were observed after antigen c

Functional versatility of synergid cells in plant reproduction.

In flowering plants (angiosperms), the Polygonum-type embryo sac accounts for approximately 70% of the female gametophytes. It contains three antipodal cells, one central cell, one egg cell, and two synergid cells. Apart from the female gamete cells, the synergid cells play a key role in sexual reproduction. This review traces the origin and research history of synergid cells and summarizes the latest research progress in understanding their core functions in the angiosperm-specific siphonogamy. In particular, we discuss the diverse functions of synergid cells in pollen tube guidance, pollen t
