Chemical modification of chlorinated microbial polyesters.

Chlorination of microbial polyesters poly(3-hydroxybutyrate) (PHB) and poly(3-hydroxyoctanoate) (PHO) was carried out by passing chlorine gas through their solutions. The chlorine contents in chlorinated PHB (PHB-Cl) and chlorinated PHO (PHO-Cl) were between 5.45 and 23.81 wt % and 28.09 and 39.09 wt %, respectively. Molecular weights of the chlorinated samples were in the range of between one-half to one-fourth of the original values because of hydrolysis during the chlorination process. Thermal properties of the PHO-Cl were dramatically changed with an increase in its glass transition (T(g)

Process-level optimization of light-driven CO2 fixation and PHB production by Synechocystis salina in scalable photobioreactor systems.

Photoautotrophic cultivation of cyanobacteria represents a sustainable platform for biomass-based carbon utilization and bioproduction, yet its scalability remains limited by light utilization, nutrient balance, and reactor design. In this study, an integrated photoautotrophic cultivation framework was established for Synechocystis salina through systematic optimization of light regime, nutrient availability, and photobioreactor configuration. Highly reproducible cultivation under inorganic BG-11 medium achieved a maximum dry cell weight of approximately 2 g L-1 within 9 days, corresponding to

An Efficient CO2-Upcycling Platform Based on Engineered Halomonas TD with Enhanced Acetate-Utilizing Capacity via Adaptive Laboratory Evolution.

Biohybrid conversion of carbon dioxide (CO2) into value-added bioproducts via engineered microbes using CO2-derived electrolytes (CDE) addresses global CO2 emissions, but most recombinants have poor saline CDE tolerance and low carbon conversion rate (CCR). Herein, Halomonas TD (salt-resistant) was adaptively evolved into TD80, which efficiently uses acetate; its aceE gene mutation (encoding pyruvate dehydrogenase) drives acetate utilization. Subsequently, different biosynthesis pathways in TD80 enabled high yields of poly-3-hydroxybutyrate (PHB), poly-3-hydroxybutyrate-co-4-hydroxybutyrate (P

Fast, inexpensive, and reliable HPLC method to determine monomer fractions in poly(3-hydroxybutyrate-co-3-hydroxyvalerate).

The determination of the monomer fractions in polyhydroxyalkanoates is of great importance for research on microbial-produced plastic material. The development of new process designs, the validation of mathematical models, and intelligent control strategies for production depend enormously on the correctness of the analyzed monomer fractions. Most of the available detection methods focus on the determination of the monomer fractions of the homopolymer poly(3-hydroxybutyrate). Only a few can analyze the monomer content in copolymers such as poly(3-hydroxybutyrate-co-3-hydroxyvalerate), which us

Degradation of Trichloroethylene by Methanol-Grown Cultures of Methylosinus trichosporium OB3b PP358.

A soluble methane monooxygenase-constitutive mutant strain of Methylosinus trichosporium OB3b, strain PP358, was grown with methanol as the carbon source, and the kinetics of trichloroethylene (TCE) degradation were determined. PP358 exhibited high TCE degradation rates under both oxygen- and carbon-limiting conditions. The optimal pseudo first-order rate constant for TCE was comparable to the values measured for cells grown with methane. We found that growth under oxygen-limiting conditions results in increased accumulation of polyhydroxybutyrate, which in turn correlates with higher transfor

Persistent low PHB accumulation during pilot-scale gas fermentation in Cupriavidus necator.

Climate change and food security are tightly linked global challenges. Gas fermentation offers a sustainable route to convert CO₂ into microbial single-cell protein (SCP), with hydrogen-oxidizing bacteria (HOB) emerging as efficient biomass producers via the Calvin-Benson-Bassham cycle. While SCP can replace protein-rich feed components, intracellular polyhydroxybutyrate (PHB) accumulation redirects carbon and reducing power away from biomass and protein synthesis, reducing process efficiency. With this aim, a low amount of PHB is needed to focus on protein production rather than PHB accumulat

Characterization of Streptomyces species with poly(3-hydroxybutyrate) degradation capabilities isolated from rice field soil.

The shift towards sustainable alternatives to petroleum-based polymers has become essential for addressing environmental challenges. Among these alternatives, bio-plastics such as poly(3-hydroxybutyrate) (PHB) have gained considerable attention due to their biodegradability into water and carbon dioxide through microbial activity. PHB is one of the most widely commercialized bio-plastics. However, its excessive accumulation in the environment due to insufficient degradation remains a significant ecological concern. This study focused on isolating and characterizing PHB-degrading bacteria from

Evaluation of the Highly Ordered Structure, Ligature, and Enzymatic Degradation of Poly[(R)-3-hydroxybutyrate-co-4-hydroxybutyrate] Elastic Porous Fibers.

We prepared biocompatible elastic fibers with high porosity and high tensile strength from poly[(R)-3-hydroxybutyrate-co-4-hydroxybutyrate], which is a microbial polyester that can be produced from renewable carbon resources by isothermal crystallization. It was possible to control the pore size by adjusting the isothermal crystallization time. Most of the pores were approximately less than 10 μm in diameter, did not penetrate, and were distributed discontinuously throughout the fibers. The elasticity of the fibers was apparently attributable to the generation of tie molecules with planar zigz

Binding-Enhanced Organic Semiconductor-Bacteria Hybrids for Efficient Visible Light-Driven CO2 Conversion to Bioplastics.

Photosynthetic semiconductor-biohybrids, which integrate excellent light-harvesting capabilities of semiconductors and high activity and selectivity of biocatalysts, have proven a promising strategy to enable solar-driven CO2 conversion to chemicals. However, broadening the light-harvesting scope and strengthening interfacial contact, which facilitates enhancing solar-to-chemical conversion efficiencies, remain challenging. Herein, we report a series of organic semiconductors (OSCs) to hybridize with nonphotosynthetic bacteria, Ralstonia eutropha, realizing efficient CO2 photoreduction to poly

Avocado seed waste bioconversion into poly(3-hydroxybutyrate) by using Cobetia amphilecti and ethyl levulinate as a green extractant.

The avocado processing industry produces up to 1.3M tons of agro-waste annually. Chemical analysis of avocado seed waste (ASW) revealed that it is rich in carbohydrates (464.7 ± 21.4 g kg-1) and proteins (37.2 ± 1.5 g kg-1). Optimized microbial cultivation of Cobetia amphilecti using an acid hydrolysate of ASW, generated poly(3-hydroxybutyrate) (PHB) in a 2.1 ± 0.1 g L-1 concentration. The PHB productivity of C. amphilecti cultivated on ASW extract was 17.5 mg L-1 h-1. The process in which a novel ASW substrate was utilized has been further augmented by using ethyl levulinate as a sustainable

Preparation, statistical optimization and characterization of poly(3-hydroxybutyrate) fermented by Cupriavidus necator utilizing various hydrolysates of alligator weed (Alternanthera philoxeroides) as a sole carbon source.

Alligator weed (Alternanthera philoxeroides) is a stoloniferous, amphibious and perennial herb which has invaded many parts of the world and led to serious environmental and ecological problems. In order to exploit cheap carbon source for poly(3-hydroxybutyrate) (PHB) production, alligator weed hydrolysates were prepared by acid and enzyme treatment and used for PHB production via Cupriavidus necator. The bacterium utilized alligator weed enzymatic hydrolysate and produced the PHB concentration of 3.8 ± 0.2 g/L at the conditions of pH 7.0, 27.5°C, 1.5 g/L of nitrogen source, and 25 g/L of carb

Distribution and selection of poly-3-hydroxybutyrate production capacity in methanotrophic proteobacteria.

Methanotrophs are known to produce poly-3-hydroxybutyrate (PHB), but there is conflicting evidence in the literature as to which genera produce the polymer. We screened type I and II proteobacterial methanotrophs that use the ribulose monophosphate and serine pathways for carbon assimilation, respectively, for both phaC, which encodes for PHB synthase, and the ability to produce PHB under nitrogen-limited conditions. Twelve strains from six different genera were evaluated. All type I strains tested negative for phaC and PHB production; all Type II strains tested positive for phaC and PHB produ

PHB Biomanufacturing in China.

China has made rapid progress from fundamental research to industrial translation in polyhydroxyalkanoates (PHA) biomanufacturing, with polyhydroxybutyrate (PHB) and its copolymers as representative products. Propelled by the national "dual carbon" strategy, Chinese researchers and enterprises have developed unconventional halophilic chassis and next-generation industrial biotechnology (NGIB) to reduce the sterilization burden of conventional fermentation and enable open, continuous, and unsterile production. This review focuses on the NGIB-centered PHA/PHB landscape in China while also noting

[Use of rifampicin for directed modification of the nucleotide composition of the cells of Bacillus megaterium and Candida utilis].

The object of this work was to study the effect of rifampicin on the physiological state of Bacillus megaterium and Candida utilis in chemostat and batch cultures. When B. megaterium is cultivated in a medium containing 1 microgram of rifampicin per litre, its metabolism changes, the pool of free nucleotides rises, and the content of RNA drops in response to the specific action of the inhibitor. The modified state is observed throughout many generations. The action of the inhibitor is also non-specific: protein content falls and polyhydroxybutyric acid is accumulated. The morphology of B. mega

Pressure Dependent Changes in CO2-Concentrations Modulate the 3-Hydroxyvalerate Fraction in Terpolymeric Polyhydroxyalkanoates.

Polyhydroxyalkanoates (PHAs), especially their copolymeric forms, such as poly(3-hydroxybutyrate-co-3-hydroxyvalerate-co-4-hydroxyvalerate) (PHBVV), have promising properties for replacing petrochemical-derived plastics. However, the research focuses mostly on producing PHA from a specific substrate or waste-stream, while the reproducibility of a precise polymer composition is seldom targeted. This study systematically focuses on the influence of pressure conditions on the production of terpolymeric PHBVV by Cupriavidus necator NCIMB 11599 using levulinic acid (LA). Three different process set

Poly(3-hydroxybutyrate) production from swine wastewater and waste vegetable oil by Cupriavidus necator in nonsterile bioreactors.

Swine wastewater (SW), rich in nitrogen and phosphorus, and waste vegetable oil (WVO) are abundant feedstocks with potential for microbial biopolymers production. However, high polyhydroxyalkanoates (PHA) yields under nonsterile conditions remain challenging due to inhibitory compounds and microbial competition. Here, we cultivated Cupriavidus necator DSM545 under nonsterile conditions with varying SW dilutions combined with WVO for PHA synthesis. Batch bioreactors containing two-thirds (v/v) non-centrifuged wastewater yielded the highest polyhydroxybutyrate titers of 6.8 ± 0.1 gPHB/L, with C.
