Histopathology of explanted AlphaCor due to keratoprosthesis extrusion.

AlphaCor keratoprosthesis (KPro) is a new-concept poly (2-hydroxyethyl methacrylate) one-piece KPro that makes possible a two-step implantation technique easy to perform with a short learning curve. In literature an 18% incidence of AlphaCor removal due to melting complications is reported. The histopathology of corneal tissue removed during a re-operation while bearing an AlphaCor KPro has previously been described in the literature only in one report. Herein, the first histological features of an AlphaCor-corneal complex explanted because of KPro extrusion is described. The histopathology of

Human histologic evaluation of HTR polymer and freeze-dried bone allograft. A case report.

This case report compares the results of placement of HTR (Hard Tissue Replacement Synthetic Bone) and freeze-dried bone allograft (FDBA) in the same patient. 2 notches were made in each of 6 teeth, 1 at the gingival margin and the other at the most apical level of calculus. Soft tissue responses with both materials included probing depth reduction, gingival shrinkage and gain in clinical attachment. Histological sections of block extraction sites 30 months after placement of either material showed no signs of new attachment. Little or no inflammation was present with both materials. There was

Photodynamic anti-biofilm PHEMA/Rose Bengal hydrogels enhanced by iodide: evidence for hydrogen peroxide and triiodide formation.

Biofilm-associated infections remain a major challenge for biomedical devices, motivating the development of light-activated antimicrobial materials. Here we report the antibiofilm activity (against Staphylococcus aureus and Pseudomonas aeruginosa) of a photodynamic hydrogel consisting of Rose Bengal (RB) physically entrapped within a crosslinked poly(2-hydroxyethyl methacrylate) (PHEMA) matrix. Under visible-light irradiation, RB-PHEMA alone produced only modest reductions (ca. 1 log), whereas addition of iodide markedly enhanced antibiofilm performance, yielding a 4.2-log reduction for S. au

Mechanically enhanced nested-network hydrogels as a coating material for biomedical devices.

UNLABELLED: Well-organized composite formations such as hierarchical nested-network (NN) structure in bone tissue and reticular connective tissue present remarkable mechanical strength and play a crucial role in achieving physical and biological functions for living organisms. Inspired by these delicate microstructures in nature, an analogous scaffold of double network hydrogel was fabricated by creating a poly(2-hydroxyethyl methacrylate) (pHEMA) network in the porous structure of alginate hydrogels. The resulting hydrogel possessed hierarchical NN structure and showed significantly improved

Surface immobilized α-1 acid glycoprotein and collagen VI modulate mouse macrophage polarization and reduce the foreign body capsule.

Macrophages are widely recognized in modulating the foreign body response, and the manner in which they do so largely depends on their activation state, often referred to as their polarization. This preliminary study demonstrates that surface immobilized α-1 acid glycoprotein (AGP), as well as collagen VI (Col6) in conjunction with AGP, can direct macrophages towards the M2 polarization state in vitro and modify the foreign body response in vivo. AGP and Col6 are immobilized onto poly(2-hydroxyethyl methacrylate) (pHEMA) surfaces using carbonyl diimidazole chemistry. Mouse bone marrow derived

Designing dapsone polymer conjugates for controlled drug delivery.

UNLABELLED: Polymer-drug conjugates have significantly influenced polymer therapeutics over the last decade via controlled pharmacokinetics. Dapsone (4,4'-diamino diphenylsulphone) is not only widely used in the treatment of leprosy but forms an essential component in the treatment of autoimmune inflammatory diseases and malaria. However, its low bioavailability and non-specific distribution in the body leads to absorption throughout organs including skin, liver, and kidneys that can cause serious side effects. Thus, in this study we report the synthesis of polymer-drug conjugates of dapsone c

[Clinical and histological evaluation of implantation of a microporous resin in periodontitis. Report of a case after 13 months].

A patient with adult periodontitis was treated for a 2 and 3-walled intrabony lesion on tooth n degrees 27. After scaling and root planing, the defect was surgically exposed and filled with a micro-porous resin. Following healing, there was a reduction in pocket depth as a result of gingival recession and a gain in probing attachment levels. This was confirmed 13 months post-operatively by clinical observation on re-entry. A biopsy of the treated site was taken at that time and undecalcified sections were studied by light microscopy and microradiography. New bone formation was noted inside and

Progressive maturation resistance to microcystin-LR cytotoxicity in two different hepatospheroidal models.

To develop three-dimensional (3D) cytotoxicity models further, microcystin-induced cytoskeletal disruption was tested in two different models of multicellular hepatocyte aggregate formation (hepatospheroids). Rat hepatocyte suspensions were seeded either onto poly(2-hydroxyethylmethacrylate)-treated culture wells (poly-HEMA) or in a rotating wall vessel (RWV) device which provides minimal shear forces and enhances differentiated 3D growth. Ninety percent of spheroids forming on poly-HEMA tended to fuse and form nonhomogeneous multilobular structures by day 4 of incubation. In contrast, spheroi

Contact lens induced peripheral ulcers during hydrogel contact lens wear.

PURPOSE: Ulcerative keratitis is the most serious adverse response associated with contact lens wear. However, some reports of microbial keratitis in the literature may refer to a less severe form of corneal ulceration, which we have termed contact lens induced peripheral ulcers (CLPU). METHODS: We present details of 11 cases of CLPU which occurred in clinical trials at our research unit. RESULTS: CLPU episodes are characterized by a single, small (< 2 mm diameter) circular lesion in the corneal mid-periphery or periphery, involving a focal loss of the full thickness of the epithelium associ

Inflammatory nodules following soft tissue filler use: a review of causative agents, pathology and treatment options.

Nodule development is a common complication following the use of fillers for soft tissue augmentation and is commonly categorized as inflammatory or non-inflammatory in nature. Inflammatory nodules may appear anywhere from days to years after treatment, whereas non-inflammatory nodules are typically seen immediately following implantation and are usually secondary to improper placement of the filler. Although inflammatory nodules are more common with permanent fillers such as silicone, inflammatory nodule development following administration of temporary fillers such as hyaluronic acid and col

Poly(2-hydroxyethyl methacrylate-co-methacrylated hyaluronan-β-cyclodextrin) hydrogel: A potential contact lens material with high hydrophilicity, good mechanical properties and sustained drug delivery.

A novel poly(2-hydroxyethyl methacrylate-co-methacrylated hyaluronan-β-cyclodextrin) [p(HEMA-co-mHA-β-CD)] hydrogel was developed as a potential contact lens for ophthalmic disease. The hydrogel was synthesized from the copolymerization of 2-hydroxyethyl methacrylate (HEMA) monomer and mHA-β-CD as a hydrophilic macromolecular crosslinker. By adjusting the methacrylate substitution degree in hyaluronan (20-29 %) and the mHA-β-CD content (5-11 %), transparent p(HEMA-co-mHA-β-CD) hydrogels were achieved. p(HEMA-co-m20HA-β-CD) hydrogels exhibited an enhanced tensile modulus (from 0.35 to 0.88 MPa)

Toxicity and photosensitizing assessment of gelatin methacryloyl-based hydrogels photoinitiated with lithium phenyl-2,4,6-trimethylbenzoylphosphinate in human primary renal proximal tubule epithelial cells.

Gelatin methacryloyl (GelMA) and lithium phenyl-2,4,6-trimethylbenzoylphosphinate (LAP) photoinitiator are commonly used in combination to produce a photosensitive polymer but there are concerns that must be addressed: the presence of unreacted monomer is well known to be cytotoxic, and lithium salts are known to cause acute kidney injury. In this study, acellular 10% GelMA hydrogels cross-linked with different LAP concentrations and cross-linking illumination times were evaluated for their cytotoxicity, photosensitizing potential, and elastic moduli. "Bioactive" Therapeutic Contact Lens Triggered by Biomimetic Chiral Helical Nanoarchitectonics for Rapid Corneal Repair.

Consistent corneal epithelial injury would cause chronic inflammation, neovascularization, and even corneal scarring, resulting in vision loss. Rapid repair is crucial for treatment, within which the use of therapeutic contact lenses presents great promise. A great challenge is how to achieve rapid repair of severely deficient corneal epithelium and regulation of the oxidative stress environment simultaneously. Herein, a "bioactive" therapeutic contact lens, mimicking the layered helical structure of the native cornea, is designed based on the assembly of cellulose nanocrystals (CNCs) inside p

An integrated lubricating and antibacterial hydrogel coating for the inner surface of medical catheters.

Silicone rubber (PDMS) catheters are prone to tissue friction and bacterial infection due to their inherent hydrophobicity, yet achieving a durable and functional coating on the inner lumen remains technically challenging. This study presents a multifunctional hydrogel coating designed for the inner wall of medical silicone tubing to address these issues. The coating, composed of PHEMA-PVP-glycerol, was applied via selective benzophenone pretreatment, followed by UV curing. Systematic optimization revealed that a 40% glycerol content achieves an optimal balance of properties, yielding strong i
