Stage-dependent inhibitory mechanism of ε-polylysine against Clostridium perfringens spores: inner membrane damage-mediated metabolic imbalance.

Clostridium perfringens (C. perfringens) spores pose a persistent food safety risk owing to their high resistance, yet the mechanisms governing their germination and outgrowth remain poorly understood. This study utilized ε-polylysine (ε-PL) as a model antimicrobial to systematically investigate its inhibitory effects and underlying mechanisms during the spore-to-vegetative cell transition. The results indicate that ε-PL had the least effect on early germination events; over 60 min, no significant differences were observed between the treatment and control groups in terms of OD₆₀₀ reduction or

ε-Polylysine/Sodium Alginate Bilayer-Modified Nanoliposomes Enhancing the Stability and In Vitro Bioavailability of Epigallocatechin Gallate.

Epigallocatechin gallate (EGCG) represents the key phenolic compound in green tea, which has been verified to possess various biological effects but suffers from low stability and poor bioavailability. To address these issues, EGCG-loaded nanoliposomes (ELs) were screened and prepared using an ethanol injection-calcium acetate gradient (EtOH-CAG) method. An encapsulation efficiency of 94.61% was achieved, involving a particle size of 118.6 nm and a polydispersity index (PDI) of 0.23. Via layer-by-layer assembly, nanoliposomes modified with either ε-polylysine (ε-PL) monolayer (ELP) or ε-polyly

High Pressure Thermal Sterilization and ε-Polylysine Synergistically Inactivate Bacillus subtilis Spores by Damaging the Inner Membrane.

ABSTRACT: This study was conducted to determine the sterilization effect of a combination of high pressure thermal sterilization (HPTS) and ε-polylysine (ε-PL) on Bacillus subtilis spores. The spores were treated with HPTS (550 MPa at 25, 65, and 75°C) and ε-PL at 0.1 and 0.3%. HPTS and ε-PL synergistically decreased the number of surviving spores and increased the release of the intracellular components in the spore suspension, with the maximal effects from treatment with 550 MPa at 75°C plus 0.3% ε-PL. Maximum fluidity and permeability of the cell inner membrane were observed with 550 MPa at

DNAzymes and Oligodeoxyribonucleotides Immobilized on Aminopropylsilanol and Polylysine-Anatase Nanoparticles as Active Agents Against Influenza A/H5N1 Virus.

The nanocomposites based on aminopropylsilanol (Si~NH2) and polylysine-anatase (Ans/PL) nanoparticles bearing DNAzymes (Dz) and oligodeoxyribonucleotides (ON) can effectively inhibit the replication of the influenza A/H5N1 virus in cell culture. Nanocomposites based on both types of nanoparticles exhibit high activity. The Dz-containing nanocomposites exhibited higher antiviral activity than those containing ON, reducing influenza A/H5N1 virus production by 3.5 log versus 2.4-3.0 log, respectively. Notably, the effect of Dz incorporated into the nanoparticles was an order of magnitude higher t

Construction and properties of antibacterial sanxan gum/gelatin pads loaded with ε-polylysine and application on bananas preservation.

This study aimed to prepare ε-polylysine (ε-PL)-loaded gelatin/sanxan gum (SG) composite aerogel pads via freeze-drying. Here, ε-PL functioned as both a crosslinking agent and an antibacterial component. Three formulations (G1S2, G1S1, G2S1) with different gelatin/SG mass ratios were fabricated. These aerogel pads were characterized by Fourier transform infrared spectroscopy (FTIR), Scanning electron microscopy (SEM), X-ray diffraction (XRD), and Thermogravimetric analysis (TGA). The results showed that G2S1 had a high-water absorption rate and achieved the highest encapsulation efficiency for

Construction of hyperbranched polylysine-modified antibacterial polyurethane foam dressings and their promotion of infected deep burn wound healing.

Foam dressings are vital for exuding wounds, yet antibacterial modification remains challenging and relies heavily on cytotoxic silver compounds. To address these challenges, in this study we developed a process for the antibacterial modification of polyurethane foam dressings using hyperbranched polylysine. The modification process consisted of a simple five-step procedure (soaking-drying-reacting-washing-drying) that did not compromise the foam's intrinsic properties, including hydrophilicity, pore structure and mechanical performance. The optimized modified foam achieved above 99.9% killing

Antibiofilm efficacy of ε-polylysine hydrochloride against Enterococcus faecalis biofilms and its biocompatibility: A promising root canal medicament.

OBJECTIVES: Enterococcus faecalis (E. faecalis) is a predominant pathogen associated with refractory apical periodontitis (RAP), owing to its strong biofilm-forming ability and alkali resistance, which may compromise the antimicrobial efficacy of conventional calcium hydroxide (Ca(OH)₂)-based intracanal medication. This study aimed to evaluate the antibacterial and antibiofilm effects of ε-polylysine hydrochloride (ε-PLH) against E. faecalis within the root canal microenvironment, as well as its cytocompatibility, effects on dentin microhardness, removability, and potential to promote periapic

Photocrosslinked polylysine/oxidized hyaluronic acid double-network hydrogel with anti-inflammatory and antibacterial functions for skin wound healing.

Conventional wound dressings often struggle to simultaneously address infection control, inflammatory regulation, and tissue regeneration during skin wound healing. To overcome these limitations, we developed a multifunctional dual-network hydrogel dressing (GE-OHA2@Rut), which integrates antibacterial, anti-inflammatory, and pro-regenerative functions. This system was constructed via UV-initiated crosslinking of methacrylated poly-l-lysine (GE) and Schiff base crosslinking of oxidized hyaluronic acid (OHA), followed by incorporation of the natural anti-inflammatory flavonoid rutin. The GE-OHA

Enhancement of waterborne pathogen removal by functionalized biochar with ε-polylysine ″dynamic arms″: Potential application in ultrafiltration system.

Widespread outbreaks of threatening infections caused by unknown pathogens and water transmission have spawned the development of adsorption methods for pathogen elimination. We proposed a biochar functionalization strategy involving ε-polylysine (PLL), a bio-macromolecular poly(amino acid)s with variable folding conformations, as a "pathogen gripper" on biochar. PLL was successfully bridged onto biochar via polydopamine (PDA) crosslinking. The extension of electropositive side chains within PLL enables the capture of both nanoscale viruses and micrometer-scale bacteria in water, achieving exc

Entirely physical crosslinking strategy: pH-tuned double-network hydrogel composed of ferulic acid-ε-polylysine and polyvinyl alcohol.

This study reports a novel pH-tuned double-network hydrogel via an entirely physical crosslinking strategy. The hydrogel consists of a first network formed between ferulic acid and ε-polylysine via hydrogen bonding and electrostatic interaction, while a second polyvinyl alcohol network established through freeze-thaw cycles. The former network confers biological activities, while the combination of the two networks ensures mechanical strength. The hydrogels exhibit pH-tuned behavior, with significant differences in mechanical performance, swelling characteristics, and antioxidant/antibacterial

Anti-Adhesion Effects and Potential Mechanisms of Polyglutamic Acid-Polylysine Crosslinked Hydrogels in a Rat Peritoneal Adhesion Model.

Peritoneal adhesions are a common complication following abdominal surgery. Their formation is associated with factors such as inflammatory responses, imbalance in the fibrinolytic system, and proliferation of fibrous tissue. Currently available anti-adhesion products on the market are primarily physical barrier membranes, which have limited functionality, can only serve as passive barriers, and have suboptimal handling and residence times. To address this issue, this study utilized 1-ethyl-3-(3-dimethylaminopropyl)carbodiimide hydrochloride/N-hydroxysuccinimide (EDC/NHS) chemical crosslinking

Injectable self-healing hydrogel fabricated from antibacterial carbon dots and ɛ-polylysine for promoting bacteria-infected wound healing.

Developing highly efficient pharmaceuticals to eradicate pathogens and facilitate wound healing is of great concern. Despite some cationic carbon dots (CDs) have been used for sterilization, hardly any anionic CDs with antimicrobial activity have appeared. In the present work, we engineered a string of anionic CDs (especially CD31) as valid broad-spectrum bactericides to kill bacteria. Furthermore, CD31 conjugated with ɛ-polylysine (Plys) to construct injectable, and self-healing hydrogel (CD-Plys) that possess the advantages of remarkable broad spectrum antibacterial activity, excellent wound

Polylysine-modified polyethylenimine polymer can generate genetically engineered mesenchymal stem cells for combinational suicidal gene therapy in glioblastoma.

Glioblastoma remains the most resistant malignant brain tumor owing to the lack of an efficient delivery system for therapeutic genes or drugs, especially in outgrowing tumor islands. Cell-based delivery systems such as mesenchymal stem cells (MSCs) are a potential candidate in this regard. Conventionally, MSCs have been genetically modified for cancer therapy by using viral vectors that can illicit oncogenicity and limit their use in clinical trials. In this study, we have used nonviral agents such as the polylysine-modified polyethylenimine (PEI-PLL) copolymer to generate genetically enginee

N epsilon-cyclosuccinyllysine: synthesis and formation from hemisuccinylated polylysine and its hapten conjugates with sulfamethoxazole during acid hydrolysis.

The novel amino acid N epsilon-cyclosuccinyllysine (4) forms as a side reaction during acid hydrolysis of N epsilon-hemisuccinylated succinylsulfamethoxazole-polylysine conjugates. The presence of 4 in hydrolysates can be obscured in amino acid analysis. Identification of 4 was based on chromatographic and electrophoretic comparisons with authentic N epsilon-cyclosuccinyllysine, which was synthesized in high yield by acid-catalyzed ring closure, under anhydrous conditions, of N epsilon-hemisuccinyl-L-lysine. The latter was prepared by an improved route, starting from N-alpha-(tert-butyloxycarb
