[Effects of neighbors with different sexuality and soil sterilization on biomass of Populus cathayana seedlings].

Although plant-plant interactions have been intensively studied, few studies examined sex-related neighbor interactions in dioecious plant species. Here, we investigated the sexual diffe-rences in biomass accumulation, and analyzed the sexual neighbor effects and yields of sexual combination in Populus cathayana seedlings under the treatments of inter- and intra-sex neighbors and soil sterilization. The results showed that biomass accumulation of P. cathayana decreased in both of sexes when grown with an intra-sex neighbor. Females increased biomass accumulation under non-sterilization conditi

The nature of the progression of drought stress drives differential metabolomic responses in Populus deltoides.

BACKGROUND AND AIMS: The use of woody crops for Quad-level (approx. 1 × 1018 J) energy production will require marginal agricultural lands that experience recurrent periods of water stress. Populus species have the capacity to increase dehydration tolerance by lowering osmotic potential via osmotic adjustment. The aim of this study was to investigate how the inherent genetic potential of a Populus clone to respond to drought interacts with the nature of the drought to determine the degree of biochemical response. METHODS: A greenhouse drought stress study was conducted on Populus deltoides 'W

A transcriptomic network underlies microstructural and physiological responses to cadmium in Populus x canescens.

Bark tissue of Populus × canescens can hyperaccumulate cadmium, but microstructural, transcriptomic, and physiological response mechanisms are poorly understood. Histochemical assays, transmission electron microscopic observations, energy-dispersive x-ray microanalysis, and transcriptomic and physiological analyses have been performed to enhance our understanding of cadmium accumulation and detoxification in P. × canescens. Cadmium was allocated to the phloem of the bark, and subcellular cadmium compartmentalization occurred mainly in vacuoles of phloem cells. Transcripts involved in microstru

The role of calcium and calmodulin in freezing-induced freezing resistance of Populus tomentosa cuttings.

To explore the role of calcium-calmodulin messenger system in the transduction of low temperature signal in woody plants, Populus tomentosa cuttings after being treated with CaCl(2) (10 mmol/L), Ca(2+) chelator EGTA (3 mmol/L), Ca(2+) channel inhibitor LaCl(3) (100 mmol/L) or CaM antagonist CPZ (50 mmol/L) were used for freezing acclimation at -3 degrees C. The changes in the calmodulin (CaM) and malonaldehyde (MDA) contents, the activities of superoxide dismutase (SOD), peroxidase (POD) and Ca(2+)-dependent adenosinetriphosphatase (Ca(2+)-ATPase) of mitochondrial membrane as well as freezing

Physiological and Transcriptome Analysis on Diploid and Polyploid Populus ussuriensis Kom. under Salt Stress.

Populus ussuriensis Kom. is a valuable forest regeneration tree species in the eastern mountainous region of Northeast China. It is known that diploid P. ussuriensis (CK) performed barely satisfactorily under salt stress, but the salt stress tolerance of polyploid (i.e., triploid (T12) and tetraploid (F20)) P. ussuriensis is still unknown. In order to compare the salt stress tolerance and salt stress response mechanism between diploid and polyploid P. ussuriensis, phenotypic observation, biological and biochemistry index detections, and transcriptome sequencing (RNA-seq) were performed on CK,

The PtoERF1-PtomiR393a-PtoFBL4 module confers drought tolerance via ABA-auxin crosstalk in Populus.

Drought stress limits forest tree growth and adaptation, with xylem vessels critical for hydraulic transport and structural integrity. However, the molecular mechanisms of abscisic acid (ABA)-auxin interaction in regulating vessel morphogenesis under water-deficit conditions remain unclear. Here, we identified PtomiR393a, a drought-responsive microRNA in Populus tomentosa that mediates crosstalk between ABA and auxin signaling pathways under drought stress. Suppressing PtomiR393 enhanced drought tolerance and growth, whereas its overexpression had the opposite effect. Under drought conditions,

The transcription factor PtoMYB142 enhances drought tolerance in Populus tomentosa by regulating gibberellin catabolism.

Drought stress caused by global warming has resulted in significant tree mortality, driving the evolution of water conservation strategies in trees. Although phytohormones have been implicated in morphological adaptations to water deficits, the molecular mechanisms underlying these processes in woody plants remain unclear. Here, we report that overexpression of PtoMYB142 in Populus tomentosa results in a dwarfism phenotype with reduced leaf cell size, vessel lumen area, and vessel density in the stem xylem, leading to significantly enhanced drought resistance. We found that PtoMYB142 modulates

Identification and functional characteristics of key regulatory genes in Populus alba in response to cold stress.

Forest crops are particularly susceptible to cold stress after dormancy release. Newly emerged shoots are especially vulnerable, and shoot mortality can cause serious loss of biomass production. Revealing the molecular mechanisms of cold stress responses in forest trees is therefore crucial. Populus alba is a frost-resistant tree species distributed worldwide. In this study, we characterized the transcriptome of P. alba in response to cold stress and observed that a large number of genes were up-regulated in both root and leaf tissues. Through Mfuzz clustering and phylogenetic analysis, four e

PtCP1 Is an Extraplastidial Cysteine Protease Involved in Leaf Protein Degradation of Populus tomentosa Carr.

Protein turnover is essential for cellular metabolism, organelle biogenesis, stress adaptation, and ultimately the viability of cells and tissues. Papain-like cysteine proteases (PLCPs) are one of the vital components in protein degradation. PLCPs have been reported to act in senescence-associated proteolysis, but their roles in vegetative growth remain unclear. We identified PtCP1, an AALP-like PLCP in Populus tomentosa, localized to the vacuole and acid-triggered activated. CRISPR/Cas9-generated loss-of-function mutant (d7) showed dwarfism and non-stomatal photosynthetic limitations. On the

Genetic composition and heterozygosity of gibberellin-induced 2n gametes evaluated by SSR markers in Populus.

BACKGROUND: The artificial induction of unreduced (2n) gametes, due to first division restitution (FDR) or second division restitution (SDR), is an important method to produce triploids in higher plants. It is crucial to evaluate the genetic composition and heterozygosity transmission of induced 2n gametes with different formation mechanisms. RESULTS: Here, we produced 110 triploids by pollinating the female inflorescence of Populus alba × P. glandulosa with gibberellin-induced 2n pollen of Populus bolleana. The genetic composition and heterozygosity of the induced 2n pollen were inferred fro

Salicylic acid in Populus tomentosa is a remote signalling molecule induced by Botryosphaeria dothidea infection.

The salicylic acid (SA) plays a critical role during the establishment of systemic acquired resistance (SAR) in uninfected plant tissues after localised exposure to a pathogen. Here, we studied SA in Populus tomentosa infected by the plant pathogen Botryosphaeria dothidea. The accumulation of SA and methyl salicylate (MeSA) occurred in chronological order in P. tomentosa. The SA and MeSA contents were greater at infected than uninfected sites. Additionally, a gene expression analysis indicated that SA might be accumulated by phenylalanine ammonialyase (PAL) and converted to MeSA by SA carboxyl

Stomatal Conductance and Sulfur Uptake of Five Clones of Populus tremuloides Exposed to Sulfur Dioxide.

Plants of five clones of Populus tremuloides Michx. were exposed to 0, 0.2 or 0.5 microliter per liter SO(2) for 8 hours in controlled environment chambers. In the absence of the pollutant, two pollution-resistant clones maintained consistently lower daytime diffusive conductance (LDC) than did a highly susceptible clone or two moderately resistant clones. Differences in LDC among the latter three clones were not significant. At 0.2 microliter per liter SO(2), LDC decreased in the susceptible clone after 8 hours fumigation while the LDC of the other clones was not affected. Fumigation with 0.5

Functional characterization and expression patterns of PnATX genes under different abiotic stress treatments in Populus.

The copper chaperone ATX1 has been investigated previously in the herbaceous plants Arabidopsis and rice. However, the molecular mechanisms of ATX1 underlying copper transport and functional characteristics in the woody plant Populus are poorly understood. In this study, PnATX1 and PnATX2 of Populus simonii × P. nigra were identified and characterized. Sequence analysis showed that PnATXs contained the metal-binding motif MXCXXC in the N-terminus and a lysine-rich region. Phylogenetic analysis of ATX protein sequences revealed that PnATXs were clustered in the same group as AtATX1. PnATX prote

Net cadmium flux and accumulation reveal tissue-specific oxidative stress and detoxification in Populus × canescens.

To characterize the dynamics of Cd²⁺ flux in the rhizosphere and to study cadmium (Cd) plant-internal partitioning in roots, wood, bark and leaves in relation to energy metabolism, reactive oxygen species (ROS) formation and antioxidants, Populus × canescens plantlets were exposed to either 0 or 50 µM CdSO₄ for up to 20 days in the nutrient solution. A strong net Cd²⁺ influx in root apex was observed after Cd exposure for 24 h, even if net Cd²⁺ influx decreased gradually in roots. A large amount of Cd was accumulated in roots. Cd ions were uploaded via the xylem to leaves and further transport

Carbon allocation and partitioning in Populus tremuloides are modulated by ectomycorrhizal fungi under phosphorus limitation.

The fate of carbon (C) captured by forest trees during photosynthesis is influenced by the supply of other resources. Fixed C may be partitioned among biomolecules within the leaf and/or allocated throughout the tree to growth, storage and maintenance activities. Phosphorus (P) availability often limits tree productivity due to its high biological demand and strong interactions with soil minerals. As ectomycorrhizal (ECM) fungi play critical roles in enhancing phosphate (Pi) acquisition by their hosts, these symbioses will influence the fate of C within trees and forested ecosystems. Using Pop

A Variable Polyglutamine Repeat Affects Subcellular Localization and Regulatory Activity of a Populus ANGUSTIFOLIA Protein.

Polyglutamine (polyQ) stretches have been reported to occur in proteins across many organisms including animals, fungi and plants. Expansion of these repeats has attracted much attention due their associations with numerous human diseases including Huntington's and other neurological maladies. This suggests that the relative length of polyQ stretches is an important modulator of their function. Here, we report the identification of a Populus C-terminus binding protein (CtBP) ANGUSTIFOLIA (PtAN1) which contains a polyQ stretch whose functional relevance had not been established. Analysis of 917
