Time- and temperature-dependent postmortem ∆9-tetrahydrocannabinol concentration changes in rabbits following controlled inhaled cannabis administration.

ostmortem redistribution (PMR), a well-known phenomenon in forensic toxicology, can result in substantial changes in drug concentrations after death, depending on the chemical characteristics of the drug, blood collection site, storage conditions of the body and postmortem interval (PMI). Limited PMR data are available for ∆9-tetrahydrocannabinol (THC), the primary psychoactive component in Cannabis sativa. PMR was evaluated after controlled cannabis inhalation via a smoking machine and exposure chamber in New Zealand white rabbits. Necropsies were performed on five control rabbits immediately

Postmortem Temporal Changes in Liver and Spleen Stiffness: Evaluation with Shear Wave Elastography in a Rat Model.

Background/Objectives: Postmortem changes in tissue stiffness and organ morphology are critical for forensic medicine and pathology. Shear wave elastography (SWE) has emerged as a non-invasive tool to assess tissue stiffness, yet its potential for postmortem interval estimation remains underexplored. While previous studies have demonstrated early postmortem alterations in tissue elasticity, the temporal progression of these changes in different organs is not fully understood. This study aims to investigate the temporal changes in liver and spleen stiffness during the postmortem period using SW

Onset and progression of postmortem histological changes in the central nervous system of RccHan™: WIST rats.

Death initiates a cascade of physiological and biochemical alterations in organs and tissues, resulting in microscopic changes that challenge the histopathological evaluation. Moreover, the brain is particularly susceptible to artifacts owing to its unique composition and its location within the cranial vault. The aim of this study was to compile and illustrate the microscopic changes in the central nervous system (CNS) of rats subjected to delayed postmortem fixation. It also scrutinizes the influence of exsanguination and cooling methods on the initiation and progression of these alterations

Influence of blood thiosulfate produced by postmortem changes for the diagnosis of hydrogen sulfide poisoning in forensic autopsy.

BACKGROUND: Thiosulfate concentration in blood is an important indicator for the diagnosis of hydrogen sulfide poisoning. It may also be detected at high levels in postmortem decomposition cases. OBJECTIVES: To determine the effect of postmortem decomposition on blood thiosulfate concentration and define precautions for diagnosing hydrogen sulfide poisoning based on thiosulfate concentration. METHODS: A total of 57 cadavers (37 males and 20 females) of non-hydrogen sulfide poisoning-related deaths that underwent forensic autopsy in our department between 2016 and 2019 were classified into th

Changes in blood vessels in fetuses 4 to 9 months intrauterine life old by postmortem angiography method.

We monitored changes in caliber, position and branching of blood vessels in fetuses of 4 - 9 months of intrauterine life. By precise dissection we prepared starting parts of common carotid artery and internal jugular vein in 40 cadaver fetuses. The vessels were injected with Telebrix and subjected to postmortem angiography at the Institute of Radiology Clinics Center in Sarajevo. Thereafter, arteries obtained were compared and analyzed. In preparations of few months old fetal material we observed arteries of fairly straight course, low caliber and with no observable ramification. When preparat

Systematic changes in gene expression in postmortem human brains associated with tissue pH and terminal medical conditions.

Studies of gene expression abnormalities in psychiatric or neurological disorders often involve the use of postmortem brain tissue. Compared with single-cell organisms or clonal cell lines, the biological environment and medical history of human subjects cannot be controlled, and are often difficult to document fully. The chance of finding significant and replicable changes depends on the nature and magnitude of the observed variations among the studied subjects. During an analysis of gene expression changes in mood disorders, we observed a remarkable degree of natural variation among 120 samp

Forensic implication of Zinc sulfide and Zinc sulfide Nanoparticles toxicity on muscle histopathology and postmortem changes in rats.

The current study aimed to investigate the molecular toxic effects of zinc sulfide (ZnS) and zinc sulfide nanoparticles (ZnS-NPs) on rat skeletal muscle tissue. It also explored the forensic implications of ZnS toxicity on postmortem carrion infestation, particularly its potential impact on postmortem interval (PMI) estimation. ZnS nanoparticles were synthesized and characterized using X-ray diffraction (XRD) and scanning electron microscopy (SEM). Thirty Wistar rats were divided into three groups: control, ZnS-treated, and ZnS-NP–treated (both administered at the LD50 dose of 2000 mg/kg). Mus

Forensic implications of early postmortem changes in skeletal, cardiac, and smooth muscles following acute colchicine intoxication in rats.

Acute colchicine (CHC) poisoning can be fatal due to its narrow therapeutic index, involvement of multiple organ systems, and the lack of a specific antidote. This study investigated early postmortem biochemical, histopathological, and immunohistochemical changes in skeletal, smooth, and cardiac muscles within 24 h of death following acute CHC intoxication in rats. Sixty male Sprague Dawley rats were divided into control and CHC-intoxicated groups (n = 30 each). Controls received saline and were euthanized humanely; CHC rats received a single oral lethal dose (4.9 mg/kg b.wt) and died naturall

Understanding Proton Magnetic Resonance Spectroscopy Neurochemical Changes Using Alzheimer's Disease Biofluid, PET, Postmortem Pathology Biomarkers, and APOE Genotype.

In vivo proton (1H) magnetic resonance spectroscopy (MRS) is a powerful non-invasive method that can measure Alzheimer's disease (AD)-related neuropathological alterations at the molecular level. AD biomarkers include amyloid-beta (Aβ) plaques and hyperphosphorylated tau neurofibrillary tangles. These biomarkers can be detected via postmortem analysis but also in living individuals through positron emission tomography (PET) or biofluid biomarkers of Aβ and tau. This review offers an overview of biochemical abnormalities detected by 1H MRS within the biologically defined AD spectrum. It include

Examination of postmortem changes in the lungs, trachea, and bronchi in a rat model imaged with small-animal computed tomography.

OBJECTIVES: As less autopsies are performed, the need for postmortem computed tomography (PMCT) as an alternative is increasing. It is important to know how postmortem changes over time are reflected on CT, in order to improve the diagnostic capability of PMCT and replace forensic pathology evaluations such as time of death estimation. METHODS: In this study, we examined temporal changes on postmortem chest CT images of a rat model. After acquiring antemortem images under isoflurane inhalation anesthesia, the rats were euthanized with a rapid intravenous injection of anesthetics. From immedia

Effects of apoptosis on quality changes of Larimichthys crocea postmortem storage with emphasis on cytochrome c-mediated lipid deterioration.

Apoptosis in fish muscle postmortem has been confirmed. Based on the apoptosis is associated with quality deterioration in fish during storage. This study aims to investigate the impact of apoptosis on the quality of Larimichthys crocea at 4 °C storage, particularly the lipid deterioration mediated by cytochrome c (cyt-c). The results indicated that the apoptosis inhibitor cyclosporin A (CsA) mitigated the decline in glycogen and ATP levels during 6-12 h, reduced the extent of mitochondrial damage during 12-72 h, especially inhibiting mitochondrial permeability transition pore (MPTP) opening,

iPSC motor neurons, but not other derived cell types, capture gene expression changes in postmortem sporadic ALS motor neurons.

Motor neuron degeneration, the defining feature of amyotrophic lateral sclerosis (ALS), is a primary example of cell-type specificity in neurodegenerative diseases. Using isogenic pairs of induced pluripotent stem cells (iPSCs) harboring different familial ALS mutations, we assess the capacity of iPSC-derived lower motor neurons, sensory neurons, astrocytes, and superficial cortical neurons to capture disease features including transcriptional and splicing dysregulation observed in human postmortem neurons. At early time points, differentially regulated genes in iPSC-derived lower motor neuron

Changes in magnetic resonance imaging relaxation time on postmortem magnetic resonance imaging of formalin-fixed human normal heart tissue.

BACKGROUND: Postmortem magnetic resonance imaging (MRI) has been used to investigate the cause of death, but due to time constraints, it is not widely applied to the heart. Therefore, MRI analysis of the heart after formalin fixation was previously performed. However, the changes in MRI signal values based on the fixation time of formalin were not investigated. The objective was to investigate changes over time in the T1- and T2-values of MRI signals in normal areas of hearts removed during autopsy, hearts subsequently fixed in formalin, and heart specimens sliced for the preparation of pathol

Determining time of death: temperature-dependent postmortem changes in calcineurin A, MARCKS, CaMKII, and protein phosphatase 2A in mouse.

While the determination of postmortem interval (PMI) is a crucial and fundamental step in any death investigation, the development of appropriate biochemical methods for PMI estimation is still in its infancy. This study focused on the temperature-dependent postmortem degradation of calcineurin A (CnA), calmodulin-dependent kinase II (CaMKII), myristoylated alanine-rich C-kinase substrate (MARCKs), and protein phosphatase 2A (PP2A) in mice. The results show that MARCKS, CaMKII, and the use of lung tissue do not appear to warrant further study for the determination of PMI in humans. In skeletal

Pathology-associated change in levels and localization of SIDT2 in postmortem brains of Parkinson's disease and dementia with Lewy bodies patients.

Parkinson's disease (PD) and dementia with Lewy bodies (DLB) are major neurodegenerative disorders that share commonalities in their pathology involving the formation of Lewy bodies, the main component of which is α-synuclein protein. Aberrancy and dysfunction in lysosomes have been suggested to play critical roles in the pathogenesis of Lewy body diseases. We recently identified a novel lysosomal degradation pathway in which various macromolecules, including α-synuclein protein, are directly imported into lysosomes and degraded. In this study, we analyzed the levels and localization of the ly

Contrasting changes in DRD1 and DRD2 splice variant expression in schizophrenia and affective disorders, and associations with SNPs in postmortem brain.

Dopamine 2 receptor (DRD2) is of major interest to the pathophysiology of schizophrenia (SCZ) both as a target for antipsychotic drug action as well as a SCZ-associated risk gene. The dopamine 1 receptor (DRD1) is thought to mediate some of the cognitive deficits in SCZ, including impairment of working memory that relies on normal dorsolateral prefrontal cortex (DLPFC) function. To better understand the association of dopamine receptors with SCZ, we studied the expression of three DRD2 splice variants and the DRD1 transcript in DLPFC, hippocampus and caudate nucleus in a large cohort of subjec
