The cytopathology and length of latency in single-step growth curves of two isolates of stomatitis papulosa virus are compared in this report. Isolate 721 was obtained from a calf with oral ulcers and isolate 8665 was obtained from a calf with respiratory disease and oral ulcers. In single-step growth curves, the latency period of isolate 721 was 8 h while that of isolate 8665 was 6 h. The cytopathic effect produced by isolate 721 in bovine lung cells was characterized by enlargement of the cell, cell-to-cell adherence and large intracytoplasmic accumulations of viral inclusion material. Monkeypox virus is a member of the Orthopoxvirus genus and the Poxviridae family. Orthopoxviruses are among the most intricate animal viruses. The pathogenicity of human monkeypox infection has been emphasized in response to its recent emergence in non-endemic countries and the threat of bioterrorism. It is always necessary to take appropriate precautions in exposure to emerging or re-emerging infections. Here, we focus on the current state of the human monkeypox infection outbreak, research & development of immune responses, and clinical interventions to prevent and treat the human monkeypox

Non-native rodents serve as bridges between anthropogenic and natural landscapes. They have expanded across the planet alongside humans while bringing competition, predation, and pathogens, such as poxviruses, to naïve ecosystems. Although rodents serve as reservoirs for multiple zoonotic poxviruses, limited research has focused on rodents for identification of unknown poxviruses. Here, we characterized a divergent metagenome-assembled poxvirus, madamurpox virus, from the oral swab of a black rat in southeastern Madagascar. While madamurpox virus shares a phylogenetic relationship with human-i

The eradication of smallpox is one of the greatest medical successes in history. Vaccinia virus was made famous by being the virus used in the live vaccine that enabled this feat. Nearly 40 years on from that success, this prototypical poxvirus continues to empower the exploration of fundamental biology and the potential to develop therapeutics against some of the major causes of death and disease in the modern world.

Viruses that successfully replicate within the host have devised strategies to subvert or evade the challenges posed by the innate and adaptive immune responses. Many investigators are now beginning to dissect the diverse and complex interactions involving chemokines, chemokine receptors and viral infection. In recent years, much attention has been focused on the role of chemokines in antiviral defense.

Light grey macules developed on the skin and in the mouths of juvenile caimans, (Caiman crocodilus fuscus) (n = 8), kept in the quarantine section of the reptile park at the National Zoological Gardens, Pretoria, Republic of South Africa. The gross, histopathological and ultrastructural features of the lesions were commensurate with pox virus infection. This outbreak closely resembled the disease described elsewhere in 3 juvenile captive caimans.

Monkeypox virus (MPVX) is a double-stranded deoxyribonucleic acid (DNA) virus from the genus Orthopoxvirus, family Poxviridae. Many cases of MPVX infection are currently seen worldwide. Although the majority of cases reported are not severe, basic precautionary measures are key to preventing this infection from turning into a catastrophic pandemic. We report a case of a 54-year-old male with no high-risk behavior who developed a sudden uncharacteristic monkeypox virus rash while on a flight from Nigeria.

Cases of pox-like lesions in horses and donkeys have been associated with poxviruses belonging to different genera of the family Poxviridae. These include the orthopoxviruses vaccinia virus (VACV), horsepoxvirus (HPXV) and cowpoxvirus (CPXV), as well as a potentially novel parapoxvirus and molluscum contagiosum virus (MOCV). However, with the exception of VACV, HPXV and CPXV, the genomic characterization of the causative agents remains largely elusive with only single short genome fragments available. Here we present the first full-length genome sequence of an equine molluscum contagiosum-like

Monkeypox virus (MPXV) belonging to poxviridae family causes chronic viral disease in various mammals including human and monkeys. Conventional vaccines developed against smallpox of poxviridae, are not specific against Mpox. Also, they can cause various side effects after vaccination. In this study, we aimed to analyze the A17L, A28L, A37R, A43R, E8L, H3L, B6R, and M1R structural proteins of MPXV and identify epitopes in them which can be used to generate vaccine antigens. Among the proteins analyzed, the M1R protein was predicted to be more appropriate for use in vaccine research due to its

Poxviruses are highly successful pathogens, known to infect a variety of hosts. The family Poxviridae includes Variola virus, the causative agent of smallpox, which has been eradicated as a public health threat but could potentially reemerge as a bioterrorist threat. The risk scenario includes other animal poxviruses and genetically engineered manipulations of poxviruses. Studies of orthologous gene sets have established the evolutionary relationships of members within the Poxviridae family. It is not clear, however, how variations between family members arose in the past, an important issue i

Twenty strains of Orthopox viruses (vaccina (3), cowpox (2), monkeypox (2), variola major and minor (7) and whitepox (6)) have been tested by electrofocusing in a granulated gel. From our preliminary results, it appears that there is a clearcut distinction between smallpox and whitepox. If our results are confirmed, it will be the first real genetic marker identified because other in vitro or in vivo test have heretofore failed to differentiate between smallpox and whitepox. Further work is now in progress, to evaluate of the test and it is hoped to find and explanation for the observations no

Mpox is caused by the mpox virus, which belongs to the Orthopoxvirus genus and Poxviridae family. Animal hosts, such as African rodents, mice, prairie dogs, and non-human primates, play important roles in the development and transmission of outbreaks. Laboratory animal infection experiments have demonstrated that some animals are susceptible to mpox virus. This review summarizes the current progress on the animal hosts for mpox virus. The surveillance of mpox virus in animal hosts will provide important insights into virus tracing, analysis of mutation evolutionary patterns, transmission mecha

Canarypox viruses (CNPV) are excellent candidates to develop recombinant vector vaccines due to both their capability to induce protective immune responses and their incompetence to replicate in mammalian cells (safety profile). In addition, CNPV and the derived recombinants can be manipulated under biosafety level 1 conditions. There is no commercially available system to obtain recombinant CNPV; however, the methodology and tools required to develop recombinant vaccinia virus (VV), prototype of the Poxviridae family, can be easily adapted. This chapter provides protocols for the generation,

The upsurge of mpox (formerly known as monkeypox) in Africa and its global spread highlight the need for improved vaccines. The development of new recombinant vaccines, including mRNA and protein nanoparticles, depends on understanding the biology of poxviruses and selecting the most protective immunogens. Animal studies demonstrate that vaccines need to target the antigens of both infectious forms - the mature virion and the enveloped virion - which display surface proteins responsible for cell entry and cell-to-cell spread, respectively. Although some of these proteins have been shown to ind

Tecovirimat (ST-246 or TPOXX) is an antiviral agent developed as part of a U.S. biodefense initiative aimed at addressing Orthopoxvirus infections, including smallpox and mpox. Although smallpox was declared eradicated in 1980, the potential for its reemergence as a biothreat persists due to illegal stockpiling and the possibility of laboratory synthesis. The F13 protein, which plays a critical role in the formation of extracellular viral particles, serves as the primary target for tecovirimat, inhibiting the transition from intracellular mature viruses (IMVs) to intracellular enveloped viruse

Illnesses caused by poxviruses have plagued humanity for millennia. The most notable are caused by viruses comprising the orthopoxvirus genus. Smallpox (caused by Variola virus) became the first eradicated disease in 1980. Devotion to smallpox eradication required significant orthopoxvirus research and discovery including vaccine and medical countermeasure development. Here we describe historical and current poxvirus epidemiology, with particular emphasis on orthopoxviruses including Monkeypox virus and Vaccinia virus.

In an outbreak of wart-like skin lesions in nine-month-old farmed Nile crocodiles (Crocodylus niloticus) the lesions were especially common on the head and neck but occurred on all parts of the body except the tail. Eosinophilic intracytoplasmic inclusion bodies were present in epithelial cells and further examination of the lesions by transmission electron microscopy revealed the presence of a poxvirus. An autogenous vaccine was produced from material from the lesions and in six vaccinated crocodiles the lesions healed more quickly than in unvaccinated controls.

Poxviridae is a diverse family of double-stranded DNA viruses, historically significant for diseases like smallpox caused by variola virus (VARV). These viruses exhibit unique cytoplasmic replication strategies, large genomes encoding numerous proteins, and the ability to cause severe cutaneous and systemic diseases. Recent attention has focused on their neurotropic potential, including mechanisms of CNS invasion, immune-mediated damage, and clinical manifestations such as encephalitis and myelitis. This review synthesises current knowledge on poxvirus neurotropism, highlighting pathophysiolog

Vaccinia virus strains from the family Poxviridae have been frequently isolated in Brazil and associated with outbreaks of exanthematic disease affecting cows and humans. An ELISA IgG was applied to evaluate the seroprevalence of orthopoxviruses in a community located in a rural settlement in the Amazon region, where no orthopoxvirus outbreaks have yet been reported. An overall seroprevalence of 27.89% was found, and it was 23.38% in the non-vaccinated population (smallpox vaccination). These results strongly suggest that orthopoxviruses circulate in this population, and it is the first findin

Although WHO-led global efforts led to eradication of smallpox over four decades ago, other poxviruses, especially monkeypox, have re-emerged to occupy the ecological niche vacated by smallpox. Many of these viruses produce similar lesions thus mandating a prompt laboratory confirmation. There has been considerable evolution in the techniques available to diagnose these infections and differentiate between them. With the 2022 multi-country outbreak of monkeypox, significant efforts were made to apprise the laboratory diagnosis of the virus and numerous real-time-PCR-based assays were made comm
