Illuminating posterior targets for TMS beyond the prefrontal cortex.

Transcranial Magnetic Stimulation (TMS) is rapidly developing as a therapeutic tool for multiple psychiatric disorders. All current FDA-cleared TMS indications for psychiatric illness target the frontal lobes. Historically, TMS target selection was based on reports of brain lesion locations that cause illness or functional neuroimaging correlates within the prefrontal cortex. In addition to prefrontal targets, however, these neuroimaging studies have also consistently implicated posterior brain regions in a variety of psychiatric disorders. To date, these posterior regions have rarely been tes

Medial prefrontal cortex circuitry and social behaviour in autism.

Autism spectrum disorder (ASD) has proven to be highly enigmatic due to the diversity of its underlying genetic causes and the huge variability in symptom presentation. Uncovering common phenotypes across people with ASD and pre-clinical models allows us to better understand the influence on brain function of the many different genetic and cellular processes thought to contribute to ASD aetiology. One such feature of ASD is the convergent evidence implicating abnormal functioning of the medial prefrontal cortex (mPFC) across studies. The mPFC is a key part of the 'social brain' and may contrib

Rodent mnemonic similarity task performance requires the prefrontal cortex.

Mnemonic similarity task performance, in which a known target stimulus must be distinguished from similar lures, is supported by the hippocampus and perirhinal cortex. Impairments on this task are known to manifest with advancing age. Interestingly, disrupting hippocampal activity leads to mnemonic discrimination impairments when lures are novel, but not when they are familiar. This observation suggests that other brain structures support discrimination abilities as stimuli are learned. The prefrontal cortex (PFC) is critical for retrieval of remote events and executive functions, such as work

Identification of prefrontal cortex protein alterations in Alzheimer's disease.

BACKGROUND: The tuberculosis (TB) case detection rate has stagnated at 60% due to disorganized case finding and insensitivity of sputum smear microscopy. Of the identified TB cases, 4% die while being treated, monitored with tools that insufficiently predict failure/mortality. OBJECTIVE: To explore the TBscore, a recently proposed clinical severity measure for pulmonary TB (PTB) patients, and to refine, validate, and investigate its place in case finding. DESIGN: The TBscore's inter-observer agreement was assessed and compared to the Karnofsky Performance Score (KPS) (paper I). The TBscore's

Cholinergic Overstimulation Attenuates Rule Selectivity in Macaque Prefrontal Cortex.

Acetylcholine is released in the prefrontal cortex (PFC) and is a key modulator of cognitive performance in primates. Cholinergic stimulation has been shown to have beneficial effects on performance of cognitive tasks, and cholinergic receptors are being actively explored as promising targets for ameliorating cognitive deficits in Alzheimer's disease. We hypothesized that cholinergic stimulation of PFC during performance of a cognitive task would augment neuronal activity and neuronal coding of task attributes. We iontophoretically applied the general cholinergic receptor agonist carbachol ont

Nature experience reduces rumination and subgenual prefrontal cortex activation.

Urbanization has many benefits, but it also is associated with increased levels of mental illness, including depression. It has been suggested that decreased nature experience may help to explain the link between urbanization and mental illness. This suggestion is supported by a growing body of correlational and experimental evidence, which raises a further question: what mechanism(s) link decreased nature experience to the development of mental illness? One such mechanism might be the impact of nature exposure on rumination, a maladaptive pattern of self-referential thought that is associated

Prefrontal cortex white matter tracts in prodromal Huntington disease.

Huntington disease (HD) is most widely known for its selective degeneration of striatal neurons but there is also growing evidence for white matter (WM) deterioration. The primary objective of this research was to conduct a large-scale analysis using multisite diffusion-weighted imaging (DWI) tractography data to quantify diffusivity properties along major prefrontal cortex WM tracts in prodromal HD. Fifteen international sites participating in the PREDICT-HD study collected imaging and neuropsychological data on gene-positive HD participants without a clinical diagnosis (i.e., prodromal) and

Increased functional selectivity over development in rostrolateral prefrontal cortex.

Relational reasoning, or the ability to identify and consider relationships between multiple mental representations, is a fundamental component of high-level cognition (Robin and Holyoak, 1995). The capacity to reason with relations enables abstract thought and may be at the core of what makes human cognition unique (Penn et al., 2008). This capacity improves throughout childhood and adolescence (Ferrer et al., 2009). Here, we sought to better understand the neural mechanisms that support its emergence. We have hypothesized previously, based on fMRI research in adults, that (1) inferior pariet

Selective control of prefrontal neural timescales by parietal cortex.

The frontal eye field (FEF) and posterior parietal cortex (PPC) are key hubs of the dorsal attention network, yet how parietal inputs shape prefrontal circuit temporal dynamics and attentional computations remains largely unknown. We measured intrinsic timescales of FEF neurons in male rhesus macaques and examined their changes during PPC inactivation. We observed two distinct classes of FEF neurons based on their intrinsic timescales: short-timescale neurons (~25 ms) and long-timescale neurons (~100 ms). Short-timescale neurons showed stronger transient visual responses, suggesting a role in

Prefrontal and somatosensory-motor cortex effective connectivity in humans.

Effective connectivity, functional connectivity, and tractography were measured between 57 cortical frontal and somatosensory regions and the 360 cortical regions in the Human Connectome Project (HCP) multimodal parcellation atlas for 171 HCP participants. A ventral somatosensory stream connects from 3b and 3a via 1 and 2 and then via opercular and frontal opercular regions to the insula, which then connects to inferior parietal PF regions. This stream is implicated in "what"-related somatosensory processing of objects and of the body and in combining with visual inputs in PF. A dorsal "action

Prefrontal cortex exhibits multidimensional dynamic encoding during decision-making.

Recent work has suggested that the prefrontal cortex (PFC) plays a key role in context-dependent perceptual decision-making. In this study, we addressed that role using a new method for identifying task-relevant dimensions of neural population activity. Specifically, we show that the PFC has a multidimensional code for context, decisions and both relevant and irrelevant sensory information. Moreover, these representations evolve in time, with an early linear accumulation phase followed by a phase with rotational dynamics. We identify the dimensions of neural activity associated with these phas

Got chocolate? Bilateral prefrontal cortex stimulation augments chocolate consumption.

BACKGROUND: Understanding the mechanisms behind exerting self-control may reveal why health behaviors are resistant to change. Activity in the right inferior frontal gyrus (rIFG) plays a role in self-control processes and may be modulated using transcranial direct current stimulation (tDCS). OBJECTIVE: In this early phase behavioral research study, we investigated whether anodal stimulation over the rIFG with cathodal stimulation over the left IFG (versus sham) reduced chocolate consumption. METHODS: Twenty-three healthy females (ages 18-35) completed two tDCS sessions (2.0 mA vs. sham; orde

Fifty Years of Prefrontal Cortex Research: Impact on Assessment.

Our knowledge of the functions of the prefrontal cortex, often called executive, supervisory, or control, has been transformed over the past 50 years. After operationally defining terms for clarification, we review the impact of advances in functional, structural, and theoretical levels of understanding upon neuropsychological assessment practice as a means of identifying 11 principles/challenges relating to assessment of executive function. Three of these were already known 50 years ago, and 8 have been confirmed or emerged since. Key themes over this period have been the emergence of the use

Prefrontal cortex controls human balance during overground ataxic gait.

PURPOSE: 1) to verify if prefrontal cortex (PFC) is activated during over ground walking in ataxic patients, 2) to correlate the clinical parameters of gait with the PFC activation patterns. METHODS: Fourteen patients and 20 healthy subjects were studied. Ataxia was assessed by the Scale for the Assessment and Rating of Ataxia (SARA). A 2-channel near-infrared system was used to investigate the changes in oxygenated ([O2Hb]t) and deoxygenated ([HHb]t) hemoglobin concentrations on the PFC during gait. [O2Hb] baseline-corrected activation values ([O2Hb]c) were calculated by the difference betwe

Prefrontal cortex activity during the discrimination of relative distance.

To compare with our previous findings on relative-duration discrimination, we studied prefrontal cortex activity as monkeys performed a relative-distance discrimination task. We wanted to know whether the same parts of the prefrontal cortex compare durations and distances and, if so, whether they use similar mechanisms. Two stimuli appeared sequentially on a video screen, one above a fixed reference point, the other below it by a different distance. After a delay period, the same two stimuli reappeared (as choice stimuli), and the monkeys' task was to choose the one that had appeared farther f

Dominance attributions following damage to the ventromedial prefrontal cortex.

Damage to the human ventromedial prefrontal cortex (VM) can result in dramatic and maladaptive changes in social behavior despite preservation of most other cognitive abilities. One important aspect of social cognition is the ability to detect social dominance, a process of attributing from particular social signals another person's relative standing in the social world. To test the role of the VM in making attributions of social dominance, we designed two experiments: one requiring dominance judgments from static pictures of faces, the second requiring dominance judgments from film clips. We

Some hypotheses concerning the functional organization of prefrontal cortex.

The paper deals with systematization of the behavioral disorders after partial prefrontal lesions. It is assumed that depending on the localization of lesions two disorders may be discerned: impairment of drive inhibition (meaning by this only preservative but not protective drives) land of motor-act inhibition. The first deficit leads to the disinhibition of no-go responses in Pavlovian differentiation, the second one to the disorder of R1-R2 differentiation due to the difficulty of switching from one response to the other. Different parts of the prefrontal cortex are engaged in directional r

Proneness to hypomania predicts EEG coherence between left motor cortex and left prefrontal cortex.

Previous research has demonstrated that hypomania is associated with approach motivation and activity in the left prefrontal cortex (PFC). Other research has linked left motor cortex excitability to approach motivation, suggesting the existence of connections between the motor cortex and PFC. The present research extends this work using unilateral hand contractions to manipulate contralateral cortical activity, and examining the relationship between motor cortex and PFC inter-electrode EEG coherence and hypomania. Within the right-hand contraction condition, hypomania related to greater connec

Stereo-seq of the prefrontal cortex in aging and Alzheimer's disease.

Aging increases the risk for Alzheimer's disease (AD), driving pathological changes like amyloid-β (Aβ) buildup, inflammation, and oxidative stress, especially in the prefrontal cortex (PFC). We present the first subcellular-resolution spatial transcriptome atlas of the human prefrontal cortex (PFC), generated with Stereo-seq from six male AD cases at varying neuropathological stages and six age-matched male controls. Our analyses revealed distinct transcriptional alterations across PFC layers, highlighted disruptions in laminar structure, and exposed AD-related shifts in layer-to-layer and ce

GABA system in the prefrontal cortex involved in psychostimulant addiction.

Drug addiction is a chronic and relapse brain disorder. Psychostimulants such as cocaine and amphetamine are highly addictive drugs. Abuse drugs target various brain areas in the nervous system. Recent studies have shown that the prefrontal cortex (PFC) plays a key role in regulating addictive behaviors. The PFC is made up of excitatory glutamatergic cells and gamma-aminobutyric acid (GABAergic) interneurons. Recently, studies showed that GABA level was related with psychostimulant addiction. In this review, we will introduce the role and mechanism of GABA and γ-aminobutyric acid receptors (GA
