It is well known that electrolytic lesions in the medial preoptic area (MPOA) and the lateral hypothalamic area (LHA) seriously impair masculine sexual behavior in the rat. We here report that bilateral infusions of the neurotoxin, ibotenic acid (IBO), in the MPOA were as effective as electrolytic lesions in eliminating copulation whereas no behavioral effects were detected following similar infusions in the LHA. Histological examination of MPOA and LHA following IBO exposure revealed extensive degeneration of neuronal cell bodies with little evidence of non-specific damage. Also, immunohistoc

Precopulatory behaviors that are preferentially directed towards opposite-sex conspecifics are critical for successful reproduction, particularly in species wherein the sexes live in isolation, such as Syrian hamsters (Mesocricetus auratus). In females, these behaviors include sexual odor preference and vaginal scent marking. The neural regulation of precopulatory behaviors is thought to involve a network of forebrain areas that includes the medial amygdala (MA), the bed nucleus of the stria terminalis (BNST), and the medial preoptic area (MPOA). Although MA and BNST are necessary for sexual o

Ascorbic acid (AA), also known as Vitamin C, enhances dopamine (DA) transmission in mesolimbic and nigrostriatal terminals and augments DA-mediated behaviors. It is not yet known whether AA has a similar influence in other DA terminals, in particular terminals of the incertohypothalamic system that modulate the function of the medial preoptic area (mPOA). In female rats, DA in the mPOA plays a critical role in the generation of appetitive sexual responses, notably solicitations, hops, and darts, and we have shown previously that the role of DA in this region on female sexual behavior changes d

Sexual receptivity in female hamsters potentially lasts for about 16 h. However, vaginal cervical stimulation (VCS) from a male during mating eventually reduces receptivity and can shorten the duration of behavioral estrus. The process by which this change in response to the male takes place is unknown. Recently, detection of the Fos protein has indicated that the medial preoptic area (POA) is one of the brain regions particularly responsive to VCS. Additionally, the POA may have an inhibitory effect on sexual receptivity. To determine if protein synthesis in the POA is required to initiate th

In our previous work, it was shown that hibernation could be enhanced by injection of 6-hydroxydopamine (6-OHDA, 200 micrograms) into the lateral cerebral ventricle of ground squirrels by destructing norepinephrine (NE) nerve terminals in brain areas including hypothalamus, cerebral cortex, hippocampus and periaqueductal gray matter of middle brain. In order to identify the specific area contributing for the modulation of NE system on hibernation, small doses of 6-OHDA (4 micrograms) were microinjected bilaterally into medial preoptic hypothalamus (MPA), lateral preoptic hypothalamus (LPA) and

It has been shown that activation of type 1 orexinergic receptors (ORX1) in several parts of the hypothalamus stimulate food intake. Orexin A receptive sites for food intake exist primarily in a narrow band of the hypothalamus that is known to be involved in control of energy homeostasis. The present study aimed to investigate the role of orexin receptors in the medial preoptic area (MPOA) on food intake in rats. Twenty-four male rats weighing 200-250g were divided into three groups (n=8 in each group). Rats were cannulated using stereotaxic coordinates above the MPOA. Normal saline was microi

The medial preoptic area is a structure located in the hypothalamic anteroventral third ventricle region, and is closely related to the olfactory brain development and sexual differentiation of the brain. The medial preoptic area surrounds the organum vasculosum of the lamina terminalis, and both structures are the main areas where synthesis of gonadotropin-releasing hormone occurs in the brain. Neurons synthesizing gonadotropin-releasing hormone migrate from the medial nasal epithelium to the rostral brain and reach the organum vasculosum of the lamina terminalis and the medial preoptic area.

The present study examined the effect of ibotenic acid lesions of the medial preoptic area (mPOA) on the expression of a conditioned place preference (CPP) for vaginocervical stimulation. Rats with bilateral lesions of the mPOA failed to display the CPP for vaginocervical stimulation shown by rats with sham or incomplete lesions. These findings provide additional support for the role of the mPOA in the neural circuitry underlying the reinforcing effects of female sexual behavior and raise the possibility that the altered pattern of approach and withdrawal behavior observed following lesions of

Electrochemical and electrical stimulation of the medial preoptic area (MPA) was shown to induce release of LH in rats. Owing to differences in cytoarchitecture and neural afferents between the medial (mMPA) and lateral (lMPA) parts of the MPA, we decided to explore whether this difference in organization would distinctly influence the secretion of gonadotropin. Both parts of the MPA were electrochemically stimulated on the day of proestrus in freely behaving rats bearing chronic implanted electrodes. An anodic direct current of 100 microA was delivered for 40 s at 11.00 h and blood samples we

Polycystic ovary syndrome (PCOS) is a common endocrine disorder with developmental origins. While the etiology is unclear, current postulates include epigenetic programming. Cell-type-specific epigenetic changes by which prenatal androgen excess programs the neuroendocrine axis have not been defined. Using single-nucleus (sn) multiome sequencing (single-nucleus ribonucleic acid sequencing [snRNAseq] + single-nucleus assay for transposase-accessible chromatin using sequencing [snATACseq]) of the mouse preoptic area, we profiled transcriptional and chromatin accessibility landscapes across 31 ce

Fluctuations in reproductive hormone levels are associated with mood disruptions in women, such as in postpartum and perimenopausal depression. However, the neural circuit mechanisms remain unclear. Here we report that medial preoptic area (MPOA) GABAergic neurons mediate multifaceted depressive-like behaviors in female mice after ovarian hormone withdrawal (HW), which can be attributed to downregulation of activity in Esr1 (estrogen receptor-1)-expressing GABAergic neurons. Enhancing activity of these neurons ameliorates depressive-like behaviors in HW-treated mice, whereas reducing their act

The study was designed to examine whether the administration of neostigmine (0.5 mg/animal), a peripheral inhibitor of acetylcholinesterase (AChE), during an immune/inflammatory challenge provoked by intravenous injection of bacterial endotoxin-lipopolysaccharide (LPS; 400 ng/kg)-attenuates the synthesis of proinflammatory cytokines in the ovine preoptic area (POA), the hypothalamic structure playing an essential role in the control of the reproduction process, and in the choroid plexus (CP), a multifunctional organ sited at the interface between the blood and cerebrospinal fluid in the ewe. The hypothalamus plays an important role in the regulation of sleep/wakefulness states. While the ventrolateral preoptic nucleus (VLPO) plays a critical role in the initiation and maintenance of sleep, the lateral posterior part of the hypothalamus contains neuronal populations implicated in maintenance of arousal, including orexin-producing neurons (orexin neurons) in the lateral hypothalamic area (LHA) and histaminergic neurons in the tuberomammillary nucleus (TMN). During a search for neurons that make direct synaptic contact with histidine decarboxylase-positive (HDC+), histaminergic neuro

BACKGROUND: Decreased estrogen levels can cause abnormal thermosensitivity of the preoptic area (POA) in the hypothalamus during menopause, which may cause hot flashes. Thermosensitive transient receptors (ThermoTRPs) affect the thermosensitivity of neurons. It is worth exploring whether ThermoTRPs change under low estrogen state and participate in the abnormal thermoregulation of POA.
METHODS: Adult female Sprague-Dawley rats were randomly divided into sham operation (SHAM), ovariectomy (OVX) and estrogen treatment after ovariectomy (OVX+E) groups. Under 10 ℃, 18 ℃, 25 ℃, 37 ℃ and 45 ℃ incub

Electrolytic lesions were placed in the preoptic area or in the middle and posterior hypothalamus of female rats within 48 hrs after birth. Vaginal opening was delayed significantly in rats having the medial preoptic area (mPOA) destructed as compared to sham-operated animals and those given lesions in the other parts of the hypothalamus. Only rats with the mPOA completely destructed developed the anovulatory syndrome. In these animals, unilateral ovariectomy was not followed by enlargement of its fellow. The present results are not in agreement with those of previous workers that electrolytic

The medial preoptic area (mPOA) in the hypothalamus is an important integrator of neuroendocrine signaling and a key regulator of both natural and drug-induced reward. Although the mPOA modulates sex differences in other behaviors, whether it also modulates sex differences in cocaine response remains unclear. To help us better understand the mPOA's role in sex differences associated with cocaine response, we examined cocaine-induced changes in locomotion and neural activity in the mPOA of male and female rats. In addition, neural activity in the striatum, a brain area known to be involved in c

It has been established that deoxycorticosterone acetate (DOCA)-salt hypertensive rats have an overactive brain angiotensin-system. The purpose of the present study was to identify the brain sites showing enhanced angiotensin-system activity responsible for the pathogenesis of hypertension in DOCA-salt hypertensive rats. The angiotensin receptor antagonist, losartan, was injected into brain ventricles or into tissues around the rostral parts of the third ventricle in conscious DOCA-salt hypertensive rats. Losartan (1 microg) injection into the lateral ventricle or into the rostral parts of the

Calcitonin receptor (Calcr) and its brain ligand amylin in the medial preoptic area (MPOA) are found to be critically involved in infant care and social contact behaviors in mice. In primates, however, the evidence is limited to an excitotoxic lesion study of the Calcr-expressing MPOA subregion (cMPOA) in a family-living primate species, the common marmoset. The present study utilized pharmacological manipulations of the cMPOA and shows that reversible inactivation of the cMPOA abolishes infant-care behaviors in sibling marmosets without affecting other social or non-social behaviors. Amylin-e

Within the central neural circuitry for thermoregulation, the balance between excitatory and inhibitory inputs to the dorsomedial hypothalamus (DMH) determines the level of activation of brown adipose tissue (BAT) thermogenesis. We employed neuroanatomical and in vivo electrophysiological techniques to identify a source of excitation to thermogenesis-promoting neurons in the DMH that is required for cold defense and fever. Inhibition of median preoptic area (MnPO) neurons blocked the BAT thermogenic responses during both PGE2-induced fever and cold exposure. Disinhibition or direct activation

Horizontal cuts between the septum and preoptic area (anterior roof deafferentation, or ARD) dramatically affect sexual behavior, and in ways that could explain a variety of differences across behavioral categories (precopulatory, copulatory), species, and the sexes. Yet little is known about how these effects develop. Such information would be useful generally and could be pivotal in clarifying the mechanism for ultrasonic vocalization in female hamsters. Ultrasounds serve these animals as precopulatory signals that can attract males and help initiate mating. Their rates can be increased by e
