Side-effects of long-term prostaglandin E(1) treatment in neonates.

BACKGROUND: In some neonates suffering from ductus arteriosus dependent congenital heart defect, a Prostaglandin E(1) (PGE1) therapy longer than 2 weeks may be needed. However, PGE1 analogue compounds may produce several adverse effects. METHODS: The authors retrospectively analyzed the data of nine patients who underwent a PGE1 treatment lasting longer than 14 days. RESULTS: The leukocyte count of the patients remained high throughout the treatment period, and the proportion of neutrophils was over 50%. Transient feeding difficulty and abdominal distension, and possible signs of gastric-out

Localization of prostaglandin E receptor subtypes in the ciliary body of mouse eye.

Prostaglandin E2 (PGE2) markedly reduces intraocular pressure (IOP) when applied topically and induces strong relaxation of pre-contracted isolated ciliary muscle through PGE2 receptor. Because the ciliary muscle relaxation reduces IOP by enhancing uveoscleral aqueous outflow, the ciliary muscle where the existence of PGE2 receptors has been demonstrated is thought to be one of the target tissues for PGE2-induced IOP reduction. To investigate the subtypes of PGE2 receptors in the ciliary muscle, the regional distribution of four PGE2 receptor subtypes (EP1, EP2, EP3 and EP4) in the mouse cilia

Studies on the pathophysiological role of thyroidal prostaglandin E (PGE) in Graves' disease.

A potential role for prostaglandin E (PGE) of thyroid origin in the pathogenesis of the hyperthyroidism of Graves' disease was investigated in vivo. The venous-arterio prostaglandin E (PGE) concentration gradient across the thyroid was measured in 5 patients undergoing subtotal thyroidectomy for Graves' disease. This gradient (161 +/- 14 pg/ml; mean /+- SE) was significantly greater than that found in 11 patients undergoing surgery for hyperparathyroidism and thyroid nodules (64 +/- 20 pg/ml; P < 0.01). This suggests that there is a net increase in PGE output by the thyroid gland in Graves' di

Is urinary flow rate a major regulator of prostaglandin E excretion in man?

Urinary PGE2 excretion is enhanced in several polyuric states in man suggesting that PGE2 synthesis could be a mediator of diuresis. To explore the alternate hypothesis that polyuria is the cause of the increased PGE2 excretion, we increased urine flow rate by intravenous administration of dextrose and water with different magnesium, calcium and potassium solutions in four normal males. Urinary PGE excretion rose in parallel with urine volume (r = 0.65 p < 0.01) independently of the electrolyte solution. To determine the effects of chronic alterations in water balance in 5 female subjects, we

Decreased prostaglandin E turnover in infants with essential fatty acid deficiency.

Sick low birth weight infants (LBWI) are prone to develop rapid onset of essential fatty acid (EFA) deficiency. EFAs serve as precursors for prostaglandins (PGs). We measured the excretion of the major urinary metabolite of prostaglandins E1 and E2, 7alpha-hydroxy-5,11-diketotetranorprostane-1,16-dionic acid (PGE-M), in three EFA-deficient and in nine thriving neonates. There was no significant difference in PGE-M excretion between the sexes among thriving infants nor did PGE-M excretion appear to be affected by postconceptual age. However, a significant difference between the PGE-M excretion

[Level of group E prostaglandins in gastric mucosa of patients with various forms of chronic gastritis].

There was studied prostaglandins E concentration in bioptats of gastric mucosa in 158 patients with various forms and localization of chronic gastritis using a radioimmunological method. Chronic gastritis without atrophy and with mild atrophy showed to have decreased levels of prostaglandins E in comparison with the control. In the patients with atrophic gastritis with "reconstruction" of epithelium there was revealed an increase of prostaglandins concentration, which was maximal in the patients with atrophic-hyperplastic forms of the disease.

Prostaglandins E and F in amniotic fluid during stretch-induced cervical softening and labor at term.

An unspecified region of the uterus of 7 healthy pregnant patients at term, not yet in labor, was mechanically distended by extraamniotic application of rubber bougies. In all cases, significant onset of labor was noted within 20 min to 5 h, cervical softening progressed with time, and delivery was achieved within 21 h. Significant elevation of prostaglandin E (PGE) and F (PGF) levels in amniotic fluid was noted with the progress of cervical dilatation. It appears that the release of PGE and PGF is dependent on uterine stretching and/or the subsequently induced uterine contractions. A possible

The effect of intravenous infusions of prostaglandins E-2 and F-2alpha on human gastric function.

The effect of intravenous infusions of prostaglandins E-2 and F(-2alpha) at various dose levels on basal, or on maximally or submaximally pentagastrin-stimulated acid secretion, was studied in 40 male subjects. Intraluminal antral pressures were also measured. Prostaglandin F (0.08 mug kg-minus 1 min-minus 1) transiently, but significantly, inhibited submaximal acid output and increased the frequency of antral contractions. Prostaglandin E(2)(0.08 mug kg-minus 1 min-minus 1) inhibited basal acid secretion.

Prostaglandin E(2) and 6-keto-prostaglandin F(1alpha) production is elevated following traumatic injury to sciatic nerve.

Sciatic nerve explants cultured either alone or in the presence of peritoneal macrophages were used to study prostaglandin E(2) (PGE(2)) and 6-keto-PGF(1alpha) production following traumatic peripheral nerve injury. Although barely detectable at early time points (1-3 h in vitro), the production of PGE(2) and 6-keto-PGF(1alpha) by sciatic nerve explants increased significantly after 18 h and remained elevated for up to 96 h. The cyclooxygenase-2 (COX-2) selective inhibitor, NS-398, inhibited PGE(2) and 6-keto-PGF(1alpha) production by injured sciatic nerve in a dose-dependent manner. Consisten

EP3 is the only prostaglandin E receptor required for lipopolysaccharide-induced fever.

Fever induced by systemic inflammation is mediated by prostaglandin E2 (PGE2), but the identity of the prostaglandin E (EP) receptor subtype responsible for the febrile response remains debated. Although EP3 receptors have been strongly implicated in fever generation, studies using pharmacological approaches and receptor-deficient mice have also suggested involvement of EP1 and EP4 receptors. Because previous studies have differed substantially with regard to experimental conditions, including ambient temperature, route of immune challenge, and injection-associated stress responses, we systema

Regulation of ocular surface inflammation by prostaglandin E receptor subtype EP3.

We first investigated whether the prostaglandin (PG) E2-PGE receptor subtype EP3 axis regulates the development of murine experimental allergic conjunctivitis because it has been reported that this pathway negatively regulates allergic reactions in a murine allergic asthma model. We observed that EP3 is constitutively expressed in mice conjunctival epithelium. EP3 knockout mice demonstrated significantly increased eosinophil infiltration in conjunctiva after ragweed challenge compared with wild-type mice. Consistently, significantly higher expression of eotaxin-1 messenger RNA was observed in

Expression and regulation of prostaglandin E synthase isoforms in human myometrium with labour.

Since the controversies regarding the use of non-steroidal anti-inflammatory drugs (NSAIDs) and selective cyclo-oxygenase (COX)-2 antagonists for the treatment of preterm labour (PTL), more emphasis has been placed on investigating the terminal synthases involved in the production of prostaglandins (PGs) to allow more targeted therapy in PTL. Prostaglandin E(2) (PGE(2)) is synthesized by one of three enzymes, cytosolic prostaglandin E synthase (cPGES), microsomal PGES-1 (mPGES-1) and microsomal PGES-2 (mPGES-2). We have determined (i) the immuno-localization of all three PGES enzymes in lower

Efficacy of the treatment with prostaglandin E-1 in venous ulcers of the lower limbs.

BACKGROUND: Venous ulcers represent an important medical problem because of their high prevalence and consequent sanitary costs. In this study, we evaluated the effect of prostaglandin E-1 (PGE-1), a drug that improves district ischemia, on the healing of venous ulcers. METHODS: We performed a randomized, placebo-controlled, single blind study in which 87 patients who had venous leg ulcers homogeneous for dimensions and characteristics were treated for 20 days with an infusion of prostaglandin E-1 or placebo, in association with topical therapy. The dimension and the number of the ulcers were

Effect of hCG injection on prostaglandin E concentrations in ram seminal plasma.

Ram and bull seminal plasma, respectively, contain 0.5-20 microg PGE/ml and 5-10 ng PGE/ml. To demonstrate that PGE concentrations in the seminal plasma are related to sperm quality and could be affected by hormonal stimulation in vivo, four rams were injected with 500 IU hCG, in and out of season. The rams responded 1 week after hCG with a 1.5- to 4-fold increase in seminal plasma PGE. The PGE peak was temporally separate from the hCG-induced rise in seminal plasma testosterone which was observed after 1 day. Using a simulated cryptochid ram, peaks in seminal fluid PGE were found to be associ

Variable responses to prostaglandin E(2) in human non-pregnant myometrium.

Cumulative concentration-effect curves for prostaglandin E(2), sulprostone and butaprost were constructed in matched strips of human non-pregnant myometrium from 14 different donors. All samples were obtained from the mid-lateral wall of the uterus. Prostaglandin E(2) produced four types of concentration-effect curves: monophasic inhibitory (n = 7), monophasic excitatory (n = 2), biphasic consisting of an excitatory phase followed by an inhibitory phase (n = 4), and biphasic consisting of an inhibitory phase followed by an excitatory phase (n = 1). Sulprostone produced excitation of spontaneou

Human glutathione dependent prostaglandin E synthase: gene structure and regulation.

A P1 clone containing the gene for human glutathione dependent PGE synthase (PGES) was isolated and characterized. The gene is divided into three exons, spans 14.8 kb and was localized to chromosome 9q34. 3. In A549 cells, the protein and activity levels of PGES were increased by interleukin-1beta. This increase was prevented by phenobarbital. Reporter constructs containing the 5'-flanking region of exon 1, which exhibited strong promoter activity, responded accordingly, except that interleukin-1beta induced a transient increase followed by a decrease. As cyclooxygenase 2 expression has been r

Prostaglandin E production by amniotic cells in relation to term and preterm labour.

The rate of prostaglandin E (PGE) production was measured in collagenase-dispersed amniotic cells obtained from 14 women after spontaneous labour at term--seven after spontaneous preterm labour, nine after delivery by elective caesarean section at term and six after induction of labour at term. Cells were incubated with and without arachidonic acid and PGE was estimated by specific radioimmunoassay. Basal PGE output (pmol/10(6) cells per 3 h) was highest in the spontaneous labour group, 27.5 (SEM 5.5) and lowest in the preterm labour group, 4 (SEM 1.2) (P less than 0.001). Values in the electi

Effect of acetylsalicylic acid on urinary excretion of prostaglandin E in stroke patients.

In 12 of 76 stroke patients complicated by the syndrome of inappropriate secretion of antidiuretic hormone (SIADH), a significant increase in urinary prostaglandin E (PGE) (p less than 0.005), and a significant positive relationship between the plasma arginine vasopressin (AVR) level and urinary PGE excretion were observed (r = 0.72, p less than 0.05). The experimental results are consistent with the view that renal PGE acts as a modulator of ADH. Nowadays acetylsalicylic acid (ASA), an inhibitor of prostaglandin biosynthesis, is widely used in ischemic stroke, it was felt necessary to study t
