AMP-activated protein kinase (AMPK) is the central component of a cellular signaling system that regulates multiple metabolic enzymes and pathways in response to reduced intracellular energy levels. The transcription factor hepatic nuclear factor 4alpha (HNF4alpha) is an orphan nuclear receptor that regulates the expression of genes involved in energy metabolism in the liver, intestine, and endocrine pancreas. Inheritance of a single null allele of HNF4alpha causes diabetes in humans. Here we demonstrate that AMPK directly phosphorylates HNF4alpha and represses its transcriptional activity. AM

The survival motor neuron (SMN) protein is an essential and highly versatile assembly factor that coordinates RNA metabolism and ribonucleoprotein (RNP) complex formation across multiple cellular compartments. Although SMN is required for the survival of virtually all cell types, its deficiency disproportionately affects α-motor neurons, causing their selective degeneration and giving rise to spinal muscular atrophy (SMA). Once viewed primarily as a motor neuron disease, SMA is now understood to be a multi-systemic disorder in which cell-intrinsic dysfunction extends to skeletal muscle, inflam

SLC45A2 encodes a putative transporter expressed primarily in pigment cells. SLC45A2 mutations cause oculocutaneous albinism type 4 (OCA4) and polymorphisms are associated with pigmentation variation, but the localization, function, and regulation of SLC45A2 and its variants remain unknown. We show that SLC45A2 localizes to a cohort of mature melanosomes that only partially overlaps with the cohort expressing the chloride channel OCA2. SLC45A2 expressed ectopically in HeLa cells localizes to lysosomes and raises lysosomal pH, suggesting that in melanocytes SLC45A2 expression, like OCA2 express

When contact lenses (CLs) are worn, they are subject to deposition of the surrounding biomolecules found in the tear film (TF) of the eye. There is a correlation between protein deposition on CLs and feelings of discomfort in patients, but it has not been well understood if these feelings of discomfort arise solely from immunogenic reactions to the protein deposits or a physical instability of the tear film on protein-fouled CLs. This study compared two hydrogel CLs: etafilcon A (polyhydroxyethylmethacrylate-based hydrogel) and senofilcon A (silicone hydrogel with internal wetting agent) to el

Protein thermodynamics are an integral determinant of viral fitness and one of the major drivers of protein evolution. Mutations in the influenza A virus (IAV) hemagglutinin (HA) protein can eliminate neutralizing antibody binding to mediate escape from preexisting antiviral immunity. Prior research on the IAV nucleoprotein suggests that protein stability may constrain seasonal IAV evolution; however, the role of stability in shaping the evolutionary dynamics of the HA protein has not been explored. We used the full coding sequence of 9,797 H1N1pdm09 HA sequences and 16,716 human seasonal H3N2

Prions are the etiological agent of fatal neurodegenerative diseases called prion diseases or transmissible spongiform encephalopathies. These maladies can be sporadic, genetic or infectious disorders. Prions are due to post-translational modifications of the cellular prion protein leading to the formation of a β-sheet enriched conformer with altered biochemical properties. The molecular events causing prion formation in sporadic prion diseases are still elusive. Recently, we published a research elucidating the contribution of major structural determinants and environmental factors in prion p

ML-IAP [melanoma IAP (inhibitor of apoptosis)] is an anti-apoptotic protein that is expressed highly in melanomas where it contributes to resistance to apoptotic stimuli. The anti-apoptotic activity and elevated expression of IAP family proteins in many human cancers makes IAP proteins attractive targets for inhibition by cancer therapeutics. Small-molecule IAP antagonists that bind with high affinities to select BIR (baculovirus IAP repeat) domains have been shown to stimulate auto-ubiquitination and rapid proteasomal degradation of c-IAP1 (cellular IAP1) and c-IAP2 (cellular IAP2). Single-strand DNA (ssDNA)-binding proteins (SSBs) are ubiquitous and essential for a wide variety of DNA metabolic processes, including DNA replication, recombination, DNA damage detection and repair. SSBs have multiple roles in binding and sequestering ssDNA, detecting DNA damage, stimulating nucleases, helicases and strand-exchange proteins, activating transcription and mediating protein-protein interactions. In eukaryotes, the major SSB, replication protein A (RPA), is a heterotrimer. Here we describe a second human SSB (hSSB1), with a domain organization closer to the archaeal SSB than to

A small-scale oil bath immersion-heating system using sealed stainless-steel process tubes was developed that has the flexibility to evaluate a wide range of holding temperatures and times. After heat treatment and rapid cooling, the liquid product was removed from each stainless-steel tube and a centrifugal protein sedimentation test and laser light scattering particle size analysis were done to determine differences among milk protein beverages in heat stability and casein micelle aggregation before and after thermal processing. The sedimentation and particle size analysis method was able to

Iron deficiency anemia (IDA) is one of the most common nutritional disorders worldwide. However, traditional iron supplement often induces adverse gastrointestinal effects (e.g. nausea, vomiting, abdominal pain) due to their gastrointestinal irritation and poor bioavailability. To overcome this defect, we designed an antioxidative whey protein hydrolysate fibril-dextran (WPHF-DEX) complex system, which can delivery low-irritant and highly biocompatible iron oxide by in-situ co-precipitation and enhancing iron bioavailability. After mixing with WPH and grafting with dextran, the antioxidant of

Protein based vaccines are the most safe and affordable strategy to combat pneumococcal disease circumventing the limitations of conventional polysaccharide-based vaccines like serotype dependence, high cost and inability to be administered to immunocompromised. SP0845 is a highly conserved vaccine candidate shown to provide protection against heterologous strains of Streptococcus pneumoniae, the primal cause of pneumonia. However, the associated poor immunogenicity warrants the need for adjuvants and multiple doses to mount desired responses. The present study relates to improve the immunogen

The severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) is a global threat to human health and has compromised economic stability. In addition to the development of an effective vaccine, it is imperative to understand how SARS-CoV-2 hijacks host cellular machineries on a system-wide scale so that potential host-directed therapies can be developed. In situ proteome-wide abundance and thermal stability measurements using thermal proteome profiling (TPP) can inform on global changes in protein activity. Here we adapted TPP to high biosafety conditions amenable to SARS-CoV-2 handling. We

ΔNp63α is finely and strictly regulated during embryogenesis and differentiation. ΔNp63α is the only p63 isoform degraded by the proteasome after Ubiquitin and SUMO (Small Ubiquitin-like MOdifier) conjugation. Here, we show that p63 ubiquitylation per se is not the signal triggering p63 proteasomal degradation. Taking advantage of natural ΔNp63α mutants isolated by patients with Split Hand and Foot Malformation IV syndrome, we found that SUMO and Ub modifications are not redundant and both are required to guarantee efficient ΔNp63α degradation. Here, we present evidence that sumoylation and ub

The key pathogenic steps leading to spinal muscular atrophy (SMA), a genetic disease characterized by selective motor neuron degeneration, are not fully clarified. The full-length SMN protein (FL-SMN), the main protein product of the disease gene SMN1, plays an established role in the cytoplasm in snRNP biogenesis ultimately leading to mRNA splicing within the nucleus. It is also involved in the mRNA axonal transport. However, to what extent the impairment of these two SMN functions contributes to SMA pathogenesis remains unknown. A shorter SMN isoform, axonal-SMN or a-SMN, with more specific

We present a molecular analysis of four dominant alleles of the pair-rule gene ftz. Three of these, the ftzUal alleles, cause anti-ftz segmentation defects and homeotic transformations of the first abdominal segment to the third. These alleles are shown to be missense changes affecting two nearby proline codons. Embryos homozygous for these mutations accumulate higher levels of ftz protein than wild type and show strong persistence of ftz protein, but not RNA. These effects appear to result from stabilization of the ftz protein, since ftz stripes decay much more slowly in mutant embryos than i

The emulsification of vegetable protein is closely related to solubility. The purpose of this study was to evaluate the effect of ultrasound on protein emulsification and to provide a prospective method for assessing the digestive properties of emulsions. In this article, we investigate the emulsion stability of ultrasonic pretreated soy protein isolate (SPI), and its three storage proteins, namely β-conglycinin (7S), lipophilic protein (LP), and glycinin (11S), under dynamic gastric conditions. The effects of these emulsions on lipolysis during digestion in the small intestine are also assess

Myofibrillar protein microgel particles (MMP) are promising Pickering stabilisers due to their structure and delivery potential. However, their fibrous, irregular shape promotes aggregation, limiting practical use. This study investigated the effect of xanthan gum (XG) concentration (0.025-0.4%) on MMP dispersion in water and its role in stabilising Pickering emulsions. FTIR and interaction analysis revealed that hydrophobic interactions dominate between XG and MMP, followed by hydrogen bonding and electrostatic forces. At higher XG concentrations (0.2-0.4%), complex particle size decreased fr

In recent years, many studies have focused on Pickering emulsions (PEs) stabilized by protein particles/nanoparticles (NPs) because of their excellent nutritional value, biodegradability, easy extraction, high surface activity, and modification capability. Despite growing interest in this area, the relationships among protein particle morphology, fabrication techniques, surface modification strategies, and stabilization performance have not always been discussed in an integrated manner, synthesizing the goal of this paper. First, a brief history of PEs is presented. Second, the article provide

The purpose of the study was to investigate the stability and oral delivery of DHA-encapsulated Pickering emulsions stabilized by soy protein isolate-chitosan (SPI-CS) nanoparticles (SPI-CS Pickering emulsions) under various conditions and in the simulated gastrointestinal (GIT) model. The stability of DHA was characterized by the retention rate under storage, ionic strength, and thermal conditions. The oral delivery efficiency was characterized by the retention and release rate of DHA in the GIT model and cell viability and uptake in the Caco-2 model. The results showed that the content of DH

Membrane curvature plays a central role in a wide range of biological processes, yet a quantitative and transferable description of its underlying energetics remains challenging. In this work, we develop a "reaction coordinate" within the Umbrella-Sampling framework to compute the free-energy cost associated with local membrane bending in both coarse-grained and atomistic simulations. The methodology is systematically validated across diverse lipid environments, including pure bilayers with varying tail unsaturation, tail length, and headgroup chemistry, where it reproduces established trends
