Proteolysis Targeting Chimera Agents (PROTACs): New Hope for Overcoming the Resistance Mechanisms in Oncogene-Addicted Non-Small Cell Lung Cancer.

Non-small cell lung cancer (NSCLC) remains a disease with a poor prognosis despite the advances in therapies. NSCLC with actionable oncogenic alterations represent a subgroup of diseases for which tyrosine kinase inhibitors (TKIs) have shown relevant and robust impact on prognosis, both in early and advanced stages. While the introduction of powerful TKIs increases the ratio of potentially curable patients, the disease does develop resistance over time through either secondary mutations or bypass activating tracks. Therefore, new treatment strategies are being developed to either overcome this

Probing class I histone deacetylases (HDAC) with proteolysis targeting chimera (PROTAC) for the development of highly potent and selective degraders.

Class I HDACs are considered promising targets for cancer due to their role in epigenetic modifications. The main challenges in developing a new, potent and non-toxic class I HDAC inhibitor are selectivity and appropriate pharmacokinetics. The PROTAC technique (Proteolysis Targeting Chimera) is a new method in drug development for the production of active substances that can degrade a protein of interest (POI) instead of inhibiting it. This technique will open the era to produce selective and potent drugs with a high margin of safety. Previously, we reported different inhibitors targeting clas

Next-Generation Proteolysis-Targeting Chimeras in Precision Oncology: Multifunctional Designs, Emerging Modalities, and Translational Prospects in Targeted Protein Degradation.

Proteolysis-targeting chimeras (PROTACs)-mediated protein degradation has been recently developed as a game-changing approach in oncology drug development. It represents a paradigm shift from traditional enzyme inhibition to selective protein degradation. PROTACs are different from regular small-molecule inhibitors because they are heterobifunctional compounds that use the ubiquitin-proteasome system to breakdown disease-causing oncogenic proteins. This review discusses the next generation of PROTAC platforms that innovate beyond traditional designs, such as dual-targeting PROTACS that present

Sequential Disruption of Tumor Camouflage and Metabolic Coercion by Cascade-Acting Nano-Proteolysis Targeting Chimera Engines to Reprogram the Immune Ecosystem.

The clinical efficacy of cancer immunotherapy is fundamentally constrained by the synergistic evasion mechanisms of antigen concealment and effector immune cell suppression. To overcome these barriers, we proposed a therapeutic paradigm that sequentially dismantles the tumor's "immune camouflage" and relieves "metabolic coercion," thereby enabling systematic remodeling of the tumor immune microenvironment. A cascade-acting nano-Proteolysis Targeting Chimera (PROTAC) engine was fabricated via phosphatidylcholine-driven self-assembly, enabling tumor-targeted delivery and pH-responsive drug relea

Research Trends and Emerging Frontiers in Proteolysis Targeting Chimeras (PROTACs): A Bibliometric Analysis of 2630 Publications (2001-2025).

Background/Objectives: Proteolysis Targeting Chimeras (PROTACs) are heterobifunctional small molecules that induce ubiquitin-proteasome-mediated degradation of target proteins and have matured from proof-of-concept chemistry to a clinically validated therapeutic modality, with the first Phase 3 readout reported in 2025. A systematic bibliometric analysis covering this pivotal-trial era, however, has been lacking. This study aimed to map the historical trajectory, current research front, and emerging frontiers of PROTAC research. Methods: We analyzed 2630 PROTAC-related publications indexed in

The androgen receptor-targeted proteolysis targeting chimera and other alternative therapeutic choices in overcoming the resistance to androgen deprivation treatment in prostate cancer.

Androgen receptor (AR) plays a vital role in prostate cancer (PCa), including castration-resistant PCa, by retaining AR signalling. Androgen deprivation treatment (ADT) has been the standard treatment in the past decades. A great number of AR antagonists initially had been found effective in tumour remission; however, most PCa relapsed that caused by pre-translational resistance such as AR mutations to turn antagonist into agonist, and AR variants to bypass the androgen binding. Recently, several alternative therapeutic choices have been proposed. Among them, proteolysis targeting chimera (PRO

Limiting Trial-and-Error: Linker-Based 2D Descriptor Analysis to Support Very Early Proteolysis-Targeting Chimeras Design.

Targeted protein degradation has emerged as a clinically validated therapeutic strategy, culminating in the recent FDA approval of the first proteolysis-targeting chimeras (PROTAC) drug (VEPPANU). Within PROTACs, the linker plays a central role in modulating degradation efficiency as well as physicochemical and in vitro absorption, distribution, metabolism, and excretion properties, yet linker optimization still largely relies on empirical "linkerology" approaches. Here, we investigated whether simple 2D linker descriptors could provide informative insights for very early-stage PROTAC design.

Fluorinated Proteolysis Targeting Chimeras-Sorafenib Nanoassembly for Epigenetic Remodeling to Combat Multi-Pathway Drug Resistance in Hepatocellular Carcinoma.

Although sorafenib (SF) is the standard first-line therapy for hepatocellular carcinoma (HCC), extended exposure frequently induces multipathway resistance driven by tumor hypoxia and an immunosuppressive microenvironment. Here we report a fluorinated Proteolysis TArgeting Chimeras (PROTAC)-sorafenib nanoassembly (FCP@SF/FPro) that boosts efficacy by degrading bromodomain-containing protein 4 (BRD4) and rewiring epigenetic programmes. This innovative system integrates fluorinated PROTAC targeting BRD4 (FPro) and sorafenib within a fluorinated poly(ethylene glycol)-conjugated poly(ethylenimine)

BET proteolysis targeted chimera-based therapy of novel models of Richter Transformation-diffuse large B-cell lymphoma.

Richter Transformation (RT) develops in CLL as an aggressive, therapy-resistant, diffuse large B cell lymphoma (RT-DLBCL), commonly clonally-related (CLR) to the concomitant CLL. Lack of available pre-clinical human models has hampered the development of novel therapies for RT-DLBCL. Here, we report the profiles of genetic alterations, chromatin accessibility and active enhancers, gene-expressions and anti-lymphoma drug-sensitivity of three newly established, patient-derived, xenograft (PDX) models of RT-DLBCLs, including CLR and clonally-unrelated (CLUR) to concomitant CLL. Proteolysis Targeting Chimera (PROTAC) Linkerology Enhances the CDK-Degradation Selectivity Profile of a Multi-CDK Inhibitor: Discovery of Potent, Anti-Leukemic CDK9 Degraders.

The degradation efficiency of a PROTACbivalent compounds that link together a ligand for a protein of interest with a ligand for an E3 ligaseis governed, in part, by the length and flexibility of its linker ("linkerology"), as well as the accessibility of ubiquitinatable surface lysines. Based on the prior serendipitous discovery of the cyclin-dependent kinase 9 (CDK9)-selective PROTAC degrader "THAL-SNS-032", developed from the multi-CDK2/CDK7/CDK9 inhibitor SNS-032, we speculated that other multi-CDK inhibitors might likewise be transformed into CDK9-selective PROTACs, establishing a "link

Discovery of CN0 as a novel proteolysis-targeting chimera (PROTAC) degrader of PARP1 that can activate the cGAS/STING immunity pathway combined with daunorubicin.

Poly ADP-ribose polymerase 1 (PARP1) plays an essential role in DNA repair signaling, rendering it an attractive target for cancer treatment. Despite the success of PARP1 inhibitors (PARPis), only a few patients can currently benefit from PARPis. Moreover, drug resistance to PARPis occurs during clinical treatment. Natural and acquired resistance to PARPis has forced us to seek new therapeutic approaches that target PARP1. Here, we synthesized a series of compounds by proteolysis-targeting chimera (PROTAC) technology to directly degrade the PARP1 protein. We found that CN0 (compound 3) with no

Carrier-Free Nano-Proteolysis Targeting Chimeras Enable Selective Protein Degradation and Enhance Photoimmunometabolic Therapy against Lung Adenocarcinoma.

The suboptimal efficacy of immunotherapy is closely associated with the immunosuppressive tumor microenvironment, especially the overexpression of bromodomain-containing protein 4 (BRD4) and the accumulation of immunosuppressive adenosine. This study aimed to develop carrier-free nanoparticles (MNC NPs) by integrating a BRD4-targeting proteolysis-targeting chimera (PROTAC), an adenosine A2A receptor (A2AR) antagonist, and a photosensitizer to enhance the efficacy of immunotherapy. Clinical data from the Gene Expression Omnibus and patient samples highlighted the therapeutic potential of simult

A Physiologically Responsive Nanocomposite Hydrogel for Treatment of Head and Neck Squamous Cell Carcinoma via Proteolysis-Targeting Chimeras Enhanced Immunotherapy.

Although immunotherapy has revolutionized oncotherapy, only ≈15% of head and neck squamous cell carcinoma (HNSCC) patients benefit from the current therapies. An immunosuppressive tumor microenvironment (TME) and dysregulation of the polycomb ring finger oncogene BMI1 are potential reasons for the failure. Herein, to promote immunotherapeutic efficacy against HNSCC, an injectable nanocomposite hydrogel is developed with a polymer framework (PLGA-PEG-PLGA) that is loaded with both imiquimod encapsulated CaCO3 nanoparticles (RC) and cancer cell membrane (CCM)-coated mesoporous silica nanoparticl

Anti-Protein-Adsorption Nano-Proteolysis Targeting Chimeras as a Potent Platform for Efficient Programmed Cell Death Ligand 1 Degradation.

Immune checkpoint blockade (ICB) therapies targeting the PD-1/PD-L1 axis have shown clinical promise, yet most patients do not achieve durable responses due to adaptive PD-L1 upregulation and recycling. While nanoparticle-based proteolysis targeting chimeras (PROTACs) offer a strategy for direct PD-L1 degradation, their therapeutic potential remains largely untapped because of a fundamental barrier: the inevitable formation of a nonspecific protein corona that severely limits target engagement. Here, we introduce a corona-free nano-PROTACs platform (CF-nPTs) that overcomes this obstacle. By in

Structural-based design of HD-TAC7 PROteolysis TArgeting chimeras (PROTACs) candidate transformations to abrogate SARS-CoV-2 infection.

Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) is responsible for about 672 million infections and 6.85 million deaths worldwide. Upon SARS-CoV-2 infection, Histone deacetylases (HDACs) hyperactivate the pro-inflammatory response resulting in stimulation of Acetyl-Coenzyme A and cholesterol for viral entry. HDAC3 inhibition results in the anti-inflammatory activity and reduction of pro-inflammatory cytokines that may restrict COVID-19 progression. Here, we have designed 44 conformational ensembles of previously known HD-TAC7 by enumerating torsions of dihedral angles tested for t

Discovery of Dihydropyrimido-Pyrimidine-Based Bone Marrow Tyrosine Kinase Gene in Chromosome X Protein Proteolysis-Targeting Chimeras for the Treatment of Prostate Cancer.

Bone marrow tyrosine kinase gene in chromosome X protein (BMX) plays a critical role in the initiation, progression, and development of castration-resistant prostate cancer. While several ATP-competitive BMX inhibitors have been developed as cellular chemical tools to investigate the function of BMX, they are unable to target the nonenzymatic roles of BMX. Proteolysis-targeting chimeras (PROTACs) represent an emerging technology that enables the rapid and complete degradation of target proteins. Here, we describe the design, synthesis, and characterization of BMX PROTACs by conjugating the BMX

Oncogene activation mechanism determines the limits of targeted protein degradation.

Protein degrader drugs such as PROTACs are being advanced as therapeutics targeted against oncogenic proteins. During tumorigenesis, oncogenic proteins can become constitutively activated via mechanisms including gene amplification, which increases protein production, and point mutations, which can extend protein half-life. Few experimental studies have addressed how disease-associated changes in target protein homeostasis influence PROTAC activity. We developed orthogonal methods to increase production or enhance stability of β-catenin, an important oncoprotein and target for degrader therape
