Artemisinin and its derivatives in treating protozoan infections beyond malaria.

Parasitic protozoan diseases continue to rank among the world's greatest global health problems, which are also common among poor populations. Currently available drugs for treatment present drawbacks, urging the need for more effective, safer, and cheaper drugs. Artemisinin (ART) and its derivatives are some of the most important classes of antimalarial agents originally derived from Artemisia annua L. However, besides the outstanding antimalarial and antischistosomal activities, ART and its derivatives also possess activities against other parasitic protozoa. In this paper we review the acti

Antibiotic resistance in prevalent bacterial and protozoan sexually transmitted infections.

The emergence of multi-drug resistant sexually transmitted infections (STIs) is causing a treatment crisis across the globe. While cephalosporin-resistant gonorrhea is one of the most pressing issues, extensively antibiotic resistant Chlamydia trachomatis and Mycoplasma hominis are also becoming commonplace. Experts have suggested that the failure of current treatment regimens are "largely inevitable" and have called for entirely new classes of antimicrobial agents. With the exception of several new classes of drugs primarily targeting nosocomial infections, progress has been slow. While pharm

Some aspects of protozoan infections in immunocompromised patients- a review.

Protozoa are among the most important pathogens that can cause infections in immunocompromised hosts. These microorganisms particularly infect individuals with impaired cellular immunity, such as those with hematological neoplasias, renal or heart transplant patients, patients using high doses of corticosteroids, and patients with acquired immunodeficiency syndrome. The protozoa that most frequently cause disease in immunocompromised patients are Toxoplasma gondii, Trypanosoma cruzi, different Leishmania species, and Cryptosporidium parvum; the first two species cause severe acute meningoencep

[Laboratory diagnosis of intestinal protozoan opportunistic infections in Goiânia, GO].

Cryptosporidiosis, isosporiasis, cyclosporiasis and microsporídiosis have become common in immunocompromised patients. The diagnosis of these diseases is important, because although they produce similar clinical pictures, they have a different therapeutic approach and prognostic. Seeking to evaluate the situation of the diagnosis of these parasites, the laboratories of the SUS network and health centers of the district of Goiânia-GO were mapped.

Intestinal protozoan infections: prevalence in the San Francisco Bay Area.

A sample of 415 members of the Kaiser Foundation Health Plan (KFHP) were tested for intestinal protozoa. This parasite survey group was found to be comparable in age distribution, sex and ethnic background with the membership at large. Gastrointestinal complaints were no more numerous in this group than in a large series of KFHP members taking multiphasic examinations. Because KFPH membership included, at the time of the survey, approximately 27 percent of all residents of the San Francisco Bay Area, the membership may be considered approximately representative of the entire area. Fourteen per

Gastrointestinal protozoan and helminth parasite infections in captive chameleons in Germany.

Within lizards, chameleons (family Chamaeleonidae) represent a distinctive and highly specialized clade of insectivorous reptiles. Chameleons have become more popular as reptile pets in recent years worldwide, due to their feasible husbandry and attractiveness. To date, not only reptile pet numbers but also health-compromising endoparasite infections are rising. In the current study, we analyzed a total of 670 faecal samples of 25 different chameleon species kept as pet animals all over Germany. In total, 223 animals (33%) were found to harbor gastrointestinal parasites. In total, 10 different

Host-Derived Extracellular Vesicles in Blood and Tissue Human Protozoan Infections.

Blood and tissue protozoan infections are responsible for an enormous burden in tropical and subtropical regions, even though they can also affect people living in high-income countries, mainly as a consequence of migration and travel. These pathologies are responsible for heavy socio-economic issues in endemic countries, where the lack of proper therapeutic interventions and effective vaccine strategies is still hampering their control. Moreover, the pathophysiological mechanisms associated with the establishment, progression and outcome of these infectious diseases are yet to be fully descri

The State of Art of Extracellular Traps in Protozoan Infections (Review).

Protozoan parasite infection causes severe diseases in humans and animals, leading to tremendous economic and medical pressure. Natural immunity is the first line of defence against parasitic infection. Currently, the role of natural host immunity in combatting parasitic infection is unclear, so further research on natural host immunity against parasites will provide a theoretical basis for the prevention and treatment of related parasitic diseases. Extracellular traps (ETs) are an important natural mechanism of immunity involving resistance to pathogens. When immune cells such as neutrophils

Clearing or subverting the enemy: Role of autophagy in protozoan infections.

The protozoan parasites are evolutionarily divergent, unicellular eukaryotic pathogens representing one of the essential sources of parasitic diseases. These parasites significantly affect the economy and cause public health burdens globally. Protozoan parasites share many cellular features and pathways with their respective host cells. This includes autophagy, a process responsible for self-degradation of the cell's components. There is conservation of the central structural and functional machinery for autophagy in most of the eukaryotic phyla, however, Plasmodium and Toxoplasma possess a de

Protozoan infections are under-recognized in Swedish patients with gastrointestinal symptoms.

In acute gastroenteritis (GE), identification of the infectious agent is important for patient management and surveillance. The prevalence of GE caused by protozoa may be underestimated in Swedish patients. The purpose was to compare the prevalence of E. histolytica, Cryptosporidium spp., G. intestinalis, and C. cayetanensis in samples from patients where the clinician had requested testing for gastrointestinal parasites only (n = 758) to where testing for bacterial GE only (n = 803) or where both parasite and bacterial testing (n = 1259) was requested and a healthy control group (n = 197). Macrophage migration inhibitory factor (MIF): a key player in protozoan infections.

Macrophage migration inhibitory factor (MIF) is a pleiotropic cytokine produced by the pituitary gland and multiple cell types, including macrophages (Mø), dendritic cells (DC) and T-cells. Upon releases MIF modulates the expression of several inflammatory molecules, such as TNF-α, nitric oxide and cyclooxygenase 2 (COX-2). These important MIF characteristics have prompted investigators to study its role in parasite infections. Several reports have demonstrated that MIF plays either a protective or deleterious role in the immune response to different pathogens. Here, we review the role of MIF

How colonization bottlenecks, tissue niches, and transmission strategies shape protozoan infections.

Protozoan pathogens such as Plasmodium spp., Leishmania spp., Toxoplasma gondii, and Trypanosoma spp. are often associated with high-mortality, acute and chronic diseases of global health concern. For transmission and immune evasion, protozoans have evolved diverse strategies to interact with a range of host tissue environments. These interactions are linked to disease pathology, yet our understanding of the association between parasite colonization and host homeostatic disruption is limited. Recently developed techniques for cellular barcoding have the potential to uncover the biology regulat

Primary Tick-Borne Protozoan and Rickettsial Infections of Animals in Turkey.

Parasitic diseases caused by ticks constitute a barrier on global animal production, mainly in tropical and subtropical regions. As a country with a temperate and subtropical climate, Turkey has topography, climate, and pasture resources, and these resources are suitable for animal breeding and parasite-host-vector relationships throughout the country. This geography restricts the regulations on animal movements in the southeastern and eastern Anatolia because of the close contact with the neighboring states. The livestock resources in Turkey are regulated by strong foundations. Almost 30% of

Update on the major imported protozoan infections in travelers and migrants.

Globalization has contributed to the emergence of specific parasitic diseases in novel geographical areas, and in these regions, these infections in travelers and immigrants may cause a considerable burden of disease. Timely diagnosis and treatment of protozoan infections to decrease mortality and prevent associated complications are essential. In this respect, the increased availability of specific DNA-detection procedures has improved the diagnosis of many imported parasitic infections. Travelers and immigrants with associated comorbidities or immunosuppression may pose a special challenge r

The role of monocytes in models of infection by protozoan parasites.

The confirmation of developmental differences between tissue macrophages and peripheral monocytes has changed our view of the functions and dynamics of these two important components of the innate immune system. It has been demonstrated conclusively that homeostasis of tissue resident macrophages is maintained by a low proliferative turn over. During an inflammatory response, bone marrow derived monocytes enter the tissue in large numbers and take part in the defense against the pathogens. After the destruction of invading pathogens, these cells disappear and tissue resident macrophages can be

[Common tropical infections with protozoans, worms and ectoparasites].

Infectious diseases of the skin have become rarer in industrialized nations, but they still affect a considerable part of the population in tropical regions. Skin diseases induced by protozoa, worms and ectoparasites are among the 17 "neglected tropical diseases" defined by the WHO (leishmaniasis, dracunculiasis, lymphatic filariasis, onchocerciasis). Skin symptoms in travellers returning from the tropics may challenge dermatologists in Germany regarding differential diagnostic assessment and therapy. Among the 12 most frequent skin diseases in travellers are cutaneous larva migrans, leishmani
