Susceptible dairy cattle were immunized with attenuated, live calf-derived, in vitro-cultured or biologically cloned Babesia bovis, with non-viable exoantigens, or with recombinant rhoptry-associated protein 1 (rRAP1). Antibody response assessed by the indirect fluorescent assay (IFA) and by the growth inhibition activity in vitro showed that seroconversion correlated with neutralization activity in vitro in all immunized groups, but not with protective immunity in vivo. The protective responses elicited by immunization with completely avirulent biologically cloned live parasites, or by the ex

Equines play a crucial role in global food security, economic development, and recreation, particularly in regions such as Central Asia, parts of Africa, and South America. However, parasitic infections significantly impact their health, productivity, and reproductive performance, leading to economic losses and reduced animal welfare. This review synthesizes the effects of parasitic infections, including protozoan, helminthic, and ectoparasitic species, on equines. These infections cause hematological alterations like anemia, leukocytosis, leukopenia, and thrombocytopenia, compromising overall

Visceral leishmaniasis is a re-emergent disease and a significant cause of morbidity worldwide. Amongst the more than 20 Leishmania species, Leishmania donovani, Leishmania infantum and more rarely Leishmania amazonensis are associated with visceral leishmaniasis. A major question in leishmaniasis research is how these species migrate to and infect visceral organs whereas other species such as Leishmania major and Leishmania braziliensis remain in the skin, causing tegumentary leishmaniasis. Here we present the more recent advances and approaches towards the identification of species-specific

Histomonosis, caused by the protozoan parasite Histomonas meleagridis, is a major concern in turkey production due to the lack of licenced drugs and vaccines. Despite various studies on experimental vaccination, the duration of immunity of such a vaccine remains unclear. This study evaluated the long-term efficacy of an attenuated clonal monoxenic H. meleagridis culture in turkeys, focusing on its protective effects. Day-old turkeys were vaccinated orally using a frozen vaccine culture directly, without additional multiplication, and challenged 12 weeks later. The vaccine caused no adverse cli

Discovery of new protective malaria antigens will enable the development of novel vaccine formulations with potentially higher efficacy. While several high-throughput experimental approaches enable the identification of novel immunogens, none so far has been designed to selectively identify protective antigens. Here, we propose that sieve analysis conducted on the whole genome (SAWG) can be used specifically for this purpose. We review available medium- to high-throughput methods for antigen identification and contextualize the need for the identification of protective antigens. We then provid

Tritrichomonas foetus is a protozoan parasite responsible for bovine trichomonosis, a venereal disease that causes reproductive failure in cows while leaving bulls asymptomatic. Currently, no vaccine is available in Australia. This study aimed to develop and assess the safety, efficacy, and immunogenicity of a locally sourced T. foetus whole-cell killed vaccine in young bulls. The TfOz5 Queensland isolate was used as the vaccine strain, and the TfOz-N36 Northern Territory isolate served as the challenge strain. The heat-inactivated vaccine, adjuvanted with Montanide ISA 61 VG, was administered

Attenuated Babesia bigemina were multiplied in vivo and in vitro to vaccinate two groups (Groups 2 and 3) of Holstein Friesian heifers. Another group (Group 1) of heifers was vaccinated twice with purified soluble antigens obtained from the supernatant of in vitro culture combined with saponin. All these heifers plus controls (Group 4) were inoculated with heterologous pathogenic B. bigemina 5 months later. Heifers vaccinated with live organisms (Group 2 and 3) were able to stand the challenge without specific treatment whereas the opposite occurred in heifers vaccinated with soluble antigens

The mRNA vaccine platform should help research on East Coast fever subunit vaccines, but it remains unexplored. A theoretical strength of this platform, namely its capacity to prime cytotoxic T lymphocyte responses, is appealing as these, rather than antibodies, are the major mediators of immunity induced by a live parasite-based vaccine. Here, I highlight knowledge on functionally relevant bovine adaptive cellular and antibody immune responses to Theileria parva and antigens targeted by them that could help to assess this vaccine platform, and in the design of a broad-spectrum subunit vaccine

Leishmaniases, caused by parasites belonging to Leishmania spp, constitute a vast variety of diseases, from cutaneous lesions (CL) to visceral leishmaniasis (VL). If untreated, leishmaniases can be fatal, and affect 12 million people in nearly 90 countries, presenting a worldwide public-health problem. Most diagnostic tools are not suitable for use in field conditions. There is no satisfactory chemotherapy for CL; chemotherapy for VL is efficient in most immunocompetent people, but not in immunocompromised individuals, and is toxic and costly; and chemotherapy-resistant leishmania strains have

It has been known for several decades that the soluble parasite antigen (SPA) of several Babesia species can be used as a vaccine against the clinical manifestations of babesiosis. Originally observed in the plasma of infected animals, SPA can also be recovered from the supernatants of in vitro cultures of these parasites. Variable success has been reported for vaccines against the bovine and canine Babesia parasites, which seems to be related to antigenic diversity within Babesia species. In this article, an overview is presented of the development of such vaccines for dogs, and additional re

Cryptosporidium spp. is a zoonotic intracellular protozoan and a significant cause of diarrhoea in humans and animals worldwide. This parasite can cause high morbidity in immunocompromised people and children in developing countries, livestock being the main reservoir. This study was aimed at performing preliminary tests on Swiss albino weaned mice (ICR) to evaluate the humoral immune response induced against peptides derived from Cryptosporidium parvum CP15 (15 kDa sporozoite surface antigen) and CSL (circumsporozoite-like antigen) proteins. Peptides were identified and characterised using bi

BACKGROUND: Chicken coccidiosis is a protozoan disease that leads to considerable economic losses in the poultry industry. Live oocyst vaccination is currently the most effective measure for the prevention of coccidiosis. However, it provides limited protection with several drawbacks, such as poor immunological protection and potential reversion to virulence. Therefore, the development of effective and safe vaccines against chicken coccidiosis is still urgently needed.
METHODS: In this study, a novel oral vaccine against Eimeria tenella was developed by constructing a recombinant Lactobacillu

At present, there is not any available accepted vaccine for prevention of Toxoplasma gondii (T. gondii) in human and animals. We conducted literature search through English (Google Scholar, PubMed, Science Direct, Scopus, EBSCO, ISI Web of Science) scientific paper databases to find the best vaccine candidates against toxoplasmosis among T. gondii antigens. Articles with information on infective stage, pathogenicity, immunogenicity and characterization of antigens were selected. We considered that the ideal and significant vaccines should include different antigens and been expressed in all in

Visceral leishmaniasis is a zoonosis commonly caused in Brazil by the parasite Leishmania infantum. This protozoan parasite can infect several species of mammals, with dogs being the main reservoir in urban areas. Several methods are used to prevent the disease, including collars impregnated with 4% deltamethrin to prevent contact between the sandfly and the animal, and vaccines. Vaccines aim to stimulate an immune response that can effectively fight the parasite, with the Th1 immune response being the most desired. There are several research groups around the world dedicated to testing new im

In developing countries, cryptosporidiosis causes moderate-to-severe diarrhea and kills thousands of infants and toddlers annually. Drinking and recreational water contaminated with Cryptosporidium spp. oocysts has led to waterborne outbreaks in developed countries. A competent immune system is necessary to clear this parasitic infection. A better understanding of the immune responses required to prevent or limit infection by this protozoan parasite is the cornerstone of development of an effective vaccine. In this light, lessons learned from previously developed vaccines against Cryptosporidi

Toxoplasma gondii is a cosmopolitan protozoan parasite that infects a wide range of mammal and bird species. Common infection leads to high economic (e.g., abortions in sheep) and human (e.g., congenital toxoplasmosis or neurotoxoplasmosis in humans) losses. With one exception (Toxovax for sheep), there are no vaccines to prevent human or animal toxoplasmosis. The paper presents the current state and challenges in the development of a vaccine against toxoplasmosis, designed for farm animals either bred for consumption or commonly kept on farms and involved in parasite transmission. So far, the

PURPOSE: Antigens expressed by Toxoplasma gondii (T. gondii) during its life cycle trigger various immune responses in the host. Recently, toxoplasma vaccine research focused on T. gondii surface antigen 1 (SAG1) and Rhoptry Protein 18 (ROP18) to establish a safe and efficacious DNA vaccine.
METHOD: We constructed two eukaryotic expression plasmids: p3 × FLAG-Myc-CMV™-24-SAG1 and p3 × FLAG-Myc-CMV™-24-ROP18. BALB/c mice were randomly divided into six groups and immunized with these DNA vaccines either separately or in combination. The combination vaccine was administered at either the full do

Buffalo babesiosis represents a major problem for the livestock industry in China. The pathogen of this disease was isolated and identified as Babesia orientalis and subsequently propagated in vitro, using the microaerophilus stationary phase culture system. The aim of this study was to determine the efficacy of exoantigens derived from B. orientalis cultures to induce a protective immunity against challenge exposure to virulent organisms in laboratory and field. The results showed that exoantigens of B. orientalis could induce a high degree of protection against challenge with a virulent B. o

The apicomplexan parasites are globally considered as the major cause of numerous infectious diseases in humans and animals. Apicomplexan parasites include Plasmodium, Toxoplasma gondii, Cryptosporidium, Eimeria and Babesia. The rise in the drug resistance have made the traditional control measures, such as chemotherapy and vector management, inadequate against them. These are the intracellular infectious agent and possess complex life cycles, antigenic variability, and immune evasion abilities. These different abilities hinder the development of vaccines against them. Hence, there is urgent n
