Synergetic Protein Factors That Improve rhGM-CSF Absorption via an Oral Route Exist in Silkworm Pupae.

Recent studies have demonstrated that recombinant human granulocyte macrophage colony-stimulating factor (rhGM-CSF) produced by the silkworm pupae bioreactor is absorbed into blood through oral administration and functions as an active cytokine. The aim of this study was to further examine and identify synergetic protein factors in silkworm pupae that improve rhGM-CSF absorption via an oral route. The concentrations of rhGM-CSF in serum were evaluated in mice after oral administration of rhGM-CSF using different chemical compositions of silkworm pupae as pharmaceutical excipients. The experime

Nutritional and Functional Enhancement of Chinese Steamed Bread Through Incorporation of Acheta domesticus and Antheraea pernyi Pupae Powders.

The study intended to assess the impacts of partially replacing wheat flour with Acheta domesticus powder (AD) and Antheraea pernyi pupae powder (AP) at 5%, 10%, and 15% levels on the proximate composition, color properties, texture profile, antioxidant capacity, starch digestibility, and flavor characteristics of Chinese steamed bread (CSB). The addition of AP and AD notably increased the levels of protein, fat, and ash and also led to an elevated chewiness and hardness. Most importantly, compared to the control, AP- and AD-fortified CSB exhibited a significantly low estimated glycemic index

Immunomodulatory Effects of L-Arginine-Modified Silkworm Pupae Protein Enteral Nutrition on Murine Intestinal Morphology and Immunity.

L-arginine, a semi-essential amino acid, is well-documented for its role in nitric oxide (NO) synthesis, its anti-inflammatory properties, and its modulation of immune responses. Studies suggest it may improve gut barrier function and reduce inflammation in conditions such as colitis or sepsis. However, its specific efficacy in diabetic enteropathy (a complication of diabetes involving intestinal dysfunction, inflammation, and neuropathy) is less studied. To verify whether L-arginine and silkworm pupae components have a role in the treatment of diabetic enteropathy via the regulation of other

Should I dive or should I float? Behavioural plasticity of Aedes mariae pupae under predation threat.

BACKGROUND: The pupal stage in holometabolous insects is a critical transition between larval and adult forms, during which feeding ceases and survival depends on stored energy reserves. Mosquito pupae exhibit active diving behaviour in response to threats, which is energetically costly due to their positive buoyancy. Whether pupae are able to adjust diving behaviour according to environmental conditions, balancing predator avoidance and energy expenditure, remains poorly understood. Here, we investigated how water salinity affects the diving behaviour of Aedes mariae pupae, a species inhabiti

Purification and characterization of immunogenic recombinant virus-like particles of porcine circovirus type 2 expressed in silkworm pupae.

Porcine circovirus type 2 (PCV2) is a primary causative agent of postweaningmultisystemic wasting syndrome (PMWS), which has a significant economic impact on the swine industry. The capsid protein (Cap) encoded by ORF2 of the viral genome has been used effectively as a vaccine against PCV2 infection. The Cap protein can spontaneously assemble into virus-like particles (VLPs) that are safe and highly immunogenic for vaccine applications. Several expression systems, including bacteria, yeast and insect cells, have been utilized to produce PCV2 VLPs. However, in some cases, the recombinant Cap (r

Graphitized Porous Carbon for Rapid Screening of Angiotensin-Converting Enzyme Inhibitory Peptide GAMVVH from Silkworm Pupa Protein and Molecular Insight into Inhibition Mechanism.

A novel hydrophobic hexapeptide with high angiotensin-converting enzyme (ACE) inhibitory activity was screened from silkworm pupa protein (SPP) hydrolysate via graphitized porous carbon and reverse-phase high-performance liquid chromatography methods. Graphitized porous carbon derived from dopamine, possessing high surface area and high graphitic carbon, was used to rapidly screen and enrich hydrophobic peptides from SPP hydrolysate. The ACE inhibition pattern and mechanism of the purified peptide were also systematically studied by the classic Lineweaver-Burk model and by molecular docking/dy

Model updating strategy study about sex identification of silkworm pupae using transfer learning and NIR spectroscopy.

This paper proposes a novel model updating strategy named SilkwormNet for the first time to address the sex discrimination problem of silkworm pupae with new species. SilkwormNet integrates a ResNet block, a multi-head attention mechanism, and a Schedule-Free optimization strategy. Initially, the preprocessed spectra from one species were input into SilkwormNet to establish an optimal primary model. Then, the feature extraction layers and classification head remained unfrozen and the optimal weight parameters from the basic model were applied for model updating to identify the new species. Fin

A knockout screen of genes expressed specifically in Ae. aegypti pupae reveals a critical role for stretchin in mosquito flight.

Aedes aegypti is a critical vector for transmitting Zika, dengue, chikungunya, and yellow fever viruses to humans. Genetic strategies to limit mosquito survival based upon sex distortion or disruption of development may be valuable new tools to control Ae. aegypti populations. We identified six genes with expression limited to pupal development; osi8 and osi11 (Osiris protein family), CPRs and CPF (cuticle protein family), and stretchin (a muscle protein). Heritable CRISPR/Cas9-mediated gene knockout of these genes did not reveal any defects in pupal development. However, stretchin-null mutati

Antipredatory Responses of Mosquito Pupae to Non-Lethal Predation Threat-Behavioral Plasticity Across Life-History Stages.

Antipredatory behavioral responses tend to be energetically expensive, and prey species thus need to resolve trade-offs between these behaviors and other activities such as foraging and mating. While these trade-offs have been well-studied across taxa, less is known about how costs and benefits vary in different life-history contexts, and associated consequences. To address this question, we compared responses of the yellow fever mosquito (Aedes aegypti [Diptera: Culicidae]) to predation threat from guppy (Poecilia reticulata [Cyprinodontiformes: Poeciliidae]) across two life-history stages-la

Differentiation of Parasitized and Non-parasitized Anastrepha fraterculus (Wiedemann, 1830) (Diptera: Tephritidae) Pupae Based on Hyperspectral Reflectance Profiles.

Biological control programs for fruit flies rely on the mass rearing and release of parasitoids to suppress pest populations. The success of this strategy depends on effective quality control, including the assessment of parasitism and insect survival. Conventional methods, such as dissection, Polymerase Chain Reaction (PCR), and adult emergence, are destructive, labor-intensive, and time-consuming. Hyperspectral imaging offers a rapid and non-destructive alternative for evaluating parasitism. Therefore, this study aimed to discriminate Anastrepha fraterculus (Wiedemann, 1830) (Diptera: Tephri

Metabolic depression drives discontinuous gas exchange in diapause pupae of Sericinus montelus (Lepidoptera: Papilionidae): a seasonal adaptation strategy.

Adaptive shifts in respiratory patterns during insect diapause represent a key strategy for coping with seasonal adversity, yet the underlying regulatory mechanisms remain poorly understood. Using the swallowtail butterfly Sericinus montelus Gray as a model, this study investigated the regulatory roles of diapause and temperature on respiratory patterns by comparing gas exchange patterns and metabolic characteristics of pupae under different photoperiods and temperature conditions. The results demonstrated that the metabolic rate of diapause-destined pupae was significantly lower than that of

Toxicity of the essential oil from Thymus serpyllum and thymol to larvae and pupae of the housefly Musca domestica L. (Diptera: Muscidae).

The essential oil (EO) of Thymus serpyllum and thymol were evaluated for their insecticidal activity against the housefly (Musca domestica) larvae and pupae. Contact toxicity and fumigation bioassays were used. Chemical composition analysis of T. serpyllum EO by gas chromatographic mass spectrometry (GC-MS) revealed that thymol (41.6%), p-cymene (21.9%), and γ-terpinene (19.2%) were the major components. For larval assays, the LC50 value of T. serpyllum EO was 0.4 μl/cm2 for contact toxicity and 20.9 μl/l for fumigation toxicity. For thymol, the contact toxicity LC50 value was 0.035 μl/cm2 and

High yield production of norovirus GII.4 virus-like particles using silkworm pupae and evaluation of their protective immunogenicity.

Noroviruses (NoVs) are one of the major causes of acute viral gastroenteritis in humans. Virus-like particles (VLPs) without genomes that mimic the capsid structure of viruses are promising vaccine candidates for the prevention of NoVs infection. To produce large amounts of recombinant protein, including VLPs, the silkworm-expression vector system (silkworm-BEVS) is an efficient and powerful tool. In this study, we constructed a recombinant baculovirus that expresses VP1 protein, the major structural protein of NoV GII.4. Expression analysis showed that the baculovirus-infected silkworm pupae

Determining the Sterilization Doses under Hypoxia for the Novel Black Pupae Genetic Sexing Strain of Anastrepha fraterculus (Diptera, Tephritidae).

A common strategy used to maintain sterile fly quality without sacrificing sterility is to irradiate the insects under an oxygen-reduced atmosphere. So far, sterilizing doses for the South American fruit fly Anastrepha fraterculus have only been determined under normoxia. Our study reports for the first time the dose-sterility response under hypoxia for two different A. fraterculus strains. The pupae were derived from a bisexual strain (a Brazilian-1 population) and a recently developed genetic sexing strain (GSS-89). Two hours prior to irradiation, pupae were transferred to sealed glass bottl

An alternative solution for α-linolenic acid supplements: in vitro digestive properties of silkworm pupae oil in a pH-stat system.

α-Linolenic acid (ALA) is recognised to have a regulatory effect on cardiovascular diseases. Due to the low bioavailability of linseed oil (LINO), which is the most common ALA supplement, it is necessary to find a replacement for ALA supplements that is more easily accepted by the human body. The content of ALA in silkworm pupae oil (SPO) is 32.60 ± 0.67%, and SPO can be substituted as a dietary lipid to meet the demand of the human body. In the present study, a pH-stat system was used to investigate the release degree of free fatty acids (FFAs) from SPO and construct a first-order kinetic mod

Targeted allergen suppression in edible insects: Caffeic acid phenethyl ester-induced conformational remodeling of honeybee pupa profilin for dual-mode allergy prevention and antioxidant amplification.

Edible insects, exemplified by honeybee pupa, represent a sustainable protein source but are associated with undercharacterized allergenic risks. This study innovatively addresses this gap by targeting honeybee pupa-derived Profilin, a cross-reactive pan-allergen, through conjugation with caffeic acid phenethyl ester (CAPE), a bioactive polyphenol derived from propolis. Structural and biochemical analyses revealed that CAPE-induced covalent polymerization and conformational remodeling (characterized by a shift from α-helix dominance to β-sheet dominance, accompanied by tertiary structural pert
