CA9 Silencing Promotes Mitochondrial Biogenesis, Increases Putrescine Toxicity and Decreases Cell Motility to Suppress ccRCC Progression.

Carbonic anhydrase IX (CA9), a pH-regulating transmembrane protein, is highly expressed in solid tumors, and particularly in clear cell renal cell carcinoma (ccRCC). The catalytic mechanisms of CA9 are well defined, but its roles in mediating cell migration/invasion and survival in ccRCC remain to be determined. Here, we confirmed that the mRNA expression of CA9 in ccRCC was significantly higher than that in para-carcinoma tissues from analysis of the datasets in The Cancer Genome Atlas. CA9 knockdown upregulated oxidative phosphorylation-associated proteins and increased mitochondrial biogene

Ammonium mitigates Cd toxicity in rice (Oryza sativa) via putrescine-dependent alterations of cell wall composition.

In plants, different forms of nitrogen (NO3- or NH4+) affect nutrient uptake and environmental stress responses. In the present study, we tested whether NO3- and NH4+ affect the ability of rice (Oryza sativa) to tolerate the toxic heavy metal cadmium (Cd). Compared with NO3-, NH4+ treatment significantly increased chlorophyll contents and reduced Cd2+ levels in rice cultivars Nipponbare (japonica) and Kasalath (indica) grown in 0.2 mM Cd2+. NH4+ significantly reduced the pectin and hemicellulose contents and inhibited the pectin methylesterase (PME) activity in rice roots, thereby reducing the

Therapeutic benefits of putrescine-modified catalase in a transgenic mouse model of familial amyotrophic lateral sclerosis.

Dominant mutations in the copper/zinc superoxide dismutase (SOD1) gene have been observed in 15-20% of familial amyotrophic lateral sclerosis (FALS) cases. The mechanism by which SOD1 mutations result in motor neuron degeneration in FALS mice partly involves oxidative damage and an increased peroxidase activity of the mutant SOD1. A new therapeutic approach designed to eliminate the substrate of this peroxidase activity was examined in two lines of transgenic mice expressing the FALS-linked mutation glycine to alanine (G93A). We investigated the ability of putrescine-modified catalase (PUT-CAT

Dietary putrescine reduces the intestinal anticarcinogenic activity of sulindac in a murine model of familial adenomatous polyposis.

The nonsteroidal antiinflammatory drug sulindac displays chemopreventive activity in patients with familial adenomatous polyposis (FAP). Sulindac metabolites induce apoptosis in colon tumor cells, in part, by a polyamine-dependent mechanism that can be suppressed with exogenous putrescine. To determine the relevance of this mechanism in animals, we treated Apc(Min/+) mice, a model of human FAP, with sulindac alone or in combination with dietary putrescine. Sulindac increased steady-state RNA levels and enzymatic activity of the polyamine catabolic enzyme spermidine/spermine N(1)-acetyltransfer

Ornithine decarboxylase activity and putrescine levels in reversible cerebral ischemia of Mongolian gerbils: effect of barbiturate.

Reversible cerebral ischemia was produced in anesthetized Mongolian gerbils by occluding both common carotid arteries. After 5 min of ischemia, brains were recirculated for 8 or 24 h. Treated animals received a single intraperitoneal injection of pentobarbital (50 mg/kg) immediately after the aneurysm clips were removed. At the end of the experiments, animals were reanesthetized and their brains frozen in situ. Tissue samples were taken from the cerebral cortex, lateral striatum, CA1 subfield of the hippocampus, thalamus, and cerebellum for measuring ornithine decarboxylase (ODC) activity and

De novo biosynthesis of putrescine from glucose using engineered Escherichia coli in a two-step process.

Putrescine, is a key monomer for the synthesis of high-performance polymer materials. However, its sustainable microbial production remains challenging in yield and efficiency. In this study, we developed a modular two-step biosynthesis process for the conversion of glucose to putrescine. In the first stage, an l-arginine-overproducing Escherichia coli ARGFM strain was constructed through metabolic engineering. Then, fermentation optimization in a 5 L fermenter achieved a titer of 97.6 g/L with a yield of 0.52 g/g, representing a 49% improvement over the initial process. Notably, by implementi

The Enterococcus-putrescine-hadh axis drives mitochondrial redox imbalance and endothelial barrier breakdown in aortic dissection.

Growing evidence indicates that gut microbiota-derived metabolites contribute to aortic dissection (AD), yet the molecular mechanisms linking microbial metabolism to endothelial barrier failure remain unclear. Untargeted metabolomics identified putrescine as a markedly elevated circulating metabolite in AD patients, and higher putrescine levels were associated with disease severity and adverse clinical characteristics. Targeted polyamine metabolomics further confirmed putrescine as the most prominently altered metabolite among the measured polyamine-related metabolites. Integrated microbiome-m

Regulation of putrescine export in lipopolysaccharide or IFN-gamma-activated murine monocytic-leukemic RAW 264 cells.

The regulation of putrescine/polyamine export out of the cell was investigated during activation of monocytic-leukemic RAW 264 cells with LPS and IFN-gamma. The RAW 264 cells exported putrescine constitutively at a significant rate into the culture medium. This export process appeared to be selective for putrescine in that only a small amount of other polyamines (spermidine and N1-acetylspermidine) was found in the culture medium. LPS and IFN-gamma alone and in combination markedly stimulated putrescine export and nitrite production throughout a 24-h period. The efflux of putrescine but not ni

Characterization of simian malarial parasite (Plasmodium knowlesi)-induced putrescine transport in rhesus monkey erythrocytes. A novel putrescine conjugate arrests in vitro growth of simian malarial parasite (Plasmodium knowlesi) and cures multidrug resistant murine malaria (Plasmodium yoelii) infection in vivo.

A stage-dependent increase in the level of putrescine, spermidine, and spermine during intraerythrocytic growth of Plasmodium knowlesi in rhesus monkey erythrocytes was observed. Further, intraerythrocytic P. knowlesi-induced putrescine influx was found in trophozoite stage-infected erythrocytes and process was time- and temperature-dependent and showed saturable kinetics. Characteristics of induced putrescine influx appears in infected erythrocytes to be close to the normal erythrocytes in terms of affinity of putrescine to the putrescine transporter (Km 34.6 +/- 3.8 microM as normal erythroc

Exogenous putrescine modulates variety-specific cadmium tolerance in wheat seedlings: synergistic roles of antioxidant defense and physiological homeostasis.

INTRODUCTION: Cadmium (Cd) contamination in farmland is a significant environmental issues affecting crop yields. Putrescine (Put), a polyamine compound, functions as a signaling molecule that actively mediates plant responses to environmental adversities. Wheat exhibits a distinctive propensity for translocating the heavy metal Cd into its seeds compared to other crops, which poses a critical environmental adaptation challenge that needs to be addressed in agricultural systems. METHODS: This study employed Changmai 4013 (Cd-tolerant) and Chang 6475 (Cd-sensitive) varieties as test materials

Putrescine protects hulless barley from damage due to UV-B stress via H2S- and H2O2-mediated signaling pathways.

In hulless barley, H 2 S mediated increases in H 2 O 2 induced by putrescine, and their interaction enhanced tolerance to UV-B by maintaining redox homeostasis and promoting the accumulation of UV-absorbing compounds. This study investigated the possible relationship between putrescence (Put), hydrogen sulfide (H2S) and hydrogen peroxide (H2O2) as well as the underlying mechanism of their interaction in reducing UV-B induced damage. UV-B radiation increased electrolyte leakage (EL) and the levels of malondialdehyde (MDA) and UV-absorbing compounds but reduced antioxidant enzyme activities and

Arginine and putrescine ameliorate heat stress-induced testicular inflammation and apoptosis via inhibiting endoplasmic reticulum stress in mice.

Heat stress can severely impair male reproductive performance. Endoplasmic reticulum stress (ERS) is recognized as an early cellular response to stress, which can subsequently induce inflammation and apoptosis, thereby reducing semen quality. In this study, arginine, putrescine, and 4-phenylbutyric acid (4-PBA; an ERS inhibitor) were administered by gavage to a heat-stressed ICR mouse model subjected to seven days of heat exposure, in order to evaluate their effects on testicular function and the underlying mechanisms. Supplementation with arginine, putrescine, or 4-PBA significantly ameliorat

Hanseniaspora uvarum FS35 degrade putrescine in wine through the direct oxidative deamination pathway of copper amine oxidase 1.

Putrescine is abundant in wine and have toxicological risks for the health of consumers. Certain microbes with oxidative deamination activity are considered to be one of the most effective ways to degrade putrescine. The characterization and possible mechanism of putrescine degradation by Hanseniaspora uvarum FS35 were studied in this work. Hanseniaspora uvarum FS35 was selected from 111 yeast strains by UPLC analysis and exhibited the ability to eliminate > 44.5 mg/L of putrescine after 12 h of culture. Transcriptome analysis showed that by adding putrescine as a nitrogen source, the gene exp

Changes in brain putrescine concentration associated with nonconvulsant behavioral patterns induced by systemic N-methyl-D-aspartate injection.

The relationship between the behavioral effects and motor activity induced by N-methyl-D-aspartate (NMDA) (150 mg/kg, ip) and brain polyamine concentration was studied in male Wistar rats. Motor activity was evaluated by an automated subtraction analysis system to measure the duration and vigor of any kind of movement. The behavioral modifications exhibited by the nonconvulsant NMDA-treated rats were evaluated according to the composition and sequence of behavioral components as: hypoactivity (pattern A), partially stereotyped activity (pattern B), and generalized stereotyped activity (pattern

Insights into the facilitation effects of putrescine in mediating the multi-pathway metabolism of bentazone to mitigate environmental risks to rice production.

BACKGROUND: Putrescine (PUT), the original by-product of polyamine biosynthesis, plays multiple roles in plant growth and defense against osmotic and environmental stresses. It may also aid in decreasing herbicide phytotoxicity, but this remains unconfirmed. RESULTS: Compared with rice treated with 2.3 mg L-1 bentazone (BNTZ) alone, co-application of 150 mg L-1 PUT restored rice morphological performance, increasing shoot and root elongation by 7.1% and 8.3%, and elevating shoot and root dry weight by 19.4% and 22.2%, respectively. Transcriptome enrichment analysis revealed a large set of sig

Interactive effects of silica nanoparticles and putrescine on morphophysiological and biochemical performance in stock (Matthiola incana L. 'Pilaster White').

Stock (Matthiola incana L. 'Pilaster White') is an economically important ornamental species whose commercial value depends largely on vigorous vegetative growth, high floral quality, and extended vase life. Silica nanoparticles (SiO₂ NPs) and putrescine have individually been reported to improve plant growth and physiological performance through different mechanisms; however, information regarding their interactive effects in stock remains limited. Therefore, this study evaluated the individual and interactive effects of foliar-applied SiO₂ NPs and putrescine on the morphophysiological, bioch

Identification of cisapride as new inhibitor of putrescine uptake in Trypanosoma cruzi by combined ligand- and structure-based virtual screening.

Nowadays, the pharmacological therapy for the treatment of Chagas disease is based on two old drugs, benznidazole and nifurtimox, which have restricted efficacy against the chronic phase of the illness. To overcome the lack of efficacy of the traditional drugs (and their considerable toxicity), new molecular targets have been studied as starting points to the discovery of new antichagasic compounds. Among them, polyamine transporter TcPAT12 (also known as TcPOT1.1) represents an interesting macromolecule, since polyamines are essential for Trypanosoma cruzi, the parasite that causes the illnes
