Quinolones sensitize gram-negative bacteria to antimicrobial peptides.

The treatment of infections caused by bacteria resistant to the vast majority of antibiotics is a challenge worldwide. Antimicrobial peptides (APs) make up the front line of defense in those areas exposed to microorganisms, and there is intensive research to explore their use as new antibacterial agents. On the other hand, it is known that subinhibitory concentrations of antibiotics affect the expression of numerous bacterial traits. In this work we evaluated whether treatment of bacteria with subinhibitory concentrations of quinolones may alter the sensitivity to APs. A 1-h treatment of Klebs

Bactericidal mechanism of gatifloxacin compared with other quinolones.

The quinolones differ in their mechanisms of bacterial killing. The rate of bacterial killing by quinolones can be influenced by the addition of bacterial protein or RNA synthesis inhibitors, and the growth phase of the bacterium. In this study, we compared the killing activities of gatifloxacin, trovafloxacin, ciprofloxacin and norfloxacin against staphylococci, pneumococci and Escherichia coli. Gatifloxacin killing of these organisms occurred regardless of the metabolic state of the microbes. Unlike the comparator quinolones, gatifloxacin killing was not influenced by the addition of bacteri

The place of quinolones in everyday clinical practice.

The new fluoroquinolones have many attributes and relatively few drawbacks. As a result, they have been rapidly adopted for the treatment of serious bacterial infections. The currently available fluoroquinolones are particularly effective for the treatment of urinary tract infections, bacterial diarrhea, enteric fever, gonorrhea, chancroid, skin and soft tissue infections, acute and chronic gram-negative osteomyelitis, gram-negative pneumonias, serious infections due to a variety of gram-negative bacilli, and in the eradication of the meningococcal carrier state. As we learn more about the pro

Long-term use of quinolones and their safety.

Although short courses of 4-quinolones are effective in routine infections, longer courses are necessary for chronic, deep-seated sepsis. Oral 4-quinolones exhibit efficacy equal to that of traditional parenteral regimens against osteomyelitis caused by gram-negative pathogens and have proved successful against chronic prostatitis and suppurative otorhinologic infections. The efficacy of these agents in the prophylaxis of urinary tract infection, travelers' diarrhea, and infections in neutropenic patients suggests other indications for potential widespread, long-term use. It is therefore impor

Quinolones in the treatment of sexually transmitted diseases.

Many of the new quinolone-carboxylic acids possess in vitro activity against a variety of sexually transmitted pathogens. Neisseria gonorrhoeae and Hemophilus ducreyi are particularly susceptible to those antimicrobial agents while the in vitro sensitivities of Chlamydia trachomatis and genital mycoplasmas vary widely amongst the quinolones and range from poor to modest. Clinically, single dose therapy with many of the quinolones has been highly effective in curing gonorrhea and appears to be promising in the management of chancroid. In contrast, however, the quinolones (even when used in mult

Quinolone Resistance in Gallibacterium anatis Determined by Mutations in Quinolone Resistance-Determining Region.

Control of the important pathogen, Gallibacterium anatis, which causes salpingitis and peritonitis in poultry, relies on treatment using antimicrobial compounds. Among these, quinolones and fluoroquinolones have been used extensively, leading to a rise in the prevalence of resistant strains. The molecular mechanisms leading to quinolone resistance, however, have not previously been described for G. anatis, which is the aim of this study. The present study combines phenotypic antimicrobial resistance data with genomic sequence data from a collection of G. anatis strains isolated from avian host

Quinolone Consumption by Mothers Increases Their Children's Risk of Acquiring Quinolone-Resistant Bacteriuria.

BACKGROUND: Quinolone resistance has been documented in the pediatric population, although their use is limited in children. This study investigated the effect of maternal quinolone use on gram-negative bacterial resistance to quinolones in their offspring. METHODS: We conducted a population-based, unmatched case-control study during 2010-2017. Cases were all children aged 0.5-17 years with community acquired, gram-negative quinolone-resistant bacteriuria. Controls were similar children with quinolone-sensitive bacteriuria. Only the first positive urine cultures for each child were included.

This study provides an insight into the prevalence of (fluoro)quinolones (FQs) and their specific quinolone qnrS resistance gene in the Avon river catchment area receiving treated wastewater from 5 wastewater treatment plants (WWTPs), serving 1.5 million people and accounting for 75% of inhabitants living in the catchment area in the South West of England.. Ofloxacin, ciprofloxacin, nalidixic acid and norfloxacin were found to be ubiquitous with daily loads reaching a few hundred g/day in wastewater influent and tens of g/day in receiving waters. This was in contrast to other FQs analysed: flu

Benzopyrone-mediated quinolones as potential multitargeting antibacterial agents.

A new type of benzopyrone-mediated quinolones (BMQs) was rationally designed and efficiently synthesized as novel potential antibacterial molecules to overcome the global increasingly serious drug resistance. Some synthesized BMQs effectively suppressed the growth of the tested strains, outperforming clinical drugs. Notably, ethylidene-derived BMQ 17a exhibited superior antibacterial potential with low MICs of 0.5-2 μg/mL to clinical drugs norfloxacin, it not only displayed rapid bactericidal performance and inhibited bacterial biofilm formation, but also showed low toxicity toward human red b

Phototoxicity in quinolones: comparison of ciprofloxacin and grepafloxacin.

Skin photosensitizing reactions have been reported during treatment with fluoroquinolone antibiotics. The incidence and severity of such reactions, however, appear to differ between agents. The photosensitizing effect of grepafloxacin 400 and 600 mg once daily was compared with that of ciprofloxacin 500 mg bd and placebo in a double-blind trial involving 32 healthy subjects. Skin photosensitivity, expressed as the minimal erythemal dose (MED), was measured before treatment and towards the end of the 7 day treatment period. Grepafloxacin showed a mild photosensitizing effect comparable to that

Phototoxicity and photoallergenicity of quinolones in guinea pigs.

Clinical reports indicate that the fluoroquinolone group of antibiotics can induce cutaneous photosensitivity reactions. In the present study, phototoxicity and photoallergenicity of quinolones including nalidixic acid (NA) norfloxacin (NFLX), ofloxacin (OFLX), enoxacin (ENX), ciprofloxacin (CPFX), lomefloxacin (LFLX), and tosufloxacin (TFLX) were experimentally examined in an in vivo system using the guinea pig. Phototoxicity of all quinolones tested was demonstrated after a single, oral administration of the drugs and subsequent exposure to long-wave ultraviolet (UVA) at a dose of 30 J/cm2.

The importance of the quinolones in antibacterial therapy.

New quinolones have obtained a definite position in the treatment of certain sexually transmitted diseases, urinary tract infections, prostatitis, gastrointestinal infections, nosocomially acquired pulmonary infections with resistant organisms, pseudomonas infections in cystic fibrosis, and osteomyelitis. The role of the new quinolones in upper respiratory tract infections, acute or chronic bronchitis and community acquired pneumonia is far less established. Their role in selective decontamination of the gastrointestinal tract in neutropenic patients is under investigation. Future modification

Biofilm Formation and Plasmid-Mediated Quinolone Resistance Genes at Varying Quinolone Inhibitory Concentrations in Quinolone-Resistant Bacteria Superinfecting COVID-19 Inpatients.

The likelihood of antimicrobial failure in COVID-19 patients with bacterial superinfection arises from both phenotypic (biofilms) and genotypic mechanisms. This cross-sectional study aimed to determine the inhibitory concentrations of quinolones-nalidixic acid, norfloxacin, ciprofloxacin, ofloxacin, and levofloxacin-in biofilm formers (minimum biofilm inhibitory concentration [MBIC]) and nonformers (minimum inhibitory concentration [MIC]) and correlate inhibitory concentrations with plasmid-mediated quinolone resistance (PMQR) genes in quinolone-resistant bacteria isolated from COVID-19 inpati

High-Level Quinolone-Resistant Haemophilus haemolyticus in Pediatric Patient with No History of Quinolone Exposure.

The prevalence of antimicrobial resistance among Haemophilus spp. is a critical concern, but high-level quinolone-resistant strains had not been isolated from children. We isolated high-level quinolone-resistant H. haemolyticus from the suction sputum of a 9-year-old patient. The patient had received home medical care with mechanical ventilation for 2 years and had not been exposed to any quinolones for >3 years. The H. haemolyticus strain we isolated, 2019-19, shared biochemical features with H. influenzae. However, whole-genome analysis found this strain was closer to H. haemolyticus. Phylog

Quinolone Antibiotics: Resistance and Therapy.

The clinical application of quinolone antibiotics is particularly extensive. In addition to their high efficiency in infectious diseases, the treatment process brings multiple hidden dangers or side effects. In this regard, drug resistance becomes a major challenge and is almost unavoidable in the clinical application of quinolones. Both genetic and phenotypic variations contribute to bacterial survival resistance under antibiotic therapy. This review is focusing on the drug discovery history, compound structure, and bactericidal mechanism of quinolone antibiotics. Recent studies bring a more
