First genetic characterisation and phylogenetic position of Aplectana hylambatis (Nematoda: Cosmocercidae), infecting Pithecopus azureus (Anura: Hylidae) in the Brazilian Pantanal.

Aplectana hylambatis (Baylis, 1927) is a widespread nematode in South America that infects different species of anurans from different families. Although the morphology of this species shows intraspecific variations and is well studied, A. hylambatis does not have genetic characterisation. Therefore, the present study aimed to provide the first genetic characterisation of this parasite, based on 18S and 28S rDNA sequences, as well as to evaluate its phylogenetic position in the family Cosmocercidae. The present specimens showed no major morphological variations and were assigned to A. hylambat

New species from the cradle of biodiversity inventories: cryptic diversity within Lamellariinae in the European Atlantic and Mediterranean (Gastropoda: Velutinidae).

The Mediterranean, despite a long tradition of naturalist observation dating back to Aristotle, is still home to undisclosed cryptic species complexes. The gastropod subfamily Lamellariinae d'Orbigny, 1841 (Velutinidae Gray, 1840) is composed of benthic sea slug-like species with a vestigial internal shell covered by the mantle. The high morphological plasticity of this group hampers species delimitation and identification. In the Mediterranean Sea, two - considered well-known - species are present: Lamellaria latens (O. F. Müller, 1776) and Marsenia perspicua (Linnaeus, 1758). This study asse

Multigene phylogenetic analysis reveals non-monophyly of Anisakis s.l. and Pseudoterranova (Nematoda: Anisakidae).

The nematode genera Anisakis s.l. and Pseudoterranova (Anisakidae) include causative agents of anisakiasis and pseudoterranovosis, parasitic diseases resulting from eating undercooked or raw fish or squid. Species in both genera have thus attracted considerable attention especially in public health and taxonomic studies. The phylogenetic relationships of these genera within the subfamily Anisakinae, however, remain to be investigated with dense taxonomic sampling. In this study, we collected an anisakid third-stage larva, and identified it morphologically and molecularly as Pseudoterranova cet

Leidynemella shahi n. sp. (Nematoda: Thelastomatidae) from wood-eating cockroaches (Insecta: Blaberidae) in the Yunnan Province, China.

Leidynemella shahi n. sp. is described based on material from Panesthia angustipennis cognata Bey-Bienko and Salganea raggei Roth in the Yunnan Province, China. The new species is very similar to L. fusiformis Cobb in Chitwood & Chitwood, 1934, but differs from the latter in the position of the nerve-ring and vulva, and in the length of the eggs. In addition, molecular analysis of partial 28S rDNA sequences also supported the distinct status of the new species.

Detection and molecular characteristics of Pyelosomum cochlear (Digenea: Pronocephalidae) in the urinary bladder of the green sea turtle (Chelonia mydas) in the Northwest Pacific Ocean.

The genus Pyelosomum consists of parasitic flukes occurring primarily in marine turtles; Pyelosomum cochlear Looss 1899 is the only species of this genus that parasitizes the urinary bladder. In this study, we detected flukes in the urinary bladders of 20 of 88 green sea turtles (Chelonia mydas) harvested in the Ogasawara Islands, in the Northwest Pacific Ocean. We identified the flukes as P. cochlear based on detailed morphological observations and comparisons of morphometric measurements of the species reported previously. Nucleotide sequences of nuclear ribosomal 18S and 28S regions and the

Joblinomyces undulatus, sp. nov., a novel anaerobic fungus from the feces of a Boer goat.

Members of the phylum Neocallimastigomycota, often referred to as anaerobic gut fungi (AGF), primarily inhabit the gut of herbivorous animals. To date, 25 AGF genera have been described, but only five harbor more than one species. Here, we report on the isolation and characterization of novel AGF isolates from the feces of a Boer goat. The isolates formed small, mostly round, dense colonies on agar roll tubes and produced flaky, loosely clumped white biomass in liquid medium. Microscopic features included monoflagellated spherical zoospores and monocentric filamentous thallus. Sporangia were s

First report of Sarcocystis bovifelis-like sporocysts in the intestine of the Canadian lynx (Lynx canadensis) in USA.

Sarcocystis bovifelis is a cattle parasite for which the domestic cat (Felis catus) is the only confirmed definitive host, and the role of wild felids in its transmission remains unknown. This study aimed to investigate the presence and identity of Sarcocystis species in the intestinal tissue of a dead Canada lynx (Lynx canadensis) from Minnesota, USA, to assess whether this wild felid could serve as a definitive host. Approximately 30 cm of terminal ileum was examined microscopically for sporocysts, and DNA from sporocyst-positive material was assayed using 18S rRNA and 28S rRNA gene markers.

Identification and characterization of the causative triatomine bugs of anaphylactic shock in Zhanjiang, China.

BACKGROUND: Two health concerns primarily related to triatomine bugs are transmission of Trypanosoma cruzi through infective feces, and allergic reactions induced by triatomine bites. In the Southwestern United States, reduviid bugs bites commonly cause insect allergy. In South China, four cases of anaphylactic shock have been reported after this bite exposure. To further classify the species of these bugs and confirm the sensitization of the triatomine saliva, we caught triatomine bugs from the region where the bites occurred and performed phylogenetic and immunohistochemical (IHC) analysis.

Optimizing preservation protocols to extract high-quality RNA from different tissues of echinoderms for next-generation sequencing.

Transcriptomic information provides fundamental insights into biological processes. Extraction of quality RNA is a challenging step, and preservation and extraction protocols need to be adjusted in many cases. Our objectives were to optimize preservation protocols for isolation of high-quality RNA from diverse echinoderm tissues and to compare the utility of parameters as absorbance ratios and RIN values to assess RNA quality. Three different tissues (gonad, oesophagus and coelomocytes) were selected from the sea urchin Arbacia lixula. Solid tissues were flash-frozen and stored at -80 °C until
