5-HT neurons of the medullary raphe contribute to respiratory control in toads.

Air-breathing vertebrates undergo respiratory adjustments when faced with disturbances in the gas composition of the environment. In mammals, the medullary raphe nuclei are involved in the neuronal pathway that mediates the ventilatory responses to hypoxia and hypercarbia. We investigate whether the serotoninergic neurons of the medullary raphe nuclei of toads (Rhinella diptycha) play a functional role in respiratory control during resting conditions (room air), hypercarbia (5% CO2), and hypoxia (5% O2). The raphe nuclei were located and identified based on the location of the serotoninergic n

Clinical correlates of raphe serotonergic dysfunction in early Parkinson's disease.

Post-mortem and neuroimaging studies suggest that the serotonergic system, which originates from the brainstem raphe nuclei, is disrupted in Parkinson's disease. This could contribute to the occurrence of non-motor symptoms and tremor, which are only partially explained by dopamine loss. However, the level of involvement of the serotonergic raphe nuclei in early Parkinson's disease is still debated. (123)I-FP-CIT single photon emission computed tomography is a marker of dopamine and serotonin transporter availability. While (123)I-FP-CIT binds primarily to dopamine transporters in the striatum

Nuclear organization of serotonergic neurons in the brainstems of a lar gibbon and a chimpanzee.

In the current study, we detail, through the analysis of immunohistochemically stained sections, the morphology and nuclear parcellation of the serotonergic neurons present in the brainstem of a lar gibbon and a chimpanzee. In general, the neuronal morphology and nuclear organization of the serotonergic system in the brains of these two species of apes follow that observed in a range of Eutherian mammals and are specifically very similar to that observed in other species of primates. In both of the apes studied, the serotonergic nuclei could be readily divided into two distinct groups, a rostr

Lateral habenula as a link between dopaminergic and serotonergic systems contributes to depressive symptoms in Parkinson's disease.

Degeneration of substantia nigra dopaminergic neurons is a key pathological change of Parkinson's disease (PD), and its motor consequences have been widely recognized. Recently, mood disorders associated with PD have begun to attract a great deal of interest, however, their pathogenesis remains unclear. PD is associated with not only degenerative changes in dopaminergic neurons in the substantia nigra but also changes in serotonergic neurons in the raphe nuclei. The abnormalities in central 5-hydroxytryptamine (5-HT) neurotransmission are thought to play a key role in the pathogenesis of depre

Serotonergic afferents to the rat olfactory bulb: I. Origins and laminar specificity of serotonergic inputs in the adult rat.

This is a study of the chemoanatomical organization of the projection from the raphe nuclei to the main olfactory bulb in the rat. A heavy projection from the dorsal and median raphe nuclei to the main olfactory bulb was shown by both retrograde and anterograde tracing techniques. Following injections of 1% wheat germ agglutinin-horseradish peroxidase (WGA-HRP) into the main olfactory bulb, up to 1300 neurons were retrogradely labeled in the dorsal and median raphe nuclei, a much larger number than has been suggested by previous studies. By combining 5-HT immunofluorescence with True blue retr

Innervation of the nucleus of the solitary tract and the dorsal vagal nucleus by thyrotropin-releasing hormone-containing raphe neurons.

The nucleus of the solitary tract and the dorsal vagal nucleus are richly innervated by thyrotropin-releasing hormone (TRH)-containing fibers arising from the caudal raphe nuclei. After transection of vertically oriented fibers by a horizontal knife-cut in the medulla oblongata, TRH-staining disappeared from the vagal nuclei while it increased in transected nerve fibers ventral to the knife-cut. TRH-containing cells are mainly located in the nucleus raphe pallidus and raphe obscurus. TRH-containing fibers run dorsally within the raphe and enter the dorsal vagal complex at its rostral tip. Then

Common and unique dysconnectivity profiles of dorsal and median raphe in Parkinson's disease.

The serotonergic (5-HT) system, which undergoes degeneration in Parkinson's disease (PD), is involved in the pathogenesis of motor and nonmotor symptoms. The dorsal raphe (DR) and median raphe (MR) nuclei are the main source of 5-HT neurons, however, brain connectivity changes in these two nuclei have not been delineated in PD. Here we used resting-state fMRI (rs-fMRI) to characterize functional connectivity profiles of DR and MR and further examine the associations between dysconnectivity of raphe nuclei and clinical phenotypes of PD. We found that DR and MR commonly hypo-connected with the s

Exercise prevents raphe nucleus mitochondrial overactivity in a rat depression model.

Monoamine deficit and mitochondrial dysfunction may underlie depression. Serotoninergic neurons from raphe nuclei project widely and may be involved in depression. This study used chronic unpredictable stress (CUS) in rats as a model of depression to assess the effects of CUS, exercise and fluoxetine on mitochondrial function and serotonin levels in the raphe nuclei. Rats were divided into 4 groups (6 per group): control (C); depression (D), CUS for 28days; depression+exercise (DE), treadmill exercises from days 11-28 of CUS; depression+fluoxetine (DF), fluoxetine (5mg/kg/d i.g.) from days 11

Modulatory effects of repeated psychophysical stress on masseter muscle nociception in the nucleus raphe magnus of rats.

Psychophysical stress can cause neural changes that increase nociception in the orofacial region, particularly the masseter muscle (MM). The nucleus raphe magnus (NRM), which is located in the brain stem, serves the crucial role of regulating nociception through descending modulatory pain control. However, it remains unclear if neural activities in the NRM are affected under psychophysical stress conditions. This study conducted experiments to assess (1) whether neural activity, indicated by Fos expression in an NRM that has experienced MM injury, is affected by the stress of repeated forced s

Distribution of striatal afferents from the mesencephalon in the cat.

In a series of cats, various quantities of horseradish peroxidase were injected in different parts of the striatum to map out the extent and the topography of the afferent projections from the mesencephalon. Following large injections in the head of the caudate nucleus and putamen, the majority of cells were retrogradely labeled in the substantia nigra pars compacta (SNc) and in its dorsal extension the retrorubral nucleus or pars dorsalis (SNd), confirming previous reports. Numerous HRP positive neurons were also present in the ventral tegmental area (VTA) and the rostral raphe nuclei, includ

Monoaminergic inputs to frog motoneurons: an anatomical study using fluorescence histochemical and silver degeneration techniques.

The monoaminergic input to spinal segment 10 of the frog, Rana pipiens, was investigated anatomically. Fluorescence histochemistry demonstrated a diffuse and overlapping distribution of catecholamine (CA) and indoleamine (IA) containing terminals within the spinal cord. Terminal fields containing high numbers of CA and IA varicosities included the marginal plexus and the anterior commissural region. IA terminals were also abundant in the dorsal portion of the dorsal horn, while CA terminals were found in high number in the ventral part of the dorsal horn. IA varicosities were regularly observe

Association between increased serotonin metabolism in rat brainstem nuclei and development of spontaneous hypertension.

Spontaneously hypertensive rats have increased brain stem serotonin turnover localized to the nuclei raphe magnus, raphe pallidus, tractus solitarius and reticularis lateralis. It occurs only in young animals coinciding with the development of hypertension. In the locus coeruleus, increased serotonin metabolism is present during both the early and the established phase of hypertension. Our results provide evidence for serotonergic overactivity in specific brainstem nuclei during the development of spontaneous hypertension in the rat.

Kainic acid injection into medullary raphe produces gastric lesions through the vagal system in rats.

There is little evidence of a role of nuclei in the brain stem, other than the dorsal motor nucleus of the vagus nerve in the medulla oblongata, in terms of central mechanisms for gastric ulcer formation. Recent reports suggest that the medullary raphe nuclei may be involved in the central regulation of gastric functions such as gastric acid secretion and motility. In the present study, we examined whether neurons in the nucleus raphe obscurus (NRO) played a significant role in the formation of gastric ulcers with the use of Sprague-Dawley rats. First, we determined whether excitation of the m

Cholinergic systems in the rat brain: IV. Descending projections of the pontomesencephalic tegmentum.

Descending projections from cholinergic neurons in the pedunculopontine and laterodorsal tegmental nuclei, collectively referred to as the pontomesencephalotegmental (PMT) cholinergic complex, were studied by use of the fluorescent retrograde tracers fluorogold, true blue, or Evans Blue in combination with choline acetyltransferase (ChAT) immunohistochemistry of acetylcholinesterase (AChE) pharmacohistochemistry. Pedunculopontine somata positive for ChAT or staining intensely for AChE were retrogradely labeled with fluorescent tracers following infusions into the motor nuclei of cranial nerves

Regional distribution and cellular expression of tryptophan hydroxylase messenger RNA in postmortem human brainstem and pineal gland.

The characterization and cellular localization of tryptophan hydroxylase mRNA in the human brainstem and pineal gland were investigated by using northern blot analysis and in situ hybridization histochemistry. Northern analysis of human pineal gland revealed the presence of two mRNA species that were absent in RNA isolated from human raphe. In situ hybridization experiments revealed very dense hybridization signal corresponding to tryptophan hydroxylase mRNA in cells throughout the pineal gland. In contrast, tryptophan hydroxylase mRNA was heterogeneously distributed in neurons in the dorsal a

The raphe nuclear organization and serotonergic system in the bat (Artibeus planirostris).

5-Hydroxytryptamine (5-HT), widely recognized as serotonin, is a multifunctional substance present across various body tissues, performing as a neurotransmitter within the nervous framework. Serotonergic neurons are predominantly localized within the raphe nuclei of the brainstem, rendering neuronal 5-HT a definitive marker for these nuclei. Research has substantiated serotonin's role in the modulation of thermoregulation, appetite, reproductive drive, circadian patterns, slumber, motoric activity, and cognitive processing. While the anatomical structure of serotonergic systems has undergone e

Changes in serotonergic neurons in the brain of pyrithiamine-induced acute thiamine-deficient mice.

We examined changes in 5-hydroxytriptamine (5-HT, serotonin) neurons in pyrithiamine-induced thiamine deficiency in mice immunohistochemically. Extensive decreases in the densities of 5-HT-immunoreactive fibers were detected in the lateral septal nucleus, the thalamus, the medial mammillary nucleus, the dorsal and the median raphe nuclei, the raphe obscurus nucleus, the tegmental area, the cerebellum and the vestibular nucleus, though only a small decrease was detected in the inferior colliculus. Most remarkably, degenerative winding fibers were detected between the deep mesencephalic nucleus

Robust and tissue-specific expression of TPH2 versus TPH1 in rat raphe and pineal gland.

BACKGROUND: Regulation of raphe serotonergic cells is fundamental to the prevailing hypothesis of major depression pathophysiology. Tryptophan hydroxylase (TPH) is the rate-limiting enzyme in serotonin biosynthesis, but brainstem TPH mRNA expression has been difficult to measure and study. Recently, a novel paralog of TPH, TPH2 (or neuronal TPH), was described, but its anatomic expression is unknown. METHODS: In situ hybridization histochemical survey was conducted across Sprague-Dawley rat brain for TPH1 and TPH2 mRNA. Semiquantitative techniques were used to estimate relative mRNA levels in

Longitudinal Trajectory of Dopamine and Serotonin Transporters in Parkinson Disease.

Parkinson disease (PD) is a multisystem disorder marked by progressive dopaminergic neuronal degeneration in the substantia nigra, as well as nondopaminergic systems. Our aim was to investigate longitudinal changes in N-(3-[18F]fluoropropyl-2β-carbomethoxy-3β-(4-iodophenyl)nortropane (18F-FP-CIT) binding at the putamen, substantia nigra, and raphe nuclei in PD. Methods: This retrospective cohort study enrolled 127 patients with PD, who underwent 18F-FP-CIT PET scans twice or more, and 71 age- and sex-matched healthy controls. A temporal trajectory model was created to estimate the longitudinal
