[Development of a real-time polymerase chain reaction method for the identification of Candida species].

Candida species are one of the major causes of nosocomial infections and are the fourth most common agent involved in bloodstream infections. The impact of non-albicans Candida species is increasing, however C.albicans is still the most common species. Since the antifungal susceptibility pattern among Candida spp. may be different, rapid diagnosis and identification of non-albicans Candida spp. are important for the determination of antifungal agents that will be used for treatment. The aim of the study was to describe a real-time polymerase chain reaction (Rt-PCR) assay that rapidly detects,

Development and validation of a real-time polymerase chain reaction for the detection of Haycocknema perplexum.

Haycocknema perplexum is a rare and emerging cause of parasitic myositis. Detection and surveillance are of growing importance given the increasing degree of climate conditions conducive to transmission. In this study, we developed a real-time polymerase chain reaction (PCR) method for direct detection of two genomic regions of H. perplexum. The real-time PCR assays, SSU-PCR and COX-1-PCR, targeted the small subunit of nuclear ribosomal RNA and cytochrome oxidase-1 genomic regions, respectively. The performance of the assays was assessed using a panel of H. perplexum samples, both fresh frozen

Foot-and-mouth disease virus detection on a handheld real-time polymerase chain reaction platform.

Foot-and-mouth disease (FMD) is a highly contagious disease of livestock that requires rapid control. Early detection is critical but transportation of samples to laboratory delays testing. Sensitive and specific field-deployable assays are therefore desirable. Real-time reverse transcription polymerase chain reaction (RRT-PCR) and RRT-loop-mediated isothermal amplification assays for FMDV on portable platforms have been described but none of these are handheld. In this report, we have evaluated a handheld Biomeme two3™ Real-Time PCR Thermocycler (two3) as a field-deployable platform for FMDV

Cost analysis of real-time polymerase chain reaction microbiological diagnosis in patients with septic shock.

Antibiotic treatment for septic shock is generally prescribed on an empirical basis using broad-spectrum antibiotics. Molecular diagnostic techniques can detect the presence of microbial DNA in blood within a few hours and facilitate early, targeted treatment. The aim of this study was to evaluate the economic impact of a real-time polymerase chain reaction technique, LightCycler SeptiFast (LSC), in patients with sepsis. A cost-minimisation study was carried out in patients admitted with a diagnosis of severe sepsis or septic shock to the intensive care unit of a university hospital. The stay

Quantitative real-time polymerase chain-reaction assay allows characterization of pneumocystis infection in immunocompetent mice.

Pneumocystis causes pneumonia in immunodeficient hosts but also likely causes infection in healthy hosts. To characterize infection in healthy mice, we developed and validated a real-time polymerase chain reaction assay for quantitation of Pneumocystis carinii f. sp. muris. In healthy mice exposed to Pneumocystis-infected animals, organisms were first detected at 2-3 weeks, peaked at 5-6 weeks, and were cleared by 7-9 weeks. The peak organism load in healthy animals was 2-3 logs lower than that in immunodeficient animals. This approach should facilitate studies of anti-Pneumocystis immune mech

Rapid enumeration of Listeria monocytogenes in artificially contaminated cabbage using real-time polymerase chain reaction.

A quantitative real-time polymerase chain reaction (PCR) detection method specific for Listeria monocytogenes was developed, and studies involving pure culture showed that the response of the assay was linear over 7 log10 (log) cycles. The method was then applied to the detection of L. monocytogenes artificially inoculated onto cabbage, a vegetable chosen because it is a major component of coleslaw, which has been associated with an outbreak of listeriosis. After being allowed to attach to the food, cells were washed from the cabbage leaf surface and recovered by centrifugation. The DNA was pu

Detection of different Ikaros isoforms in human leukaemias using real-time quantitative polymerase chain reaction.

The Ikaros gene is an essential regulator in development and haematopoiesis. Dysregulated Ikaros gene expression participates in leukaemic processes, as evidenced in animal models, and by analyses of blast-cell populations from leukaemic patients. We used real-time quantitative polymerase chain reaction (PCR) to evaluate the relative abundance of several Ikaros transcript isoforms in a variety of leukaemic-cell samples. Total RNA was isolated from bone-marrow or blood-cell samples collected at diagnosis in children or adult patients, 18 of whom had acute myeloblastic leukaemia (AML), 61 of who

Real-Time Polymerase Chain Reaction in Detecting Active Hepatitis C Virus Infection: A Retrospective Analysis.

Introduction Hepatitis C virus (HCV) infection is a major worldwide health concern. It is a major cause of cirrhosis, hepatocellular carcinoma, and chronic hepatitis. Testing for anti-HCV antibodies is common for screening purposes, but it does not distinguish between an active and a past infection. A real-time polymerase chain reaction (RT-PCR)-based detection of HCV ribonucleic acid (RNA) is used to confirm active infection following initial serological screening. The current study aimed to assess the proportion of HCV RNA positivity, the distribution of viral load, and its association with

Real-time polymerase chain reaction (PCR) for Pneumocystis jirovecii detection in lower respiratory tract samples.

To date, the gold standard for Pneumocystis jirovecii pneumonia (PjP) diagnosis is direct microscopic examination (ME). However, several studies have shown that the DNA copy number of the P. jirovecii, measured by quantitative real-time PCR (qPCR), is significantly higher in patients with PjP than in colonized individuals. Nevertheless, this approach is not standardized yet and requires further investigation. In this study, we aimed to compare the results obtained using the Pneumocystis ELITe MGB Kit (ELITech Group S.p.a., Italy) with those from ME, evaluating specificity, sensitivity, and the

A Novel Nested Real-time Polymerase Chain Reaction for Treponema pallidum DNA in Syphilis Biospecimens.

BACKGROUND: Early diagnosis of Treponema pallidum infection is helpful for disease management, and conventional PCR is suitable for lesion swabs of patients with probable early syphilis. We thus tested nested and real-time PCR (NR-PCR) in various biosamples from syphilitic patients. METHODS: Samples were collected from syphilis patients before treatment. Specific primer sequences targeting the T. pallidum gene polA were designed for NR-PCR. RESULTS: Across syphilis types, most samples assayed with NR-PCR returned a positive result, including earlobe blood (92.0%), cerebrospinal fluid (CSF) (
