Ca2+ -permeable AMPA receptors and their auxiliary subunits in synaptic plasticity and disease.

AMPA receptors are tetrameric glutamate-gated ion channels that mediate a majority of fast excitatory neurotransmission in the brain. They exist as calcium-impermeable (CI-) and calcium-permeable (CP-) subtypes, the latter of which lacks the GluA2 subunit. CP-AMPARs display an array of distinctive biophysical and pharmacological properties that allow them to be functionally identified. This has revealed that they play crucial roles in diverse forms of central synaptic plasticity. Here we summarise the functional hallmarks of CP-AMPARs and describe how these are modified by the presence of auxi

Ketamine may exert antidepressant effects via suppressing NLRP3 inflammasome to upregulate AMPA receptors.

Recent studies have indicated that sub-anesthetic dose of ketamine exerts rapid antidepressant effects. Upregulation of α-amino-3-hydroxy-5-methylisoxazole-4-propionic acid (AMPA) receptors contributes to rapid antidepressant effects of ketamine. A recent study indicated that the increasing expression of AMPARs is related to the inhibition of NLRP3 inflammasome. Thus, we postulated that NLRP3 inflammasome might play an important role in ketamine's antidepressant effects. We found that sub-anesthesia dose of ketamine (10 ml/kg) ameliorated LPS-induced depressive-like behaviors, including decrea

A human PET study of [11C]HMS011, a potential radioligand for AMPA receptors.

BACKGROUND: α-Amino-3-hydroxy-5-methyl-4-isoxazole propionate (AMPA) receptor is a primary mediator of fast glutamatergic excitatory signaling in the brain and has been implicated in diverse neuropsychiatric diseases. We recently developed a novel positron emission tomography (PET) ligand, 2-(1-(3-([11C]methylamino)phenyl)-2-oxo-5-(pyrimidin-2-yl)-1,2-dihydropyridin-3-yl) benzonitrile ([11C]HMS011). This compound is a radiolabelled derivative of perampanel, an antiepileptic drug acting on AMPA receptors, and was demonstrated to have promising in vivo properties in the rat and monkey brains. In

S-nitrosylation of N-ethylmaleimide sensitive factor mediates surface expression of AMPA receptors.

Postsynaptic AMPA receptor (AMPAR) trafficking mediates some forms of synaptic plasticity that are modulated by NMDA receptor (NMDAR) activation and N-ethylmaleimide sensitive factor (NSF). We report that NSF is physiologically S-nitrosylated by endogenous, neuronally derived nitric oxide (NO). S-nitrosylation of NSF augments its binding to the AMPAR GluR2 subunit. Surface insertion of GluR2 in response to activation of synaptic NMDARs requires endogenous NO, acting selectively upon the binding of NSF to GluR2. Thus, AMPAR recycling elicited by NMDA neurotransmission is mediated by a cascade i

Rapid spine delivery and redistribution of AMPA receptors after synaptic NMDA receptor activation.

To monitor changes in alpha-amino-3-hydroxy-5-methyl-4-isoxazole propionate (AMPA) receptor distribution in living neurons, the AMPA receptor subunit GluR1 was tagged with green fluorescent protein (GFP). This protein (GluR1-GFP) was functional and was transiently expressed in hippocampal CA1 neurons. In dendrites visualized with two-photon laser scanning microscopy or electron microscopy, most of the GluR1-GFP was intracellular, mimicking endogenous GluR1 distribution. Tetanic synaptic stimulation induced a rapid delivery of tagged receptors into dendritic spines as well as clusters in dendri

Responses of directionally selective retinal ganglion cells to activation of AMPA glutamate receptors.

Previous studies in the rabbit retina have shown that drugs which block AMPA glutamate receptors abolish directional selectivity in ON-OFF directionally selective (DS) ganglion cells. The effects of activation of AMPA receptors on the directionally selective responses of these ganglion cells had not been studied. In the present study, extracellular recordings of the responses of ON-OFF DS ganglion cells to a moving bar of light were made in an in vitro rabbit retinal preparation. In control solution, bath application of AMPA (7-10 microM) abolished the light responses of most ON-OFF DS ganglio

Dorsomedial Striatum Calcium Permeable AMPA Receptors in the Development of Aversion-Resistant Alcohol Drinking.

RATIONALE: Alcohol use disorder is defined by drinking alcohol despite knowledge of negative consequences, often referred to as aversion-resistant drinking (ARD). The dorsomedial (DMS) and dorsolateral striatum (DLS) are necessary for goal-directed and habitual action selection, respectively. Leading hypotheses posit that once drug use becomes compulsive, DMS dependence degrades while DLS dependence increases. This shift may be mediated by changes in synaptic weights from glutamatergic inputs. OBJECTIVES: Using a combination of western-blot, micro-injections, and ex-vivo electrophysiology, we

Calcium permeable AMPA receptors and autoreceptors in external tufted cells of rat olfactory bulb.

Glomeruli are functional units of the olfactory bulb responsible for early processing of odor information encoded by single olfactory receptor genes. Glomerular neural circuitry includes numerous external tufted (ET) cells whose rhythmic burst firing may mediate synchronization of bulbar activity with the inhalation cycle. Bursting is entrained by glutamatergic input from olfactory nerve terminals, so specific properties of ionotropic glutamate receptors on ET cells are likely to be important determinants of olfactory processing. Particularly intriguing is recent evidence that AMPA receptors o

The role of AMPA and metabotropic glutamate receptors on morphine withdrawal in infant rats.

Glutamate receptors, especially N-methyl-d-aspartate (NMDA) receptors, are hypothesized to play key roles in opiate tolerance and withdrawal. There is also accumulating evidence that alpha-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid (AMPA) receptor antagonists and group II metabotropic glutamate receptor (mGluR) agonists attenuate opiate withdrawal. However, most existing data are derived from adult animal models. Glutamate receptor types undergo dramatic developmental changes during early life. Thus, the pharmacological effects on opiate withdrawal of NMDA receptor, AMPA receptor, and

Tunable AMPA receptor function via recurrent evolution of heterotetramers.

Most ionotropic neurotransmitter receptors are hetero-oligomers of three to five homologous subunits assembled into membrane-spanning ion channels. Mammalian AMPA-type glutamate receptors (AMPARs), which emerged in animals with centralized nervous systems and now mediate most of the excitatory synaptic signaling in our brain, readily assemble as homotetramers, yet often occur as heterotetramers of two to three different AMPAR subunits. Here, we looked for unifying functional properties of the AMPAR family using molecular phylogenetics, together with electrophysiological and pharmacological cha

Structural basis for activation and conformational plasticity of the GluA4 AMPA receptor.

AMPA-subtype glutamate receptors (AMPARs) mediate excitatory synaptic transmission. AMPAR ion channels exhibit multiple subconductance states that tune neuronal responses to glutamate. GluA4 is the rarest subunit in the brain but is enriched in interneurons and the cerebellum. Rising evidence points to GluA4 AMPARs in the development of neurological diseases, but the structural mechanisms of GluA4 function remain enigmatic. Here, we show the distinct features of GluA4 that tune AMPAR function. We find that GluA4 AMPARs have a canonical "Y" shaped architecture where local dimer pairs are domain

AMPA/kainate glutamate receptor antagonists prevent posttraumatic osteoarthritis.

Musculoskeletal disorders represent the third greatest burden in terms of death and disability in the developed world. Osteoarthritis is the single greatest cause of chronic pain, has no cure, and affects 8.5 and 27 million people in the UK and US, respectively. Osteoarthritis is most prevalent in older people, but as it commonly occurs after joint injury, young people with such injuries are also susceptible. Painful joints are often treated with steroid or hyaluronic acid (HA) injections, but treatments to prevent subsequent joint degeneration remain elusive. In animals, joint injury increase

AMPA receptor GluA2 subunit defects are a cause of neurodevelopmental disorders.

AMPA receptors (AMPARs) are tetrameric ligand-gated channels made up of combinations of GluA1-4 subunits encoded by GRIA1-4 genes. GluA2 has an especially important role because, following post-transcriptional editing at the Q607 site, it renders heteromultimeric AMPARs Ca2+-impermeable, with a linear relationship between current and trans-membrane voltage. Here, we report heterozygous de novo GRIA2 mutations in 28 unrelated patients with intellectual disability (ID) and neurodevelopmental abnormalities including autism spectrum disorder (ASD), Rett syndrome-like features, and seizures or deve

AMPA Receptor-Dependent Glutamatergic Signaling is Present in the Carotid Chemoreceptor.

Exposure to both sustained and intermittent hypoxia for as little as a day produces sustained augmentation of carotid chemoreceptor sensitivity; however, the molecular basis for this chemoreflex plasticity remains uncertain. We previously reported that NMDA receptor-dependent glutamatergic signaling in rat carotid body played a role in altered hypoxic sensitivity after exposure to cyclic intermittent hypoxia (CIH). Here we found that mRNAs of multiple AMPA and Kainate glutamate receptors were expressed in rat carotid body. The AMPA receptor subunit GluR1 showed intense immunoreactivity in the

Anti-AMPA receptor encephalitis associated with thymomatous myasthenia gravis.

No thymomatous myasthenia gravis (TMG) has been reported in patients with anti-α-amino-3-hydroxy-5-methyl-4-isoxazole-propionic acid receptor encephalitis (AMPAR-E). We described clinical presentation and autoimmune profile of the first case with both AMPAR-E and TMG. Clinical information was obtained from original medical records. Antibodies against AMPAR and confounding antigens were examined on transfected human embryonic kidney 293 cells by the indirect immunofluorescence method. In this case, anti-AMPAR antibodies and anti-acetylcholine receptor (AchR) antibodies were both positive and co

Argiotoxin in the closed AMPA receptor channel: experimental and modeling study.

Binding of argiotoxin in the closed state of Ca(2+)-permeable AMPA receptor channels was studied using electrophysiological and molecular modeling approaches. Experimental study unambiguously revealed that argiotoxin is trapped in the closed AMPA receptor channels after agonist dissociation. Docking of the argiotoxin to the channel model based on recently published X-ray structure demonstrated that the drug can be effectively accommodated in the cavity of the closed channel only if the terminal moiety of the molecule penetrates in the narrow portion of the pore below the selectivity filter. AMPA Receptors in NG2 Glia Differently Affect Signal Transduction in the Hippocampus and Cerebellum.

Gray matter NG2 glia constitute a heterogeneous population of cells whose functions remain incompletely understood. In the hippocampus, Schaffer collaterals activate AMPA receptors (AMPARs) in NG2 glia, giving rise to small excitatory post-synaptic currents (EPSCs). Climbing fibers of the cerebellum also form synapses with NG2 glia, although producing much larger EPSCs. We aimed to identify mechanisms generating these regional differences in the efficacy of neuron-glia synapses. Combined patch-clamp and RT-PCR analyses allowed for determining structural and functional differences of AMPARs exp

Granulocyte colony-stimulating factor acts through calcium-permeable AMPA receptors to potentiate cocaine reward.

Neuroimmune interactions have emerged as critical modulators of substance use disorders and may represent promising translational therapeutic targets. In prior work, we demonstrated that the cytokine granulocyte colony stimulating factor (G-CSF) is elevated in mice following cocaine exposure, with circulating levels correlating with cocaine intake and locomotor sensitization. Additionally, exogenous G-CSF enhances cocaine reward and increases low-dose cocaine self-administration. We have further shown that repeated G-CSF administration alters expression of glutamatergic synapse-associated prot

Understanding the role of AMPA receptors in autism: insights from circuit and synapse dysfunction.

Autism spectrum disorders represent a diverse etiological spectrum that converge on a syndrome characterized by discrepant deficits in developmental domains often highlighted by concerns in socialization, sensory integration, and autonomic functioning. Importantly, the incidence and prevalence of autism spectrum disorders have seen sharp increases since the syndrome was first described in the 1940s. The wide etiological spectrum and rising number of individuals being diagnosed with the condition lend urgency to capturing a more nuanced understanding of the pathogenic mechanisms underlying the
