Central Glucagon-like Peptide-1 Receptor Signaling via Brainstem Catecholamine Neurons Counteracts Hypertension in Spontaneously Hypertensive Rats.

Glucagon-like peptide-1 receptor (GLP-1R) agonists, widely used to treat type 2 diabetes, reduce blood pressure (BP) in hypertensive patients. Whether this action involves central mechanisms is unknown. We here report that repeated lateral ventricular (LV) injection of GLP-1R agonist, liraglutide, once daily for 15 days counteracted the development of hypertension in spontaneously hypertensive rats (SHR). In parallel, it suppressed urinary norepinephrine excretion, and induced c-Fos expressions in the area postrema (AP) and nucleus tractus solitarius (NTS) of brainstem including the NTS neuron

Beyond catecholamine escalation: Receptor desensitization and endogenous vasopressor failure, and extracorporeal circuit biocompatibility after cardiopulmonary bypass-A narrative review.

BACKGROUND: Vasoplegic syndrome following cardiopulmonary bypass (CPB) is a major cause of postoperative circulatory failure in cardiac surgery and is associated with increased morbidity and mortality. Traditionally, vasoplegia has been attributed to systemic inflammation, endothelial dysfunction, excessive nitric oxide production, and pathological vasodilation. However, these mechanisms alone do not fully explain the marked variability in vasopressor responsiveness observed in clinical practice. Increasing evidence suggests that vasoplegia may result not only from depletion of endogenous vaso

Altered plasma catecholamines and numbers of alpha- and beta-adrenergic receptors in platelets and leucocytes in hyperthyroid patients normalized under antithyroid treatment.

We measured plasma catecholamines, alpha- and beta-adrenoreceptor numbers and the accumulation of cyclic adenosine monophosphate (cAMP) in the unstimulated state and in response to 10 mumol/l (-) isoproterenol in blood cells from 29 euthyroid controls and from 18 patients with spontaneous hyperthyroidism. In the thyrotoxic patients plasma norepinephrine (1.14 +/- 0.5 nmol/l) and epinephrine (0.3 +/- 0.14 nmol/l) were significantly decreased compared with plasma norepinephrine (1.87 +/- 0.7 nmol) and epinephrine (0.41 +/- 0.19 nmol/l) in the controls (P less than 0.01 and P less than 0.05, resp

Plasma catecholamine concentration in hypertensive subjects with chronic renal failure before and during treatment with the beta-receptor blocking drug metipranolol.

In seven male hypertensive patients with chronic renal failure (serum creatinine level 6.9 +/- 4.1 mg%) plasma catecholamine levels were in the same range as in five normotensive healthy volunteers. Treatment with the beta-receptor blocker metipranolol (2 x 20 mg/day orally for 2 weeks) significantly elevated plasma catecholamine levels in the hypertensive patients with chronic renal failure, but not in the healthy volunteers. Blood pressure and heart rate were reduced in both groups.

Ectopic beta-adrenergic receptor binding sites. possible molecular basis of aberrant catecholamine responsiveness of an adrenocortical tumor adenylate cyclase.

The molecular basis for the aberrant catecholamine responsiveness of the adenylate cyclase of adrenocortical carcinoma 494 was explored. The adenylate cyclase of this corticosteroid-producing, transplanted, adrenal cancer of the rat was stimulated not only by adrenocorticotropic hormone and fluoride, but also by the beta-adrenergic agonist, isoproterenol. The adenylate cyclase of normal adrenal tissue was unresponsive to isoproterenol. Direct binding studies with the specific high affinity B-adrenergic ligand, (-)[3H]dihydroalprenolol, demonstrated the pressure of 0.094 pmol of specific bindin

Enhanced adenosine A2b receptor signaling facilitates stimulus-induced catecholamine secretion in chronically hypoxic carotid body type I cells.

Chronic hypoxia (CHox) augments chemoafferent activity in sensory fibers innervating carotid body (CB) chemoreceptor type I cells; however, the underlying mechanisms are poorly understood. We tested the hypothesis that enhanced paracrine signaling via adenosine (Ado) A2b receptors is involved. Dissociated rat CB cultures were exposed for 24 h to normoxia (Nox, 21% O2) or CHox (2% O2) or treated with the hypoxia mimetic deferoxamine mesylate (DFX), and catecholamine secretion from type I cells was monitored by amperometry. Catecholamine secretion was more robust in CHox and DFX type I cells tha

Catecholamine metabolism inhibitors and receptor blockades only partially suppress cardiac hypertrophy of juvenile visceral steatosis mice with systemic carnitine deficiency.

To clarify the mechanism of cardiac hypertrophy in carnitine-deficient JVS mice, we studied the possible role of catecholamine metabolism. Cardiac hypertrophy occurs 2 weeks after birth. The turnover of norepinephrine in the ventricles of JVS mice at 2 weeks was 3 times that of control, but it was not different from control at 5 days when the heart weight was not changed. To evaluate the accelerated norepinephrine turnover, we examined the effects of catecholamine metabolism inhibitors (alpha-methyltyrosine and 6-hydroxydopamine) and catecholamine receptor blockades (propranolol, prazosin and

Cardiac Somatostatin-receptor Expression Demonstrated by 68 Ga DOTANOC PET/CT in a Child Presenting With Neuroblastoma and Presumed Catecholamine-induced Cardiomyopathy.

Hypertension in pediatric neuroblastoma can be challenging to manage due to its multifactorial etiology. We present a 3-year-old girl who presented with cardiogenic shock, echocardiographic evidence of dilated cardiomyopathy, right renal artery stenosis, and elevated catecholamines. Somatostatin receptor (SSTR) PET/CT revealed intense tracer uptake in a right adrenal mass and a low-grade diffuse myocardial uptake, suggestive of inflammatory myocarditis. This unusual finding highlights the potential role of SSTR PET/CT in detecting signs of myocarditis, with myocardial SSTR uptake as a possible

Receptor for catecholamines responding to catechol which potentiates voltage-dependent calcium current in single cells from guinea-pig taenia caeci.

1. Single isolated cells were obtained from the taenia of the guinea-pig's caecum by enzymic digestion and held under voltage clamp. The effects of various catecholamines, sympathomimetics and related compounds were tested for their ability to potentiate the voltage-dependent calcium current (ICa) evoked in these cells by a depolarizing step. 2. ICa was potentiated by up to 60% by isoprenaline, adrenaline, and noradrenaline which were equipotent. The EC50 for isoprenaline was about 40 nM. 3. Ikarisoside A inhibits acetylcholine-induced catecholamine secretion and synthesis by suppressing nicotinic acetylcholine receptor-ion channels in cultured bovine adrenal medullary cells.

Ikarisoside A is a natural flavonol glycoside derived from plants of the genus Epimedium, which have been used in Traditional Chinese Medicine as tonics, antirheumatics, and aphrodisiacs. Here, we report the effects of ikarisoside A and three other flavonol glycosides on catecholamine secretion and synthesis in cultured bovine adrenal medullary cells. We found that ikarisoside A (1-100 μM), but not icariin, epimedin C, or epimedoside A, concentration-dependently inhibited the secretion of catecholamines induced by acetylcholine, a physiological secretagogue and agonist of nicotinic acetylcholi

The role of the guanine nucleotide exchange reaction in the regulation of the beta-adrenergic receptor and in the actions of catecholamines and cholera toxin on adenylate cyclase in turkey erythrocyte membranes.

Several changes were noted in the characteristics of the turkey erythrocyte beta-adrenergic receptor and in the kinetic properties of adenylate cyclase following pretreatment of erythrocyte membranes with isoproterenol and GMP, and thorough washing to remove these agents. The changes include modifications in the binding of agonist (isoproterenol) and in revelation of marked effects of GTP on agonist binding; reduction in the lag in Gpp(NH)p activation of adenylate cyclase; short lived activation by GTP which is lengthened by treatment with cholera toxin and NAD prior to pretreatment with isopr

The A(2B)-D(2) receptor interaction that controls carotid body catecholamines release locates between the last two steps of hypoxic transduction cascade.

We have recently demonstrated that adenosine controls the release of catecholamines (CA) from carotid body (CB) acting on A(2B) receptors. Here, we have investigated the hypothesis that this control is exerted via an interaction between adenosine A(2B) and dopamine D(2) receptors present in chemoreceptor cells and if it is, the location of this interaction on the CB hypoxic transduction cascade. Experiments were performed in vitro in CB from 3 months rats. The effect of adenosine A(2B) and dopamine D(2) receptor agonists applied alone or conjunctly, was studied on the basal and evoked release

Catecholamines and pituitary function. VII: Effects of acute and chronic dopamine-receptor blockade on pituitary response to TRH-GNRH in normal women and in patients with hyperprolactinemic amenorrhea.

To investigate whether an enhanced dopamine (DA) inhibition on pituitary thyrotrophs and gonadotrophs may account for the abnormal TSH and LH dynamics in pathological hyperprolactinemia, we examined the effect of an acute lysis of the putative DA overinhibition, as obtained with continuous domperidone (DOM) infusion, on both basal and TRH-GnRH stimulated PRL, TSH and LH release in both normal cycling women and patients with pathological hyperprolactinemia. Dual effects of nobiletin, a citrus polymethoxy flavone, on catecholamine secretion in cultured bovine adrenal medullary cells.

Nobiletin, a compound of polymethoxy flavones found in citrus fruits, possesses a wide range of pharmacological activities. Here we report the effects of nobiletin on catecholamine secretion in cultured bovine adrenal medullary cells. Nobiletin (1.0-100 microM) concentration-dependently stimulated catecholamine secretion and (45)Ca(2+) influx. Its stimulatory effect of nobiletin on catecholamine secretion was abolished by deprivation of extracellular Ca(2+) and partially inhibited by specific inhibitors of voltage-dependent Ca(2+) channels and Na(+)/Ca(2+) exchangers. On the other hand, nobile

Cyclical antiidiotypic response to anti-hormone antibodies due to neutralization by autologous anti-antiidiotype antibodies that bind hormone.

Antiidiotype antibodies were raised against anti-catecholamine ligand antibodies. The antiidiotype response was shown to be cyclical and to correspond to the production of antibodies that could bind to catecholamine beta-adrenergic receptors and stimulate adenylate cyclase. Disappearance of these antibodies from the serum could be correlated with the appearance of a catecholamine ligand-binding activity corresponding to the synthesis of autologous anti-antiidiotype antibodies directed against the induced antiidiotypic molecules. Comparison of the injected versus the induced anti-ligand antibod

Effects of the potassium channel openers cromakalim and pinacidil on catecholamine secretion and calcium mobilization in cultured bovine adrenal chromaffin cells.

The effects of two K+ channel openers, cromakalim and pinacidil, on voltage-dependent and receptor-mediated catecholamine secretion and Ca2+ mobilization in bovine adrenal chromaffin cells were studied to determine the role of membrane K+ channels in the regulation of a Ca(2+)-dependent secretory process. Both cromakalim and pinacidil stimulated the efflux of 86Rb (used to monitor K+ permeability) from preloaded cells. Cromakalim and pinacidil did not affect the catecholamine secretion induced by excessive depolarization with 56 mM K+, but inhibited that induced by moderate depolarization with

Circulating and local catecholamines are elevated in high fat diet induced adipose catecholamine resistance.

Catecholamines such as epinephrine and norepinephrine regulate white adipose tissue lipolysis through β-adrenergic receptors (β-AR). In metabolic syndrome, adipose tissue downregulates β-AR and becomes catecholamine resistant, a finding recapitulated under high-fat diet. In many receptor-ligand systems, chronic stimulation induces desensitization, but limited data explores whether this relationship holds true for adipose tissue catecholamine resistance. Catecholamine signaling in adipose tissue is also regulated by clearance through monoamine oxidases and catechol-O-methyltransferases, but alm
