Molecular and morphological data support the existence of a sexual cycle in species of the genus Paracoccidioides.

The genus Paracoccidioides includes the thermodimorphic species Paracoccidioides brasiliensis and P. lutzii, both of which are etiologic agents of paracoccidioidomycosis, a systemic mycosis that affects humans in Latin America. Despite the common occurrence of a sexual stage among closely related fungi, this has not been observed with Paracoccidioides species, which have thus been considered asexual. Molecular evolutionary studies revealed recombination events within isolated populations of the genus Paracoccidioides, suggesting the possible existence of a sexual cycle. Comparative genomic ana

Aplysia temptin - the 'glue' in the water-borne attractin pheromone complex.

Temptin, a component of the complex of water-borne protein pheromones that stimulate attraction and mating behavior in the marine mollusk Aplysia, has sequence homology to the epidermal growth factor (EGF)-like domains of higher organisms that mediate protein-cell surface contact during fertilization and blood coagulation. In this work, recombinant temptin for structural and functional studies was produced in Escherichia coli using a cold shock promoter and purified by RP-HPLC. CD spectra confirmed a predominantly beta-sheet structure. Two disulfide bonds were determined via limited proteolysi

Structure and function of the vomeronasal organ.

The vomeronasal organ (VNO) is a complex of different structures that forward specific chemical signals commonly called pheromones to the central nervous system. In some macrosmatic animals, e.g. rodents, the VNO consists of vomeronasal receptor neurons located in a sensory epithelium of the vomeronasal duct, their afferent axons connecting the duct with the accessory olfactory bulb, associated glands and ganglionic cells in the nasal septal mucosa. The organ's main task is to influence mating and social behavior. In humans, the VNO does not exist, at least not in its complexity. Although deve

Male and female mice scent mark during social communication regardless of sexual motivation or partner identity.

Many species communicate via pheromone signals that orchestrate key social and survival behaviors. In laboratory assays, only male mice are observed to broadcast pheromones through urine scent marking. This has contributed to the view that males have specialized circuits and anatomy to intentionally transmit urine pheromones and actively participate in socio-chemical communication, while females remain mute recipients in laboratory settings. Here, we find that simple modifications to standard assays reveal exuberant urine marking by laboratory females, enabling reexamination of its dimorphic f

Semiochemical and Communication Ecology of the Emerald Ash Borer, Agrilus planipennis (Coleoptera: Buprestidae).

Knowledge of buprestid chemical ecology is sparse but the appearance of the invasive pest Agrilus planipennis Fairmaire in North America has provided the impetus to study in detail the semiochemistry and ecology of this important buprestid. The macrocyclic lactone (3Z)-12-dodecenolide [(3Z)-lactone] is identified as a key antennally-active compound that is produced by females and attracts males. Though a weak trap attractant alone, when combined with the host kairomone (3Z)-hexenol and the important visual cue of a green canopy trap, significant increases in male trap capture occur, thus defin

Synaptotagmin I, a molecular target for steroid hormone signaling controlling the maturation of sexual behavior in an insect.

UNLABELLED: As in vertebrates, the insect steroid hormones, especially 20-hydroxyecdysone (20E), initiate and regulate sexual behavior by acting on the central nervous system. This 20E action is, in part, triggered by transcriptional events mediated through the binding of 20E to a heterodimer comprising the ecdysone receptor (EcR) and ultraspiracle (USP). However, to date, our knowledge about this genomic steroid pathway remains incomplete. In moths, males detect female sex pheromones, eliciting stereotyped sexual behavior. In Agrotis ipsilon males, the behavioral response and the neuronal sen

The RGS protein Crg2 regulates both pheromone and cAMP signalling in Cryptococcus neoformans.

G proteins orchestrate critical cellular functions by transducing extracellular signals into internal signals and controlling cellular responses to environmental cues. G proteins typically function as switches that are activated by G protein-coupled receptors (GPCRs) and negatively controlled by regulator of G protein signalling (RGS) proteins. In the human fungal pathogen Cryptococcus neoformans, three G protein alpha subunits (Gpa1, Gpa2 and Gpa3) have been identified. In a previous study, we identified the RGS protein Crg2 involved in regulating the pheromone response pathway through Gpa2 a

Sublethal indoxacarb exposure alters pheromone production and ovarian development in the yellow peach moth, Conogethes punctiferalis.

The Conogethes punctiferalis, a major pest of peach and other fruit trees, causes significant damage by boring into fruits during its larval stage. Indoxacarb, a sodium channel blocker insecticide, has been widely applied in agriculture and horticulture for pest control, particularly against larval pests. However, its effects on adult insects remain largely unexplored. Present study employed C. punctiferalis as model to investigate the toxic effects of indoxacarb on adult moths. The results showed that sublethal exposure to indoxacarb significantly reduced the release of sex pheromones, reduce

Evidences of G Coupled-Protein Receptor (GPCR) Signaling in the human Malaria Parasite Plasmodium falciparum for Sensing its Microenvironment and the Role of Purinergic Signaling in Malaria Parasites.

The nucleotides were discovered in the early 19th century and a few years later, the role of such molecules in energy metabolism and cell survival was postulated. In 1972, a pioneer work by Burnstock and colleagues suggested that ATP could also work as a neurotransmitter, which was known as the "purinergic hypothesis". The idea of ATP working as a signaling molecule faced initial resistance until the discovery of the receptors for ATP and other nucleotides, called purinergic receptors. Among the purinergic receptors, the P2Y family is of great importance because it comprises of G proteincouple

Gustatory receptors interact with the serotonin pathway to regulate silkmoth stereotyped courtship behavior.

BACKGROUND: Gustatory receptors (GRs) are known to mediate responses to various chemical stimuli including sugars and bitter compounds and sex pheromones. Although numerous types of GRs have been identified across insect species, the physiological roles of most GRs remain largely unknown. The silkworm, Bombyx mori, is representative of the order Lepidoptera, which includes over 70% of agriculturally important pest species. A notable feature of GRs in B. mori is that most GRs are organized in clusters, making them highly suitable for large-scale genetic studies. RESULTS: Here we developed a tr

Bmal1 Is Required for Normal Reproductive Behaviors in Male Mice.

Circadian rhythms synchronize physiological processes with the light-dark cycle and are regulated by a hierarchical system initiated in the suprachiasmatic nucleus, a hypothalamic region that receives direct photic input. The suprachiasmatic nucleus then entrains additional oscillators in the periphery. Circadian rhythms are maintained by a molecular transcriptional feedback loop, of which brain and muscle aryl hydrocarbon receptor nuclear translocator-like protein 1 (BMAL1) is a key member. Disruption of circadian rhythms by deletion of the BMAL1 gene (Bmal1 knockout [KO]) induces a variety o

Sensory processing in the main and accessory olfactory systems: comparisons and contrasts.

The vomeronasal organ (VNO) and accessory olfactory system (AOS) are present in most terrestrial vertebrates except birds and higher primates. The receptor neurons of the AOS are sequestered inside the VNO, away from the main airflow to the main olfactory receptor neurons. Mechanisms of stimulus access to the sensory neurons vary across species but in most cases there is a system for delivering stimuli faster than would be possible by diffusion. Vomeronasal (VN) receptor neurons typically lack cilia, the site of most of the transduction apparatus in the main olfactory receptors. The VN recepto

SNMP1 Contributes to Sensitive Pheromone and Plant Volatile Detection in the Locust Olfactory System.

In holometabolous insects, such as flies and moths, the sensory neuron membrane protein 1 (SNMP1) has been primarily linked to the olfactory detection of long-chain aliphatic pheromones, but whether it plays an important role in receiving other chemical cues is largely unknown. Here, comparing electroantennogram (EAG) recordings of wild-type (WT) and SNMP1-deficient (SNMP1-/-) hemimetabolous desert locusts (Schistocerca gregaria), we found that mutant animals showed significantly reduced responses to several pheromones and plant odorants belonging to different classes of aromatic and aliphatic

Juvenile Hormone Involved in the Defensive Behaviors of Soldiers in Termite Reticulitermes aculabialis.

Eusocial insects have evolved specific defensive strategies to protect their colonies. In termite colonies, soldiers perform a colony-level defense by displaying mechanical biting, head-banging and mandible opening-closing behaviors. However, few studies have been reported on the factors modulating defensive behaviors in termites. Owing to JH (juvenile hormone) being involved in soldier differentiation, JH was speculated to affect defensive behaviors in termite soldiers. To determine the effect of JH on the defensive behaviors of termite soldiers, we performed a JHA-feeding and RaSsp1-silencin

Insulin Signaling in the Peripheral and Central Nervous System Regulates Female Sexual Receptivity during Starvation in Drosophila.

Many animals adjust their reproductive behavior according to nutritional state and food availability. Drosophila females for instance decrease their sexual receptivity following starvation. Insulin signaling, which regulates many aspects of insect physiology and behavior, also affects reproduction in females. We show that insulin signaling is involved in the starvation-induced reduction in female receptivity. More specifically, females mutant for the insulin-like peptide 5 (dilp5) were less affected by starvation compared to the other dilp mutants and wild-type flies. Knocking-down the insulin

Measuring Physiological Responses of Drosophila Sensory Neurons to Lipid Pheromones Using Live Calcium Imaging.

Unlike mammals, insects such as Drosophila have multiple taste organs. The chemosensory neurons on the legs, proboscis, wings and ovipositor of Drosophila express gustatory receptors(1,2), ion channels(3-6), and ionotropic receptors(7) that are involved in the detection of volatile and non-volatile sensory cues. These neurons directly contact tastants such as food, noxious substances and pheromones and therefore influence many complex behaviors such as feeding, egg-laying and mating. Electrode recordings and calcium imaging have been widely used in insects to quantify the neuronal responses ev

Chemosensory adaptations in Caenorhabditis males during the establishment of androdioecy.

Caenorhabditis elegans has evolved from its dioecious ancestors to adopt an androdioecious reproductive strategy. In this process, ancestral female C. elegans acquired genetic modifications that enabled self-sperm generation, self-sperm activation and a reduced reliance on sexual reproduction. However, how males have adapted during this transition from dioecy to androdioecy is less explored. Using Caenorhabditis species, we demonstrated that androdioecious hermaphrodites exhibit attenuated sex pheromone potency, while androdioecious males show heightened olfactory habituation and diminished ma

An electrostatic interaction between BlpC and BlpH dictates pheromone specificity in the control of bacteriocin production and immunity in Streptococcus pneumoniae.

UNLABELLED: The blp locus of Streptococcus pneumoniae secretes and regulates bacteriocins, which mediate both intra- and interspecific competition in the human nasopharynx. There are four major alleles of the gene blpH, which encodes the receptor responsible for activating the blp locus when bound to one of four distinct peptide pheromones (BlpC). The allelic variation of blpH is presumably explained by a need to restrict cross talk between competing strains. The BlpH protein sequences have polymorphisms distributed throughout the sequence, making identification of the peptide binding site dif

Buffalo nasal odorant-binding protein (bunOBP) and its structural evaluation with putative pheromones.

Pheromones are odoriferous volatile chemical cues produced by animals for communication among conspecifics so as to regulate their social behaviors. In general, the odor compounds are recognized by receptors in the nasal cavity. Odorant-binding protein (OBP), a lipocalin family protein, mediates the air-borne odor cues to nasal receptors through nasal mucus. The presence of OBP in several mammalian species is well documented but to-date there is no report of a nasal OBP in buffalo. Hence, the present study was undertaken to investigate if OBP is present in buffalo nasal mucus. Uni- and two-dim

A comparison of the effects of male pheromone priming and optogenetic inhibition of accessory olfactory bulb forebrain inputs on the sexual behavior of estrous female mice.

Previous research has shown that repeated testing with a stimulus male is required for ovariectomized, hormone-primed female mice to become sexually receptive (show maximal lordosis quotients; LQs) and that drug-induced, epigenetic enhancement of estradiol receptor function accelerated the improvement in LQs otherwise shown by estrous females with repeated testing. We asked whether pre-exposure to male pheromones ('pheromone priming') would also accelerate the improvement in LQs with repeated tests and whether optogenetic inhibition of accessory olfactory bulb (AOB) projection neurons could in
