Effects of pentobarbital on purinergic P2X receptors of rat dorsal root ganglion neurons.

Purinergic P2X receptors are ligand-gated ion channels that are activated by extracellular adenosine triphosphate (ATP) and are widely expressed not only in the central and peripheral nervous system but also in tissues throughout the body, playing an important role in the transfer of nociceptive information. Since the influence of barbiturates on P2X receptor subtypes is not known, we studied the effects of pentobarbital sodium (PB) on ATP responses in dorsal root ganglion (DRG) neurons. DRG neurons were dissected from 10- to 14-day-old rats and dissociated after enzyme treatment. Electrical m

Serum Levels of Glutamatergic (GRIN1) and Purinergic (P2RX1/P2RY2) Receptors in Patients with Fibromyalgia.

Fibromyalgia (FM) is a chronic pain syndrome characterized by central sensitization, in which glutamatergic and purinergic signaling pathways are thought to play critical roles. This study aimed to evaluate the diagnostic potential of serum glutamate ionotropic receptor N-methyl-D-aspartate type subunit 1 (GRIN1), purinergic receptor P2X 1 (P2RX1), and purinergic receptor P2Y 2 (P2RY2) levels in patients with FM. A total of 93 newly diagnosed FM patients and 93 age- and sex-matched healthy controls were included in the study. Serum levels of GRIN1, P2RX1, and P2RY2 were measured using enzyme-l

Purinergic signaling in dengue infection: modulation of P2 receptors as a potential therapeutic strategy.

Dengue is a globally widespread arboviral disease characterized by an intense inflammatory response that may progress to severe clinical manifestations, such as dengue shock syndrome and hemorrhagic fever. The lack of effective treatments underscores the urgent need to identify new therapeutic targets. In this context, purinergic signaling-mediated by molecules such as ATP and adenosine-has emerged as a central regulator of immune, inflammatory, and vascular responses during dengue virus (DENV) infection. Activation of purinergic receptors, particularly P2X7, directly influences viral replicat

Altered purinergic P2X7 and A2B receptors signaling limits macrophage-mediated host defense in schistosomiasis.

BACKGROUND: The occurrence of co-infections during schistosomiasis, a neglected tropical disease, with other parasites have been reported suggesting an impaired host immune defense. Macrophage purinergic P2X7 receptor (P2X7R) plays an important role against intracellular pathogens. Therefore, we investigated the P2X7R-mediated phagocytosis and killing capacity of Leishmania amazonensis by macrophages during schistosomiasis in vitro and in vivo. METHODS: Swiss and C57BL/6 (Wild type) and P2X7R-/- were randomized in two groups: control (uninfected) and Schistosoma mansoni-infected. Alternativel

Purinergic Receptors in Basal Ganglia Diseases: Shared Molecular Mechanisms between Huntington's and Parkinson's Disease.

Huntington's (HD) and Parkinson's diseases (PD) are neurodegenerative disorders caused by the death of GABAergic and dopaminergic neurons in the basal ganglia leading to hyperkinetic and hypokinetic symptoms, respectively. We review here the participation of purinergic receptors through intracellular Ca2+ signaling in these neurodegenerative diseases. The adenosine A2A receptor stimulates striatopallidal GABAergic neurons, resulting in inhibitory actions on GABAergic neurons of the globus pallidus. A2A and dopamine D2 receptors form functional heteromeric complexes inducing allosteric inhibiti

Purinergic receptors in the carotid body as a new drug target for controlling hypertension.

In view of the high proportion of individuals with resistance to antihypertensive medication and/or poor compliance or tolerance of this medication, new drugs to treat hypertension are urgently needed. Here we show that peripheral chemoreceptors generate aberrant signaling that contributes to high blood pressure in hypertension. We discovered that purinergic receptor P2X3 (P2rx3, also known as P2x3) mRNA expression is upregulated substantially in chemoreceptive petrosal sensory neurons in rats with hypertension. These neurons generate both tonic drive and hyperreflexia in hypertensive (but not

Epigallocatechin gallate attenuates interstitial cystitis in human bladder urothelium cells by modulating purinergic receptors.

BACKGROUND: Epigallocatechin gallate (EGCG) has exhibited antitumor properties against bladder cancer. However, its effects in interstitial cystitis (IC) have not been investigated. METHODS: Here, we performed repeated cystoscopy and re-biopsy of bladder mucosa before and after intravesical irrigation of EGCG in eight patients diagnosed with IC based on clinical and histopathologic assessments. Six normal bladder tissue samples were obtained from age-, race-, and sex-matched asymptomatic control subjects. IC symptom index was used to compare the therapeutic effect in IC patients. Contribution of two ionotropic purinergic receptors to ATP responses in submandibular gland ductal cells.

The effect of extracellular ATP on salivary gland function was compared in wild-type (WT) and P2X(7) knockout (KO) mice. The increase in the intracellular concentration of calcium ([Ca(2+)](i)) in response to carbachol was similar in submandibular ductal cells of WT and KO mice. ATP and its analog, benzoyl-ATP, induced a sustained increase in the [Ca(2+)](i) in WT animals. In KO mice, ATP slightly and transiently increased the [Ca(2+)](i) and benzoyl-ATP had no effect. The response to ATP of WT but not KO mice was blocked by KN-62, Coomassie blue and magnesium. The small response of ATP observ

Joannesia princeps Vell. inhibits purinergic P2Y receptors and presents antinociceptive and anti-inflammatory effects.

P2Y2, P2Y4, and P2Y6 are purinergic receptors physiologically activated by uridine nucleotides. These metabotropic Gq-coupled receptors induce intracellular calcium mobilization and are expressed in various cell types, particularly leukocytes. Their activation triggers proinflammatory responses, playing a critical role in the progression of inflammatory diseases, nociception, and cancer. However, the absence of selective antagonists limits their clinical application, encouraging the search for novel compounds. In this study, we investigated the antagonistic effects of dichloromethane-methanol

Increased Purinergic Responses Dependent on P2Y2 Receptors in Hepatocytes from CCl4-Treated Fibrotic Mice.

Inflammatory and wound healing responses take place during liver damage, primarily in the parenchymal tissue. It is known that cellular injury elicits an activation of the purinergic signaling, mainly by the P2X7 receptor; however, the role of P2Y receptors in the onset of liver pathology such as fibrosis has not been explored. Hence, we used mice treated with the hepatotoxin CCl4 to implement a reversible model of liver fibrosis to evaluate the expression and function of the P2Y2 receptor (P2Y2R). Fibrotic livers showed an enhanced expression of P2Y2R that eliminated its zonal distribution. H

Purinergic receptors and regulatory volume decrease in seabream (Sparus aurata) hepatocytes: a videometric study.

The response of isolated hepatocytes of Sparus aurata to hypotonic stress was studied by the aid of videometric methods with the aim to investigate the possible involvement of ATP in the regulatory volume decrease (RVD). This study confirms our previous observations showing the ability of these cells to undergo RVD. In addition, it shows that the homeostatic response was inhibited by apyrase, an ATP scavenger, thus suggesting the involvement of extracellular ATP in the RVD response. Experiments performed in the presence of ATPγS or adenosine, agonists of P(2) and P(1) receptors respectively, a

Spontaneous Ca2+ oscillations via purinergic receptors elicit transient cell swelling in rat parotid ducts.

Rat parotid ductal cells were found to exhibit spontaneous Ca(2+) oscillations. These oscillatory Ca(2+) responses were observed during continuous perfusion with physiological salt solution at 37 degrees C in the absence of calcium mobilizing agonist stimulation. These Ca(2+) oscillations were completely blocked by the purinergic receptor inhibitors, pyridoxal phosphate-6-azo (benzene-2,4-disulfonic acid) (PPADS) and suramin, but were not blocked by the muscarinic antagonist, atropine, nor the alpha-adrenergic antagonist, phentolamine. Simultaneous observation with fura-2 fluorescence and diff

Nonselective cation conductance activated by muscarinic and purinergic receptors in rat spiral ganglion neurons.

The present study characterizes the ionic conductances activated by acetylcholine (ACh) and ATP, two candidate neuromodulators, in isolated spiral ganglion neurons (SGNs). Brief application (1 s) of ACh evoked in a dose-dependent manner (EC(50) = 4.1 microM) a reversible inward current with a long latency (average 1.3 s), at holding potential (V(h)) = -50 mV. This current was reversibly blocked by atropine and mimicked by muscarine. Application of ATP also evoked a reversible inward current at V(h) = -50 mV, but the current showed two components. A fast component with a short latency was large

Purinergic receptor activation rectifies autism-associated endothelial dysfunction.

Recent evidence in a 16p11.2 deletion mouse model of autism spectrum disorder (ASD) revealed brain endothelial abnormalities postnatally, but the endothelial alterations eliciting these changes remained unknown. Using 14-day-old and adult 16p11.2-deficient and wild-type male mice, we now show that the 16p11.2 deletion causes a bioenergetic failure selectively in endothelial cells (ECs) with reduced intracellular ATP levels. Intra- or extracellular ATP supplementation rescued the function of 16p11.2-deficient ECs in vitro via P2 purinergic receptor activation, specifically P2Y2 receptors. Activ

Possible Roles for Purinergic Receptor P2RX4 in Breast and Prostate Cancers.

The purinergic receptor P2RX4 contributes to the malignant behavior of breast and prostate cancers. It is upregulated in these malignancies and promotes tumor progression through mechanisms involving EMT, autophagy, and the release of pro-malignant lysosomal contents. P2RX4 also influences cellular signaling pathways by interacting with oncogenes and tumor suppressors. Certain genetic variants of P2RX4 are associated with an increased risk of developing these cancers. Hence, targeting P2RX4 represents a promising therapeutic approach, with potential strategies including antibody and CAR-T cell

Purinergic signaling through the P2Y2 receptor regulates osteocytes' mechanosensitivity.

Osteocytes' response to dynamic loading plays a crucial role in regulating the bone mass but quickly becomes saturated such that downstream induction of bone formation plateaus. The underlying mechanisms that downregulate osteocytes' sensitivity and overall response to loading remain unknown. In other cell types, purinergic signaling through the P2Y2 receptor has the potential to downregulate the sensitivity to loading by modifying cell stiffness through actin polymerization and cytoskeleton organization. Herein, we examined the role of P2Y2 activation in regulating osteocytes' mechanotransduc

The Purinergic Receptor P2X4 Promotes Th17 Activation and the Development of Arthritis.

Purinergic signaling plays a major role in T cell activation leading to IL-2 production and proliferation. However, it is unclear whether purinergic signaling contributes to the differentiation and activation of effector T cells. In this study, we found that the purinergic receptor P2X4 was associated with human Th17 cells but not with Th1 cells. Inhibition of P2X4 receptor with the specific antagonist 5-BDBD and small interfering RNA inhibited the development of Th17 cells and the production of IL-17 by effector Th17 cells stimulated via the CD3/CD28 pathway. Our results showed that P2X4 was

The P2X7 purinergic receptor: An emerging therapeutic target in cardiovascular diseases.

The P2X7 purinergic receptor, a calcium permeable cationic channel, is activated by extracellular ATP. Most studies show that P2X7 receptor plays an important role in the nervous system diseases, immune response, osteoporosis and cancer. Mounting evidence indicates that P2X7 receptor is also associated with cardiovascular disease. For example, the P2X7 receptor activated by ATP can attenuate myocardial ischemia-reperfusion injury. By contrast, inhibition of P2X7 receptor decreases arrhythmia after myocardial infarction, prolongs cardiac survival after a long term heart transplant, alleviates t

Pirt reduces bladder overactivity by inhibiting purinergic receptor P2X3.

Pirt is a transmembrane protein predominantly expressed in peripheral neurons. However, the physiological and pathological roles of Pirt in hollow viscus are largely unknown. Here we show that Pirt deficiency in mice causes bladder overactivity. The density of α,β-meATP-induced currents is significantly reinforced in Pirt-deficient dorsal root ganglion (DRG) neurons. Pirt and P2X3 receptor co-localize in bladder nerve fibres and heterologous Pirt expression significantly reduces P2X3-mediated currents. Pirt interacts with P2X3 through the N-terminal 14 amino-acid residues. TAT-conjugated Pirt(

Functional expression of purinergic P2 receptors and transient receptor potential channels by the human urothelium.

In addition to its role as a physical barrier, the urothelium is considered to play an active role in mechanosensation. A key mechanism is the release of transient mediators that activate purinergic P2 receptors and transient receptor potential (TRP) channels to effect changes in intracellular Ca²⁺. Despite the implied importance of these receptors and channels in urothelial tissue homeostasis and dysfunctional bladder disease, little is known about their functional expression by the human urothelium. To evaluate the expression and function of P2X and P2Y receptors and TRP channels, the human
