Reports of fingertip regrowth in children are interpreted in the perspective of epimorphic regeneration in lower forms and in relation to a few other examples in mammals. The latter include cases of antler replacement in deer as well as ingrowth from the margins of holes cut in bat wing membranes and in the external ears of rabbits, pikas, cats, and echolocating bats. It is suggested that the relative inadequacy of regeneration in warm-blooded vertebrates may be attributed to the precocity with which they tend to form dermal scars in healing wounds, scars that are believed to preclude blastema

In mammals, multi-tissue regeneration is largely restricted to the distal portion of the digit tip and involves the formation of a blastema, a transient, proliferating cell mass that reforms the diverse tissues of the digit. Historically little was known about the mammalian blastema but with recent advances in single cell transcriptomic approaches and genetic lineage tracing, a more precise understanding of this critical structure has begun to emerge. In this review we summarise the cellular mechanisms underlying adult mammalian digit tip regeneration. We posit that understanding how some mamm

Zebrafish have the ability to regenerate skeletal structures, including the fin, skull roof, and jaw. Although fin regeneration proceeds by epimorphic regeneration, it remains unclear whether this process is involved in other skeletal regeneration in zebrafish. Initially in epimorphic regeneration, the wound epidermis covers the wound surface. Subsequently, the blastema, an undifferentiated mesenchymal mass, forms beneath the epidermis. In the present study, we re-examined the regeneration of the zebrafish lower jaw in detail, and investigated whether epimorphic regeneration is involved in thi

The peripheral branch of sensory dorsal root ganglion (DRG) neurons regenerates readily after injury unlike their central branch in the spinal cord. However, extensive regeneration and reconnection of sensory axons in the spinal cord can be driven by the expression of α9 integrin and its activator kindlin-1 (α9k1), which enable axons to interact with tenascin-C. To elucidate the mechanisms and downstream pathways affected by activated integrin expression and central regeneration, we conducted transcriptomic analyses of adult male rat DRG sensory neurons transduced with α9k1, and controls, with

Pediatric intestinal failure resulting from short bowel syndrome remains a significant source of morbidity and mortality despite advances in parenteral nutrition and multidisciplinary intestinal rehabilitation. While native intestinal adaptation provides a foundation for recovery in many patients, those with the most severe forms of short bowel syndrome face limited therapeutic options, including lifelong parenteral nutrition dependence, surgical lengthening procedures, and intestinal transplantation-each associated with substantial complications. The field of bowel regeneration has expanded d

Regenerative endodontics (REs) has redefined the management of immature permanent teeth with necrotic pulps by shifting the objective from simple disinfection to biologically driven root maturation and restoration of the dentine-pulp complex. A critical component of this process is the selection of intracanal medicaments that can effectively eliminate microbial infection while preserving the viability of stem cells required for regeneration. This narrative review evaluates the biological and antimicrobial performance of commonly used medicaments, particularly calcium hydroxide (Ca(OH)₂) and tr

BACKGROUND: Alopecia is a prevalent condition correlated with significant psychosocial challenges, while contemporary treatments exhibit variable efficacy, require extended use, and may cause adverse effects. This review aimed to comprehensively assess exosome-mediated interventions as a novel cell-free regenerative approach for hair regeneration across multiple alopecia subtypes.
METHODS: A narrative-scoping review was performed utilizing PubMed, Scopus, and Web of Science databases, identifying 80 preclinical and clinical studies. Eligible studies evaluated exosome-based mechanisms, deliver

After an injury, adult mammals do not regenerate or repair their central nervous system in general, and their spinal cord in particular, leading to life-long disabilities. However, many aquatic species such as zebrafish, salamanders and frogs, as well as some land reptiles (turtles and lizards) have the remarkable ability to regenerate their spinal cords. In the case of anurans such as Xenopus, this is a transient ability that disappears after metamorphosis, making it an interesting model with accessible regenerative and nonregenerative stages within the same species. Here, we will summarize o

The thymus, as a vital immune organ in the human body, provides the essential microenvironment for the development, differentiation, and maturation of T lymphocytes. With advancing age, the thymus gradually undergoes atrophy and degeneration, primarily characterized by structural disruption of thymic epithelial cells, leading to slowed T cell development and reduced output. Experimental studies have identified multiple regenerative pathways, including IL-7, KGF, BMP4, IL-22, RANKL, regulatory T cells, FOXN1-directed approaches, and sex-steroid blockade. Recent human observational studies also

Peptide nanofibers have been attractive targets for regenerative medicine applications due to their tailorability to be easily functionalized for specific bioactivity, biocompatibility, ease of synthesis, adjustability of their physicochemical characteristics, and lack of biological contamination. Research groups have investigated their use for the regeneration of various tissues, such as bone, cartilage, brain, peripheral nerves, cardiac tissue, vascular tissues, endocrine cells, muscles, etc., for the treatment of degenerative diseases or tissue loss due to accidents or aging. Wound healing

There are high expectations regarding heart regeneration for refractory heart failure (HF). Transplantation of human pluripotent stem cell (hPSC)-derived cardiomyocytes (CMs) is expected to replace CMs lost due to HF, and various studies have been conducted to apply this therapy clinically. Though issues such as arrhythmias and immune rejection remain, the mass production of purified hPSC-derived CMs, their efficient transplantation, and methods to improve their engraftment pushed up the transplantation of hPSC-derived CMs to the clinical stage. In contrast, a direct cardiac reprogramming meth

Skeletal muscle has an innate regenerative capacity to restore their structure and function following acute damages and injuries. However, in congenital muscular dystrophies, large volumetric muscle loss, cachexia, or aging, the declined regenerative capacity of skeletal muscle results in muscle wasting and functional impairment. Recent studies indicate that muscle mass and function are closely correlated with morbidity and mortality due to the large volume and location of skeletal muscle. However, the options for treating neuromuscular disorders are limited. Biomedical engineering strategies

The mortality due to heart diseases remains highest in the world every year, with ischemic cardiomyopathy being the prime cause. The irreversible loss of cardiomyocytes following myocardial injury leads to compromised contractility of the remaining myocardium, adverse cardiac remodeling, and ultimately heart failure. The hearts of adult mammals can hardly regenerate after cardiac injury since adult cardiomyocytes exit the cell cycle. Nonetheless, the hearts of early neonatal mammals possess a stronger capacity for regeneration. To improve the prognosis of patients with heart failure and to fin

Liver cirrhosis is the final stage of continuous hepatic inflammatory activity derived by viral, metabolic or autoimmune origin. In the last years, cirrhosis was considered a unique and static condition; recently was accepted some patients subgroups with different liver injury degrees that coexist under the same diagnosis, with implications about the natural disease history. The liver growth factor (LGF) is a potent in vivo and in vitro mitogenic agent and an inducer of hepatic regeneration (HR) through the hepatocytes DNA synthesis. The clinical implications of the LGF levels in cirrhosis, ar

Uniquely among adult tissues, the human endometrium undergoes cyclical shedding, scar-free repair and regeneration during a woman's reproductive life. Therefore, it presents an outstanding model for study of such processes. This Review examines what is known of endometrial repair and regeneration following menstruation and parturition, including comparisons with wound repair and the influence of menstrual fluid components. We also discuss the contribution of endometrial stem/progenitor cells to endometrial regeneration, including the importance of the stem cell niche and stem cell-derived extr

INTRODUCTION: A sound and vital pulp is an essential prerequisite for long-term tooth survival and preservation. However, current endodontic treatment concepts are based on the removal of inflamed or necrotic pulp tissue and the replacement by a synthetic biomaterial. Recently, total or partial pulp regeneration has been proposed as an alternative treatment concept. The aim of this review was to evaluate the current options of pulp treatment and regenerative approaches, both for immature and mature teeth, in a clinical context.
METHODS: Clinical success rates of classic treatment options such

Stem cell research plays a central role in the future of medicine, which is mainly dependent on the advances on regenerative medicine (RM), specifically in the disciplines of tissue engineering (TE) and cellular therapeutics. All RM strategies depend upon the harnessing, stimulation, or guidance of endogenous developmental or repair processes in which cells have an important role. Among the most clinically challenging disorders, cartilage degeneration, which also affects subchondral bone becoming an osteochondral (OC) defect, is one of the most demanding. Although primary cells have been clini

PURPOSE OF REVIEW: The goal of this review is to summarize the unique regenerative milieu within mature mammalian extraocular muscles (EOMs). This will aid in understanding disease propensity for and sparing of EOMs in skeletal muscle diseases as well as the recalcitrance of the EOM to injury.
RECENT FINDINGS: The EOMs continually remodel throughout life and contain an extremely enriched number of myogenic precursor cells that differ in number and functional characteristics from those in limb skeletal muscle. The EOMs also contain a large population of Pitx2-positive myogenic precursor cells
