Effects of acute and chronic aripiprazole treatment on choice between cocaine self-administration and food under a concurrent schedule of reinforcement in rats.

RATIONALE: Dopamine D2-like partial agonists such as aripiprazole have received some attention as potential pharmacotherapies for the treatment of psychostimulant addiction. However, the preclinical evaluations so far have focused on acute effects of aripiprazole. OBJECTIVES: We tested the hypothesis that aripiprazole, both as acute and as chronic treatment, would preferentially decrease cocaine self-administration while sparing behavior maintained by a natural reinforcer, resulting in a shift in the allocation of behavior from cocaine-taking towards the alternative reinforcer. MATERIALS AND

Selective dopamine D3 receptor partial agonist (±)VK4-40 reduces the reinforcing strength of d-amphetamine but not cocaine in rhesus monkeys responding under a progressive-ratio schedule of reinforcement.

BACKGROUND: Although countless studies have aimed to identify and test novel therapeutics for stimulant misuse, there are still no FDA-approved pharmacotherapies for stimulant use disorders. One potential treatment target is the dopamine D3 receptor (D3R) and studies in rodents have suggested that the novel D3R partial agonist (±)VK4-40 may be effective at decreasing cocaine self-administration. However, no previous studies have examined the efficacy of (±)VK4-40 in reducing cocaine self-administration in nonhuman primates nor the generality of effects by examining self-administration of other

Timing and space usage are disrupted by amphetamine in rats maintained on DRL 24-s and DRL 72-s schedules of reinforcement.

RATIONALE: A differential-reinforcement-of-low-rate schedule (DRL) delivers reinforcement only when the interresponse time (IRT) exceeds a fixed time interval, thereby shaping rats to discriminate the timing of their responses. However, little is known about the motor behavior and location of the rats in the chamber during the IRTs that lead to reinforcement. Although amphetamine is known to disrupt DRL timing behavior, the effects of this drug on non-operant motor behavior during DRL performance has not yet been quantified. OBJECTIVE: The purpose of this research was to measure the motor beh

Does the learning deficit observed under an incremental repeated acquisition schedule of reinforcement in Ts65Dn mice, a model for Down syndrome, change as they age?

The Ts65Dn mouse is partly trisomic at chromosome 16 and is considered to be a valid mouse model of human Down syndrome. Prior research using an incremental repeated acquisition (IRA) schedule of reinforcement has revealed that there is a significant learning deficit in young, adult Ts65Dn mice compared to littermate controls. The purpose of this study was to examine whether this deficit changes during the life-span of these mice. In order to determine if changes in the deficit were caused by motoric or motivational deficiencies, a second group of mice was trained to respond under a performanc

Quantitative analysis of performance on a progressive-ratio schedule: effects of reinforcer type, food deprivation and acute treatment with Δ⁹-tetrahydrocannabinol (THC).

Rats' performance on a progressive-ratio schedule maintained by sucrose (0.6M, 50 μl) and corn oil (100%, 25 μl) reinforcers was assessed using a model derived from Killeen's (1994) theory of schedule-controlled behaviour, 'Mathematical Principles of Reinforcement'. When the rats were maintained at 80% of their free-feeding body weights, the parameter expressing incentive value, a, was greater for the corn oil than for the sucrose reinforcer; the response-time parameter, δ, did not differ between the reinforcer types, but a parameter derived from the linear waiting principle (T0), indicated th

Deep Reinforcement Learning-Based Energy-Efficient Resource Allocation and Scheduling in 6G-Enabled UAV-Assisted IoT Wireless Networks.

Unmanned Aerial Vehicles (UAVs) have emerged as a flexible, cost-effective solution for connecting Internet of Things (IoT) devices where traditional infrastructure falls short. However, managing their limited energy alongside the diverse demands of densely deployed devices makes resource allocation a genuinely hard problem. This paper presents a Deep Reinforcement Learning (DRL) framework that jointly optimizes user scheduling, IoT device transmit power, bandwidth, and UAV movement in a 6G-enabled UAV-relay uplink network, using a deterministic large-scale air-to-ground path-loss channel mode

60 Hz electric fields and incandescent light as aversive stimuli controlling the behavior of rats responding under concurrent schedules of reinforcement.

Several reports have shown that animals will sometimes engage in behaviors that reduce their exposure to a 60 Hz electric field (E-field). The field, therefore, can function as an aversive stimulus. In other studies, the E-field at equivalent strengths failed to function as an aversive stimulus. The present experiment, using rats, demonstrates how factors other than field strength can influence whether a subject engages in behavior that reduces field exposure. The general design consisted of giving the rat a choice between two alternatives, one of which sometimes included an added stimulus. Ea

The reinforcing efficacy of the dopamine reuptake inhibitor 2beta-propanoyl-3beta-(4-tolyl)-tropane (PTT) as measured by a progressive-ratio schedule and a choice procedure in rhesus monkeys.

The present series of experiments was undertaken to investigate the variables that influence the reinforcing efficacy of psychostimulants. The time of onset for dopamine transporter (DAT) occupancy of the long-acting, high-affinity DAT blocker 2beta-propanoyl-3beta-(4-tolyl)-tropane (PTT) was measured using an ex vivo binding assay in rodents and was determined to be significantly longer than for cocaine (30 min versus 2 min). To assess the reinforcing efficacy of PTT relative to cocaine, a discrete-trials drug-drug choice procedure (n = 3) and a progressive-ratio (PR) schedule (n = 4) were us

Access to Arbitrary Stimuli During Fixed-Lean Schedule Thinning.

UNLABELLED: Fixed-lean schedule thinning involves programming the terminal reinforcement schedule from the onset of schedule thinning rather than gradually reducing the delay to reinforcement over time in a dense-to-lean approach. This study extended Sumter et al. (2020) and investigated the effect of continuous access to attention (an arbitrary stimulus; not functionally related to challenging behavior) during fixed-lean schedule thinning for individuals who engaged in challenging behavior maintained by tangible reinforcement or a combination of tangible reinforcement and escape. In some sess

Schedule-induced licking during multiple schedules.

Schedule-induced polydipsia was studied in rats bar pressing under two-component multiple schedules of food reinforcement. The first component of the multiple schedule was a variable-interval 1-min schedule throughout the experiment. The schedule comprising the second component was varied over blocks of sessions in terms of rate and magnitude of reinforcement, and was either variable-interval 3-min (one pellet), variable-interval 3-min (three pellets), variable-interval 1-min (one pellet), or extinction. Water intake per session varied with the rate of reinforcement in the schedule comprising

Interlocking schedules: the relationship between response and time requirements.

Rats were exposed to an interlocking fixed-ratio 150 fixed-interval 5-minute schedule of food reinforcement and then to yoked variable-ratio schedules in which individual ratios corresponded exactly to the ratios of responses to reinforcement obtained on the interlocking schedule. After additional training with the interlocking schedule, the rats were exposed to yoked variable-interval schedules in which intervals corresponded to the intervals between successive reinforcements obtained on the second interlocking schedule. Response rates were highest in the yoked VR condition and lowest in the

Effects of monoamine oxidase inhibitors on performance during differential reinforcement of low response rate.

The effects of three monoamine oxidase inhibitors (MAOI) on performance under a differential-reinforcement-of-low-rate 72-s schedule (DRL 72-s) for water reinforcement were determined. All three drugs (isocarboxazid, iproniazid, phenelzine) reduced response rate and increased reinforcement rate in rats performing under the DRL schedule. Drugs from other classes (alcohol, chlordiazepoxide, morphine, pentobarbital) did not produce similar effects. The ability of MAOI to increase reinforcement rate under a DRL 72-s schedule is similar to that recently reported for tricyclic antidepressants and th

Sequential contrast effects with retarded subjects after discrimination learning with and without errors.

Four retarded adults were exposed to a multiple variable interval-extinction schedule of reinforcement. The 2 min components were presented in a random order. Two subjects acquired the discrimination with an errorless procedure. The two other subjects learned the discrimination with errors. The schedule generated sequential contrast effects under both training procedures: response rates during periods of reinforcement were higher when a reinforcement period followed an extinction period than when it followed another reinforcement period. These results were confirmed in a second observation whe

Conditioned reinforcement value and resistance to change.

Three experiments examined the effects of conditioned reinforcement value and primary reinforcement rate on resistance to change using a multiple schedule of observing-response procedures with pigeons. In the absence of observing responses in both components, unsignaled periods of variable-interval (VI) schedule food reinforcement alternated with extinction. Observing responses in both components intermittently produced 15 s of a stimulus associated with the VI schedule (i.e., S+). In the first experiment, a lower-valued conditioned reinforcer and a higher rate of primary reinforcement were ar

Concurrent fixed-interval variable-ratio schedules and the matching relation.

Five rats responded under concurrent fixed-interval variable-ratio schedules of food reinforcement. Fixed-interval values ranged from 50-seconds to 300-seconds and variable-ratio values ranged from 30 to 360; a five-second changeover delay was in effect throughout the experiment. The relations between reinforcement ratios obtained from the two schedules and the ratios of responses and time spent on the schedules were described by Baum's (1974) generalized matching equation. All subjects undermatched both response and time ratios to reinforcement ratios, and all subjects displayed systematic bi

Concurrent ratio schedules: Fixed vs. variable response requirements.

Rats were trained on concurrent fixed-ratio variable-ratio or concurrent fixed-ratio mixed-ratio schedules of food reinforcement. The variable-ratio schedule was composed of an arithmetic sequence of 11 ratios that averaged 50; the mixed-ratio schedule consisted of equiprobable ratios of 1 and 99. Fixed-ratio values, varied over experimental conditions, included 25, 35, 50, 60, and 99. The proportion of responses and time allocated to the variable- or mixed-ratio schedule increased as the size of the fixed ratio increased. For most subjects, higher proportions of responses and time were mainta

Effects of ethanol on behavior under fixed-ratio, fixed-interval, and multiple fixed-ratio fixed-interval schedules in the pigeon.

Behavior of pigeons was controlled by fixed-ratio, fixed-interval and multiple fixed-ratio fixed-interval schedules of reinforcement. When each thirtieth peck on a response key produced food (fixed-ratio schedule) high rates of responding were maintained; low to intermediate doses of ethanol left unchanged or increased those rates. Under the fixed-interval schedule the first response after 5 min produced food and comparatively lower rates of responding were maintained that gradually increased up to food presentation. Low to intermediate doses of ethanol only decreased overall rates of respondi

Pituitary-adrenal correlates of schedule-induced polydipsia and wheel running in rats.

To investigate the pituitary-adrenal correlates of schedule-induced behaviours, rats were submitted to a fixed-time 60-s schedule of food reinforcement, with free access to either a water spout or a running wheel. Schedule-induced water drinking (polydipsia) was accompanied by an important drop in plasma corticosterone levels, while wheel running resulted in a significant increase in plasma corticosterone concentrations. This contrast between the pituitary-adrenal consequences of schedule-induced drinking and wheel running was not due to functional differences between the two activities since

The effects of variable-time delivery of food items and praise on problem behavior reinforced by escape.

Prior research indicates that reinforcement of an appropriate response (e.g., compliance) can produce concomitant reductions in problem behavior reinforced by escape when problem behavior continues to produce negative reinforcement (e.g., Lalli et al., 1999). These effects may be due to a preference for positive over negative reinforcement or to positive reinforcement acting as an abolishing operation, rendering demands less aversive and escape from demands less effective as negative reinforcement. In the current investigation, we delivered a preferred food item and praise on a variable-time 1

5-HT1B receptor agonist enhances breakpoint for cocaine on a progressive ratio (PR) schedule during maintenance of self-administration in female rats but reduces breakpoint for sucrose.

Background: Previous research showed that the 5-HT1B receptor agonist CP94253 enhanced cocaine reinforcement rate during maintenance of daily self-administration (SA), but inhibited reinforcement rate after 21 days of abstinence in male rats. Here we examined whether female rats show similar effects of CP94253 during maintenance as males across estrous cycle phases. Methods: Female rats trained on a fixed ratio 5 (FR5) cocaine reinforcement schedule were tested for the effects of CP94253 (5.6 mg/kg, s.c.) on cocaine reinforcement rate during each phase of the estrous cycle, with access to eith
