Optimization of metals and rare earth elements leaching from spent Ni-MH batteries by response surface methodology.

The rechargeable battery market has almost doubled in 15 years. Regardless of the type of batteries, their limited lifespan means that sooner or later they will constitute a mass of waste whose management is problematic as their content is high in elements and metals of high economic interest, but also toxic to the environment. This project is to optimize the solubilization conditions for rare earth elements (REEs) and other metals from waste nickel-metal hydride (Ni-MH) batteries. The Ni-MH battery powder used contained the following main elements: Ni (548 g/kg), La (45 g/kg), Co (32 g/kg), Z

Altered gene expression in late-onset Alzheimer's disease due to SNPs within 3'UTR microRNA response elements.

Late-onset Alzheimer's disease (LOAD) is a progressive and fatal neurodegenerative disease found in people older than 65years of age. Disease etiology is complex, as susceptibility has been linked to multiple gene variants conferred by single nucleotide polymorphisms (SNPs). However, the molecular mechanisms by which SNPs contribute to LOAD pathogenesis have not been extensively studied, particularly for SNPs within the 3' untranslated regions (3'UTRs), the hubs for microRNA binding. Therefore, we screened for SNPs within the 3'UTRs of LOAD-associated genes that may create or destroy microRNA

Linking polymorphic p53 response elements with gene expression in airway epithelial cells of smokers and cancer risk.

Chronic cigarette smoking exposes airway epithelial cells to thousands of carcinogens, oxidants and DNA-damaging agents, creating a field of molecular injury in the airway and altering gene expression. Studies of cytologically normal bronchial epithelial cells from smokers have identified transcription-based biomarkers that may prove useful in early diagnosis of lung cancer, including a number of p53-regulated genes. The ability of p53 to regulate transcription is critical for tumor suppression, and this suggests that single-nucleotide polymorphisms (SNPs) in functional p53 binding sites (p53

Effect of Europium and Gadolinium Alloying Elements on the Tribological Response of Low Hydrogen Content Amorphous Carbon.

Dopants and alloying elements are commonly introduced in amorphous carbon (a-C) materials to tailor their mechanical and tribological properties. While most published studies have focused on doping and alloying a-C coatings with metals or metalloids, doping a-C films with rare-earth elements has only recently been explored. Notably, our understanding of the shear-induced structural changes occurring in rare-earth-element-containing a-C films is still elusive, even in the absence of any liquid lubricants. Here, the friction response of Eu- and Gd-containing a-C films with low hydrogen content d

An octamer element functions as a regulatory element in the differentiation-responsive CD11c integrin gene promoter: OCT-2 inducibility during myelomonocytic differentiation.

The integrin CD11c/CD18 mediates leukocyte adhesion to endothelium and other cell types and is a receptor for LPS, iC3b, and fibrinogen. CD11c expression is restricted to myeloid and activated B cells, is regulated during leukocyte differentiation, and constitutes a diagnostic tool for hairy cell leukemia. Mapping of in vivo DNA-protein interactions in the CD11c proximal promoter revealed three adjacent myeloid-specific interactions, one of which lies on an octamer consensus sequence, ATTT GCAT (Oct185). Oct185 disruption increased the CD11c promoter activity while decreasing its myeloid diffe

Identification of positive and negative placenta-specific basal elements and a cyclic adenosine 3',5'-monophosphate response element in the human gene for P450scc.

The chronic regulation of steroiodgenesis is mediated principally by transcriptional regulation of the genes encoding the various steroidogenic enzymes. The cholesterol side-chain cleavage enzyme, P450scc, is rate limiting and hormonally regulated in a tissue-specific fashion. Human placental steroidogenesis is regulated by LH and hCG through increased intracellular cAMP, and forskolin and 8-bromo-cAMP increase the abundance of human P450scc mRNA in human JEG-3 choriocarcinoma cells. We transfected JEG-3 cells with 24 promoter/reporter constructions to examine the tissue-specific and hormonall

Alterations in phosphorylated cAMP response element-binding protein (pCREB) signaling: an endophenotype of lithium-responsive bipolar disorder?

OBJECTIVES: Abnormalities of signal transduction are considered among the susceptibility factors for bipolar disorder (BD). These include changes in G-protein-mediated signaling and subsequent modification of gene expression via transcription factors such as cAMP response element-binding protein (CREB). METHODS: We investigated levels of CREB in lymphoblasts from patients with BD, all responders to lithium prophylaxis (n = 13), and healthy control subjects (n = 15). Phosphorylated CREB (pCREB) was measured by immunoblotting in subjects with BD (n = 15) as well as in their affected (n = 17) an

Biomechanical response of human skull to impact by three-dimensional finite element analysis.

Traumatic Brain Injury resulting from the impact of the human skull is one of the leading causes of death worldwide. The knowledge of the biomechanics of such injuries is essential for the effective design of protective headgear. This study investigates the biomechanical responses of a human skull subjected to an impact with a rigid wall. Finite element analyses were performed by impacting various skull locations, and the utility concept approach was introduced to rank skull vulnerability. Time history plots of the velocity, acceleration and energy were extracted to determine the dynamic respo

A multi-body finite element model for hydrogel packings: linear response to shear.

We study a multi-body finite element model of a packing of hydrogel particles using the Flory-Rehner constitutive law to model the deformation of the swollen polymer network. We show that while the dependence of the pressure, Π, on the effective volume fraction, ϕ, is virtually identical to a monolithic Flory material, the shear modulus, μ, behaves in a non-trivial way. μ increases monotonically with Π from zero and remains below about 80% of the monolithic Flory value at the largest Π we study here. The local shear strain in the particles has a large spatial variation. Local strains near the

Molecular characterization and identification of the E2/P4 response element in the porcine HOXA10 gene.

Homeobox A10 (HOXA10), a well-known transcriptional factor that can be regulated by estrogen (E(2)) and progesterone (P(4)), is necessary not only for endometrial differentiation but also for establishing the conditions required for implantation. However, little research has focused on the regulation of the porcine HOXA10 gene. In this study, we aimed to (1) characterize the genetic structure of the porcine HOXA10 gene, (2) analyze the tissue expression pattern and differential expression levels in the endometrium of HOXA10 in different developmental stages of Meishan and commercial Yorkshire

Trace element concentrations and bioindicator responses in tree swallows from northwestern Minnesota.

Extremely high concentrations of cadmium (3.5 microg/g dry wgt.) and elevated concentrations of chromium (>10 microg/g dry wgt.) and mercury (1.6 microg/g dry wgt.) were reported in waterbird tissues at Agassiz National Wildlife Refuge in northwestern Minnesota in 1994. Tree swallows (Tachycineta bicolor) were studied during 1998-2001 at three drainages into the Refuge, two pools on the Refuge, and at a nearby reference location to document whether high levels of contaminants were still present, and if so to quantify the source and severity of the contamination. Trace elements were measured in

Characterization of the human PPARalpha promoter: identification of a functional nuclear receptor response element.

PPARalpha is a nuclear receptor that controls lipid and glucose metabolism and exerts antiinflammatory activities. The factors regulating human PPARalpha (hPPARalpha) gene expression remain largely unexplored. To study the mechanisms controlling hPPARalpha expression, the hPPARalpha gene promoter was identified and characterized. First, an alternatively spliced exon within the 5'-untranslated region of the hPPARalpha gene was identified by RT-PCR. Next, the transcription start site was mapped and the hPPARalpha gene promoter was cloned and functionally analyzed. Because PPARalpha levels are el

Expression pattern of proteins that bind to the ultraviolet-responsive element (TGACAACA) in human keratinocytes.

We previously identified an ultraviolet (UV)-responsive element (URE; TGACAACA) that plays a role in both transcription and replication of polyoma sequences. Mouse polyclonal antibodies were raised against affinity-purified URE-bound proteins to characterize their expression patterns. These antibodies specifically recognized two of four URE-bound proteins, of 40 and 68 kDa. The 68-kDa protein was constitutively expressed in human keratinocytes, while the expression of the 40-kDa protein was induced by UV irradiation. Of the two, the 68-kDa protein bound to the URE with greater affinity than th

Putative cis-Regulatory Elements Predict Iron Deficiency Responses in Arabidopsis Roots.

Plant iron deficiency (-Fe) activates a complex regulatory network that coordinates root Fe uptake and distribution to sink tissues. In Arabidopsis (Arabidopsis thaliana), FER-LIKE FE DEFICIENCY-INDUCED TRANSCRIPTION FACTOR (FIT), a basic helix-loop-helix (bHLH) transcription factor (TF), regulates root Fe acquisition genes. Many other -Fe-induced genes are FIT independent, and instead regulated by other bHLH TFs and by yet unknown TFs. The cis-regulatory code, that is, the cis-regulatory elements (CREs) and their combinations that regulate plant -Fe-responses, remains largely elusive. Using A

The Nrf2-Antioxidant Response Element Signaling Pathway Controls Fibrosis and Autoimmunity in Scleroderma.

Systemic sclerosis (SSc) is an autoimmune disease with fibrosis of the skin and internal organs and vascular alterations. Dysregulations in the oxidant/antioxidant balance are known to be a major factor in the pathogenesis of the disease. Indeed, reactive oxygen species (ROS) trigger neoepitopes leading to a breach of immune tolerance and autoimmune responses, activate fibroblasts to proliferate and to produce excess of type I collagen. ROS also alter endothelial cells leading to vascular dysfunction. Glutathione (GSH) is the most potent antioxidant system in eukaryotic cells. Numerous studies

A hypermorphic antioxidant response element is associated with increased MS4A6A expression and Alzheimer's disease.

Late onset Alzheimer's disease (AD) is a multifactorial disorder, with AD risk influenced by both environmental and genetic factors. Recent genome-wide association studies (GWAS) have identified genetic loci associated with increased risk of developing AD. The MS4A (membrane-spanning 4-domains subfamily A) gene cluster is one of the most significant loci associated with AD risk, and MS4A6A expression is correlated with AD pathology. We identified a single nucleotide polymorphism, rs667897, at the MS4A locus that creates an antioxidant response element and links MS4A6A expression to the stress

The role of the carbohydrate response element-binding protein in male fructose-fed rats.

By 2030, nearly half of Americans will have nonalcoholic fatty liver disease. In part, this epidemic is fueled by the increasing consumption of caloric sweeteners coupled with an innate capacity to convert sugar into fat via hepatic de novo lipogenesis. In addition to serving as substrates, monosaccharides also increase the expression of key enzymes involved in de novo lipogenesis via the carbohydrate response element-binding protein (ChREBP). To determine whether ChREBP is a potential therapeutic target, we decreased hepatic expression of ChREBP with a specific antisense oligonucleotide (ASO)

A finite element model of region-specific response for mild diffuse brain injury.

It is well known that rotational loading is responsible for a spectrum of diffuse brain injuries spanning from concussion to diffuse axonal trauma. Many experimental studies have been performed to understand the pathological and biomechanical factors associated with diffuse brain injuries. Finite element models have also been developed to correlate experimental findings with intrinsic variables such as strain. However, a paucity of studies exist examining the combined role of the strain-time parameter. Consequently, using the principles of finite element analysis, the present study introduced

The identification and characterization of a FOXL2 response element provides insights into the pathogenesis of mutant alleles.

The Forkhead transcription factor FOXL2 plays a crucial role in ovarian development and maintenance. In humans, its mutations lead to craniofacial abnormalities, isolated or associated with ovarian dysfunction. Using a combinatorial approach, we identified and characterized a FoxL2 response element (FLRE) and showed that it is highly specific and that it diverges from that of other Forkhead transcription factors. This specificity should prevent aberrant regulation of FOXL2 targets by other members of the family and should prevent ectopic activation of the ovarian differentiation program in tes
