Molecular virology of hepatitis B virus and targets for antiviral intervention.

The members of the viral family Hepadnaviridae comprise one of the smallest enveloped DNA viruses and cause acute and chronic infections in mammals and birds, leading to large virus and antigen loads in the blood. They have a restricted host range and depend on differentiated hepatocytes for replication. Hepatitis B virus (HBV) is the prototype of the Hepadnaviridae. HBV can persist in infected hepatocytes and has evolved elaborate strategies to evade the immune system. HBV replicates like HIV (family of Retroviridae) via reverse transcription. Drugs licensed for inhibition of HIV reverse tran

Lacunar Infarction as a Neurological Manifestation of Human Immunodeficiency Virus (HIV): A Case Report.

Human immunodeficiency virus (HIV) belongs to the Lentivirus genus of the Retroviridae family. The virus primarily attacks CD4+ T-lymphocyte helper cells, which results in severe immunological suppression and ongoing cell death. HIV has several neurological manifestations, including meningitis, epileptic seizures, and stroke. A 35-year-old male presented with an eight-month history of fever, convulsion, and weight loss. Neurological exam was significant for bilateral upgoing plantar response and neck stiffness. The enzyme-linked immunosorbent assay (ELISA) was positive for HIV, and his brain M

Complement Evasion Strategies of Viruses: An Overview.

Being a major first line of immune defense, the complement system keeps a constant vigil against viruses. Its ability to recognize large panoply of viruses and virus-infected cells, and trigger the effector pathways, results in neutralization of viruses and killing of the infected cells. This selection pressure exerted by complement on viruses has made them evolve a multitude of countermeasures. These include targeting the recognition molecules for the avoidance of detection, targeting key enzymes and complexes of the complement pathways like C3 convertases and C5b-9 formation - either by enco

The Analysis of Oral and Fecal Virome Detects Multiple Novel Emerging Viruses in Snakes.

Wild animals are considered reservoirs for emerging and reemerging viruses, such as the novel severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). Previous studies have reported that bats and ticks harbored variable important pathogenic viruses, some of which could cause potential diseases in humans and livestock, while viruses carried by reptiles were rarely reported. Our study first conducted snakes' virome analysis to establish effective surveillance of potential transboundary emerging diseases. Consequently, Adenoviridae, Circoviridae, Retroviridae, and Parvoviridae were identifie

[Retroviruses and their importance in neurology].

Retroviruses comprise spumaviruses, lentiviruses and oncoviruses. After these taxonomic remarks neurologic-psychiatric-syndromes (multifocal leucencephalopathy, dementia-complex, vacuolar myelopathy and others) are described, that could be observed as manifestations of the "acquired immune deficiency syndrome". Some therapeutic measures are discussed, especially the administration of Zidovudine that shows good temporary results. Essentially Zidovudine works by misleading the Reserve Transkriptase by offering a "Fake-thymidine"--so misleading replication. Lastly the hypotheses concerning multip

Polymyositis in an immunodeficiency disease in monkeys induced by a type D retrovirus.

Fifty percent of primates with acquired immunodeficiency caused by a well-characterized type D retrovirus (SAIDS D) developed clinical, laboratory, and histologic features of polymyositis. By use of specific antisera and immunochemical techniques, we found the virus in the lymphoid cells surrounding muscle fibers and invading the endomysia septa. SAIDS D virus was isolated from the involved muscles and infected myotubes of normal muscle in tissue culture. These results suggest that retroviruses, a group of viruses increasingly associated with human diseases, can cause polymyositis with immunod

Review and future perspectives on the integration characteristics for equine lentivirus in the host genome.

Despite over 40 years of research on the human immunodeficiency virus type 1 (HIV-1) vaccine, we still lack a considerable progress. Equine infectious anemia virus (EIAV) is a lentivirus in the Retroviridae family, akin to HIV-1 in genome structure and antigenicity. EIA is an important infectious disease in equids, characterized by anemia, persistent infection, and repeated fevers. The EIAV attenuated vaccine in China is the only lentiviral vaccine used on a large scale. Elucidating the mechanism of waning and induction of protective immunity from this attenuated vaccine strain will provide a

No Increased Detection of Nucleic Acids of CNS-related Viruses in the Brains of Patients with Schizophrenia, Bipolar Disorder, and Autism Spectrum Disorder.

BACKGROUND AND HYPOTHESIS: Viral infections are increasingly recognized in the etiology of psychiatric disorders based on epidemiological and serological studies. Few studies have analyzed viruses directly within the brain and no comprehensive investigation of viral infection within diseased brains has been completed. This study aims to determine whether viral infection in brain tissues is a risk factor for 3 major psychiatric disorders, including schizophrenia, bipolar disorder, and autism spectrum disorder. STUDY DESIGN: This study directly evaluated the presence of viral DNA or RNA in 1569

Influence of Immunogenetic Biomarkers in the Clinical Outcome of HTLV-1 Infected Persons.

Human T-lymphotropic virus 1, a member of the Retroviridae family, causes a neglected, silent, persistent infection affecting circa 5 to 10 million people around the world, with biology, immune pathology, clinical diseases, epidemiology, and laboratory issues still unsolved. Most of the infected subjects are asymptomatic, but severe clinical disorders appear as a neurodegenerative disease (HTLV-1 associated myelopathy-HAM) or a lymphoprolipherative disorder (Adult T Leukemia/Lymphoma-ATLL) and in other target organs of the human body. HTLV-1 infections are frequently asymptomatic, but there is

Infection-Induced Telomere Length Variation: Insights into Pathogenesis of Koala Retrovirus.

The pathogenesis of koala retrovirus (KoRV) has been explored in various contexts, yet its role in tumorigenesis remains incompletely understood. Unlike acute transforming retroviruses, KoRV lacks a viral oncogene but may contribute to oncogenesis via indirect mechanisms. However, the relationship between KoRV and telomere length, as a potential indicator of telomerase activity, has not been examined. This study investigates the effect of KoRV infection on telomere length in 47 samples from Southern Australian koalas in a novel telomere length quantification method. Telomere lengths of 30 KoRV

Transmission of Koala Retrovirus from Parent Koalas to a Joey in a Japanese Zoo.

Koala retrovirus (KoRV) is of an interest to virologists due to its currently active endogenization into the koala (Phascolarctos cinereus) genome. Although KoRV has frequently been isolated in wild and captive koala populations, its pathogenesis and transmission remain to be fully characterized, and most previous research has concentrated on adult koalas rather than on joeys. Here, we characterized KoRV isolates obtained from a deceased male joey and its parents (animals reared in a Japanese zoo) to investigate KoRV transmission mode and pathogenesis. We sequenced the KoRV long terminal repea

Evidence for a retroviral trigger in Graves' disease.

An apparently high frequency of Graves' disease encountered in New Orleans, Louisiana, prompted an investigation for a possible infectious agent that might be triggering the disease in genetically susceptible individuals. We studied 40 patients with Graves' disease, and compared them to the following groups of controls: age and gender matched healthy subjects; patients with multinodular goiter (non-autoimmune thyroid controls); patients with chronic lymphocytic thyroiditis (autoimmune thyroid disease controls) and additional organ or tissue specific autoimmune controls exclusive of thyroid aut

Deceiving the gatekeepers: virus modulation of gap junctions.

SUMMARYGap junctions (GJs) are specialized intercellular channels that mediate the direct exchange of ions, metabolites, and signaling molecules between adjacent cells, playing essential roles in tissue homeostasis and immune coordination. Their function is tightly controlled by connexin isoform composition, trafficking and turnover, and post-translational modifications, particularly phosphorylation and ubiquitination. This review synthesizes current knowledge on the diverse strategies employed by DNA and RNA viruses, including members of the Herpesviridae, Adenoviridae, Papillomaviridae, Poly

A Textbook Case of Human T-lymphotropic Virus-1 (HTLV-1)-Induced Adult T-cell Leukemia Treated With Cyclophosphamide, Hydroxydaunorubicin, Oncovin, and Prednisone/Prednisolone (CHOP).

Human T-lymphotropic virus-1 (HTLV-I) is an enveloped, single-stranded RNA virus of the Retroviridae family. The virus causes two well-recognized disease associations: adult T-cell leukemia/lymphoma (ATL) and HTLV-I-associated myelopathy (HAM), also known as tropical spastic paraparesis (TSP). We report a case of HTLV-1-induced adult T-cell lymphoma/leukemia in a 45-year-old female who presented with complaints of swelling on the right side of her neck and rash on her upper and lower extremities and abdomen. The patient also had a history of strongyloidiasis infection and Crohn's disease. She

A high frequency of detection of koala retrovirus fragments in Victorian koalas suggests historic integration of KoRV.

Recombinant koala retrovirus (recKoRV) is a recently discovered variant of koala retrovirus (KoRV), which likely emerged due to recombination with another retrovirus (such as Phascolarctos endogenous retrovirus). KoRV spread and endogenization in Australia were thought to be ongoing in a north to south direction given the low prevalence of the virus in southern koala populations, based on molecular detection of the pol gene. However, recKoRV has highlighted that fragments of KoRV with the pol region missing are present within southern koalas. In this study, a new 5'-region-based KoRV PCR assay

Koala retrovirus (KoRV) subtypes and their impact on captive koala (Phascolarctos cinereus) health.

Koala retrovirus (KoRV), a major pathogen of koalas, exists in both endogenous (KoRV-A) and exogenous forms (KoRV-B to J). However, the impact of infection with multiple subtypes is not well understood. Accordingly, in this study, we surveyed a representative sample from a Japanese zoo population to determine the infection status for three KoRV subtypes (KoRV-A, B, and C) and to investigate the proviral and RNA load profiles in animals with single- and multiple-subtype infections, using peripheral blood mononuclear cells (PBMCs) and plasma. Six koalas were evaluated in the study; all were infe

Immunotherapy and Gene Therapy for Oncoviruses Infections: A Review.

Immunotherapy has been shown to be highly effective in some types of cancer caused by viruses. Gene therapy involves insertion or modification of a therapeutic gene, to correct for inappropriate gene products that cause/may cause diseases. Both these types of therapy have been used as alternative ways to avoid cancers caused by oncoviruses. In this review, we summarize recent studies on immunotherapy and gene therapy including the topics of oncolytic immunotherapy, immune checkpoint inhibitors, gene replacement, antisense oligonucleotides, RNA interference, clustered regularly interspaced shor

Characterization of enzootic nasal tumor virus 1-associated enzootic nasal adenocarcinoma in sheep and development of a tumor-derived organoid model.

Enzootic nasal adenocarcinoma (ENA) is a contagious neoplasm of the ethmoid turbinate mucosa in sheep, caused by enzootic nasal tumor virus 1 (ENTV1; family Retroviridae, unclassified Betaretrovirus). We report an outbreak of ENTV1-associated ENA in a sheep flock in South Dakota, USA. Affected animals had dyspnea, unilateral nasal discharge, and progressive weight loss. Postmortem examination revealed unilateral nasal masses that were diagnosed histologically as invasive nasal adenocarcinoma. PCR amplification followed by Sanger sequencing of the gag and env gene regions confirmed the presence
