Effect of horse sleep behavior on performance in a field-side spatial reversal learning test.

Reduced rapid-eye-movement (REM) sleep is associated with impaired learning in many species. We developed a reversal learning test (RLT) suitable for field conditions to explore this association in 16 healthy horses. Nocturnal REM-like sleep behavior was recorded five times for 48 h over a six-week period. The horses performed a target training task followed by an RLT using two objects. When the horses reached a predefined frequency of touching the object, the spatial location was reversed. Mean test parameters were statistically analysed using GENLIN models, longitudinal continuous variables

Continuous theta burst stimulation to the medial posterior cerebellum impairs reversal learning in healthy volunteers.

The role of the cerebellum in associative learning and context-updating implies involvement in learning reward-punishment contingencies. This study examined the direct contribution of the cerebellum to reward- and punishment-based reversal learning. A total of 111 healthy right-handed adult volunteers received continuous theta burst stimulation (cTBS) to either the medial posterior cerebellum (n = 37), right posterolateral cerebellum (n = 37), or right occipital lobe (n = 37) in this single-blind between-subjects study. A gambling task with two changing reward-punishment contingencies (reversa

Parsing decision making processes in prefrontal cortex: response inhibition, overcoming learned avoidance, and reversal learning.

Reversal learning refers to the ability to inhibit or switch responding to an object when the object-reward contingency changes. Deficits in this process are related to social abnormalities, impulsiveness, and a number of psychiatric disorders. A range of neural regions play a role in this process, including dorsolateral prefrontal cortex (dlPFC), dorsomedial prefrontal cortex (dmPFC), and inferior frontal gyrus (IFG). However, determining the specific functional contribution of each region has proved difficult, in part because reversal learning involves multiple cognitive subprocesses such as

Medial prefrontal administration of MK-801 impairs T-maze discrimination reversal learning in weanling rats.

Several executive functions rely on the medial prefrontal cortex (mPFC) in the rat. Aspiration and neurotoxic lesions of the mPFC impair reversal learning in adult rats. Systemic administration of MK-801, an NMDA-receptor antagonist, impairs T-maze reversal learning in weanling rats but the role of mPFC NMDA-receptor antagonism in this effect is not known in either adult or young animals. This set of studies showed that mPFC NMDA receptors are specifically involved in T-maze discrimination reversal in weanling rats. In Experiment 1, 26-day-old rats (P26) demonstrated a dose-dependent impairmen

Hemicholinium-3 impairs spatial learning and the deficit is reversed by cholinomimetics.

The effects of hemicholinium-3 (HC-3) on spatial discrimination learning were studied. Rats were equipped with indwelling cannulae in the right lateral ventricle and, following recovery, were trained on a two platform spatial discrimination task in a water maze. In this task a visible escape platform remains in a fixed position in the pool during a single training session, whilst the location of an identical "float" (which affords no escape) is randomly varied. For each session the location of the fixed escape platform was changed and the rats were retrained to criterion following pretreatment

[Reverse learning in WAG/Rij rats with depression-like behavior].

Learning and reverse learning in a complex maze, behavior in the open field test, novelty-suppressed feeding test, and forced swimming test were studies in WAG/Rij and Wistar rats. As compared with Wistar rats, WAG/Rij rats more slowly learned the spatial task, more slowly performed in the learning and reverse learning tasks, and made more errors in the complex maze (18% of WAG/Rij rats didn't reach learning criterion). Moreover, WAG/Rij rats exhibited reduced grooming reactions in the open field test, longer latency of approaching to food in the novel open field, reduced amount of food consum

Acute stress impairs reversal learning in mice via neurochemical and transcriptional mechanisms in the orbitofrontal cortex.

The effects of acute stress on cognitive flexibility are significant but inconsistent across studies. To address this inconsistency, we investigated how varying durations of acute restraint stress affect reversal learning in mice with different innate cognitive flexibility, across tasks of varying difficulty. Results revealed that 1-hour acute restraint stress impaired reversal learning in a 4-choice task but not in a 2-choice task. In the 2-choice task, 2-hour stress led to reversal learning deficits in mice with low baseline cognitive flexibility but not in those with high baseline cognitive

Slitrk5 deficiency preserves reversal learning but alters orbitofrontal-striatal signaling and task-related responding in mice.

Obsessive-compulsive disorder (OCD) is a debilitating condition associated with repetitive behaviors and cognitive inflexibility, often linked to abnormalities in cortico-striato-thalamo cortical (CSTC) circuitry. Although the Slitrk5 knockout (Slitrk5-/-, KO) mouse has been proposed as an OCD-related model, it remains unclear how Slitrk5 deficiency influences adaptive decision making, task-related behavioral organization, and orbitofrontal-dorsomedial striatum (OFC-DMS) signaling during reversal learning. To address this, Slitrk5-/- and wild-type (WT) mice were examined using probabilistic re

Central medial and paraventricular thalamic afferents to the orbitofrontal cortex have differential effects on reversal learning.

Cognitive impairments are prominent in stress-related psychiatric disorders, such as depression and PTSD. While current therapies may improve mood symptoms, many patients experience persistent deficits in cognitive flexibility. One form of cognitive flexibility disrupted in such conditions is reversal learning, dependent on the orbitofrontal cortex (OFC). Chronic stress causes OFC dysfunction and reversal learning deficits. However, the circuit mechanisms by which this happens are not understood. We hypothesized that two thalamic afferents to the OFC, the central medial (CM) and paraventricula

Deep brain stimulation of the subthalamic nucleus restores spatial reversal learning in patients with Parkinson's disease.

Spatial learning and navigation are supported by distinct memory systems in the human brain such as the hippocampus-based navigational system and the striatum-cortex-based system involved in motor sequence, habit and reversal learning. Here, we studied the role of subthalamic circuits in hippocampus-associated spatial memory and striatal-associated spatial reversal learning formation in patients with Parkinson's disease, who underwent a deep brain stimulation of the subthalamic nucleus. Deep brain stimulation patients (Parkinson's disease-subthalamic nucleus: n = 26) and healthy subjects (n =

Optogenetically-induced long term depression in the rat orbitofrontal cortex ameliorates stress-induced reversal learning impairment.

Cognitive flexibility is a higher-order executive function that requires plasticity in neuronal circuits of the prefrontal cortex. Deficits in cognitive flexibility are prominent in a variety of psychiatric disorders, such as major depression, obsessive-compulsive disorder, and posttraumatic stress disorder. Chronic stress induces deficits in cognitive flexibility, perhaps through effects on plasticity, but the mechanism is not well understood. Previous work has demonstrated that stress reduces activity and dendritic elaboration in the medial prefrontal cortex (mPFC). In contrast, stress appea

Measuring discrimination- and reversal learning in mouse models within 4 days and without prior food deprivation.

Many neurological and psychiatric disorders are characterized by deficits in cognitive flexibility. Modeling cognitive flexibility in mice enables the investigation of mechanisms underlying these deficits. The majority of currently available behavioral tests targeting this cognitive domain are reversal learning tasks that require scheduled food restriction, extended training periods and labor-intensive, and stress-inducing animal handling. Here, we describe a novel 4-day (4-d) continuously running task measuring discrimination- and reversal learning in an automated home cage (CognitionWall DL/

Reward-related reversal learning after surgical excisions in orbito-frontal or dorsolateral prefrontal cortex in humans.

Neurophysiological studies in primates and neuroimaging studies in humans suggest that the orbito-frontal cortex is involved in representing the reward value of stimuli and in the rapid learning and relearning of associations between visual stimuli and rewarding or punishing outcomes. In the present study, we tested patients with circumscribed surgical lesions in different regions of the frontal lobe on a new visual discrimination reversal test, which, in an fMRI study (O'Doherty, Kringelbach, Rolls, Hornak, & Andrews, 2001), produced bilateral orbito-frontal cortex activation in normal subjec

Mice with exonic RELN deletion identified from a patient with schizophrenia have impaired visual discrimination learning and reversal learning in touchscreen operant tasks.

The Reelin gene (RELN) encodes a large extracellular protein, which has multiple roles in brain development and adult brain function. It activates a series of neuronal signal transduction pathways in the adult brain that function in synaptic plasticity, dendritic morphology, and cognitive function. To further investigate the roles of Reln in brain function, we generated a mouse line using the C57BL/6 J strain with the specific Reln deletion identified from a Japanese patient with schizophrenia (Reln-del mice). These mice exhibited abnormal sociality, but the pathophysiological significance of

Role of superficial grey layer of superior colliculus in reversal learning: importance of nitric oxide synthase.

Reversal learning is the ability to adapt behavior to changing circumstances. It is essential for goal-directed actions and survival, and the superior colliculus (SC) is known to be involved in visual cue-guided reversal learning. The superficial grey layer (SuG) of the SC contributes to the refinement of the retinocollicular pathway and visual processing. Although this layer of the SC is rich in neuronal nitric oxide synthase (nNOS), the role of the enzyme or nitric oxide (NO) in mediating reversal behavior driven by visual cues remains unclear. We aimed to investigate the participation of ni

A multidimensional learning curve analysis of totally laparoscopic ileostomy reversal using a single surgeon' s experience.

PURPOSE: Recently, totally laparoscopic ileostomy reversal (TLAP) has received increasing attention and exhibited promising short-term outcomes. The aim of this study was to detail the learning process of the TLAP technique. METHODS: Based on our initial experience with TLAP from 2018, a total of 65 TLAP cases were enrolled. Demographics and perioperative parameters were assessed using cumulative sum (CUSUM), moving average, and risk-adjusted CUSUM (RA-CUSUM) analyses. RESULTS: The overall mean operative time (OT) was 94 min and the median postoperative hospitalization period was 4 days, and

Cortical hemodynamic mechanisms of reversal learning using high-resolution functional near-infrared spectroscopy: A pilot study.

OBJECTIVES: Reversal learning is widely used to analyze cognitive flexibility and characterize behavioral abnormalities associated with impulsivity and disinhibition. Recent studies using fMRI have focused on regions involved in reversal learning with negative and positive reinforcers. Although the frontal cortex has been consistently implicated in reversal learning, few studies have focused on whether reward and punishment may have different effects on lateral frontal structures in these tasks. METHODS: During this pilot study on eight healthy subjects, we used functional near infra-red spec

Ciliary neurotrophic factor signaling in the rat orbitofrontal cortex ameliorates stress-induced deficits in reversal learning.

Deficits in cognitive flexibility, i.e. the ability to modify behavior in response to changes in the environment, are present in several psychiatric disorders and are often refractory to treatment. However, improving treatment response has been hindered by a lack of understanding of the neurobiology of cognitive flexibility. Using a rat model of chronic stress (chronic intermittent cold stress, CIC) that produces selective deficits in reversal learning, a form of cognitive flexibility dependent on orbitofrontal cortex (OFC) function, we have previously shown that JAK2 signaling is required for

Mechanisms underlying prelimbic prefrontal cortex mGlu3/mGlu5-dependent plasticity and reversal learning deficits following acute stress.

Stress can precipitate or worsen symptoms of many psychiatric illnesses. Dysregulation of the prefrontal cortex (PFC) glutamate system may underlie these disruptions and restoring PFC glutamate signaling has emerged as a promising avenue for the treatment of stress disorders. Recently, we demonstrated that activation of metabotropic glutamate receptor subtype 3 (mGlu3) induces a postsynaptic form of long-term depression (LTD) that is dependent on the activity of another subtype, mGlu5. Stress exposure disrupted this plasticity, but the underlying signaling mechanisms and involvement in higher-

Reversal-learning deficits in childhood-onset bipolar disorder across the transition from childhood to young adulthood.

BACKGROUND: Bipolar disorder (BD) is a severe mental illness that can have high costs for youths (<18 years old) and adults. Relative to healthy controls (HC), individuals with BD often show impaired attention, working memory, executive function, and cognitive flexibility (the ability to adapt to changing reward/punishment contingencies). In our study of youths and young adults with BD, we investigated 1) how cognitive flexibility varies developmentally in BD, and 2) whether it is independent of other executive function deficits associated with BD. METHODS: We measured errors on a reversal-le
