Capture-SELEX of DNA Aptamers for Highly Selective Binding of Folate.

Folate (vitamin B9) is an essential micronutrient for human health, particularly during pregnancy. Aptamers have emerged as attractive recognition elements for small-molecule detection, as demonstrated by numerous natural riboswitches. Although an RNA aptamer for folate has been reported, the development of a DNA aptamer is highly desirable for biosensing applications due to its superior chemical stability and ease of modification. Herein, we employed a library-immobilization-based selection strategy to isolate a family of high-affinity DNA aptamers for folate. Isothermal titration calorimetry

Plasma-SELEX for Acute Myocardial Infarction Biomarker Discovery and Diagnosis.

Early diagnosis of acute diseases is fundamentally constrained by the lack of chemically tractable methods for biomarker discovery directly from a complex plasma. Here, we report PSABD (plasma-SELEX-enriched aptamer-based biomarker discovery and diagnosis), a chemical platform that integrates carboxylate magnetic-bead-mediated plasma protein capture within situ aptamer SELEX, enabling molecular-recognition-driven identification of disease-associated biomarkers. With this strategy, enriched aptamers not only serve as affinity probes but also function as structurally programmable molecular handl

Advances in aptamer technology and its updated applications in gastric cancer.

Gastric cancer (GC) is one of the leading causes of cancer-related mortality worldwide, with late-stage diagnosis and limited treatment efficacy. The major challenges in chemotherapy and radiotherapy for GC include drug resistance, systemic toxicity, limited tumor selectivity, adverse side effects, tumor heterogeneity, and the development of radioresistance, all of which hinder treatment efficacy and patient outcomes. Aptamers, single-stranded DNA or RNA molecules with high specificity and affinity for molecular targets, offer a promising alternative to traditional diagnostic and therapeutic a

From cell-SELEX to tissue-SELEX for targeted drug delivery and aptamer nanomedicine.

Aptamers are nucleic acid-based ligands that can selectively bind to target molecules. Because of their unique target-binding properties, the use of aptamers for targeting cell surface molecules has attracted broad research interest. The field has evolved from selecting aptamers against purified surface proteins to using whole cells (cell-SELEX) as targets. To further advance the field, the concept of tissue-SELEX was later proposed to ensure that selected aptamers possess optimal binding properties in more native in vivo environments. In this article, we review recent progress made for tissue

Sensing Levofloxacin with an RNA Aptamer as a Bioreceptor.

To combat the growing threat of antibiotic resistance, environmental testing for antibiotic contamination is gaining an increasing role. This study aims to develop an easy-to-use assay for the detection of the fluoroquinolone antibiotic levofloxacin. Levofloxacin is used in human and veterinary medicine and has been detected in wastewater and river water. An RNA aptamer against levofloxacin was selected using RNA Capture-SELEX. The 73 nt long aptamer folds into three stems with a central three-way junction. It binds levofloxacin with a Kd of 6 µM and discriminates the closely related compound

The Development and Characterisation of ssDNA Aptamers via a Modified Cell-SELEX Methodology for the Detection of Human Red Blood Cells.

Blood is one of the most commonly found biological fluids at crime scenes, with the detection and identification of blood holding a high degree of evidential value. It can provide not only information about the nature of the crime but can also lead to identification via DNA profiling. Presumptive tests for blood are usually sensitive but not specific, so small amounts of the substrate can be detected, but false-positive results are often encountered, which can be misleading. Novel methods for the detection of red blood cells based on aptamer-target interactions may be able to overcome these is

In vitro selection of RNA aptamers that inhibit the activity of type A botulinum neurotoxin.

The category A agent, botulinum neurotoxin (BoNT), is the most toxic molecule known to mankind. The endopeptidase activity of light chain domain of BoNT is the cause for the inhibition of the neurotransmitter release and the flaccid paralysis that leads to lethality in botulism. Currently, antidotes are not available to reverse the flaccid paralysis caused by BoNT. In the present study, we have identified three RNA aptamers through SELEX-process, which bind strongly to the light chain of type A BoNT (BoNT/A) and inhibit the endopeptidase activity, with IC(50) in low nM range. Inhibition kineti

Development of a quality control test for diphtheria vaccine based on an electrochemical aptamer-based biosensor.

At every stage of the vaccine's development, including research and development together with the routine production (in-process control of batch release), quality control testing is a crucial operation for ensuring the efficacy, safety and consistency of final products. Conventional quality control tests rely on a combination of biochemical, physicochemical, and cell-based assays. In this study, we report a quality control test based on an electrochemical aptamer-based sensor with a specific application to Diphtheria vaccine. First, specific aptamers for Diphtheria are successfully selected a

RNA aptamers developed against tRip: A preliminary approach targeting tRNA entry in Plasmodium.

Malaria is caused by Plasmodium parasites that multiply inside host cells and can be lethal when P. falciparum is involved. We identified tRip as a membrane protein that facilitates the import of exogenous transfer RNA (tRNA) into the parasite. tRip encompasses a tRNA binding domain exposed on the parasite surface. We used the SELEX approach to isolate high-affinity and specific tRip-binding RNA motifs from a library of random 25 nucleotide-long sequences. In five rounds of combined negative and positive selections, an enriched pool of aptamers was obtained; sequencing revealed that they were

DNA aptamers specific for Legionella pneumophila: systematic evolution of ligands by exponential enrichment in whole bacterial cells.

Legionella pneumophila is the major causative agent of Legionnaires' disease and Pontiac fever, which pose major public health problems. Rapid detection of L. pneumophila is important for global control of these diseases. Aptamers, short oligonucleotides that bind to targets with high affinity and specificity, have great potential for use in pathogenic bacterium detection, diagnostics, and therapy. Here, we used a whole-cell SELEX (systematic evolution of ligands by exponential enrichment) method to isolate and characterize single-stranded DNA (ssDNA) aptamers against L. pneumophila. A total o

A Blautia producta specific gFET-based aptasensor for quantitative monitoring of microbiome quality.

The use of health-relevant bacteria originating from human microbiomes for the control or therapy of diseases, including neurodegenerative disorders or diabetes, is currently gaining increasing importance in medicine. Directed and successful engineering of microbiomes via probiotic supplementation requires subtle, precise as well as, more importantly, easy, fast and convenient monitoring of its success, e.g., in patients' gut. Based on a previously described polyclonal SELEX aptamer library evolved against the human gut bacterium Blautia producta, we finally isolated three individual aptamers

Development of a label-free electrochemical aptasensor for Rift Valley fever virus detection.

In this research, we describe the first aptasensor for the detection of the Rift Valley Fever virus (RVFV). The process involved the selection of aptamers through the systematic evolution of ligands by the exponential enrichment (SELEX) technique. After 12 rounds of selection, 6 aptamers were selected and the corresponding binding affinities were assessed using fluorescence binding assays, revealing dissociation constants ranging from 15.45 to 40.98 nM. Notably, among the aptamers, RV2 and RV3 exhibited the highest binding affinities toward RVFV, with dissociation constants of 15.45 and 18.62

Chemically modified nucleic acid aptamers for in vitro selections: evolving evolution.

Combinatorial library selections through the systematic evolution of ligands by exponential enrichment (SELEX) technique identify so-called nucleic acid aptamers that bind with high-affinity and specificity to a wide range of selected molecules. However, the modest chemical functionality of nucleic acids poses some limits on their versatility as binders and catalysts, and, furthermore, the sensitivity of pure RNA- and DNA-based aptamers to nucleases restricts their use as therapeutic and diagnostic agents. Here we review synthetic chemistries for modifying nucleotides that have been developed

Selection and application of aptamers targeting the Brucella BP26 protein.

Rapid and specific detection of Brucella remains a challenge due to the limitations of conventional serological and molecular methods. In this study, we developed an aptamer magnetic capture (AMC) qPCR platform for the sensitive detection of Brucella. The recombinant outer membrane protein BP26 was expressed, purified, and utilized as the target for aptamer selection via a magnetic cross-linking precipitation (MCP) SELEX strategy. After eight rounds of selection, two DNA aptamers (B1 and B3) were identified, exhibiting high affinities with dissociation constants (Kd) of 69.42 nM and 47.40 nM,

Selection and characterization of DNA aptamers targeting the surface Borrelia protein CspZ with high-throughput cross-over SELEX.

Lyme borreliosis (LB) is the most prevalent tick-borne illness, with an estimated 700 000 cases annually in the United States and Europe. The LB diagnosis based on a two-tiered serology remains controversial due to its indirect nature and low sensitivity during the early stage of the disease. Aptamers are single-stranded DNA or RNA oligonucleotides that exhibit high selectivity and specificity for their target due to their unique three-dimensional structure. By applying cross-over-SELEX process, an enrichment of DNA oligonucleotide sequences against a surface protein of Borrelia, named CspZ, h

Selection and characterization of structure-switching DNA aptamers for the salivary peptide histatin 3.

In this study, single-stranded DNA aptamers that switch structural conformation upon binding to the salivary peptide histatin 3 have been reported for the first time. Histatin 3 is an antimicrobial peptide that possesses the capability of being a therapeutic agent against oral candidiasis and has recently been linked as a novel biomarker for acute stress. The aptamers were identified through a library immobilization version of an iterative in vitro process known as the Systematic Evolution of Ligands by EXponential enrichment (SELEX). Through the SELEX process, four unique aptamer candidates s
