Clinical and Molecular Findings of Tunisian Patients with RASopathies.

Noonan syndrome (NS) and related disorders, which are now summarized under the term RASopathies, are caused by germline mutations in genes encoding protein components of the Ras/mitogen-activated protein kinase pathway. In this study, we evaluated the clinical and molecular spectrum of 21 Tunisian patients, recruited by a cardiology unit, for whom RASopathy diagnosis was suspected by clinical geneticists. Overall, 19 patients had a clinical diagnosis of NS and 2 were classified as having Cardiofaciocutaneous (CFC) syndrome. In 52% (n = 11) of patients, a RASopathy has been molecularly confirme

Proteomic analyses identify targets, pathways, and cellular consequences of oncogenic KRAS signaling.

Mutations that activate the small GTPase KRAS are a frequent genetic alteration in cancer, and drug discovery efforts have led to inhibitors that block KRAS activity. We sought to better understand oncogenic KRAS signaling and the cytostatic effects of drugs that target this system. We performed proteomic analyses to investigate changes in protein abundance and posttranslational modifications in inhibitor-treated human KRAS-mutant pancreatic (KRAS G12C and G12D) and lung cancer (KRAS G12C) cells. The inhibitors used target these mutant forms of KRAS, the downstream effectors MEK and ERK, and t

Decreased expression of FGFR1, SOS1, RAF1 genes in cryptorchidism.

BACKGROUND: In recent years, several genes were found to be involved in the process of epididymo-testicular descent, the most frequently cited ones include INSL3, HOXA10, GNRHR, and KAL1. In this study, we analyzed the differences in gene expression profiles between cryptorchid and descended testes. In particular, we analyzed expression of all recently published genes known to be associated with undescended testis. PATIENTS AND METHODS: Twenty-two testicular biopsies from 18 boys were analyzed. We analyzed gene expression in 16 cryptorchid and 6 descended testes using Affymetrix Human Genome

Research progress of direct and indirect KRAS-targeting protein degraders in KRAS-mutant cancers.

The RAS gene, being the most frequently activated proto-oncogene, belongs to the GTPase family. Its primary physiological function is to regulate cell proliferation. The main members of the RAS gene are H-RAS, K-RAS, and N-RAS. Approximately 16 % of human cancers are related to RAS proteins. KRAS is the most common oncogene. About 17 % of solid tumors have KRAS mutations, which account for around 50 %, 80 %, and 25 % of colorectal cancer, pancreatic cancer, and lung adenocarcinoma, respectively. Currently, significant advances have been made in the development of small-molecule inhibitors targ

Unraveling sex differences in Alzheimer's disease and related endophenotypes with brain proteomes.

INTRODUCTION: Sex differences exist in Alzheimer's disease (AD), but the underlying mechanisms remain unclear. METHODS: We examined brain proteomes profiled from the dorsolateral prefrontal cortex of 770 donors (66.2% female). RESULTS: Proteome-wide differential expression analysis in males and females jointly identified many significant proteins for AD dementia (n = 1228), amyloid beta (n = 1183), tangles (n = 1309), and global cognitive trajectory (n = 2325) at a false discovery rate of <0.05. Sex-stratified analyses also identified many proteins associated with AD or its endophenotypes. F

New Mutations Associated with Rasopathies in a Central European Population and Genotype-Phenotype Correlations.

We performed the genetic analysis of Rasopathy syndromes in patients from Central European by direct sequencing followed by next generation sequencing of genes associated with Rasopathies. All 51 patients harboured the typical features of Rasopathy syndromes. Thirty-five mutations were identified in the examined patients (22 in PTPN11, two in SOS1, one in RIT1, one in SHOC2, two in HRAS, three in BRAF, two in MAP2K1 and two in the NF1 gene). Two of them (p.Gly392Glu in the BRAF gene and p.Gln164Lys in the MAP2K1 gene) were novel with a potentially pathogenic effect on the structure of these pr

Germline BRAF mutations in Noonan, LEOPARD, and cardiofaciocutaneous syndromes: molecular diversity and associated phenotypic spectrum.

Noonan, LEOPARD, and cardiofaciocutaneous syndromes (NS, LS, and CFCS) are developmental disorders with overlapping features including distinctive facial dysmorphia, reduced growth, cardiac defects, skeletal and ectodermal anomalies, and variable cognitive deficits. Dysregulated RAS-mitogen-activated protein kinase (MAPK) signal traffic has been established to represent the molecular pathogenic cause underlying these conditions. To investigate the phenotypic spectrum and molecular diversity of germline mutations affecting BRAF, which encodes a serine/threonine kinase functioning as a RAS effec

Hereditary gingival fibromatosis: characteristics and novel putative pathogenic mechanisms.

Fertilization calcium waves are introduced, and the evidence from which we can infer general mechanisms of these waves is presented. The two main classes of hypotheses put forward to explain the generation of the fertilization calcium wave are set out, and it is concluded that initiation of the fertilization calcium wave can be most generally explained in invertebrates by a mechanism in which an activating substance enters the egg from the sperm on sperm-egg fusion, activating the egg by stimulating phospholipase C activation through a src family kinase pathway and in mammals by the diffusion

A Novel Homozygous Loss-of-Function Variant in SPRED2 Causes Autosomal Recessive Noonan-like Syndrome.

Noonan syndrome is an autosomal dominant developmental disorder characterized by peculiar facial dysmorphisms, short stature, congenital heart defects, and hypertrophic cardiomyopathy. In 2001, PTPN11 was identified as the first Noonan syndrome gene and is responsible for the majority of Noonan syndrome cases. Over the years, several other genes involved in Noonan syndrome (KRAS, SOS1, RAF1, MAP2K1, BRAF, NRAS, RIT1, and LZTR1) have been identified, acting at different levels of the RAS-mitogen-activated protein kinase pathway. Recently, SPRED2 was recognized as a novel Noonan syndrome gene wi

Two cases of Noonan syndrome with severe respiratory and gastroenteral involvement and the SOS1 mutation F623I.

Noonan syndrome (NS) is an autosomal dominant, inherited disorder characterized by facial dysmorphism, congenital heart defects, and reduced postnatal growth. Dysregulated RAS-MAPK signalling is the common molecular basis for NS, a genetically heterogeneous disease. Germline mutations in genes encoding small GTPases of the RAS family (KRAS and NRAS), modulators of RAS function (PTPN11, SOS1 and SHOC2) or downstream signal transducers (RAF1) are causative for NS. SOS1 is the second major gene for NS after PTPN11. Compared to patients with mutations in other genes, SOS1 mutation-positive individ

Characterization of a rare EGFR resistance mutation in a lung cancer patient with a long response to osimertinib: case report.

INTRODUCTION: Osimertinib is the standard of care in first-line therapy for advanced EGFR-mutant NSCLC, but resistance invariably develops through EGFR-dependent or independent mechanisms. CASE DESCRIPTION: Here, we report the case of a stage IVA adenocarcinoma patient with several metastases harboring an EGFR exon 19 deletion, which progressed locally after five years on osimertinib. Sequencing of the re-biopsy revealed the emergence of an acquired exon 20 mutation, p.(Cys797Ala), not previously characterized and absent from variant databases. Immunohistochemical analysis (IHC) showed phosph

PTPN11 and KRAS gene analysis in patients with Noonan and Noonan-like syndromes.

Noonan and Noonan-like syndromes are disorders of dysregulation of the rat sarcoma viral oncogene homolog (RAS)-mitogen-activated protein kinase signaling pathway. In Noonan syndrome (NS), four genes of this pathway (PTPN11, SOS1, RAF1, and KRAS) are responsible for roughly 70% of the cases. We analyzed PTPN11 and KRAS genes by bidirectional sequencing in 95 probands with NS and 29 with Noonan-like syndromes, including previously reported patients already screened for PTPN11 gene mutations. In the new patients with NS, 20/46 (43%) showed a PTPN11 gene mutation, two of them novel. In our total

PI3K Regulates Wild-type RAS Signaling to Confer Resistance to KRAS Inhibition.

UNLABELLED: Despite the availability of RAS inhibitors and the dependence of >90% of pancreatic ductal adenocarcinomas (PDAC) on oncogenic KRAS mutations, resistance to KRAS inhibition remains a serious obstacle. We showed here that phosphoinositide 3-kinase (PI3K) plays a major role in this resistance through upstream activation of wild-type (WT) RAS signaling-beyond its known KRAS effector function. The combination of proximity labeling, CRISPR screening, live-cell imaging, and functional assays revealed that PI3K orchestrates phosphoinositide-mediated GAB1 recruitment to the plasma membrane

Nintedanib inhibits the VEGFR-ERK signaling pathway in human KRAS-mutated cancer cells.

KRAS mutations are significant drivers in various cancers, and existing drug discovery attempts targeting these mutations have largely been unsuccessful, emphasizing the need for more effective therapies. In this study, the screening library, which contains 1,374 chemical compounds, identified nintedanib, a VEGFR inhibitor, as exerting a potent and selective antiproliferative effect against KRAS-mutant cells, surpassing other VEGFR inhibitors. Nintedanib effectively suppressed tumor growth in xenografted mice with KRAS mutations and significantly inhibited phosphorylated VEGFR2 levels and its

NMR-based functional profiling of RASopathies and oncogenic RAS mutations.

Defects in the RAS small G protein or its associated network of regulatory proteins that disrupt GTPase cycling are a major cause of cancer and developmental RASopathy disorders. Lack of robust functional assays has been a major hurdle in RAS pathway-targeted drug development. We used NMR to obtain detailed mechanistic data on RAS cycling defects conferred by oncogenic mutations, or full-length RASopathy-derived regulatory proteins. By monitoring the conformation of wild-type and oncogenic RAS in real-time, we show that opposing properties integrate with regulators to hyperactivate oncogenic R

Overexpression of Lotus NnSWEET4a Alters Sugar Homeostasis and Induces Salt Hypersensitivity in Arabidopsis.

SWEET (Sugars Will Eventually be Exported Transporter) proteins constitute a conserved family of sugar transporters that play pivotal roles in carbohydrate allocation and stress responses. In this study, we systematically identified 14 SWEET homologs in the genome of sacred lotus (Nelumbo nucifera) and validated their transport activity for both hexoses and sucrose. Subsequent analysis revealed that stress-responsive elements are the most enriched promoter sequences of NnSWEET genes. Quantitative expression profiling of the members found that NnSWEET4a was strongly upregulated under salt stres

Comprehensive analysis of eccDNA characteristics and associated genes expression in peripheral blood of ASLE and ISLE patients.

To explore SLE staging markers, we analyzed eccDNA in plasma using circular sequencing, comparing healthy controls (HC), active SLE (ASLE), and inactive SLE (ISLE) patients. We found higher eccDNA levels and lower GC content in ASLE and ISLE compared to healthy controls, with a negative correlation between GC content and anti-daDNA, C3, and C4 levels in SLE and HC samples. Differential expression of exon-derived eccGenes in ASLE and ISLE suggests their role in SLE development, with KEGG analysis showing enrichment in SLE-related pathways for these differentially expressed genes.
