Sample size estimation in locomotion kinematics and electromyography for statistical parametric mapping.

In biomechanics, kinematic and electromyographic data can be represented as one-dimensional (1D) waveforms and compared by using 1D hypothesis tests. These statistical techniques are increasingly applied in the study of locomotion. However, although widely agreed as a key step to obtain reliable and replicable findings, no a priori sample size estimation is usually conducted. This can also be done in 1D tests by calculating the statistical power - i.e., the probability of rejecting the null hypothesis when it is false - by using statistical parametric mapping. With the present study we charact

Sample size and power calculations for open cohort longitudinal cluster randomized trials.

When calculating sample size or power for stepped wedge or other types of longitudinal cluster randomized trials, it is critical that the planned sampling structure be accurately specified. One common assumption is that participants will provide measurements in each trial period, that is, a closed cohort, and another is that each participant provides only one measurement during the course of the trial. However some studies have an "open cohort" sampling structure, where participants may provide measurements in variable numbers of periods. To date, sample size calculations for longitudinal clus

Sample size for K 2x2 tables in equivalence studies using Cochran's statistic.

This paper presents a new sample size formula for Cochran's test that uses additional information on stratum-specific success rates and requires fewer subjects for an equivalence study. Equivalence studies are common in clinical trials, where unlike superiority studies, the goal is to show whether a new drug therapy is as effective as a standard one. Stratification is typically used to adjust for differences among individual clinical trial centers with different success rates. The sample size is derived for a clinical trial design where two independent binomial proportions are compared within

Robust power and sample size calculations in quasi-likelihood models: methods and practice.

BACKGROUND: Accurate power and sample size calculations are essential in study planning, yet they are often difficult to carry out for quasi-likelihood (QL) models. Traditional power and sample size (PSS) approaches often rely on restrictive distributional assumptions, limiting their applicability when responses have non-standard distributions, variance functions are misspecified, or when covariates exhibit complex dependence structures. METHODS: We examine whether two effect size measures-2 Standard Deviations in the Linear Predictor (2SLiP) and Pseudo-Partial [Formula: see text] (P2R2)-orig

Sample size estimation for recurrent event data using multifrailty and multilevel survival models.

In epidemiology and clinical research, recurrent events refer to individuals who are likely to experience transient clinical events repeatedly over an observation period. Examples include hospitalizations in patients with heart failure, fractures in osteoporosis studies and the occurrence of new lesions in oncology. We provided an in-depth analysis of the sample size required for the analysis of recurrent time-to-event data using multifrailty or multilevel survival models. We covered the topic from the simple shared frailty model to models with hierarchical or joint frailties. We relied on a W

Platelet function investigation by flow cytometry: Sample volume, needle size, and reference intervals.

Flow cytometry is an increasingly used method for platelet function analysis because it has some important advantages compared with other platelet function tests. Flow cytometric platelet function analyses only require a small sample volume (3.5 mL); however, to expand the field of applications, e.g., for platelet function analysis in children, even smaller volumes are needed. Platelets are easily activated, and the size of the needle for blood sampling might be of importance for the pre-activation of the platelets. Moreover, to use flow cytometry for investigation of platelet function in clin

Estimated ventricle size using Evans index: reference values from a population-based sample.

BACKGROUND AND PURPOSE: Evans index is an estimate of ventricular size used in the diagnosis of idiopathic normal-pressure hydrocephalus (iNPH). Values >0.3 are considered pathological and are required by guidelines for the diagnosis of iNPH. However, there are no previous epidemiological studies on Evans index, and normal values in adults are thus not precisely known. We examined a representative sample to obtain reference values and descriptive data on Evans index. METHODS: A population-based sample (n = 1235) of men and women aged ≥70 years was examined. The sample comprised people living

Sample size determination in group-sequential clinical trials with two co-primary endpoints.

We discuss sample size determination in group-sequential designs with two endpoints as co-primary. We derive the power and sample size within two decision-making frameworks. One is to claim the test intervention's benefit relative to control when superiority is achieved for the two endpoints at the same interim timepoint of the trial. The other is when superiority is achieved for the two endpoints at any interim timepoint, not necessarily simultaneously. We evaluate the behaviors of sample size and power with varying design elements and provide a real example to illustrate the proposed sample

White matter lesion progression in LADIS: frequency, clinical effects, and sample size calculations.

BACKGROUND AND PURPOSE: White matter lesion (WML) progression has been advocated as a surrogate marker in intervention trials on cerebral small vessel disease. We assessed the rate of visually rated WML progression, studied correlations between lesion progression and cognition, and estimated sample sizes for clinical trials with pure WML progression vs combined WML progression-cognitive outcomes. METHODS: Those 394 participants of the Leukoaraiosis and Disability Study (LADIS) study with magnetic resonance imaging scanning at baseline and 3-year follow-up were analyzed. Power of pairwise comparisons in the equal variance and unequal sample size case.

A Monte Carlo simulation was conducted to compare five, pairwise multiple comparison procedures. The number of means varied from 4 to 6 and the sample size ratio varied from 1 to 60. Procedures were evaluated on the basis of Type I errors, any-pair power and all-pairs power. Four procedures were shown to be conservative, while the fifth provided adequate control of Type I errors only for restricted values of sample size ratios. No procedure was found to be uniformly most powerful. The Tukey-Kramer procedure was found to provide the best any-pair power provided it is applied without requiring a

Effects of aggregation and sample size on composite faecal egg counts in sheep.

Composite faecal egg counts (FEC) are increasingly used to support strategic anthelmintic treatment decisions in grazing livestock. However, their accuracy as estimators of group mean FEC is affected by the number of individual samples included, how thoroughly they are mixed, and the underlying degree of parasite aggregation between individual hosts. This paper uses a Negative Binomial model for parasite aggregation, and a Poisson model for egg distribution within faecal suspensions, in order to optimise composite FEC protocol for commercial sheep flocks. Our results suggest that faecal egg de

Effects of sample size on the noise floor and distortion product otoacoustic emissions.

This study investigated the effects of sample size on the noise floor and distortion product otoacoustic emissions (DPOAEs) in 55 normal-hearing subjects as a function of intensity. More specifically, we investigated the effects of sample size (12-400 sweeps) as a function of intensity (L1 = L2 = 35, 45 and 55 dB SPL), firstly, on the identifiability of DPOAEs (2F1-F2), secondly, on the noise floor adjacent to DPOAEs, and thirdly, on the magnitude of DPOAEs centred around geometric means of 531 Hz, 1,000 Hz, 2,000 Hz and 4,000 Hz. Testing was conducted with a commercially available system for

Minimum Sample Size Requirements for Machine Learning: A Study on Diabetic Neuropathy Prediction.

Early detection of diabetic neuropathy (DN) remains challenging due to its asymptomatic progression. Machine learning models hold promise for identifying patients at risk, yet most existing studies are limited by small sample sizes that hinder generalizability. In this study, we evaluated minimum sample size requirements for machine learning using a population-based dataset of 77,724 individuals with diabetes. We generated balanced subsets with varying sample sizes (n=100-25,000) and numbers of features (3-46). We trained random forest models on each configuration and evaluated performance usi

Principle of sample size calculation in gastroenterology research: a practical guide for clinicians.

INTRODUCTION: A proper sample size calculation enables to conduct adequately powered randomized controlled trials (RCTs) and to provide a valid assessment of a specific clinical question. AREAS COVERED: In the current manuscript, we tried to provide the reader with an easy guide on the principles of sample size calculation in RCTs, tailored specifically to the context of gastroenterology and hepatology. The basics of sample size calculation were commented with a description of some of the main methods, including the calculation of the non-inferiority margin for non-inferiority RCTs and the ca

Sample size planning for estimating the global win probability with precision and assurance.

Randomized controlled trials commonly employ multiple endpoints to collectively assess the intended effects of the new intervention on multiple aspects of the disease. Focusing on the estimation of the global win probability (WinP), defined as the (weighted) mean of the WinPs across the endpoints that a treated participant would have a better outcome than a control participant, we propose a closed-form sample size formula incorporating pre-specified precision and assurance, with precision denoted by the lower limit of confidence interval and assurance denoted by the probability of achieving th

Minimum sample size for estimating the Net Promoter Score under a Bayesian approach.

At some moment in our lives, we are probably faced with the following question: How likely is it that you would recommend [company X] to a friend or colleague?. This question is related to the Net Promoter Score (NPS), a simple measure used by several companies as indicator of customer loyalty. Even though it is a well-known measure in the business world, studies that address the statistical properties or the sample size determination problem related to this measure are still scarce. We adopt a Bayesian approach to provide point and interval estimators for the NPS and discuss the determination

Design and sample size considerations for Alzheimer's disease prevention trials using multistate models.

BACKGROUND/AIMS: Clinical trials for Alzheimer's disease have been aimed primarily at persons who have cognitive symptoms at enrollment. However, researchers are now recognizing that the pathophysiological process of Alzheimer's disease begins years, if not decades, prior to the onset of clinical symptoms. Successful intervention may require intervening early in the disease process. Critical issues arise in designing clinical trials for primary and secondary prevention of Alzheimer's disease including determination of sample sizes and follow-up duration. We address a number of these issues thr

Sample size calculations for clinical trials targeting tauopathies: a new potential disease target.

Disease-modifying therapies are being developed to target tau pathology, and should, therefore, be tested in primary tauopathies. We propose that progressive apraxia of speech should be considered one such target group. In this study, we investigate potential neuroimaging and clinical outcome measures for progressive apraxia of speech and determine sample size estimates for clinical trials. We prospectively recruited 24 patients with progressive apraxia of speech who underwent two serial MRI with an interval of approximately 2 years. Detailed speech and language assessments included the Apraxi
