Increased mitochondrial Ca2+ and decreased sarcoplasmic reticulum Ca2+ in mitochondrial myopathy.

Genetic mutations that affect mitochondrial function often cause skeletal muscle dysfunction. Here, we used mice with skeletal-muscle-specific disruption of the nuclear gene for mitochondrial transcription factor A (Tfam) to study whether changes in cellular Ca(2+) handling is part of the mechanism of muscle dysfunction in mitochondrial myopathy. Force measurements were combined with measurements of cytosolic Ca(2+), mitochondrial Ca(2+) and membrane potential and reactive oxygen species in intact, adult muscle fibres. The results show reduced sarcoplasmic reticulum (SR) Ca(2+) storage capacit

[Measuring real-time sarcoplasmic reticulum calcium pumping in skinned muscle fibers].

A method and a device for direct measurements of accumulation of calcium in the sarcoplasmic reticulum (SR) and its release from SR as a function of free Ca2+ in bath have been developed. About 30% of the volume inside muscle fibers of swimbladder of Opsanus tau is occupied by SR. A set of solutions was prepared for fiber dissection and making holes in outer membrane without destruction of membranes of the sarcoplasmic reticulum. Calcium was unloaded from SR using EGTA as a pCa buffer. Then solutions with 50-100 microM CaFURA2 or CabisFURA2 were used as pCa-buffer and fluorescent Ca-indicators

Identification of dantrolene binding sites in porcine skeletal muscle sarcoplasmic reticulum.

Dantrolene, an intracellularly acting skeletal muscle relaxant, inhibits Ca2+ release from the sarcoplasmic reticulum during excitation-contraction coupling by an unknown mechanism. The drug is used to treat malignant hyperthermia, a genetic sensitivity to volatile anesthetics which results in the massive release of intracellular Ca2+ from affected skeletal muscle. We hypothesize that determination of the site of action of dantrolene will lead to further understanding of the regulation of sarcoplasmic reticulum calcium release. We report the identification of specific dantrolene binding sites

Caffeine stimulation of malignant hyperthermia-susceptible sarcoplasmic reticulum Ca2+ release channel.

The altered caffeine sensitivity of malignant hyperthermia-susceptible (MHS) muscle contracture is one basis of the diagnostic test for this syndrome. To determine whether the Arg615-to-Cys615 mutation of the porcine sarcoplasmic reticulum (SR) Ca2+ release channel is directly responsible for this altered caffeine sensitivity, the single-channel kinetics of purified MHS and normal pig Ca2+ release channels were examined. Initial studies demonstrated that decreasing the pH of the medium in either the cis- or trans-chamber decreased the Ca2+ release channel percent open time (Po). The half-inhib

ATP regulation of calcium transport in back-inhibited sarcoplasmic reticulum vesicles.

At high concentrations of ATP, ATP hydrolysis and Ca2+ transport by the (Ca2+ + MG2+)-ATPase of intact sarcoplasmic reticulum vesicles exhibit a secondary activation that varies with the extent of back-inhibition by Ca2+ accumulated within the vesicles. When the internal ionized Ca2+ is clamped at low and intermediate levels by the use of Ca-precipitating anions, the apparent Km values for activation by ATP are lower than in fully back-inhibited vesicles (high internal Ca2+). In leaky vesicles unable to accumulate Ca2+, raising Ca2+ in the assay medium from 20-30 microM to 5 mM abolishes the a

Regulation of Ca2+ transport by sarcoplasmic reticulum Ca2+-ATPase at limiting [Ca2+].

The factors regulating Ca2+ transport by isolated sarcoplasmic reticulum (SR) vesicles have been studied using the fluorescent indicator Fluo-3 to monitor extravesicular free [Ca2+]. ATP, in the presence of 5 mM oxalate, which clamps intravesicular [Ca2+] at approximately 10 microM, induced a rapid decline in Fluo-3 fluorescence to reach a limiting steady state level. This corresponds to a residual medium [Ca2+] of 100 to 200 nM, and has been defined as [Ca2+]lim, whilst thermodynamic considerations predict a level of less than 1 nM. This value is similar to that measured in intact muscle with

Control of heat production by the Ca2+-ATPase of rabbit and trout sarcoplasmic reticulum.

The sarcoplasmic reticulum Ca2+-ATPase of rabbit skeletal muscle can convert the energy derived from a Ca2+ gradient into heat (L. de Meis, M. L. Bianconi, and V. A. Suzano. FEBS Lett. 406: 201-204, 1997). In this report, it is shown that this conversion varies depending on the temperature and on whether rabbit (endotherm) or trout (poikilotherm) sarcoplasmic reticulum vesicles are used. The gradient doubled the yield of heat produced during ATP hydrolysis and the calorimetric enthalpy of ATP hydrolysis (DeltaHcal) value found with both rabbit and trout varied between -10 and -12 kcal/mol in l

Activation of the Ca2+ release channel of cardiac sarcoplasmic reticulum by volatile anesthetics.

BACKGROUND: Depression of myocardial contractility associated with the volatile anesthetics is well established clinically and experimentally. The molecular mechanisms underlying this effect, however, have not been completely characterized. Whereas the Ca2+ release channel of cardiac sarcoplasmic reticulum (SR) has been implicated as a potential target contributing to anesthetic-induced myocardial depression, the effect of the volatile anesthetics on this protein have not been characterized at the isolated, single-channel level. The authors sought to identify changes in channel gating and cond

Effects of ischemia on sarcoplasmic reticulum and contractile myofilament activity in human myocardium.

The effects of global ischemia on the contractile system and on sarcoplasmic reticulum (SR) function were studied by measuring the isometric tension and the SR Ca2+ release activity of chemically skinned cardiac fiber preparations from seven patients undergoing open-heart surgery. Ten minutes of ischemia caused 1) a decrease in the myofilament sensitivity to Ca2+ (expected Ca2+ concentration giving half-maximal tension; from 0.69 +/- 0.04 to 1.38 +/- 0.06 microM, n = 7) and in the cooperativity index (Hill coefficient; from 2.61 +/- 0.45 to 0.92 +/- 0.15, n = 7), 2) a decrease in myosin light

Adenylate cyclase in sarcoplasmic reticulum of skeletal muscle: distribution, orientation, and regulation.

We found specific activity of adenylate cyclase (AC) to be as high in rat skeletal muscle sarcoplasmic reticulum (SR) as in sarcolemma (SL) (39 +/- 5 pmol/mg per min and 34 +/- 5 pmol/mg per min). Detection of AC in SR could not be due to SL contamination. Activity in SR was similar in triads (heavy SR) and longitudinal reticulum (light SR), despite virtual absence of surface membrane markers in preparations of light SR. Also, AC of SR and SL may differ biochemically. In the presence of 10(-5) M 5'-guanylylimidodiphosphate, 10(-4) M isoproterenol increased SL activity 508%, crude SR 46.4%, hea

[Changes in spatial organization in sarcoplasmic reticulum membranes in rabbits with experimental thyrotoxicosis].

The structure of sarcoplasmic reticulum membranes (SR) of skeletal muscles from normal and thyrotoxic rabbits was studied with the use of the fluorescent probe pyrene. It was found that protein globules in SR preparations of thyrotoxic animals were submerged into the lipid bilayer to a greater extent than in the reticulum of normal animals. Electrophoresis in polyacrylamide gel revealed no detectable differences between SR of normal and thyrotoxic rabbits. The ratio protein/lipid in SR membranes remained 2:1 in rabbits with thyrotoxicosis.

[Structural and functional characteristics of the sarcoplasmic reticulum membranes of the skeletal muscles in thyrotoxicosis].

A rate of CA2+ accumulation and efficiency of Ca2+-pump were decreased in the fractions of skeletal muscle sarcoplasmic reticulum isolated from rabbits with thyrotoxicosis. Temperature of phase transition in membranes of sarcoplasmic reticulum was changed by 4-5 degrees towards low temperatures. Under conditions of thyrotoxicosis fatty acid composition of total lipids from the sarcoplasmic reticulum fractions was altered as follows: amount of palmitic acid was increased 1.4-fold but oleic and arachidonic acids were decreased 21.8% and 40.5%, respectively. Intercellular NETwork-facilitated sarcoplasmic reticulum targeting for myocardial ischemia-reperfusion injury treatment.

Myocardial ischemia-reperfusion injury (MIRI) often leads to irreversible myocardium dysfunction, while existing therapies are palliatives that transiently alleviate the disease symptoms. Repairing sarcoplasmic reticulum Ca2+-ATPase (SERCA) could reverse MIRI, which, however, requires precise drug delivery to the sarcoplasmic reticulum (SR). To this end, we leverage cell-cell "NETwork" of neutrophils to deliver SERCA activator-loaded SR-localized nanoparticles (L-P-NPs) to the damaged myocardial cells, following a hierarchical targeting process: (i) chemotactic neutrophils deliver L-P-NPs to i

Abnormal human sarcoplasmic reticulum Ca2+ release channels in malignant hyperthermic skeletal muscle.

Single sarcoplasmic reticulum (SR) Ca2+ release channels were reconstituted from normal and malignant hyperthermic (MH) human skeletal muscle biopsies (2-5 g samples). Conduction, gating properties, and myoplasmic Ca2+ dependence of human SR Ca2+ release channels were similar to those in other species (rabbit, pig). The MH diagnostic procedure distinguishes three phenotypes (normal, MH-equivocal, and MH-susceptible) on the basis of muscle contracture sensitivity to caffeine and/or halothane. Single channel studies reveal that human MH muscles (both MH phenotypes) contain SR Ca2+ release channe

Abnormal rapid Ca2+ release from sarcoplasmic reticulum of malignant hyperthermia susceptible pigs.

Using the rapid filtration technique to investigate Ca2+ movements across the sarcoplasmic reticulum (SR) membrane, we compare the initial phases of Ca2+ release and Ca2+ uptake in malignant hyperthermia susceptible (MHS) and normal (N) pig SR vesicles. Ca2+ release is measured from passively loaded SR vesicles. MHS SR vesicles present a 2-fold increase in the initial rate of calcium release induced by 0.3 microM Ca2+ (20.1 +/- 2.1 vs. 6.3 +/- 2.6 nmol mg-1 s-1). Maximal Ca2+ release is obtained with 3 microM Ca2+. At this optimal concentration, rate of Ca2+ efflux in absence of ATP is 55 and

MYOM1 deficiency leads to dilated cardiomyopathy by disrupting the homeostasis of sarcoplasmic reticulum.

MYOM1, a major component of the vertebrate myofibrillar M band, binds myosin, titin, and light meromyosin, has been linked to cardiomyopathy and unexpected sudden death, yet the pathogenic mechanisms remain unclear. Leveraging the UK Biobank, we identified a significant association between dilated cardiomyopathy (DCM) and loss-of-function (LoF) MYOM1 variants. Functional studies showed that MYOM1 deficiency precipitates DCM with overt heart failure, accompanied by sarcomeric disorganization, pathological structural remodeling, and mitochondrial abnormalities. Sarcolipin is a Conformation-Dependent Regulator of the Sarcoplasmic Reticulum Calcium Pump SERCA.

Muscle relaxation is enabled by the sarco-endoplasmic reticulum Ca2+-ATPase (SERCA), which removes calcium from the cytosol and returns it to the lumen of the sarcoplasmic reticulum. During transport, SERCA adopts a variety of conformational states which differ in their structure and affinity for substrates. The transition between these states is mediated by calcium and ATP binding and the formation of an aspartyl-phosphate intermediate, which enables the transport of calcium across the membrane. SERCA function is highly regulated because of the importance of calcium in processes such as muscl

Sarcoplasmic reticulum calcium leak contributes to arrhythmia but not to heart failure progression.

Increased sarcoplasmic reticulum (SR) Ca2+ leak via the cardiac ryanodine receptor (RyR2) has been suggested to play a mechanistic role in the development of heart failure (HF) and cardiac arrhythmia. Mice treated with a selective RyR2 stabilizer, rycal S36, showed normalization of SR Ca2+ leak and improved survival in pressure overload (PO) and myocardial infarction (MI) models. The development of HF, measured by echocardiography and molecular markers, showed no difference in rycal S36- versus placebo-treated mice. Reduction of SR Ca2+ leak in the PO model by the rycal-unrelated RyR2 stabiliz
