Discovery of small molecule ACC inhibitors: Potential treatment for excessive sebum secretion.

Overproduction of sebum can lead to various skin disorders, including acne and seborrheic dermatitis. Acetyl-CoA carboxylase is a key enzyme in the de novo synthesis of sebum. Consequently, inhibiting acetyl-CoA carboxylase is a feasible strategy to reduce sebum production, thereby providing therapeutic benefits for associated skin conditions. This review described the de novo synthesis of sebum and discusseed the various isoforms, functions and catalytic mechanisms of acetyl-CoA carboxylase. Additionally, it offered a comprehensive overview of the research advancements and structural-activity

Association between sebum secretion and cardiac sympathetic dysfunction in Parkinson's disease.

BACKGROUND: Patients with Parkinson's disease (PD) have α-synuclein (α-Syn) deposition in the skin, and decreased sebum secretion due to epidermal dysfunction. However, the relationship between sebum secretion and autonomic neuropathies is unknown. METHODS: Using the medical records in our facility, we identified patients newly diagnosed with PD on admission from August 2020 to December 2023. We analyzed whether sebum secretory ability at multiple sites was associated with cardiac sympathetic nerve function that was assessed by cardiac 123I-metaiodobenzylguanidine scintigraphy. RESULTS: Fort

Identifying robust and reliable volatile organic compounds in human sebum for biomarker discovery.

Sebum from sebaceous glands is a rich source of volatile organic compounds (VOCs) that can readily be sampled non-invasively from the surface of skin. The VOC profiles of sebum can then be used to obtain information regarding different medical conditions including diabetes and Parkinson's Disease. However, the effects of sampling approaches and environmental factors on sebum VOC profiles are not established and the confident attribution of VOCs to disease states needs to be free of extraneous influences such as sampling materials and preparatory conditions. Here, we investigated a more standar

Novel Demodex detection method involving non-invasive sebum collection and next-generation sequencing.

INTRODUCTION: Demodex mites are common human ectoparasites found across a broad geographical range. They reside in pilosebaceous units of the skin and feed on sebum, epithelial and glandular cells. D. folliculorum is the more common mite, inhabiting the upper end of the pilosebaceous unit while D. brevis resides deeper in the skin and meibomian glands. Until now, Demodex mites have been obtained by various techniques such as skin scraping, cellophane tape, plucking eyelashes, and also by invasive biopsies. AIM: To assess whether non-invasively collected sebum samples of patients suspected of

Green Tea and Other Tea Polyphenols: Effects on Sebum Production and Acne Vulgaris.

Polyphenols are antioxidant molecules found in many foods including nuts, fruits, vegetables, chocolate, wine, and tea. Polyphenols have antimicrobial, anti-inflammatory, and antineoplastic properties. Recent studies suggest that tea polyphenols may be used for reducing sebum production in the skin and for treatment of acne vulgaris. This review examines the evidence for use of topically and orally ingested tea polyphenols against sebum production and for acne treatment and prevention. The PubMed database was searched for studies on tea polyphenols, sebum secretion, and acne vulgaris. Of the 5

Detailed composition of wax esters in mouse sebum and the involvement of FAR2 and AWAT2.

Wax esters (WEs) are lipids characteristic of sebaceous glands and are classified into wax monoesters (WmEs) and wax diesters (WdiEs). Here, we analyzed WEs in mouse sebum via liquid chromatography-tandem mass spectrometry to characterize their composition and structural diversity. We found that mouse sebum contained type 2α and type 2ω WdiEs and revealed the detailed species composition of these WdiEs together with WmEs. In knockout mice of the acyl-CoA reductase Far2, the quantities of WEs with very-long-chain fatty alcohols were reduced relative to wild-type mice. In knockout mice of the ac

Inhibition of Sebum Production with the Acetyl Coenzyme A Carboxylase Inhibitor Olumacostat Glasaretil.

Olumacostat glasaretil (OG) is a small molecule inhibitor of acetyl coenzyme A (CoA) carboxylase (ACC), the enzyme that controls the first rate-limiting step in fatty acid biosynthesis. Inhibition of ACC activity in the sebaceous glands is designed to substantially affect sebum production, because over 80% of human sebum components contain fatty acids. OG inhibits de novo lipid synthesis in primary and transformed human sebocytes. TrueMass Sebum Panel analyses showed a reduction in saturated and monounsaturated fatty acyl chains across lipid species, including di- and triacylglycerols, phospho

Acne is an inflammatory disease and alterations of sebum composition initiate acne lesions.

Hyperseborrhoea has been considered as a major aetiopathogenetic factor of acne. However, changes in sebaceous gland activity not only correlate with seborrhoea but also with alterations in sebum fatty acid composition. Current findings indicate that sebum lipid fractions with proinflammatory properties and inflammatory tissue cascades are associated in the process of the development of acne lesions. The oxidant/antioxidant ratio of the skin surface lipids and alterations of lipid composition are the main players in the induction of acne inflammation. Nutrition may influence the development of

The effect of ketoconazole on sebum secretion in patients suffering from acne and seborrhoea.

Ketoconazole, an imidazolic antimycotic derivative, has proved to have an antiandrogenous effect, which may be observed to inhibit the adrenal and gonadal steroid synthesis. The authors wished to verify this effect of the drug in men and women suffering from acne and seborrhoea, conditions believed to indicate a heightened androgenous effect in the sebaceous gland. They aimed to do this by studying the cutaneous sebum, which is considered a reliable indirect indicator of androgenous activity. They found a statistically significant decrease in the rate of sebum production in all patients admini

Sebum-suppressing activity of the nonpolar arotinoid Ro 15-0778 in rodents.

Retinoids are known to modulate sebaceous gland activity in humans and animals. The nonpolar arotinoid Ro 15-0778 [(E)-1,2,3,4-tetrahydro-1,1,4,4-tetramethyl-6-(1-methyl-2-phenylethen yl) naphthalene] does not contain a polar end group and is devoid of the classical retinoid side effects of hypervitaminosis A. The favorable toxicological profile stimulated the evaluation of this arotinoid in animal models of sebum production. In castrated, testosterone-stimulated male rats, Ro 15-0778 is 50 times more potent than 13-cis-retinoic acid in inhibiting the production and subsequent secretion of seb

Lipidomic Insights into Seborrheic Dermatitis: Clinical Evaluation of Sebum Changes Using SpiderMass.

INTRODUCTION: Seborrheic dermatitis (SD) is a skin disorder in which chronic inflammation of the scalp is linked to excessive sebum production and overgrowth of Malassezia fungi. These fungi break down sebum lipids, releasing pro-inflammatory unsaturated fatty acids. Lipidomic and metabolomic sebum analysis is thus essential to better understand SD. SpiderMass, an innovative ambient ionization mass spectrometry (AIMS) technology, allows for fast, direct, and non-invasive biochemical profiling of sebum. This study aimed to assess the potential of SpiderMass for evaluating the skin metabolome an

Ozenoxacin suppresses sebum production by inhibiting mTORC1 activation in differentiated hamster sebocytes.

Acne vulgaris is a complex condition involving factors that affect the pilosebaceous unit. A primary manifestation of acne pathology is the development of comedones, often linked to the overproduction of sebum resulting from 5α-dihydrotestosterone (5α-DHT) and insulin activity. Ozenoxacin is a topical quinolone that exhibits potent antibacterial activity against Cutibacterium acnes (C. acnes). It is commonly used to treat acne associated with this bacterium; however, its effect on sebum production within the sebaceous glands remains unclear. In this study, the effects of ozenoxacin on sebum pr

Sebum Components Dampen the Efficacy of Skin Disinfectants against Cutibacterium acnes Biofilms.

At Canadian Blood Services, despite the use of 2% chlorhexidine and 70% isopropyl alcohol (standard disinfectant, SD) prior to venipuncture, Cutibacterium acnes evades eradication and is a major contaminant of platelet concentrates (PCs). Since C. acnes forms bacterial aggregates known as biofilms in the sebaceous niches of the skin, this study aimed to assess whether sebum-like components impact disinfectant efficacy against C. acnes leading to its dominance as a PC contaminant. C. acnes mono-species and dual-species biofilms (C. acness and a transfusion-relevant Staphylococcus aureus isolate

Sebum levels are associated with the relationship between skin properties and microbiota in Japanese women.

The diversity of the skin microbiota in Japanese women increases with skin aging and is accompanied by decreased sebum content. However, the impact of changes in skin microbiota on skin aging remains unclear. We conducted a cross-sectional subgroup analysis based on age, menopausal status, and sebum content in a cohort of Japanese women aged 30-60 years to elucidate the differences in the relationship between skin properties and microbiota under various conditions. As a fundamental characteristic of this cohort, we confirmed that older participants had lower sebum content and a higher alpha-di
