Comparative Study of the Antennal Phenotype in Species of the Belminus (Hemiptera: Triatominae) Genus Using Optical and Scanning Electron Microscopy.

The genus Belminus Stål, 1859 is distinguished by markedly entomophagous species. This genus is a poorly studied group of the Triatominae (Jeannel, 1919), which includes hematophagous species that are vectors of Trypanosoma cruzi (Chagas, 1909), the etiological agent of Chagas disease. This study reports for the first time the description of the antennal sensilla of Belminus corredori (Galvão & Angulo, 2003), Belminus herreri (Lent & Wygodzinsky, 1979), and Belminus ferroae (Sandoval, Pabón, Jurberg & Galvão, 2007) nymphs and adults throughout scanning electron microscopy and optical microscop

The microfiber texture in a specialized plastic cuticle area within a sensillum field on the cockroach maxillary palp as revealed by freeze fracturing.

A field of sensilla extends across the ventral surface of the terminal segment of the maxillary palps of Periplaneta americana. The sensilla project from a sheet of pliable cuticle. Ultrathin sections of the cuticle in this area reveal a clear-cut parabolic microfiber pattern. Microfibers can also be seen from freeze fracture faces running parallel to the cuticular surface. These microfibers have a diameter of 80 A and may consist of chitin crystallites surrounded by a matrix coat. The number of straight parallel microfibers visible in a fracture face increases the more closely parallel to the

Discrete roles of Ir76b ionotropic coreceptor impact olfaction, blood feeding, and mating in the malaria vector mosquito Anopheles coluzzii.

Anopheline mosquitoes rely on their highly sensitive chemosensory apparatus to detect diverse chemical stimuli that drive the host-seeking and blood-feeding behaviors required to vector pathogens for malaria and other diseases. This process incorporates a variety of chemosensory receptors and transduction pathways. We used advanced in vivo gene-editing and -labeling approaches to localize and functionally characterize the ionotropic coreceptor AcIr76b in the malaria mosquito Anopheles coluzzii, where it impacts both olfactory and gustatory systems. AcIr76b has a broad expression pattern in fem

Patterning an epidermal field: Drosophila lozenge, a member of the AML-1/Runt family of transcription factors, specifies olfactory sense organ type in a dose-dependent manner.

Sense organ development in the Drosophila antenna is initiated by the selection of a founder cell from an epidermal field. This cell is believed to recruit neighbours to form a cluster of cells which then divides to form a mature sense organ. In most systems so far studied, sense organ type appears to be specified by the identity of proneural genes involved in the selection of precursors. The regulation of proneural gene expression is, in turn, controlled by the prepatterning genes. In the antenna, the only known proneural function is that of atonal, a gene that is involved in founder cell cho

Ultrastructural comparison of external morphology of immature stages of Lutzomyia (Nyssomyia) intermedia and Lutzomyia (Nyssomyia) whitmani (Diptera: Psychodidae), vectors of cutaneous leishmaniasis, by scanning electron microscopy.

Lutzomyia (Nyssomyia) intermedia (Lutz & Neiva 1912) and Lutzomyia (Nyssomyia) whitmani (Antunes & Coutinho 1939) (Diptera: Psychodidae) are vectors of American cutaneous leishmaniasis in several endemic regions of Brazil. We analyzed the external morphological aspects of the immature stages of these two vectors by using scanning electron microscopy. In general, the larval stages of the two species are morphologically similar, although some differences were noted. Detailed examination of the eggs of both species revealed similar exchorionic ornamentations of unconnected parallel ridges. The la

Identification and analysis of odorant receptors expressed in the two main olfactory organs, antennae and palps, of Schistocerca americana.

Locusts depend upon their sense of smell and provide useful models for understanding olfaction. Extending this understanding requires knowledge of the molecular and structural organization of the olfactory system. Odor sensing begins with olfactory receptor neurons (ORNs), which express odorant receptors (ORs). In insects, ORNs are housed, in varying numbers, in olfactory sensilla. Because the organization of ORs within sensilla affects their function, it is essential to identify the ORs they contain. Here, using RNA sequencing, we identified 179 putative ORs in the transcriptomes of the two m

Sample Preparation Method of Scanning and Transmission Electron Microscope for the Appendages of Woodboring Beetle.

This report described sample preparation methods that scanning and transmission electron microscope observations, demonstrated by preparing appendages of the woodboring beetle, Chlorophorus caragana Xie & Wang (2012), for both types of electron microscopy. The scanning electron microscopy (SEM) sample preparation protocol was based on sample chemical fixation, dehydration in a series of ethanol baths, drying, and sputter-coating. By adding Tween 20 (Polyoxyethylene sorbitan laurate) to the fixative and the wash solution, the insect body surface of woodboring beetle was washed more cleanly in S

Larvae and adult flies of Rhinoestrus purpureus and R. usbekistanicus: morphology and pupation (Diptera: Oestridae).

Rhinoestrus species larvae are considered obligatory parasites of the nasal cavities of equine. This type of myiasis is characterised by sneezing, coughing, olfactory nerve damage and encephalomyelitis. Also, it has a zoonotic importance as the larvae might cause ophthalmomyiasis and conjunctivitis in human. While few studies describing R. purpureus adult fly antennal sensillae are available, the R. usbekistanicus antennal sensillae have never been described. Also, scanty data are available on the adult flies of Rhinoestrus species morphology. For this reason, the current study aimed at identi

Comparative study of mouthparts of four species of Culicoides from Tibet.

Mouthparts of four species of Culicoides from Tibet were studied by light and scanning electron microscopy (SEM). Photomicrographs were made of the mandible, labrum, hypopharynx, lacinia, and the sensory pit of the third palpal segment for each of the four species. Values for eighteen characters of five structures are presented graphically. Mean values for ten of the eighteen characters evaluated were significantly greater for C. circumscriptus and C. mamaensis than for C. maculatus and C. tibetensis. Photomicrographs revealed specific differences in camber and pitch of the fifth mandibular to

The TRP-channel painless mediates substrate stiffness sensing in the legs during Drosophila oviposition.

The distinct textural properties of fruits in varying stages of ripening present unique ecological opportunities for several species of fruit flies, resulting, over evolutionary times, in specialized egg-laying behaviors. In this study we identified a TrpA channel-dependent mechanosensory pathway in the legs, through the gene painless, that modulates the discernment of softer patches for oviposition in gravid D. melanogaster females. We report that the stiffness-sensing role of tarsi is mediated through external sensory organs, namely ventral mechanosensory bristles and subsets of campaniform

Antennal and maxillary palp morphology, and sensillar equipment, of the spruce bark beetle predators, Medetera signaticornis and Medetera infumata (Diptera: Dolichopodidae).

Many long-legged Medetera flies are natural enemies of bark beetle pests, which they detect using olfactory cues, likely through olfactory sensilla on the antennae and maxillary palps. Morphological characterisation of olfactory sensilla among insects can provide a basis for future taxonomic, phylogenetic or electrophysiological studies. Scanning electron microscopy was used to describe the morphology of olfactory organs and sensillar equipment of Medetera signaticornis and M. infumata. Three different olfactory sensillum types were found in both fly species, sensilla trichodea, s. basiconica

Molecular identification of cDNA, immunolocalization, and expression of a putative odorant-binding protein from an Asian honey bee, Apis cerana cerana.

An odorant-binding protein cDNA (Acer-ASP2) was cloned and characterized from antennae of adult workers of an Asian honey bee, Apis cerana cerana F. (Hymenoptera: Apidae). The full-length open reading frame of Acer-ASP2 cDNA was 429 bp, encoding 142 amino acids. Protein signature analyses revealed that it contained six conserved cysteines with an N-terminal signal sequence of 19 amino acids. The deduced protein sequence shares high homology with Amel-ASP2 from the honey bee, Apis mellifera L., and low similarity with odorant-binding proteins from other species of insects. Ultramorphological and molecular characteristics of the larval stages of the horse nasal-myiasis fly, Rhinoestrus sp. (Diptera: Oestridae) from Mongolian horse in China.

Rhinoestrus sp. (Diptera: Oestridae) is an economically important parasite that can cause severe nasal myiasis in equids and can also affect humans. The ultrastructure of all Rhinoestrus sp. larval instars from Mongolian horse was examined by light and scanning electron microscopy to characterize the features of Rhinoestrus. The structure of the anterior region, posterior region, and the spines of the third segment was analyzed for 10 specimens in each larval stage. Additionally, 34 third-instar (L3) larvae of Rhinoestrus sp. from Mongolian horse were subjected to molecular characterization by

Morphological and genetic differences in legs of a polygamous beetle between sexes, Glenea cantor (Coleopter: Cerambycidae: Lamiinae).

The legs of insects play an important role in their daily behaviour, especially reproduction. Entomologists have performed much research on the role of the leg in different behaviours of beetles, an important group in the insect family, but relatively little has been done to study the ultrastructure and transcriptome of their legs. Hence, we systematically studied the ultrastructure and gene expression of the leg of G. cantor, a polygynous beetle, and compared its male and female diversity. In this study, we found the fore-leg, mid-leg and hind-leg of the female were significantly longer than

Distribution and specific central projections of mechanoreceptors in the thorax and proximal leg joints of locusts.

Campaniform sensilla (CS) of the locust proximal leg segments are arranged predominantly as homologous groups on the trochantera and femora. Their number is reduced in the more specialized jumping legs. The specific projections of the primary afferent axons from single groups of CS are visualized after backfilling them with CoCl2 introduced through the dendritic area in the cuticle. The majority of central branching patterns reveal a high degree of uniformity if compared within a population, between populations of one segment, and between populations of different legs. All projections bifurcat

Ultrastructure of antennal sensory organs in Wohlfahrtia magnifica (Diptera: Sarcophagidae).

Wohlfahrtia magnifica (Schiner 1861) is the sole causative agent of vaginal myiasis in Bactrian camels, and its antennal sensilla serve as essential sensory organs for detecting environmental chemical cues. However, no systematic study has been reported on the morphology of antennal sensilla in male and female. Here, we examined and compared the antennal morphology of male and female W. magnifica using scanning electron microscopy. The antennae consist of 3 main segments, the scape, pedicel, and funiculus, with an arista arising from the funiculus. Four sensillum types were identified: chaetic

Stomodaeal valve ultrastructure in the sandfly Lutzomyia longipalpis (Diptera: Psychodidae).

The stomodaeal valve and adjacent cardia region of the alimentary canal of the phlebotomine sandfly Lutzomyia longipalpis have been studied using electron microscopy. With the scanning electron microscope (SEM), numerous pit-like structures of different sizes were found in the surface of the valve of male and female flies. Under the transmission electron microscope (TEM), the cells adjacent to the pores were shown to have numerous mitochondria and to lack neural innervation. It is suggested that the pit-like structures are probably secretory. A typical basiconic sensilla was found on the inner

Gustatory receptors in the abdominal terminus contribute to oviposition decisions in Anoplophora glabripennis.

BACKGROUND: Anoplophora glabripennis (Asian longhorned beetle, ALB) is a wood-boring forest pest causing tree decline and mortality. The abdominal terminus is essential for oviposition and bears numerous chemosensilla, but its role in gustatory perception and host selection remains unclear. RESULTS: Eight sensilla were identified on the female abdominal terminus using scanning electron microscopy, including three putative gustatory sensilla. RNA-sequencing of the abdominal terminus identified 30 gustatory receptors (AglaGR1-30). Reverse transcription quantitative polymerase chain reaction pro

Pheromone receptors for japonilure in Anomala corpulenta and Popillia japonica.

Using gas chromatography-electroantennographic detection (GC-EAD), gas chromatography-mass spectrometry (GC-MS), and chiral capillary GC with synthetic stereoisomers, followed by laboratory and field validation, we identified the sex pheromone of the copper green chafer (Anomala corpulenta) as (R)-japonilure, with its antipode, (S)-japonilure, acting as a behavioral antagonist. Single sensillum recordings (SSR) revealed two types of pheromone-responsive sensilla, one narrowly tuned to the sex pheromone, and another that responded to both (R)- and (S)-japonilure, albeit with lower sensitivity t
