Aoudad (Ammotragus lervia) as disease reservoirs for bighorn sheep (Ovis canadensis): experimental and epidemiological evidence.

This study evaluates the role of aoudad (Ammotragus lervia) as a maintenance host and reservoir for significant respiratory pathogens hindering bighorn sheep (Ovis canadensis) conservation; Mycoplasma ovipneumoniae and leukotoxigenic Pasteurellaceae. An experimental commingling trial and regional disease surveillance revealed significant interspecies transmission risks and epidemiological patterns. Experimentally, 80% of bighorn sheep exposed to aoudad inoculated with M. ovipneumoniae and leukotoxigenic Pasteurellaceae died of pneumonia under both indirect and direct contact conditions. Conver

Identification of novel immunogenic Mycoplasma ovipneumoniae proteins recognized by sera from domestic and bighorn sheep.

Domestic and bighorn sheep can both be infected by the bacterium Mycoplasma (Mesomycoplasma) ovipneumoniae. Disease presentation differs between these hosts, with bighorn sheep experiencing more severe outcomes; however, both species mount a humoral immune response following infection. This serological response has yet to be fully characterized; however, study of other species of Mycoplasma has demonstrated that disease severity can vary depending on the presence of antibodies, suggesting an important role of antibody production in disease manifestation. In the present study, a domestic sheep

Clearance of Mycoplasma ovipneumoniae in Captive Bighorn Sheep (Ovis canadensis) Following Extended Oral Doxycycline Treatment.

Respiratory disease is a significant barrier for bighorn sheep (Ovis canadensis) conservation, and a need remains for management options in both captive and free-ranging populations. We treated Mycoplasma ovipneumoniae infection in six bighorn lambs and five bighorn yearlings at two captive research facilities with twice daily oral doxycycline for 8 wk or longer. Doses of 5 mg/kg twice daily mixed in formula for lambs and 10 mg/kg twice daily mixed in moistened pellets for older lambs and yearlings were tolerated well with minimal side effects. All animals in this case report remain Mycoplasma

Role of Bibersteinia trehalosi, respiratory syncytial virus, and parainfluenza-3 virus in bighorn sheep pneumonia.

Pneumonic bighorn sheep (BHS) have been found to be culture- and/or sero-positive for Bibersteinia trehalosi, respiratory syncytial virus (RSV), and parainfluenza-3 virus (PI-3). The objective of this study was to determine whether these pathogens can cause fatal pneumonia in BHS. In the first study, two groups of four BHS each were intra-tracheally administered with leukotoxin-positive (Group I) or leukotoxin-negative (Group II) B. trehalosi. All four animals in Group I developed severe pneumonia, and two of them died within 3 days. The other two animals showed severe pneumonic lesions on eut

Whole-genome resequencing uncovers molecular signatures of natural and sexual selection in wild bighorn sheep.

The identification of genes influencing fitness is central to our understanding of the genetic basis of adaptation and how it shapes phenotypic variation in wild populations. Here, we used whole-genome resequencing of wild Rocky Mountain bighorn sheep (Ovis canadensis) to >50-fold coverage to identify 2.8 million single nucleotide polymorphisms (SNPs) and genomic regions bearing signatures of directional selection (i.e. selective sweeps). A comparison of SNP diversity between the X chromosome and the autosomes indicated that bighorn males had a dramatically reduced long-term effective populati

Brucellosis in captive Rocky Mountain bighorn sheep (Ovis canadensis) caused by Brucella abortus biovar 4.

Nine (four female, five male) captive adult Rocky Mountain bighorn sheep (Ovis canadensis) contracted brucellosis caused by Brucella abortus biovar 4 as a result of natural exposure to an aborted elk (Cervus elaphus) fetus. Clinical signs of infection were orchitis and epididymitis in males and lymphadenitis and placentitis with abortion in females. Gross pathologic findings included enlargement of the testes or epididymides, or both, and yellow caseous abscesses and pyogranulomas of the same. Brucella abortus biovar 4 was cultured in all bighorn sheep from a variety of tissues, including test

Exposure to Psoroptes sp. mites is common among bighorn sheep (Ovis canadensis) populations in California.

Sera (n = 806) from 50 populations of bighorn sheep (Ovis canadensis) in California (USA) were evaluated for antibodies to Psoroptes sp. mites using a kinetic enzyme-linked immunosorbent assay (ELISA). Test values for each sample were determined to be either positive or negative at each of two ELISA cutoff values that provided either 100% sensitivity (low cutoff) or 100% specificity (high cutoff), respectively. One hundred sixty-eight (20.8%) sera were seropositive at the low cutoff value, and 87 (10.8%) of these sera also were seropositive at the high cutoff value. Eleven populations were des

Experimental infections of free-ranging Rocky Mountain bighorn sheep with lungworms (Protostrongylus spp.; Nematoda: Protostrongylidae).

Twelve free-ranging Rocky Mountain bighorn lambs (Ovis canadensis canadensis), each exposed experimentally to 125-1,000 infective third-stage larvae of Protostrongylus stilesi and P. rushi, shed significantly more first-stage larvae in their feces than did control lambs, but showed no clinical signs of illness and had equivalent summer and overwinter survival as control lambs. Two adult ewes, each exposed to 925 infective larvae, showed no increase in numbers of first-stage larvae in their feces; both survived at least 14 mo postexposure. Experimentally exposed lambs did not differ from contro

Transmission of Mannheimia haemolytica from domestic sheep (Ovis aries) to bighorn sheep (Ovis canadensis): unequivocal demonstration with green fluorescent protein-tagged organisms.

Previous studies demonstrated that bighorn sheep (Ovis canadensis) died of pneumonia when commingled with domestic sheep (Ovis aries) but did not conclusively prove that the responsible pathogens were transmitted from domestic to bighorn sheep. The objective of this study was to determine, unambiguously, whether Mannheimia haemolytica can be transmitted from domestic to bighorn sheep when they commingle. Four isolates of M. haemolytica were obtained from the pharynx of two of four domestic sheep and tagged with a plasmid carrying the genes for green fluorescent protein (GFP) and ampicillin res

Natural history of a bighorn sheep pneumonia epizootic: Source of infection, course of disease, and pathogen clearance.

A respiratory disease epizootic at the National Bison Range (NBR) in Montana in 2016-2017 caused an 85% decline in the bighorn sheep population, documented by observations of its unmarked but individually identifiable members, the subjects of an ongoing long-term study. The index case was likely one of a small group of young bighorn sheep on a short-term exploratory foray in early summer of 2016. Disease subsequently spread through the population, with peak mortality in September and October and continuing signs of respiratory disease and sporadic mortality of all age classes through early Jul

Effects of intrinsic and extrinsic factors on ruminating, grazing, and bedding time in bighorn sheep (Ovis canadensis).

Rumination is the repeated process of regurgitation of a food bolus, followed by chewing, swallowing, and regurgitation, which enhance nutrient assimilation. Time spent in lateral recumbency (i.e., bedded, lying) has often been used as a proxy for time spent ruminating due to difficulties of observing detailed rumination behavior in the field. The actual proportion of time spent ruminating, or other activities, will in turn be affected by the age and sex of an individual but also with changes in food quality. We studied the effects of intrinsic and extrinsic factors on time spent ruminating, b

The isolation and partial characterization of a Babesia sp. from desert bighorn sheep (Ovis canadensis nelsoni).

A novel Babesia parasite of desert bighorn sheep was isolated. Its taxonomic description, host range, pathogenicity and antigenic relatedness were investigated. The parasite was infective for black-tailed and white-tailed deer, but with host-specific differences compared to that of bighorn sheep. A splenectomized calf and domestic sheep were refractory to infection. A comparative immunofluorescence assay detected antigens cross-reactive with Babesia odocoilei, B. divergens, B. equi and B. caballi, but not with B. bovis or B. bigemina. Babesia odocoilei was also infective for bighorn sheep, all

Structure-property relationships of velar bone tissue from the energy absorbing horncore of bighorn sheep rams.

Velar bone is the material that fills the horncore of bighorn sheep rams. The architectural dimensions of velar bone are orders of magnitude larger than trabecular bone, and velae are more sail-like compared to strut-like trabeculae. Velar bone is important for energy absorption and reduction of brain cavity accelerations during high energy head impacts, but velar bone material properties were previously unknown. It was hypothesized that velar bone tissue would have properties that are beneficial for increased energy absorption at the material level. Solid velar bone beams were tested using dy

Effects of delivery method on serological responses of bighorn sheep to a multivalent Pasteurella haemolytica supernatant vaccine.

The safety and efficacy of a remotely delivered multivalent Pasteurella haemolytica supernatant vaccine (serotypes A2 and T10) were examined in captive Rocky, Mountain bighorn sheep (Ovis canadensis canadensis). Twenty bighorn sheep were grouped according to baseline leukotoxin neutralizing antibody titers (< or =2 or >2 log2(-1)) and vaccination history (previously vaccinated or unvaccinated). Within these groups, animals were randomly assigned to one of two delivery treatments: hand injection (control) or biobullet implantation. All bighorns received a single dose from the same lot of vaccin

How Respiratory Pathogens Contribute to Lamb Mortality in a Poorly Performing Bighorn Sheep ( Ovis canadensis ) Herd.

We evaluated bighorn sheep ( Ovis canadensis ) ewes and their lambs in captivity to examine the sources and roles of respiratory pathogens causing lamb mortality in a poorly performing herd. After seven consecutive years of observed December recruitments of <10%, 13 adult female bighorn sheep from the remnant Gribbles Park herd in Colorado, US were captured and transported to the Thorne-Williams Wildlife Research Center in Wyoming in March 2013. Ewes were sampled repeatedly over 16 mo. In April 2014, ewes were separated into individual pens prior to lambing. Upon death, lambs were necropsied a

Spontaneous pasteurellosis in captive Rocky Mountain bighorn sheep (Ovis canadensis canadensis): clinical, laboratory, and epizootiological observations.

We observed clinical signs, compared adrenal responses, and performed diagnostic tests on 12 captive Rocky Mountain bighorn sheep (Ovis canadensis canadensis) during a spontaneous outbreak of pasteurellosis. Cortisol in urine and feces was measured for bighorns sampled three times between 20 October and 1 November 1986. By 6 November, four of these had developed pneumonia, four showed only mild rhinitis, and four remained clinically normal. Bighorns that ultimately developed pneumonia showed elevated mean urinary (P = 0.003) and fecal (P = 0.046) cortisol excretion over the 12-day sampling per

Disease Ecology of a Low-Virulence Mycoplasma ovipneumoniae Strain in a Free-Ranging Desert Bighorn Sheep Population.

Infectious pneumonia associated with the bacterial pathogen Mycoplasma ovipneumoniae is an impediment to bighorn sheep (Ovis canadensis) population recovery throughout western North America, yet the full range of M. ovipneumoniae virulence in bighorn sheep is not well-understood. Here, we present data from an M. ovipneumoniae introduction event in the Zion desert bighorn sheep (Ovis canadensis nelsoni) population in southern Utah. The ensuing disease event exhibited epidemiology distinct from what has been reported elsewhere, with virtually no mortality (0 adult mortalities among 70 animals tr
