Anisotropic silk fibroin/gelatin scaffolds from unidirectional freezing.

Recent studies have underlined the importance of matching scaffold properties to the biological milieu. Tissue, and thus scaffold, anisotropy is one such property that is important yet sometimes overlooked. Methods that have been used to achieve anisotropic scaffolds present challenges such as complicated fabrication steps, harsh processing conditions and toxic chemicals involved. In this study, unidirectional freezing was employed to fabricate anisotropic silk fibroin/gelatin scaffolds in a simple and mild manner. Morphological, mechanical, chemical and cellular compatibility properties were

Gentamicin promotes Staphylococcus aureus biofilms on silk suture.

BACKGROUND: Communities of bacteria, termed biofilms, develop on biotic and abiotic surfaces, including medical devices and surgical suture. Biofilm-associated bacteria are typically recalcitrant to antibiotic therapy, and the effects of antibiotics on microbial biofilms are not clearly understood. There is emerging evidence that under specific conditions, aminoglycosides may actually promote biofilm development. Experiments were designed to study the effects of gentamicin on suture-associated Staphylococcus aureus biofilms. MATERIALS AND METHODS: S. aureus biofilms were formed after 24 h inc

[Wild silk: a strong inhalation allergen (author's transl)].

In the course of 18 months, allergic bronchial asthma due to wild silk was diagnosed in 26 patients (19 women, 7 men) of the relatively high mean age at onset of 34 years. In 25 patients the allergy started 1-36 months after acquiring a so-called wild-silk rug, which contains wild-silk waste; one patient fell ill after occupational contact with the waste of wild silk. In all patients the scratch test with wild-silk waste was strongly positive. Elimination of the rug or the occupational exposure made all the symptoms disappear. Wild-silk waste, product of wild-growing oak silkworms, is thus a r

Cytoprotection of Human Progenitor and Stem Cells through Encapsulation in Alginate Templated, Dual Crosslinked Silk and Silk-Gelatin Composite Hydrogel Microbeads.

Susceptibility of mammalian cells against harsh processing conditions limit their use in cell transplantation and tissue engineering applications. Besides modulation of the cell microenvironment, encapsulation of mammalian cells within hydrogel microbeads attract attention for cytoprotection through physical isolation of the encapsulated cells. The hydrogel formulations used for cell microencapsulation are largely dominated by ionically crosslinked alginate (Alg), which suffer from low structural stability under physiological culture conditions and poor cell-matrix interactions. Here the fabri

Fabrication Scheme for Obtaining Transparent, Flexible, and Water-Insoluble Silk Films from Apparently Dissolved Silk-Gland Fibroin of Bombyx mori Silkworm.

Films from silk fibroin protein are one of the most promising biomaterials because of their exquisite balance between mechanical properties and biocompatibility. Numerous schemes have been proposed for processing fibroin film, utilizing liquid silk fibroin (LSF) or regenerated silk fibroin (RSF). The films cast from LSF or RSF in the solution state are water-soluble, and therefore require postproduction treatment inducing β-sheet formation, to render them insoluble in water. Many kinds of postproduction treatments, using alcohol-water solution, water vapor, or controlled temperature, have been

Silk-Based Matrices and c-Kit-Positive Cardiac Progenitor Cells for a Cellularized Silk Fibroin Scaffold: Study of an in vivo Model.

The production of a cellularized silk fibroin scaffold is very difficult because it is actually impossible to differentiate cells into a well-organized cardiac tissue. Without vascularization, not only do cell masses fail to grow, but they may also exhibit an area of necrosis, indicating a lack of oxygen and nutrients. In the present study, we used the so-called tyrosine protein kinase kit (c-Kit)-positive cardiac progenitor cells (CPCs) to generate cardiac cellularized silk fibroin scaffolds, multipotent cells isolated from the adult heart to date that can show some degree of differentiation

A Multiscale Characterization of Two Tropical Embiopteran Species: Nano- and Microscale Features of Silk, Silk-Spinning Behavior, and Environmental Correlates of their Distributions.

Embioptera display the unique ability to spin silk with their front feet to create protective domiciles. Their body form is remarkably uniform throughout the order, perhaps because they all live within the tight confines of silken tubes. This study contributes to an understanding of the ecology of Embioptera, an order that is rarely studied in the field. We conducted a census to quantify the habitats of two species with overlapping distributions on the tropical island of Trinidad in a search for characteristics that might explain their distinct ecologies. One species, Antipaluria urichi (Sauss

Surface Modification and Characterisation of Silk Fibroin Fabric Produced by the Layer-by-Layer Self-Assembly of Multilayer Alginate/Regenerated Silk Fibroin.

Silk-based medical products have a long history of use as a material for surgical sutures because of their desirable mechanical properties. However, silk fibroin fabric has been reported to be haemolytic when in direct contact with blood. The layer-by-layer self-assembly technique provides a method for surface modification to improve the biocompatibility of silk fibroin fabrics. Regenerated silk fibroin and alginate, which have excellent biocompatibility and low immunogenicity, are outstanding candidates for polyelectrolyte deposition. In this study, silk fabric was degummed and positively cha

Fabrication of a Silk Sericin Hydrogel System Delivering Human Lactoferrin Using Genetically Engineered Silk with Improved Bioavailability to Alleviate Chemotherapy-Induced Immunosuppression.

Chemotherapy is one of the main treatments for cancer; however, it usually causes severe atrophy of immune organs and self-immunity damage to patients. Human lactoferrin (hLF) is a multiple biofunctional protein in regulating the immune response and thus holds great promise to alleviate chemotherapy-caused immunosuppression. However, a sufficient hLF resource and efficient delivery of hLF remain a challenge. Here, we provide a useful strategy to simultaneously solve these two problems. A silk sericin hydrogel system delivering recombinant hLF (SSH-rhLF) was fabricated to alleviate the chemothe

Silk Sericin Functionalized with Carboxy Drugs for Dermocosmetic Applications.

Silk sericin protein, a textile industry by-product showing no toxicity on oral/topical administration, has recently been exploited as a drug delivery system. The high serine content in sericin makes its functionalization through covalent ester-type bonds feasible; however, only in a few cases have covalent conjugates of sericin with the drug to be delivered been developed. In this paper, we report the preparation of sericin conjugates with carboxy drugs of dermatological interest, i.e., naproxen, retinoic acid, and azelaic acid. Functional Performance of Silk Nanoparticles for Oral Drug Delivery.

Oral delivery of therapeutics, particularly peptides, proteins, and nucleic acids, remains a major challenge due to multiple biological barriers within the gastrointestinal (GI) tract. There is a pressing need for systems to stabilize and orally deliver peptides with high bioavailability. Current nanoparticle systems, such as metal-based, lipid-based, and synthetic polymers, have had limited success toward oral delivery due to toxicity, poor mucus penetration, and instability in acidic and enzymatic conditions. Silk nanoparticles (SNPs) offer an effective alternative, combining biodegradabilit

Silk-based materials: Emerging for tendon/tendon-bone regeneration.

The tendon and tendon-bone interface injuries have long been an issue for clinical practice. Autologous or allogeneic transplantation has been found to be a good method for repairing injuries; however, many factors limit the application of autologous and allogeneic transplants, which urges researchers to find a new type of strategy for regeneration. Silk fibroin (SF) scaffolds serve as promising exogenous biomaterials for alternative tendon and bone regeneration strategies. As its excellent biocompatibility, satisfactory mechanical properties, controllable biodegradability, and structural tuna

Thermoplastic silk-plasticizer membranes for biodegradable artificial corneal endothelium.

Corneal endothelial dysfunction (CED) induces stromal edema through aqueous influx, disrupting collagen organization and causing corneal opacity, with chronic cases progressing to fibrosis. As a major cause of corneal blindness, CED currently requires corneal transplantation for treatment. However, the scarcity of human donors necessitates the urgent development of artificial corneal endothelial transplant materials. Previously reported non-biodegradable polymer materials suffer from inadequate adhesion and poor water barrier efficiency, with long-term implantation potentially causing foreign-

Application of Silk-Fibroin-Based Hydrogels in Tissue Engineering.

Silk fibroin (SF) is an excellent protein-based biomaterial produced by the degumming and purification of silk from cocoons of the Bombyx mori through alkali or enzymatic treatments. SF exhibits excellent biological properties, such as mechanical properties, biocompatibility, biodegradability, bioabsorbability, low immunogenicity, and tunability, making it a versatile material widely applied in biological fields, particularly in tissue engineering. In tissue engineering, SF is often fabricated into hydrogel form, with the advantages of added materials. SF hydrogels have mostly been studied for

Silk Fibroin-Based Coatings for Pancreatin-Dependent Drug Delivery.

Triggerable coatings, such as pH-responsive polymethacrylate copolymers, can be used to protect the active pharmaceutical ingredients contained within oral solid dosage forms from the acidic gastric environment and to facilitate drug delivery directly to the intestine. However, gastrointestinal pH can be highly variable, which can reduce delivery efficiency when using pH-responsive drug delivery technologies. We hypothesized that biomaterials susceptible to proteolysis could be used in combination with other triggerable polymers to develop novel enteric coatings. Bioinformatic analysis suggest

Anderson light localization in biological nanostructures of native silk.

Light in biological media is known as freely diffusing because interference is negligible. Here, we show Anderson light localization in quasi-two-dimensional protein nanostructures produced by silkworms (Bombyx mori). For transmission channels in native silk, the light flux is governed by a few localized modes. Relative spatial fluctuations in transmission quantities are proximal to the Anderson regime. The sizes of passive cavities (smaller than a single fibre) and the statistics of modes (decomposed from excitation at the gain-loss equilibrium) differentiate silk from other diffusive structu

Nanofeatured silk fibroin membranes for dermal wound healing applications.

As an effort to create the next generation of improved skin graft materials, in this study, we modified the surfaces of a previously investigated material, silk fibroin, using a NaOH alkaline treatment to obtain a biologically inspired nanofeatured surface morphology. Such surfaces were characterized for roughness, energy, and chemistry. In addition, keratinocyte (skin-forming cells) adhesion and proliferation on such nanofeatured silk fibroin wound dressings were studied in an initial attempt to determine the promotion of an epidermal cover on the wound bed to form a new epidermal barrier.
