This study aimed to explore associations between social isolation, loneliness, and hypothyroidism risk, and whether genetic susceptibility modifies these links. It enrolled 434,628 hypothyroidism-free participants from the UK Biobank. Social isolation was assessed via 3 dichotomous questions, loneliness via 2 revised UCLA Loneliness Scale questions, and hypothyroidism cases via hospital/death records. Hypothyroidism polygenic risk scores (PRSs) were calculated using related genetic loci, with Cox proportional hazards models for analysis. Over a median 12.3-year follow-up, 9,873 hypothyroidism

Adversities associated with isolation during adolescence, including the lack of appropriate emotional and social experiences, can jeopardize the individual development leading to the onset of mental illnesses such as major depressive disorder. Girls have higher rates of depression compared to boys; however, the relative contribution of biological and cultural factors to such a gender-dependent difference remains unclear. To identify the role of biological factors in this distinct susceptibility, we exposed adolescent C57BL/6 male and female mice (n = 12-14) to social isolation and we evaluated

In humans, social isolation is a known risk factor for disorders such as substance use disorder and depression. In rodents, social isolation is a commonly used environmental manipulation that increases the occurrence of behaviors related to these disorders. Age is thought to influence the effects of social isolation, but this predictive relationship is not well-understood. The present study aimed to determine the effects of social isolation on mesolimbic dopamine release at different developmental age points in mice. The experimental ages and their corresponding comparison to human age stages

Exposure to adversity during early childhood and adolescence increases an individual's vulnerability to developing substance use disorder. Despite the knowledge of this vulnerability, the mechanisms underlying it are still poorly understood. Excitatory afferents to the nucleus accumbens (NAc) mediate responses to both stressful and rewarding stimuli. Understanding how adolescent social isolation alters these afferents could inform the development of targeted interventions both before and after drug use. Here, we used social isolation rearing as a model of early life adversity which we have pre

Depression is a mood disorder that is more prevalent in women and has been closely associated with chronic stress. Many models of depression have been suggested that consider different forms of stress. In fact, stress is present in the life of every human being, but only a few develop depression. Accordingly, it seems wrong to consider all stressed animals to be depressed, emphasizing the importance of predisposition for this mood disorder. Based on this finding, we evaluated a predisposition to depressive behavior of female rats on the forced swim test (FST), and the more immobile the animal

Social relationships are crucial for the development and maintenance of normal behavior in non-human primates. Animals that are raised in isolation develop abnormal patterns of behavior that persist even when they are later reunited with their parents. In rodents, social isolation is a stressful event and is associated with a decrease in hippocampal neurogenesis but considerably less is known about the effects of social isolation in non-human primates during the transition from adolescence to adulthood. To investigate how social isolation affects young marmosets, these were isolated from other

Obesity and metabolic dysfunction are risk factors for a number of chronic diseases, such as type 2 diabetes, hypertension, heart disease, stroke, and certain forms of cancers. Both animal studies and human population-based and clinical studies have suggested that chronic stress is a risk factor for metabolic disorders. A good social support system is known to exert positive effects on the mental and physical well-being of an individual. On the other hand, long-term deprivation of social contacts may represent a stressful condition that has negative effects on health. In the present study, we

Exposure to stress during puberty can lead to long-term behavioral alterations in adult rodents coincident with sex steroid hormone-dependent brain remodeling and reorganization. Social isolation is a stress for social animals like mice, but little is known about the effects of such stress during adolescence on later reproductive behaviors. The present study examined sexual behavior of ovariectomized, estradiol and progesterone primed female mice that were individually housed from 25 days of age until testing at approximately 95 days, or individually housed from day 25 until day 60 (during pub

UNLABELLED: In early August 2007, the Medical Advisory Secretariat began work on the Aging in the Community project, an evidence-based review of the literature surrounding healthy aging in the community. The Health System Strategy Division at the Ministry of Health and Long-Term Care subsequently asked the secretariat to provide an evidentiary platform for the ministry's newly released Aging at Home Strategy. After a broad literature review and consultation with experts, the secretariat identified 4 key areas that strongly predict an elderly person's transition from independent community living

The opioid systems play an important role in mediating both physical pain and negative affects (eg, the pain of social isolation). From an evolutionary perspective, it is not surprising that the neurocircuitry and neurochemistry of physical pain would overlap with that involved in complex social emotions. Exposure to trauma as well as a range of gene variants in the opioid system may be associated with alterations in opioid systems function, with changes in reward processing, and with vulnerability to substance abuse. A role for interventions with opioid agents in depression and anxiety disord

This research examines the connection between physical abuse and social isolation. Using data from the National Youth Survey, a measure of self-perceived social isolation was constructed indicating the extent to which respondents feel detached from their friends and from school. Those who had experienced violence were predicted to be more isolated than those who had not. Results strongly supported the hypothesis, controlling for theoretically relevant variables. Explanation is provided in terms of damage to attachment skills, social competence, and self-esteem concomitant to being a victim of

Social isolation is a potent but little understood risk factor for morbidity and mortality, and its negative consequences are most profound among the elderly, the poor, and minorities, some of the fastest growing segments of the U.S. population. A steadily increasing number of people are living alone and are therefore more likely to experience social isolation. We discuss four mechanisms--attractiveness, health behavior, stress, and repair and maintenance--by which perceived social isolation might affect health. Our studies show that neither attractiveness nor health behaviors differ as a func

The purpose of this study was to explore the prevalence of social isolation and its relationship to sexual trauma in a sample of Black women who smoke crack cocaine. Using a convenience sample of 115 Black women with a history of smoking crack cocaine, participants were interviewed for 2 to 4 hours and asked a variety of questions about their health, relationships, sexuality, and drug use. Bivariate and multivariate logistical regressions were used to predict whether the women reported being socially isolated. While social isolation was not necessarily a common experience among the sample, it

Juveniles deprived of social contact for an extended period of time demonstrate a broad range of adverse effects on behavior, including increased social aversion. Employing our recently developed integrative assay to assess social aversion, we discovered that social isolation is sufficient to induce decreases in social motivation and increases in social fear and hesitancy. Unsupervised behavioral analyses revealed that social aversion in isolated mice is largely driven by an inflexible, anxiety-like state marked by heightened social vigilance. Using intersectional, projection-specific neural c

Post-weaning social isolation (PSI) has been shown to increase aggressive behavior and alter medial prefrontal cortex (mPFC) function in rats. The present study sought to determine whether this phenotype would be normalized by increasing levels of the endocannabinoid 2-arachidonoylglycerol (2-AG) using pharmacological inhibition of monoacylglycerol lipase (MAGL). Male and female Sprague-Dawley rats were exposed to either 4 weeks of PSI or social rearing (SR) starting on postnatal day 21, then underwent a 15 min trial of social interaction with a novel, same-sex juvenile rat. Rats were administ

Social isolation is a pervasive stressor that disrupts social cognition, yet its neural mechanisms in adulthood remain unclear. Here, we showed that four weeks of social isolation in adult male mice selectively impaired social novelty memory without affecting general sociability, object recognition, working memory, or anxiety- and depression-like behaviors. This deficit was sex-specific, as female mice remained unaffected. Mechanistically, c-Fos mapping and in vivo calcium imaging revealed that social isolation induced a maladaptive shift in prefrontal excitatory-inhibitory (E/I) balance, char

OBJECTIVE: Social withdrawal is described as the condition in which an individual experiences a desire to make social contact, but is unable to satisfy that desire. It is an important issue for patients with motor neurone disease who are likely to experience severe physical impairment. This study aims to reassess the psychometric and scaling properties of the MND Social Withdrawal Scale (MND-SWS) domains and examine the feasibility of a summary scale, by applying scale data to the Rasch model.
METHODS: The MND Social Withdrawal Scale was administered to 298 patients with a diagnosis of MND, a

Pair-bonded relationships form during periods of close spatial proximity and high sociosexual contact. Like other monogamous species, marmosets form new social pairs after emigration or ejection from their natal group resulting in periods of social isolation. Thus, pair formation often occurs following a period of social instability and a concomitant elevation in stress physiology. Research is needed to assess the effects that prolonged social isolation has on the behavioral and cortisol response to the formation of a new social pair. We examined the sociosexual behavior and cortisol during th

OBJECTIVES: Most smokers desire to quit, yet many rely on unassisted attempts that frequently result in relapses. Facilitating early self-disclosure and support-seeking is a critical first step toward effective cessation. This study examines how message format and a recipient's individual context impact their readiness to engage with various support sources.
METHODS: In an experiment, adult smokers (N = 386) were assigned to a control or one of three message conditions: non-narrative, first-person narrative, or chatbot-delivered narrative. The study evaluated intentions to disclose smoking-re

INTRODUCTION: This study aims to examine the prevalence and factors associated with social isolation and loneliness among U.S. adults with self-reported hospitalization.
METHODS: Cross-sectional survey data from adults aged ≥18 years in the 2021 Medical Expenditure Panel Survey who reported at least 1 hospitalization were obtained and analyzed in 2025-2026. Weighted descriptive analyses and multivariable logistic regression models were conducted to identify factors independently associated with social isolation and loneliness.
RESULTS: Social isolation and loneliness were present in 22.6% an
