Sodium (±)-5-bromo-2-(α-hydroxypentyl) benzoate ameliorates pressure overload-induced cardiac hypertrophy and dysfunction through inhibiting autophagy.

Sodium (±)-5-bromo-2-(a-hydroxypentyl) benzoate (generic name: brozopine, BZP) has been reported to protect against stroke-induced brain injury and was approved for Phase II clinical trials for treatment of stroke-related brain damage by the China Food and Drug Administration (CFDA). However, the role of BZP in cardiac diseases, especially in pressure overload-induced cardiac hypertrophy and heart failure, remains to be investigated. In the present study, angiotensin II stimulation and transverse aortic constriction were employed to induce cardiomyocyte hypertrophy in vitro and in vivo, respec

Insights into membrane disruption and oxidative stress in Gram-positive and Gram-negative bacteria: role of sodium benzoate and phenoxyethanol.

This study investigates the antibacterial effects of two widely used preservatives in foods, pharmaceuticals, and personal care products, sodium benzoate and phenoxyethanol, on Gram-positive bacteria Staphylococcus aureus (ATCC 6538) and Gram-negative bacteria Pseudomonas aeruginosa (MTCC 1688). Despite their widespread usage, the exact antibacterial mechanisms of these preservatives are not yet fully understood. Our study demonstrated that the presence of SB and POE lead to extensive cellular rupture, membrane damage, and intracellular content leakage in bacteria. The results were validated t

Siderophore-producing bacteria from a sand dune ecosystem and the effect of sodium benzoate on siderophore production by a potential isolate.

Bioremediation in natural ecosystems is dependent upon the availability of micronutrients and cofactors, of which iron is one of the essential elements. Under aerobic and alkaline conditions, iron oxidizes to Fe(+3) creating iron deficiency. To acquire this essential growth-limiting nutrient, bacteria produce low-molecular-weight, high-affinity iron chelators termed siderophores. In this study, siderophore-producing bacteria from rhizosphere and nonrhizosphere areas of coastal sand dunes were isolated using a culture-dependent approach and were assigned to 8 different genera with the predomina

Sodium benzoate induces cardiotoxicity by suppressing the PI3K/AKT/GSK3β signaling axis: An integrated network toxicology and experimental study.

Sodium benzoate (SB), a common food preservative, has raised concerns regarding multi-organ toxicity, but its cardiovascular effects remain unclear. Here, we combined network toxicology, molecular docking, and experimental validation to investigate SB-induced cardiotoxicity. The toxicity profile of SB was predicted using ProTox 3.0 and ADMETlab 3.0, which identified potential cardiotoxicity signals. Network analysis identified AKT1 as a hub target, with functional enrichment implicating the PI3K-AKT apoptotic cascade, and molecular dynamics (MD) simulations confirmed stable SB-AKT1 binding. To

Inhibition by propofol of [3H]-batrachotoxinin-A 20-alpha-benzoate binding to voltage-dependent sodium channels in rat cortical synaptosomes.

1. Propofol (2,6 di-isopropylphenol), an intravenous general anaesthetic, blocks voltage-dependent Na+ channels (Na+ channels). In this study the interaction between propofol and Na+ channels was analysed by examining its effects on neurotoxin binding to various receptor sites of the Na+ channel in rat cerebrocortical synaptosomes. 2. Propofol (10-200 microM) exhibited concentration-dependent inhibition of equilibrium binding of [3H]-batrachotoxinin-A 20-alpha-benzoate ([3H]-BTX-B) to receptor site 2 of the Na+ channel (mean IC50 = 26 microM; 6.5 microM free). Scatchard analysis revealed that

Influence of high-dose sodium benzoate on lipopolysaccharide-induced neurobehavioral impairment, oxido-inflammatory brain damage, and cholinergic dysfunction in rats.

Sodium benzoate (SB) is a commonly utilized food preservative in the food business. Nonetheless, apprehensions regarding its impact on the brain have garnered worldwide attention. Consequently, we examined the effect of SB on lipopolysaccharide (LPS)-induced neurotoxicity in rats. Twenty-eight male Wistar rats were randomly assigned to four groups: Group 1 (Control, distilled water), Group 2 (SB, 600 mg/kg), Group 3 (LPS, 250 μg/kg/day), and Group 4 (LPS + SB; LPS, 250 μg/kg + SB, 600 mg/kg). SB was administered orally for 14 days, whereas LPS was injected intraperitoneally for 7 days. Upon co

Chronic sodium benzoate therapy in children with inborn errors of urea synthesis: effect on carnitine metabolism and ammonia nitrogen removal.

Sodium benzoate (SB) therapy is known to increase ammonia (NH3) nitrogen elimination via conjugation with glycine and excretion as urinary hippurate. In 16 children with inborn errors of urea synthesis we studied two issues: (1) the effect of chronic SB administration upon carnitine metabolism and (2) the efficacy of chronic SB therapy as measured by the molar ratio of hippurate excretion to SB intake. Measurements were performed during elective hospitalizations when the patients were in stable metabolic condition. We found that chronic SB therapy is not associated with a constant level of hip

In Vitro Effects of Sodium Benzoate on the Expression of T Cells-related Cytokines and Transcription Factors in Adjuvant-induced Arthritis Model.

Though the exact etiology of rheumatoid arthritis (RA) is unknown, the contribution of immune cells in the disease process is completely acknowledged. T helper (Th) 1 and Th17-related cytokines are required for the disease development and progression, while Th2 and regulatory T cells (Tregs)-derived cytokines are protective. Studies have shown that sodium benzoate (NaB) can switch the balance of Th cell subsets toward Th2 and Tregs. The present study aimed to evaluate the possible effects of NaB on the expression of CD4+T cells-related cytokines and transcription factors in splenocytes derived

The sodium channel in membranes of electroplax. Binding of batrachotoxinin-a [(3)H]benzoate to particulate preparations from electric eel (electrophorus).

Batrachotoxinin-A [(3)H]benzoate ([(3)H]BTX-B) binds specifically and with high affinity (K(D) 48 nM) to sites (B(max) 2.1 pmol/mg protein) associated with voltage-dependent sodium channels in rodent brain vesicular preparations. High affinity binding requires the presence of scorpion (Leiurus) venom and a membrane potential. Local anesthetics antagonize the binding. Nonspecific binding is defined in the presence of veratridine. In particulate preparations from electroplax of the eel Electrophorus electricus, [(3)H]BTX-B binds with a K(D) of about 140 nM and a B(max) of 2.5 pmol/mg protein in
