Multi-development-HPTLC method for quantitation of hyoscyamine, scopolamine and their biosynthetic precursors in selected solanaceae plants grown in natural conditions and as in vitro cultures.

INTRODUCTION: Hyoscyamine and scopolamine, anti-cholinergic agents widely used in medicine, are typically obtained from plants grown under natural conditions. Since field cultivation entails certain difficulties (changeable weather, pests, etc.), attempts have been made to develop a plant in vitro culture system as an alternative source for the production of these compounds. During experiments to locate the limiting steps in the biotechnological procedure, it is important to monitor not only the levels of the final products but also the changes in the concentration of their precursors. Seasonal variation effect on different Physalis peruviana L. (Solanaceae) waste extracts and investigation of their efficacy against Culex pipiens and Musca domestica.

Disease-carrying insects transmit many of the most serious human diseases. After decades of repeated use of insecticides, all of these vector species have demonstrated the ability to develop resistance to insecticides. This has necessitated the development of more efficient and environmentally safe alternatives in the form of biopesticides. Plants contain a wide range of potential phytochemicals that target a specific target, are rapidly biodegradable, environmentally friendly, and have a variety of therapeutic effects, making them a treasure trove of biological materials. Moreover, this has l

Laboratory evaluation of a phytosteroid compound of mature leaves of Day Jasmine (Solanaceae: Solanales) against larvae of Culex quinquefasciatus (Diptera: Culicidae) and nontarget organisms.

Crude mature leaves extract of Day Jasmine, Cestrum diurnum (Solanaceae: Solanales), was investigated for larvicidal activities against Culex quinquefasciatus, the vector of human filariasis. All the graded concentrations (0.1%, 0.2%, 0.5%, 1%, 1.5%, 2%, 2.5%, and 3% v/v) showed significant (P < 0.05) larval mortality, and results of regression equations revealed that the mortality rates were positively correlated with the concentrations of the extract (R (2) close to 1). LC(50), LC(95), and LC(99) values were calculated at different time intervals, and the lowest value was recorded at the 72-

Antioxidant and anti-inflammatory effects of fractions from ripe fruits of Solanum lycocarpum St. Hil. (Solanaceae) and putative identification of bioactive compounds by GC-MS and LC-DAD-MS.

Brazilian biodiversity includes species of the genus Solanum that have several biological activities, in addition to their relevance to agriculture, economics and popular medicine. The ripe fruits of Solanum lycocapum are an important nutritional food source, since they have levels of vitamin C, total soluble sugars, sucrose, phosphorus, and iron comparable or exceed the levels present in fruits such as pineapples, bananas, oranges, and mangoes. The pulp of the fruit is consumed by the population, and it is also used to produce jellies; to make marmalade, replacing the quince, and it can also

Solanidine and tomatidine trigger scar pruritus.

Scar pruritus is frequently encountered in clinical practice (particularly in burn patients) owing to its poorly known pathogenesis and difficult treatment. In previous work, we demonstrated the usefulness of a diet excluding edible solanaceae (viz., potatoes, tomatoes, peppers and aubergines) in patients with antihistamine-resistant scar pruritus. We hypothesized that alkaloids in solanaceae (particularly their secondary metabolites or aglycones) might be the actual pruritogens. In order to test this hypothesis, we conducted a single-blind prospective study on patients responding favourably t

An evolutionary view of vanillylamine synthase pAMT, a key enzyme of capsaicinoid biosynthesis pathway in chili pepper.

Pungent capsaicinoid is synthesized only in chili pepper (Capsicum spp.). The production of vanillylamine from vanillin is a unique reaction in the capsaicinoid biosynthesis pathway. Although putative aminotransferase (pAMT) has been isolated as the vanillylamine synthase gene, it is unclear how Capsicum acquired pAMT. Here, we present a phylogenetic overview of pAMT and its homologs. The Capsicum genome contained 5 homologs, including pAMT, CaGABA-T1, CaGABA-T3, and two pseudogenes. Phylogenetic analysis indicated that pAMT is a member of the Solanaceae cytoplasmic GABA-Ts. Comparative genome

The Evolutionary Pattern of Cocaine and Hyoscyamine Biosynthesis Provides Strategies To Produce Tropane Alkaloids.

Cocaine and hyoscyamine are two tropane alkaloids (TA) from Erythroxylaceae and Solanaceae, respectively. These famous compounds possess anticholinergic properties that can be used to treat neuromuscular disorders. While the hyoscyamine biosynthetic pathway has been fully elucidated allowing its de novo synthesis in yeast, the cocaine pathway remained only partially elucidated. Recently, the Huang research group has completed the cocaine biosynthetic route by characterizing its two missing enzymes. This allowed the whole pathway to be transferring into Nicotiana benthamiana to achieve cocaine

A Comprehensive Review on the Biosynthesis of Tropane Alkaloids.

Tropane alkaloids (TA) constitute a class of plant specialized metabolites with important pharmaceutical applications, including the anticholinergic agents hyoscyamine and scopolamine and the local anesthetic cocaine. Over the past decade, advances in genomics, structural biology, and synthetic biology have substantially revised our understanding of TA biosynthesis, leading to the identification of numerous key biosynthetic enzymes and evolutionary mechanisms. This review comprehensively summarizes current knowledge of TA biosynthesis from precursor formation to structurally diverse end produc

Elucidation of tropane alkaloid biosynthesis in Erythroxylum coca using a microbial pathway discovery platform.

Tropane alkaloids (TAs) are heterocyclic nitrogenous metabolites found across seven orders of angiosperms, including Malpighiales (Erythroxylaceae) and Solanales (Solanaceae). Despite the well-established euphorigenic properties of Erythroxylaceae TAs like cocaine, their biosynthetic pathway remains incomplete. Using yeast as a screening platform, we identified and characterized the missing steps of TA biosynthesis in Erythroxylum coca. We first characterize putative E. coca polyamine synthase- and amine oxidase-like enzymes in vitro, in yeast, and in planta to show that the first tropane ring

Mycosis fungoides masquerading as tinea of the axilla.

UNLABELLED: Angel's trumpets (Brugmansia spp.) are important medicinal and ornamental plants belonging to the Solanaceae family, domesticated in the Andean region of South America. Indigenous communities in the Colombian Andes have created unique medicinal cultivars exhibiting a variety of unusual shapes in leaves, and specific uses for ritual or medicinal procedures. These cultivars exhibit leaf symptoms typically associated with viral infections, including mosaic, vein chlorosis, and morphological deformities. Colombian datura virus (CDV) is one of the most studied viruses in Brugmansia, whi

Withalutin, a new cytotoxic withanolide from Athenaea velutina (Sendtn.) D'Arcy.

Bioassay-guided fractionation of dichloromethane extract from Athenaea velutina leaves led to the isolation of three withanolides, all being reported for the first time in this species. They were identified as withacnistin (1), withacnistin acetate (2) and a new withanolide, designated as withalutin (3). The structures were established by spectral data analysis, including MS, 1D and 2D NMR. In addition, in silico ADMET studies were employed to understand the pharmacokinetic properties of these withanolides. The withanolides isolated from A. velutina reduced cancer cell viability with IC50 valu

Associations of the Potato Psyllid and "Candidatus Liberibacter solanacearum" in Idaho with the Noncrop Host Plants Bittersweet Nightshade and Field Bindweed.

Zebra chip disease (ZC) in potato (Solanum tuberosum L. [Polemoniales: Solanaceae]) can produce unmarketable tubers with striped necrotic patterns. ZC is associated with the bacterium "Candidatus Liberibacter solanacearum" (Lso), which is transmitted by the potato psyllid, Bactericera cockerelli (Šulc) (Hemiptera: Triozidae). Potato psyllids are associated with numerous noncrop host plants, especially from the Solanaceae and Convolvulaceae; however, the contribution and importance of these hosts to ZC epidemiology in potato is poorly understood. To clarify seasonal phenologies on two such host

Adaptability Analysis of Tuta absoluta to Different Hosts and Related Salivary Genes Identification.

Tuta absoluta is a significant agricultural pest primarily affecting Solanaceae plants, resulting in substantial economic losses in agriculture. Insect saliva is an intermediary between insects and plants, playing a crucial role in modulating host adaptability and plant defense. This study analyzed the adaptive differences of T. absoluta on four plants using the two-sex life table method. Results indicated that the host adaptability of T. absoluta to tobacco is worse than its adaptability to the other three varieties of tomatoes. The salivary gland transcriptome analysis and signal peptide pre

Evaluation of glycoalkaloids and their aglycones in vegetable-based baby foods using a validated mesostructured silica-based MSPD protocol assisted by bead homogenization and UHPLC-MS/MS.

Glycoalkaloids (GAs) are a food safety concern in Solanaceae-derived products, but their occurrence in baby foods remains poorly documented. This study presents a sensitive, sustainable methodology for simultaneous determination of GAs and their aglycones in vegetable-based baby foods containing potato, eggplant, or tomato. Comparative evaluation of miniaturized ultrasound assisted (UA-MSPD), vortex assisted (VA-MSPD), and homogenizer assisted (HA-MSPD) matrix solid phase dispersion identified HA-MSPD as the most efficient strategy. Performance was improved using mesostructured silica MSU-2. B

Narcosis and nightshade.

Although this year marks the 150th anniversary of the discovery of modern surgical anaesthesia, surgery itself has a much longer history. It is well known that extracts from the opium poppy, Papaver somniferum, were used to dull the pain of surgery during ancient times but less well known that extracts from plants with sedative powers often accompanied them, producing primitive anaesthesia. Most of these sedative plants were members of a large botanical family, the Solanaceae. This paper describes some of them and discusses the ways in which they were administered. It also explains why, during
