Prediction of Dhurrin Metabolism by Transcriptome and Metabolome Analyses in Sorghum.

Sorghum (Sorghum bicolor (L.)) Moench is an important food for humans and feed for livestock. Sorghum contains dhurrin which can be degraded into toxic hydrogen cyanide. Here, we report the expression patterns of 14 candidate genes related to dhurrin ((S)-4-Hydroxymandelnitrile-β-D-glucopyranoside) metabolism and the effects of the gene expression on specific metabolite content in selected sorghum accessions. Dhurrin-related metabolism is vigorous in the early stages of development of sorghum. The dhurrin contents of most accessions tested were in the range of approximately 6-22 μg mg-1 fresh

Proximate, structural, textural, sensory and microbiological properties of non-gluten extrudate using Sorghum (Sorghum bicolor L. Moench) and a sprouted legume (Phaseolus lunatus L.).

The inclusion of legumes as functional ingredients in a gluten-free extrusion process has been gaining attention in recent times. In this study, sorghum and germinated lima bean flour (100, 90:10, 80:20, 70:30, 60:40 and 50:50) was extruded (feed moisture - 18%, screw speed - 250 rpm, barrel temperatures 50 °C-120 °C-120 °C, die hole diameter - 3 mm) and analysed for functional, proximate, textural, structural, pasting, microbiological and sensory properties. With 100% sorghum used as control, lima beans addition significantly (p < 0.05) improved the loose (0.37-0.44 g/ml) and packed (0.63-0.7

Enhancing Sorghum Growth: Influence of Arbuscular Mycorrhizal Fungi and Sorgoleone.

The low availability of phosphorus (P) in soil is one of the main constraints on crop production. Plants have developed several strategies to increase P use efficiency, including modifications in root morphology, the exudation of different compounds, and associations with microorganisms such as arbuscular mycorrhizal fungi (AMF). This study aimed to investigate the effect of sorgoleone compound on AMF colonization and its subsequent impact on P uptake, rhizosphere microbiota, and sorghum growth. The experiment was conducted in a greenhouse using the sorghum genotype P9401, known for low sorgol

Do cyanogenic glucosides help sorghum manage a fluctuating nitrogen supply?

Sorghum (Sorghum bicolor [L.] Moench) is an important forage crop that contains the cyanogenic glucoside dhurrin that releases hydrogen cyanide when tissue is damaged. The acyanogenic (dhurrin-free) sorghum line tcd1 was developed to eliminate the risk of cyanide poisoning from sorghum forage. However, dhurrin may also play a role in nitrogen accumulation and storage. We tested whether dhurrin offers the cyanogenic sorghum line BTx623 a growth advantage relative to tcd1 , when nitrogen is limiting and variable. BTx623 and tcd1 were grown under two 42-day nitrogen treatments: high dose, low fre

Regulation of dhurrin pathway gene expression during Sorghum bicolor development.

Developmental and organ-specific expression of genes in dhurrin biosynthesis, bio-activation, and recycling offers dynamic metabolic responses optimizing growth and defence responses in Sorghum. Plant defence models evaluate the costs and benefits of resource investments at different stages in the life cycle. Poor understanding of the molecular regulation of defence deployment and remobilization hampers accuracy of the predictions. Cyanogenic glucosides, such as dhurrin are phytoanticipins that release hydrogen cyanide upon bio-activation. In this study, RNA-seq was used to investigate the exp

Sweet sorghum is one of the important crops that has been widely reported to be recalcitrant to genetic manipulation endeavors. It can serve as biofuel, food, and food per production systems toward energy sources as well as human and animal sustenance. To deploy biotechnology tools in advancement of this crop of such agronomic importance, genetic and molecular investigation for understanding is necessary. Underdeveloped cell walls are a significant physiochemical state for microspores per their developmental stages, which makes it ideal for DNA isolation, being readily lysed as natural protopl

Brown midrib mutants in sorghum and their applications in renewable energy production.

Lignin is a complicated phenolic polymer that is found in the secondary cell walls of plants. It is necessary for plant mechanical strength, vascular integrity, and stress tolerance. In sorghum (Sorghum bicolor L. Moench), the amount and composition of lignin greatly affect forage digestibility, biomass recalcitrance to bioenergy production and biochemical conversion process efficiency. Brown midrib (bmr) mutants, which exhibit reddish-brown pigmentation of the vasculature, have been a classic and potent tool for identifying lignin biosynthesis and engineering cell wall traits. Over the last 6

Unraveling transcriptomics of sorghum grain carotenoids: a step forward for biofortification.

BACKGROUND: Sorghum (Sorghum bicolor [L.] Moench) is a promising target for pro-vitamin A biofortification as it is a global staple crop, particularly in regions where vitamin A deficiency is prevalent. As with most cereal grains, carotenoid concentrations are low in sorghum, and breeding could be a feasible strategy to increase pro-vitamin A carotenoids to biologically relevant concentrations. However, there are knowledge gaps in the biosynthesis and regulation of sorghum grain carotenoids, which can limit breeding effectiveness. The aim of this research was to gain an understanding of the tr

Effects of mutual shading on the regulation of photosynthesis in field-grown sorghum.

In the field, close planting inevitably causes mutual shading and depression of leaf photosynthesis. To clarify the regulative mechanisms of photosynthesis under these conditions, the effects of planting density on leaf structure, gas exchange and proteomics were carefully studied in field-grown sorghum. In the absence of mineral deficiency, (1) close planting induced a significant decrease in light intensity within populations, which further resulted in much lower stomatal density and other anatomical characteristics associated with shaded leaves; (2) sorghum grown at high planting density ha

FESW-UNet: A Dual-Domain Attention Network for Sorghum Aphid Segmentation.

Current management strategies for sorghum aphids heavily rely on indiscriminate chemical application, leading to severe environmental consequences and impacting food safety. While precision spraying offers a viable remediation for pesticide overuse, its effectiveness depends on accurately locating and classifying pests. To address the critical challenge of segmenting small, swarming aphids in complex field environments, we propose FESW-UNet, a dual-domain attention network that integrates Fourier-enhanced attention, spatial attention, and wavelet-based downsampling into a UNet backbone. We int

Indirect plant defense may provide economically important pest suppression in sorghum.

BACKGROUND: A promising strategy to optimize biological control of insect pests is selecting crop varieties with indirect defense traits. Indirect plant defenses recruit natural enemies to kill pests and include chemical attractants like herbivore-induced plant volatiles. In prior laboratory assays, we found sorghum (Sorghum bicolor L.) cultivar ATx3409/RTx436 infested with sorghum aphid (Melanaphis sorghi Theobald) was attractive to natural enemies and emitted more chemical attractants than two other cultivars. In this field study, we manually infested 9-week-old sorghum plants with aphids an

Categories of Resistance to Sugarcane Aphid (Hemiptera: Aphididae) Among Sorghum Genotypes.

The sugarcane aphid Melanaphis sacchari (Zehnter) (Hemiptera: Aphididae) has emerged as a potential threat to sorghum (Sorghum bicolor (L.) Moench) production in the United States. Since the late summer of 2013, finding and advancing M. sacchari-resistant germplasm has been a priority for all stakeholders involved. We evaluated 23 sorghum genotypes for resistance to the sugarcane aphid by testing for tolerance, and antixenosis. In addition, nine sorghum germplasm were evaluated for the expression of antibiosis. Free-choice and no-choice tests were conducted to explore the functional categories

Glutathione transferases catalyze recycling of auto-toxic cyanogenic glucosides in sorghum.

Cyanogenic glucosides are nitrogen-containing specialized metabolites that provide chemical defense against herbivores and pathogens via the release of toxic hydrogen cyanide. It has been suggested that cyanogenic glucosides are also a store of nitrogen that can be remobilized for general metabolism via a previously unknown pathway. Here we reveal a recycling pathway for the cyanogenic glucoside dhurrin in sorghum (Sorghum bicolor) that avoids hydrogen cyanide formation. As demonstrated in vitro, the pathway proceeds via spontaneous formation of a dhurrin-derived glutathione conjugate, which u

Plant regeneration from cultured immature embryos of Sorghum bicolor (L.) Moench.

Immature embryos of 20 sorghum genotypes were cultured on MS 5 medium containing MS mineral salts supplemented with 2,4-D, zeatin, glycine, niacinamide, Ca-pantothenate, L-asparagine, and vitamins. For regeneration, calli were transferred onto the same medium with the exception that IAA was substituted for 2,4-D. In general, immature embryos obtained 9-12 days after pollination resulted in the best redifferentiation. Ability of calli to regenerate varied among genotypes; cultivars C401-1 and C625 had the highest redifferentiation frequencies. Ability to redifferentiate was heritable and acted

Salt stress activates tyrosine metabolism and dhurrin-associated SbCYP79A1 in sorghum.

Potential toxicity to livestock limits the widespread adoption of forage sorghum as a higher-yielding, more nutritious, and more climate-resilient alternative to maize. Although low-dhurrin cultivars are available, their safety under saline conditions remain insufficiently understood. Here, we investigated the salt-stress molecular basis of dhurrin-associated toxicity in sorghum, with emphasis on tyrosine metabolism and the dhurrin-pathway genomics. We demonstrate that exposure to medium salt stress enriches 56 genes in the tyrosine metabolism pathway, including the dhurrin-specific cytochrome

Allelopathic Effects of Extracts from Different Sorghum Parts on Giant Foxtail.

Giant foxtail is a common weed in foxtail millet fields, and continuous cropping of foxtail millet increases its occurrence. Rotating foxtail millet with sorghum is a major practice in the dry farming regions of northern China. Given this situation, this study combined Petri dish germination tests with pot experiments to clarify the allelopathic effects of water extracts from different sorghum parts on giant foxtail. Distilled water (SCK) was used as the control, and treatments included stock solution (S1), as well as 10× dilution (S2), 50× dilution (S3), and 100× dilution (S4) of water extrac

Defensive capabilities of contrasting sorghum genotypes against Atherigona soccata (Rondani) infestation.

Plants are equipped with a wide range of defensive mechanisms such as morphophysiological, biochemical, molecular, and hormonal signaling for protecting against insect-pest infestation. The infestation of a devastating pest shoot fly [Atherigona soccata (Rodani)] at seedling stage causes huge loss of sorghum crop productivity. In morphophysiological screening ICSV700, ICSV705, and IS18551 have been categorized as resistant, PSC-4 moderately resistant, SL-44 and SWARNA as susceptible. The present study focused on the role of defensive gene expression and its products viz: superoxide dismutase (
