Calcified Artery Preparation and Processing with Preserved Morphology and RNA for Digital Spatial Profiling.

Spatial transcriptomics maps whole-transcriptome profiles directly onto tissue sections, correlating between cellular neighborhoods and gene expression and how these activities drive health and disease. Although the approach has transformed oncology, neuroscience, immunology, developmental sciences, and many fields, it has been less widely used in vascular biology, even though vascular disease remains a leading cause of death worldwide. Two obstacles stand out: arteries and veins offer little flat surface for sectioning, and diseased specimens, often atherosclerotic, thrombosed, plaque-laden,

How does otitis media with effusion history affect spatial auditory processing and listening effort?

PURPOSE: To evaluate the impact of otitis media with effusion (OME) history on spatial auditory processing and listening effort (LE) in children with normal hearing thresholds. METHODS: Seventeen children with a history of OME, whose hearing thresholds had returned to normal (mean age 7.8 years), and 22 age-matched controls (mean age 8 years) participated in the study. Assessments included audiometric evaluations, an adaptive speech-in-noise task under spatial and non-spatial conditions, pupillometry to measure changes in pupil dilation during sentence recognition in noise, and an auditory di

Aberrantly integrated adult-born immature neurons disrupt brain-wide networks during spatial memory processing.

Memory deficits observed in various neurological and psychiatric disorders may, in part, arise from dysregulated adult-born immature neurons (ABNs) in the dentate gyrus (DG). However, the mechanisms by which these aberrant neurons contribute to brain-wide network dysfunction and memory impairment remain poorly understood. Using a well-established mouse model with aberrantly integrated ABNs and associated memory deficits, we employed resting-state functional magnetic resonance imaging (rs-fMRI) and found that a few hundred dysregulated ABNs (<0.1% of total DG granule neurons) were sufficient to

Processing and cryopreservation of human ureter tissues for single-cell and spatial transcriptomics assays.

Characterizing the cellular heterogeneity of human ureter tissues using single-cell RNA sequencing (scRNA-seq) and spatial transcriptomics provides a detailed atlas of cell types, signaling networks, and potential cell-cell cross talk underlying developmental and regenerative pathways. We describe an optimized protocol for generating, cryopreserving, and thawing single-cell suspensions from ureter tissues isolated post-cystectomy for scRNA-seq. In addition, we describe an optimized protocol for cryopreserving human ureter tissues for 10x Genomics Visium spatial gene expression platform. For co

Temporal and spatial dynamics of spinal sensorimotor processing in an intersegmental cutaneous nociceptive reflex.

The cutaneus trunci muscle (CTM) reflex produces a skin "shrug" in response to pinch on a rat's back through a three-part neural circuit: 1) A-fiber and C-fiber afferents in segmental dorsal cutaneous nerves (DCNs) from lumbar to cervical levels, 2) ascending propriospinal interneurons, and 3) the CTM motoneuron pool located at the cervicothoracic junction. We recorded neurograms from a CTM nerve branch in response to electrical stimulation. The pulse trains were delivered at multiple DCNs (T6-L1), on both sides of the midline, at two stimulus strengths (0.5 or 5 mA, to activate Aδ fibers or A

External and internal facial features modulate processing of vertical but not horizontal spatial relations.

Some years ago an asymmetry was reported for the inversion effect for horizontal (H) and vertical (V) relational face manipulations (Goffaux & Rossion, 2007). Subsequent research examined whether a specific disruption of long-range relations underlies the H/V inversion asymmetry (Sekunova & Barton, 2008). Here, we tested how detection of changes in interocular distance (H) and eye height (V) depends on cardinal internal features and external feature surround. Results replicated the H/V inversion asymmetry. Moreover, we found very different face cue dependencies for both change types. Performan

Neural substrates of visuospatial processing in distinct reference frames: evidence from unilateral spatial neglect.

There is evidence for different levels of visuospatial processing with their own frames of reference: viewer-centered, stimulus-centered, and object-centered. The neural locus of these levels can be explored by examining lesion location in subjects with unilateral spatial neglect (USN) manifest in these reference frames. Most studies regarding the neural locus of USN have treated it as a homogenous syndrome, resulting in conflicting results. In order to further explore the neural locus of visuospatial processes differentiated by frame of reference, we presented a battery of tests to 171 subjec

Distinct and convergent visual processing of high and low spatial frequency information in faces.

We tested for differential brain response to distinct spatial frequency (SF) components in faces. During a functional magnetic resonance imaging experiment, participants were presented with "hybrid" faces containing superimposed low and high SF information from different identities. We used a repetition paradigm where faces at either SF range were independently repeated or changed across consecutive trials. In addition, we manipulated which SF band was attended. Our results suggest that repetition and attention affected partly overlapping occipitotemporal regions but did not interact. Changes

[A case of pure anarithmetia associated with disability in processing of abstract spatial relationship].

A 35-year-old right handed man developed pure anarithmetia after an left parieto-occipital subcortical hemorrhage. His intelligence, memory, language, and construction ability were all within normal limits. No hemispatial neglect, agraphia, finger agnosia, or right-left disorientation were noted. He showed no impairments in reading numbers aloud, pointing to written numbers, writing numbers to dictation, decomposition of numbers, estimation of numbers of dots, reading and writing of arithmetic signs, comprehension of arithmetic signs, appreciation of number values, appreciation of dots' number

Cortical areas with enhanced activation during object-centred spatial information processing. A PET study.

The phenomenon of object-centred unilateral neglect suggests that some neural networks process spatial information relative to reference objects. To examine object-centred information processing, we measured regional cerebral blood flow in 11 normal subjects with PET. During each PET scan, a subject viewed a sample stimulus followed by a cue on a video screen. The sample consisted of two polygons, termed 'objects', each located in a corner of the screen. A small target spot appeared in a corner of each polygon. There were two tasks: the visuomotor task and the matching-to-sample task. In the v

Benzodiazepines and synaptic processing in the spatial domain within the cat's primary somatosensory cortex.

In the primary somatosensory cortex of cats, the size of the receptive fields (RFs) of cutaneously responsive neurones is under the control of gamma-aminobutyric acid (GABA) mediated inhibition when the cells are situated in rapidly adapting (RA) background regions. Cells located in slowly adapting (SA) or low-velocity rapidly adapting (LVRA) background regions do not appear to be affected by GABA significantly in the spatial domain, although other response properties such as threshold and firing pattern are under the influence of bicuculline methiodide (BMI) sensitive processes. The GABA rece

Holistic processing is robust in the face of task-context-induced spatial attention biases.

One account of the characteristic holistic processing of faces and objects of expertise posits that it arises from a learned attention to the whole, rendering it difficult to attend only to parts of stimuli. We tested whether task-context-induced attentional biases for the top or bottom part of a stimulus alter holistic processing of faces. We induced attentional biases by manipulating the probability (75% or 25%) that the top or bottom part would be task-relevant in a modified composite part-judgement task. Manipulating the proportion of trials in which the top/bottom region was task-relevant

Spatial and temporal processing difficulties in Chinese children with developmental dyslexia: An ERP study.

Magnocellular (M) deficit theory indicates that individuals with developmental dyslexia (DD) have low sensitivity to stimuli with high temporal frequencies (HTF) and low spatial frequencies (LSF). However, some studies found that temporal processing and spatial processing were correlated with different reading-related skills. Chinese is a logographic language, and visual skills are particularly important for reading in Chinese. It is necessary to investigate the temporal and spatial processing abilities in the M pathway of Chinese children with DD. Using electrophysiological recordings, the pr

Non-invasive neurostimulation modulates processing of spatial frequency information in rapid perception of faces.

This study used high-frequency transcranial random noise stimulation (tRNS) to examine how low and high spatial frequency filtered faces are processed. Response times were measured in a task where healthy young adults categorised spatially filtered hybrid faces, presented at foveal and peripheral blocks, while sham and high-frequency random noise was applied to a lateral occipito-temporal location on their scalp. Both the Frequentist and Bayesian approaches show that in contrast to sham, active stimulation significantly reduced response times to peripherally presented low spatial frequency inf

Altered neural dynamics in occipital cortices serving visual-spatial processing in heavy alcohol users.

BACKGROUND: Visual-spatial processing deficits have been previously linked to heavy alcohol use, but the underlying neurological mechanisms are poorly understood. Neuroimaging studies have shown alcohol-related aberrations in occipital cortices that appear to be associated with these neuropsychological deficits in visual-spatial processing, however the neural dynamics underlying this altered processing remains unknown. METHODS: Twenty-three adults with high scores on the Alcohol Use Disorders Identification Test - Consumption (male: ⩾5, female: ⩾4) were compared to 30 demographically-matched

Auditory Localization and Spatial Release From Masking in Children With Suspected Auditory Processing Disorder.

OBJECTIVES: We sought to investigate whether children referred to our audiology clinic with a complaint of listening difficulty, that is, suspected of auditory processing disorder (APD), have difficulties localizing sounds in noise and whether they have reduced benefit from spatial release from masking. DESIGN: Forty-seven typically hearing children in the age range of 7 to 17 years took part in the study. Twenty-one typically developing (TD) children served as controls, and the other 26 children, referred to our audiology clinic with listening problems, were the study group: suspected APD (s

Image processing methods to elucidate spatial characteristics of retinal microglia after optic nerve transection.

Microglia are the mononuclear phagocytes with various functions in the central nervous system, and the morphologies of microglia imply the different stages and functions. In optical nerve transection model of the retina, the retrograde degeneration of retinal ganglion cells induces microglial activations to a unique morphology termed rod microglia. A few studies described the rod microglia in the cortex and retina; however, the spatial characteristic of rod microglia is not fully understood. In this study, we built a mathematical model to characterize the spatial trait of rod microglia. In add

Functional specialization for auditory-spatial processing in the occipital cortex of congenitally blind humans.

The study of the congenitally blind (CB) represents a unique opportunity to explore experience-dependant plasticity in a sensory region deprived of its natural inputs since birth. Although several studies have shown occipital regions of CB to be involved in nonvisual processing, whether the functional organization of the visual cortex observed in sighted individuals (SI) is maintained in the rewired occipital regions of the blind has only been recently investigated. In the present functional MRI study, we compared the brain activity of CB and SI processing either the spatial or the pitch prope

Functionally distinct regions for spatial processing and sensory motor integration in the planum temporale.

There has been much debate recently over the functional role played by the planum temporale (PT) within the context of the dorsal auditory processing stream. Some studies indicate that regions in the PT support spatial hearing and other auditory functions, whereas others demonstrate sensory-motor response properties. This multifunctionality has led to the claim that the PT is performing a common computational pattern matching operation, then routing the signals (spatial, object, sensory-motor) into an appropriate processing stream. An alternative possibility is that the PT is functionally subd
