The Interplay of Sulfur and Selenium Enabling Variations in Micronutrient Accumulation in Red Spinach.

Aside from its importance in human and animal health, low levels of foliar-applied selenate (SeO4) can be advantageous in the presence of sulfur (S), contributing to improved growth, nutrient uptake, and crop quality. A hydroponic experiment in a growth chamber explored the interactive influence of Se and S on micronutrients and several quality indices, such as soluble sugars, organic acids, and total protein concentrations in spinach (Spinacia oleracea L.). Three levels of S (deprivation, adequate, and excessive) with varying quantities of Se (deficient, moderate, and higher) were examined in

Magnesium affects spinach carotenoid bioaccessibility in vitro depending on intestinal bile and pancreatic enzyme concentrations.

Magnesium may reduce carotenoid bioavailability by forming insoluble complexes with bile salts/fatty acids, inhibiting micelle formation. Here, we investigated whether altering bile/pancreatin concentration influenced potential negative effects of magnesium on carotenoid bioaccessibility. Spinach (4g) was digested in vitro with added magnesium (0, 200, 400mg/L) and canola oil/coffee creamer, at varying bile extract (1 or 8mM) and pancreatin (100 or 990mg/L) concentrations. Bioaccessibility was determined for β-carotene, lutein, and total carotenoids via HPLC. Additionally, lipolysis, particle

Improved procedure for the isolation of a double-strand-specific ribonuclease and its application to structural analysis of various 5S rRNAs and tRNAs.

An improved method for the isolation of a double-strand-specific RNase from snake venom is presented. This RNase, called CSV, was used to cleave yeast tRNAPhe and tRNA2Glu and tRNAfMet from Escherichia coli. In addition these RNAs and E. coli tRNAPhe were examined with the single-strand-specific nuclease S1. The results are discussed in terms of the specificity of CSV RNase and the structure of tRNAs. S1 nuclease digestions at increasing temperatures allowed the melting of tertiary and secondary structure to be monitored. 5S rRNA from E. coli, Thermoplasma acidophilum and the chloroplasts of S

It takes two to attach - endo-1,3-β-d-glucanase as a potential receptor of mannose-independent, FimH-dependent Salmonella Typhimurium binding to spinach leaves.

Currently, fresh, unprocessed food has become a relevant element of the chain of transmission of enteropathogenic infections. To survive on a plant surface and further spread the infections, pathogens like Salmonella have to attach stably to the leaf surface. Adhesion, driven by various virulence factors, including the most abundant fim operon encoding type 1 fimbriae, is usually an initial step of infection, preventing physical removal of the pathogen. Adhesion properties of Salmonella's type 1 fimbriae and its FimH adhesin were investigated intensively in the past. However, there is a lack o

The reductive modulation of chloroplast fructose-1,6-bisphosphatase by tributylphosphine and sodium borohydride.

The cleavage of disulfide bonds is the major modification of chloroplast fructose-1,6-bisphosphatase when the light-mediated ferredoxin-thioredoxin system enhances the activity of the enzyme. In vitro, only thiol-bearing compounds are functional in the stimulation of fructose 1,6-bisphosphate hydrolysis. This investigation was undertaken to determine the effectivity of other reductants for enhancing the catalytic capacity. In the presence of 1 mM fructose 1,6-bisphosphate and 0.1 mM Ca2+, the five-fold activation triggered by 3.5 mM tributylphosphine is further potentiated by 15% (v/v) 2-propa

Pesticide Screening in Spinach Leaf Wash Water Using Microhematocrit Packed Capillary Tube Chromatography.

The widespread and excessive application of pesticides on food crops presents a critical, long-standing threat to the global public health. Although advanced laboratory methods like gas chromatography (GC) and electrochemical assays offer high sensitivity, their reliance on expensive infrastructure and specialized reagents creates a dangerous bottleneck for decentralized, on-site food safety screening. To bridge this urgent diagnostic gap, this study introduces low-complexity, low-cost screening platforms utilizing capillary tube chromatography (CTC) to detect active pesticide ingredients in p

Monochromatic LeafAdaptNet (MLAN): an adaptive approach to spinach leaf disease detection using monochromatic imaging.

A country's economic growth heavily relies on agricultural productivity, specifically nutrition derived from vegetables and leafy greens. Spinach, abundant in iron, vitamins, and other essential nutrients, plays a vital role in maintaining the health of human tissues, cartilage, and hair. However, extreme summer heat and plant diseases can significantly reduce spinach yields, making it less nutritious and harder to obtain. Implementing improved detection and classification of bacterial and fungal diseases affecting spinach leaves is crucial for minimizing pesticide use and enhancing agricultur

Colloidal-Bound Polyphosphates and Organic Phosphates Are Bioavailable: A Nutrient Solution Study.

Colloidal forms of Fe(III) minerals can be stabilized in solution by coatings of organic or poly-phosphate (P), which reduce the zeta-potential. This opens up a route toward the development of nanoforms of P fertilizers. However, it is unclear if such P forms are bioavailable. To address this question, spinach (Spinacia oleracea) was grown in nutrient solutions, at equal total P, using three different forms of P (orthophosphate = Pi; hexametaphosphate = HMP; myo-inositol hexaphosphate = IHP), free or bound to goethite/ferrihydrite colloids. After 10 days, P uptake was determined with a dose-re

Spectroscopic evidence for site specific chemistry at a unique iron site of the [4Fe-4S] cluster in ferredoxin:thioredoxin reductase.

Ferredoxin:thioredoxin reductase (FTR) catalyzes the reduction of the disulfide in thioredoxin in two one-electron steps using an active site comprising a [4Fe-4S] in close proximity to a redox active disulfide. Mössbauer spectroscopy has been used to investigate the ligation and electronic properties of the [4Fe-4S] cluster in as-prepared FTR which has the active-site disulfide intact and in the N-ethylmaleimide (NEM)-modified form which provides a stable analogue of the one-electron-reduced heterodisulfide intermediate and has one of the cysteines of the active-site disulfide alkylated with

Bacterial diversity, quantitative risk assessment of spring waterborne pathogens and their biocontrol through phyto-biosorbents.

Bacterial contamination of drinking water is a significant cause of water-borne illnesses in many developing countries, where water sources are commonly shared by the community. Therefore, the recent research was designed to screen the bacterial diversity associated with spring water, to assess the bacterial risk in both adults and children, and to remediate bacterial contaminants using green remediation technology via Brassica rapa and Spinacia oleracea. Microbial growth media, microscopic techniques, biochemical tests, ribotyping, antibiogram, and resistogram analysis were used to characteri

Sulfur dioxide inhibition of photosynthesis in isolated spinach chloroplasts.

Photosynthetic oxygen evolution by isolated spinach (Spinacia oleracea L.) chloroplasts approached complete inhibition in the presence of a 5 mm concentration of sulfur dioxide. A similar inhibition was observed in the presence of equimolar concentrations of bisulfite ions, suggesting a parallel mode of action. In contrast, an equimolar concentration of sulfite ions was markedly less inhibitory and sulfate ions caused negligible inhibition of apparent photosynthesis. The mode of action of sulfur dioxide and related sulfur anions in inhibiting photosynthesis was found to be essentially independ

Isolation and compositional analysis of galactoglycerolipids from perilla [Perilla frutescens (L.) Britton] leaves and comparison to the galactoglycerolipids from spinach and parsley.

The aim of this study was to isolate monogalactosyldiacylglycerols (MGDGs) and digalactosyldiacylglycerols (DGDGs) from perilla [Perilla frutescens (L.) Britton] and to investigate their fatty acid profiles. Perilla displayed the greatest total MGDG and DGDG content among the three types of leaf vegetables tested, that is, spinach, parsley, and perilla, containing 0.16 g/100 g MGDG and 0.04 g/100 g DGDG (on wet weight basis). High purity MGDG (approximately 97 g/100 g) and DGDG (approximately 86 g/100 g) were isolated from perilla chloroform/methanol (2:1, v/v) extracts by two-step silica gel

Expansion microscopy reveals thylakoid organisation alterations due to genetic mutations and far-red light acclimation.

The thylakoid membrane is the site of the light-dependent reactions of photosynthesis. It is a continuous membrane, folded into grana stacks and the interconnecting stroma lamellae. The CURVATURE THYLAKOID1 (CURT1) protein family is involved in the folding of the membrane into the grana stacks. The thylakoid membrane remodels its architecture in response to light conditions, but its 3D organisation and dynamics remain incompletely understood. To resolve these details, an imaging technique is needed that provides high-resolution 3D images in a high-throughput manner. Recently, we have used expa

The mechanism behind tenuazonic acid-mediated inhibition of plant plasma membrane H+-ATPase and plant growth.

The increasing prevalence of herbicide-resistant weeds has led to a search for new herbicides that target plant growth processes differing from those targeted by current herbicides. In recent years, some studies have explored the use of natural compounds from microorganisms as potential new herbicides. We previously demonstrated that tenuazonic acid (TeA) from the phytopathogenic fungus Stemphylium loti inhibits the plant plasma membrane (PM) H+-ATPase, representing a new target for herbicides. In this study, we further investigated the mechanism by which TeA inhibits PM H+-ATPase and the effe
