First record of the nematode Echinocephalus pseudouncinatus (Gnathostomatidae, Spirurida) in an edible, commercial host, the pen shell Atrina maura (Bivalvia: Pinnidae).

We describe a nematode infestation of the Atrina maura, the most valuable pen shell fished in Mexico, for the first time. Observations by Mexican authorities indicated that parasites were found encysted in pen shell gonads. The aims of this study included: (i) to identify the parasite morphologically; (ii) to quantify the infestation in different tissues of the pen shell; and (iii) to establish the seasonal variation in gonadal development. During 2015, 10 to 40 pen shells were randomly collected each month from the same commercial bed, and gonads were preserved and processed histologically to

Development of Novel Management Tools for Phortica variegata (Diptera: Drosophilidae), Vector of the Oriental Eyeworm, Thelazia callipaeda (Spirurida: Thelaziidae), in Europe.

Lachryphagous males of Phortica variegata (Fallén, 1823) are gaining increasing attention in Europe, as they act as vectors of the nematode Thelazia callipaeda Railliet & Henry, 1910, causal agent of thelaziosis, an emergent zoonotic disease. Currently, there are no effective control strategies against the vector, and surveillance and monitoring rely on time-consuming and nonselective sampling methods. Our aim was to improve the knowledge about the population dynamics and the chemical ecology of the species. A total of 5,726 P. variegata flies (96.4% males and 3.6% females, mostly gravid) were

Spirocerca vulpis sp. nov. (Spiruridae: Spirocercidae): description of a new nematode species of the red fox, Vulpes vulpes (Carnivora: Canidae).

Previous studies have reported nematodes of the Spirocercidae family in the stomach nodules of red foxes (Vulpes vulpes) described as Spirocerca sp. or Spirocerca lupi (Rudolphi, 1819). We characterized spirurid worms collected from red foxes and compared them to S. lupi from domestic dogs by morphometric and phylogenetic analyses. Nematodes from red foxes differed from S. lupi by the presence of six triangular teeth-like buccal capsule structures, which are absent in the latter. Additionally, in female worms from red foxes, the distance of the vulva opening to the anterior end and the ratio o

Redescription of Cercopithifilaria rugosicauda (Böhm & Supperer, 1953) (Spirurida: Onchocercidae) of roe deer, with an emended diagnosis of the genus Cercopithifilaria and a genetic characterisation.

Newly collected material of Cercopithifilaria rugosicauda from roe deer Capreolus capreolus was analysed and compared to descriptions of C. rugosicauda from Austria and Hungary. The present specimens were assigned to the genus Cercopithifilaria using both morphological and molecular analyses. Complementary morphological data on the males and microfilariae of C. rugosicauda were described. The main morphological characters of different species of Cercopithifilaria were outlined and an emended generic diagnosis proposed. A genetic characterisation based on the analyses of cox1 and 12S rDNA seque

Redescription and Revised Classification Status of Abbreviata phrynocephali comb. nov. (Spirurida: Physalopteridae) from the Yarkand Toad-Headed Agama Phrynocephalus axillaris (Lacertilia: Agamidae).

The genus Abbreviata (Travassos 1920) is distributed worldwide, with lizards serving as the primary hosts. Although Pentadentoptera phrynocephali was previously classified within Pentadentoptera, its original description lacked conclusive morphological and molecular evidence. Notably, key diagnostic features (such as oral dentition structure and spicule ratio) conform to the taxonomic criteria of Abbreviata. These ambiguous characteristics necessitate taxonomic reassessment to resolve systematic uncertainties. In this study, specimens of P. phrynocephali were collected from the stomachs of Phr

Expression of defensin, cecropin, and transferrin in Aedes aegypti (Diptera: Culicidae) infected with Wuchereria bancrofti (Spirurida: Onchocercidae), and the abnormal development of nematodes in the mosquito.

The temporal expression of defensin, cecropin and transferrin was assessed in Aedes aegypti naturally refractory to Wuchereria bancrofti upon infection with this worm, in parallel to analysis of filarial development in the insect. Compared to controls, transcription of defensin and cecropin was higher in infected mosquitoes as soon as 2h post infection and peaked before 48h. Transferrin transcription was higher in infected mosquitoes at 24h, and at 48h was almost leveled to controls. At 72h and 7 days post infection, levels of all transcripts in infected insects decreased gradually and were si

Metabolomic analysis of larval stages of Onchocerca japonica (Spirurida: Onchocercidae), raised in black fly (Diptera: Simuliidae) vectors, by gas chromatography-tandem mass spectrometry.

To monitor and prevent the spread of zoonotic onchocerciasis, identification of the natural vectors (blood-sucking insects) of its causative agents, Onchocerca species, is crucial. To date, vector identification depends on the detection of infective third-stage larvae in insects by traditional dissection. We aimed to develop a novel, more efficient method for the discrimination of the four larval stages, i.e. microfilariae (Mf), first-stage larvae (L1), second-stage larvae (L2), and third-stage larvae (L3), of O. japonica by metabolomic analysis. Microfilariae of O. japonica, the causative age

Filaria martis Gmelin 1790 (Spirurida, Filariidae) affecting beech marten (Martes foina): morphological description and molecular characterisation of the cytochrome oxidase c subunit I.

Filaria martis causes a poorly known subcutaneous filariosis in mustelids. Few information is available about lesions that F. martis causes in beech martens, on its morphology, biology and the occurrence of the infection. From 1997 to 2006, 29 beech martens from two sites of southern Italy (Sites A and B) have been necropsied. Ectoparasites and nematodes were collected and morphologically identified. A variable region of the cytochrome c oxidase subunit 1 (cox1) of F. martis has been characterised to compare females presenting caudal tips smooth without spines (i.e. Morphotype 1-Mrph. 1) and w

Ultrastructural changes in the advanced third larval stage of Gnathostoma binucleatum Almeyda-Artigas, 1991 (Nematoda: Spirurida) following albendazole and albendazole sulfoxide in vitro exposure.

We report here the ultrastructural changes in the third advanced larval stage (AL3) of Gnathostoma binucleatum Almeyda-Artigas, 1991, induced by in vitro treatment with albendazole and the metabolite albendazole sulfoxide. During the whole period of the experiment, the controls remained active and vital throughout the 108-h exposure period. The primary site of action of both drugs appears to be the mitochondria and myofibrils of the polymyarian musculature. Degenerative changes in the secretory portion of the esophagus and somatic musculature were clearly observed. These degenerative changes,

Actin protein up-regulated upon infection and development of the filarial parasite, Wuchereria bancrofti (Spirurida: Onchocercidae), in the vector mosquito, Culex quinquefasciatus (Diptera: Culicidae).

Detection and identification of humoral proteins, which are up-regulated in Culex quinquefasciatus upon infection by Wuchereria bancrofti, is important in tracing out the biochemical consequences of the filarial parasite development in the vector mosquito. Analysis of the haemolymph of infected mosquitoes through SDS-PAGE and RP-HPLC showed up-regulation of five proteins of molecular weights 40, 66, 22, 14, and 7-kDa. Among these, only the 40-kDa was unknown and the others were comparable with those already reported as transferrin, attacin, lysozyme, and defensin, respectively. In the present

Transferrin in the mosquito, Culex quinquefasciatus Say (Diptera: Culicidae), up-regulated upon infection and development of the filarial parasite, Wuchereria bancrofti (Cobbold) (Spirurida: Onchocercidae).

Transferrin is a defence protein known to be up-regulated upon infection of parasites/pathogens in Aedes aegypti mosquito. However, no information is available on its up-regulation in Culex quinquefasciatus, the vector of bancroftian filarial parasite. In the present study, enhancement of transferrin in C. quinquefasciatus infected with Wuchereria bancrofti is demonstrated through amplification of the specific mosquito transcript, its sequencing, cloning, and expression. By using two oligonucleotide primers, a 950-bp polymerase chain reaction product was obtained from the first strand cDNA mad

The influence of larval temperature and nutrients on dog heartworm (Dirofilaria immitis (Spirurida: Onchocercidae)) transmission in Aedes albopictus (Diptera: Culicidae) and Aedes triseriatus (Diptera: Culicidae).

The filarial nematode, Dirofilaria immitis (Leidy) (Spirurida: Onchocercidae), is an obligate mosquito-borne pathogen transmitted by at least 26 species of mosquitoes in the United States. Due to its size and migration through mosquito tissues during its first 3 larval stages (L1-L3), it can damage the mosquito to the point of death, thereby hindering transmission to its vertebrate host (eg canids and other carnivores). Mosquito species vary in competency for D. immitis, where some species facilitate the development of more viable, infectious L3 worms (host tolerance), while others produce few

An assessment of genetic variability in the mitochondrial cytochrome c oxidase subunit 1 gene of Cercopithifilaria sp. (Spirurida, Onchocercidae) from dog and Rhipicephalus sanguineus populations.

This study investigates sequence variation in mitochondrial cytochrome c oxidase subunit 1 gene within Cercopithifilaria sp. recorded recently in Italy. Fourteen sequence types (haplotypes) were characterized for 163 (7.7%) amplicons from 2111 Genomic DNA samples prepared from skin samples from dogs and from Rhipicephalus sanguineus (ticks) from different geographical areas of the Mediterranean basin (i.e., Italy, Spain and Greece). The most prevalent sequence types represented haplotypes I (70.5%) and X (16.0%), followed by haplotype VIII (4.9%) and other 11 haplotypes (8.6%). Three haplotype

Further morphological features, molecular characterization, and Wolbachia endosymbionts of Dirofilaria ursi Yamaguti, 1941 (Spirurida: Onchocercidae) from the Japanese black bear Ursus thibetanus japonicus Schlegel (Carnivora: Ursidae).

Dirofilaria ursi Yamaguti, 1941 (Spirurida: Onchocercidae) was originally described from the Japanese black bear Ursus thibetanus japonicus and later reported from the American black bear U. americanus in North America. Both D. ursi and D. ursi-like species have been implicated in human zoonosis in Japan and North America. This study aimed to revise the morphological characteristics of D. ursi in Japan and determine its phylogeny using multi-locus sequence analyses. We investigated the genotypes of Wolbachia endosymbionts harbored by Japanese D. ursi and compared them with those of American D.

Morphology and genetic characterization of Physaloptera sibirica Petrow & Gorbunov, 1931 (Spirurida: Physalopteridae), from the hog-badger Arctonyx collaris Cuvier (Carnivora: Mustelidae), with molecular phylogeny of Physalopteridae.

BACKGROUND: Nematodes of the family Physalopteridae (Spirurida: Physalopteroidea) commonly parasitize the alimentary canal of all major vertebrate groups. However, many physalopterid species are not adequately described, especially regarding the detailed morphology of the cephalic end. The current genetic database for Physaloptera species is still very limited, which seriously hampers molecular-based species identification. Additionally, the systematic status of some genera and the evolutionary relationships of the subfamilies in the Physalopteridae remain under debate. METHODS: New morpholog
